Vicinal fluorohydrins are useful building blocks as well as important targets for synthesis of bioactive compounds or new materials. This review gives an overview on regio- and stereoselective, particularly enantioselective, syntheses of such compounds by building block methods, reduction of α-fluoro carbonyl compounds and ring opening of epoxides with hydrofluorinating reagents. Resolution of racemic vicinal fluorohydrins using enzyme-catalyzed reactions will also be discussed in detail.  相似文献

Titanocene(II)-promoted cross-coupling between (Z)-alkenyl methyl sulfones and terminal allenes produced 1,4-dienes regioselectively via the formation of 2-alkylidenetitanacyclopentanes. Two different routes for the regio- and stereoselective synthesis of aziridino epoxides from cyclic dienes have been explored. The first strategy involves regiospecific aziridination of cyclic diene derivatives and subsequent epoxidation with m-CPBA to yield cis-aziridino epoxides as major products. The second strategy utilizes regiospecific epoxidation of cyclic diene derivatives followed by Sharpless aziridination to provide exclusively trans-aziridino epoxides. Synthesis of both enantiomers of cis-aziridino epoxides from (R)-(-)- and (S)-(+)-carvones are also reported.  相似文献

We describe the synthesis of novel 5-haloimidazole ribonucleosides as precursors of modified cobalamins. A regio- and stereoselective glycosylation of protected ribose with silylated 4(5)-haloimidazoles produces 5-haloimidazole ribonucleosides predominantly in the alpha-configuration (60-75%) without any 4-substituted imidazole ribonucleoside. The structure of the 5-fluoroimidazole ribonucleoside was confirmed by X-ray crystallography and 2D NMR spectroscopy.  相似文献

[reaction: see text] A variety of substituted isoindolin-1-ones are readily prepared in good to excellent yields under very mild reaction conditions by the reaction of o-(1-alkynyl)benzamides with ICl, I(2), and NBS. In a few cases, substituted isoquinolin-1-ones were obtained as the major product instead. This methodology accommodates various alkynyl amides and functional groups and has been successfully extended to heterocyclic starting materials. This chemistry has been successfully applied to the synthesis of a biologically interesting alkaloid, cepharanone B.  相似文献

Regio- and stereo-controlled synthesis of boryl-substituted allylsilanes via transition metal-catalyzed additions of silylboranes to unsaturated organic compounds is described. Nickel-catalyzed reactions of (dimethylphenylsilyl)pinacolborane with 1,3-dienes, vinylcyclopropanes, and vinylcyclobutanes yielded 4-, 5-, and 6-boryl-substituted allylsilanes, respectively. Palladium-catalyzed addition of the silylborane to allenes took place at the more substituted C=C bond to yield 2-borylallylsilane selectively. The 2-borylallylsilanes served as useful allylation reagents in Lewis acid-mediated reactions with acetals and aldehydes. In addition to the simple allylation reactions, a cascade reaction to form the trans-9-boryl-1,2-benzooxadecalin skeleton and a cyclization reaction to form cyclic alkenylboranes were achieved by the use of 2-borylallylsilanes as key reagents. Reactions of methylenecyclopropanes were catalyzed by palladium and platinum catalysts. The reaction course, however, depended upon the substrate structure and the catalyst employed. For instance, cycloalkylidenecyclopropanes yielded 2-cycloalkylidene-3-boryl-1-silylpropanes selectively in the presence of a palladium catalyst, while 3-cycloalkylidene-3-boryl-1-silylpropanes were obtained selectively in the corresponding platinum-catalyzed reactions.  相似文献

The regio- and stereospecific addition of chlorosulfonyl isocyanate to cis-δ-pinene enantiomers has furnished monoterpene-fused β-lactams. The observed regioselectivity can be explained by ab initio DFT modeling of transition state structures. In contrast with the less reactive α-pinane-fused β-lactam 4, the resulting β-lactams 5 and 13 containing an amino group connected to a secondary carbon possess similar reactivity to the cycloalkane-fused analogues and can be easily converted to the β-amino acid and its protected derivatives. The base-catalyzed isomerization of the cis-amino ester afforded the corresponding trans-amino ester in moderate yield.  相似文献

We present here a simple and mild method of hydrothiolation of internal and terminal alkynes under PhSeBr-catalyzed reaction in the absence of solvent at room temperature. 3-Substituted cis-cyclooctenes (3RCOEs, R = methyl, ethyl, hexyl, and phenyl) were synthesized and polymerized, and the polymers therefrom were hydrogenated to prepare model linear low density polyethylene (LLDPE) samples. The ring-opening metathesis polymerization (ROMP) of the 3RCOEs using Grubbs' catalyst proceeded in a regio- and stereoselective manner to afford polyoctenamers [poly(3RCOE)] exhibiting remarkably high head-to-tail regioregularity and high trans-stereoregularity. The overall selectivity increases with the increasing size of the R substituent. Hydrogenation of poly(3RCOE)s afforded precision LLDPEs with R substituents on every eighth backbone carbon.  相似文献

[Chemical reaction: See text] A systematic approach to the regio- and stereoselective synthesis of aminoinositols and 1,2-diaminoinositols arising from tetra-O-benzylconduritol B epoxide (9) and its aziridine analogue 22, respectively, is described. In all cases, the synthetic methodologies rely on the regio- and stereocontrolled azidolysis of the starting precursors to give the corresponding trans regioadducts. Subsequent functional group manipulation under strict configurational control affords the isomeric cis adducts. Chemoselective functionalization of the diamine moiety in 1,2-diaminoinositol derivatives can be achieved by the proper design of the reaction sequence and choice of reagents. The described protocols allow efficient access to each of the eight possible configurations of the 1,2-diamino and 1,2-amino alcohol moieties from chemical modifications of the epoxide moiety on the common precursor 9.  相似文献

An efficient and versatile approach for the synthesis of 2-deoxy-C-aryl glycosides is reported. This strategy is based on a palladium-catalyzed decarboxylative Heck coupling reaction of benzoic acids and glycals. A wide variety of glycals and benzoic acids have been screened, and all these reactions could afford the desired C-aryl glycoside products in moderate to good yields with exclusive regio- and stereoselectivities.  相似文献
