A new kind of magnetic dextran microsphere (MDMS) with uniform shape and narrow diameter distribution has been prepared from magnetic iron nanoparticles and dextran. Horseradish peroxidase (HRP) was successfully immobilized on the surface of an MDMS-modified glassy-carbon electrode (GCE), and the immobilized HRP displayed excellent electrocatalytic activity in the reduction of H2O2 in the presence of the mediator hydroquinone (HQ). The effects of experimental variables such as the concentration of HQ, solution pH, and the working potential were investigated for optimum analytical performance. This biosensor had a fast response to H2O2 of less than 10 s and an excellent linear relationship was obtained in the concentration range 0.20 μmol L−1–0.68 mmol L−1, with a detection limit of 0.078 μmol L−1 (S/N = 3) under the optimum conditions. The response showed Michaelis–Menten behavior at larger H2O2 concentrations, and the apparent Michaelis–Menten constant was estimated to be 1.38 mmol L−1. Moreover, the selectivity, stability, and reproducibility of the biosensor were evaluated, with satisfactory results. Thermally two-dimensional lattice graphene (GR) and biocompatibility chitosan (CS) act as a suitable support for the deposition of palladium nanoparticles (PdNPs). A novel hydrogen peroxide (H2O2) biosensor based on immobilization of hemoglobin (Hb) in thin film of CS containing GR and PdNPs was developed. The surface morphologies of a set of representative membranes were characterized by means of scanning electron microscopy and showed that the PdNPs are of a sphere shape and an average diameter of 50 nm. Under the optimal conditions, the immobilized Hb showed fast and excellent electrocatalytic activity to H2O2 with a small Michaelis–Menten constant of 16 μmol L−1, a linear range from 2.0 × 10−6 to 1.1 × 10−3 mol L−1, and a detection limit of 6.6 × 10−7 mol L−1. The biosensor also exhibited other advantages, good reproducibility, and long-term stability, and PdNPs/GR–CS nanocomposites film would be a promising material in the preparation of third generation biosensor.  相似文献

Sensitive fluorescent probes for the determination of hydrogen peroxide and glucose were developed by immobilizing enzyme horseradish peroxidase (HRP) on Fe3O4/SiO2 magnetic core–shell nanoparticles in the presence of glutaraldehyde. Besides its excellent catalytic activity, the immobilized enzyme could be easily and completely recovered by a magnetic separation, and the recovered HRP-immobilized Fe3O4/SiO2 nanoparticles were able to be used repeatedly as catalysts without deactivation. The HRP-immobilized nanoparticles were able to activate hydrogen peroxide (H2O2), which oxidized non-fluorescent 3-(4-hydroxyphenyl)propionic acid to a fluorescent product with an emission maximum at 409 nm. Under optimized conditions, a linear calibration curve was obtained over the H2O2 concentrations ranging from 5.0 × 10−9 to 1.0 × 10−5 mol L−1, with a detection limit of 2.1 × 10−9 mol L−1. By simultaneously using glucose oxidase and HRP-immobilized Fe3O4/SiO2 nanoparticles, a sensitive and selective analytical method for the glucose detection was established. The fluorescence intensity of the product responded well linearly to glucose concentration in the range from 5.0 × 10−8 to 5.0 × 10−5 mol L−1 with a detection limit of 1.8 × 10−8 mol L−1. The proposed method was successfully applied for the determination of glucose in human serum sample.  相似文献

The aluminium alloy Al-12Si has been polarized by potentiodynamic method at 25 °C under magnetic stirring and in an aerated solution. Its electrochemical behaviour was tested first by varying the concentration of NaI or NaCl (10−4, 10−3, 10−2) added respectively to NaCl or NaI (10−3 M), and the pH of NaCl 10−3 M (pH = 2.3, 7.3, 10) when adding HCl or NaOH (i.e. the composition of the solution), then by incorporating different ions familiar to an industrial atmosphere (Cu2+, Zn2+, , , ) at 10−6 M to NaCl 10−3 M (i.e. the electrolyte nature). The use of the electrokinetic curves obtained allowed the access to the passivation (i pass , E rup and E rep ) and to the electrokinetic parameters (i corr , R p and P). They prove the behaviour dependence of the above alloy on the composition and nature of the electrolyte.  相似文献

Biogas is produced by biological processes under anaerobic conditions and may contain up to 20,000 ppmv hydrogen sulfide (H2S), a corrosive substance that attacks power engines and can affect the health of the industrial staff. H2S must be removed from the biogas, especially in co-generation facilities where the biogas is burnt for energy production. Nowadays, biofiltration is being studied and considered as an interesting alternative for removing H2S from the biogas besides classical chemical processes. The novelty of this work is the design and construction of an automated H2S on-line analyser to assess the composition of the liquid and gas phases of gas-phase bioreactors. The analyser is made of two parallel flow configurations which share the same detection device. The first configuration is a single-channel flow injection analyser (FIA) to detect S2− in the liquid phase. The second configuration is a continuous flow analyser (CFA) with a gaseous diffusion step (GD–CFA) for detecting H2S in the gas phase. The diffusion step enables separation of the H2S(g) from the sample and its conversion into a detectable chemical species (S2−). S2− detection was performed with an Ag2S ion-selective electrode (ISE) selective to . The main response parameters of the FIA system are a linear range between 3 × 10−5 and 1 × 10−1 mol L−1 S2− (0.61–3,200 mg L−1), with a sensitivity of 27.9 mV decade−1 and a detection limit of 1.93 × 10−5 mol L−1 S2−. The GD–CFA configuration presents a linear range between 400 and 10,000 ppmv H2S(g) with a sensitivity of 26.1 mV decade−1 and a detection limit of 245 ppmv H2S. The proposed analyser was used by analysing real gas and liquid samples with optimal results at a full-scale biotrickling filter for biogas treatment at a municipal wastewater treatment plant. A capacitive biosensor for the detection of bacterial endotoxin has been developed. Endotoxin-neutralizing protein derived from American horseshoe crab was immobilized to a self-assembled thiol layer on a biosensor transducer (Au). Upon injection of a sample containing endotoxin, a decrease in the observed capacitive signal was registered. Endotoxin could be determined under optimum conditions with a detection limit of 1.0 × 10−13 M and linearity ranging from 1.0 × 10−13 to 1.0 × 10−10 M. Good agreement was achieved when applying endotoxin preparations purified from an Escherichia coli cultivation to the capacitive biosensor system, utilizing the conventional method for quantitative endotoxin determination, the Limulus amebocyte lysate test as a reference. The capacitive biosensor method was statistically tested with the Wilcoxon signed rank test, which proved the system is acceptable for the quantitative analysis of bacterial endotoxin (P < 0.05). The kinetics of the reaction between [ReN(H2O)-(CN)4]2− with different κ2 N,O-donor ligands (quin and 2,3-dipic, respectively) have been studied in the pH 4–12 range in aqueous solution. Two consecutive reaction steps with the formation of the [ReN(η1-quin)(CN)4]3− and [ReN(μ2-quin) (CN)3]2− complexes, respectively, were spectrophotometrically observed and kinetically investigated. The same reaction mechanism is proposed for these two ligands. The first fast reaction (for quin) is attributed to the aqua substitution of [ReN(H2O)(CN)4]2− with forward and reverse rate constants of 1.96(5) × 10−1 M−1 s−1 and 5.6(3) × 10−2 s−1, while a rate of 2.64(3) M−1 s−1 was observed for the reaction between the conjugate base [ReN(OH)(CN)4]3− and quin at 40.2 °C. Due to small absorbance changes, it was difficult to obtain any good quality data for the fast reactions for 2,3-dipic. The second, slower reaction is attributed to cyano substitution with rate constants (k 3 K 1) of 4.17(4) × 10−3 for quin and 4.68(7) × 10−3 M−1 s−1 for 2,3-dipic, at 80.02 °C, respectively. The acid dissociation constant for the aqua complex was spectrophotometrically determined as 11.58(3) and 11.54(2) and kinetically as 11.51(8) and 11.41(1), at 80.4 °C, respectively. Negative values of −83.5(2) and −144.1(2) J K−1 mol−1 as well as the of 71.4(3) and 47.3(3) kJ mol−1, for the slow quin and 2,3-dipic reactions, respectively, point to an ordered transition state where bond formation is responsible for the major driving force of the reaction. The and for the fast forward reaction of quin is indicative of expected associative activation in the transition state. The kinetics of hexacyanoferrate(III) reduction by hydrogen peroxide in strongly alkaline media leading to hexacyanoferrate(II) ion have been studied spectrophotometrically within the wavelength range 300–500 nm. The reaction obeys a simple pseudo-first-order rate expression under the applied conditions, namely, a large excess of the reductant and OH anion concentrations, and a low oxidant concentration. The linear dependences of the pseudo-first-order rate constant on OH and H2O2 concentrations are consistent with the rate law of the form: where and are the second- and the pseudo-third-order rate constants for the electron transfer from HO2 and O2 2− to [Fe(CN)6]3−, respectively. The apparent activation parameters determined at 0.4 M NaOH are as follows: ΔH # = (18.0 ± 1.0) kJ mol−1 and ΔS # = (−155 ± 3.5) J K−1 mol−1. A novel sensor for the determination of nitrite anion () was fabricated by electrodeposition of toluidine blue. The sensor exhibited good catalytic activity toward the electrochemical oxidation of nitrite. Amperometry was carried out to determine the concentration of . A linear response in the range from 1.0×10−7 to 1.52×10−5 M with a substantially low detection limit of 5×10−8 M (S/N=3) was obtained. The proposed sensor had a feature of high sensitivity of 4.7×105 μA M−1 cm−2. The possible interference from several common ions was tested. This sensor was applied to the amperometric determination of nitrite in food samples, and the results were consistent with those obtained with the standard spectrophotometric procedure.  相似文献

A novel hydrogen peroxide (H2O2) biosensor was developed by immobilizing hemoglobin on the gold colloid modified electrochemical pretreated glassy carbon electrode (PGCE) via the bridging of an ethylenediamine monolayer. This biosensor was characterized by UV-vis reflection spectroscopy (UV-vis), electrochemical impendence spectroscopy (EIS) and cyclic voltammetry (CV). The immobilized Hb exhibited excellent electrocatalytic activity for hydrogen peroxide. The Michaelis–Menten constant (K m) was 3.6 mM. The currents were proportional to the H2O2 concentration from 2.6 × 10−7 to 7.0 × 10−3 M, and the detection limit was as low as 1.0 × 10−7 M (S/N = 3).  相似文献

A performant reagentless electrochemiluminescent (ECL) detection system for H2O2 is presented, based on an electropolymerized polyluminol film prepared under near-neutral conditions. Such an original polyluminol electrodeposition is reported for the first time and on a screen-printed electrode (SPE) surface. Electropolymerized luminol acts as an active luminophore of the electrochemiluminescent reaction, as the monomer does. Polymerization conditions have been optimized in order to obtain the best ECL responses to H2O2. By performing electrodeposition in a potentiostatic mode, at 425 mV vs. Ag|AgCl, in 0.1 mol L−1 phosphate/0.1 mol L−1 KCl pH 6 and 1 mmol L−1 luminol, with a total charge of 0.5 mC, the linear range for H2O2 detection extends from 7.9 × 10−8 mol L−1 to 1.3 × 10−3 mol L−1. Such performant disposable reagentless easy-to-use miniaturized systems based on SPEs should be applicable to the electrochemiluminescent detection of many oxidase-substrate compounds. Figure An original polyluminol electrodeposition process on a screen-printed electrode surface is reported for the first time. The polymeric structure is demonstrated to behave as an electrochemiluminescent luminophore, allowing disposable reagentless easy-to-use optical sensors for hydrogen peroxide detection to be designed.  相似文献

In this work, an enzyme biosensor based on the immobilization of horseradish peroxidase (HRP) on SiO2/BSA/Au/thionine/nafion-modified gold electrode was fabricated successfully. Firstly, nafion was dropped on the surface of the gold electrode to form a nafion film followed by chemisorption of thionine (Thi) as an electron mediator via the ion-exchange interaction between the Thi and nafion. Subsequently, the SiO2/BSA/Au composite nanoparticles were assembled onto Thi film through the covalent bounding with the amino groups of Thi. Finally, HRP was immobilized on the SiO2/BSA/Au composite nanoparticles due to the covalent conjugation to construct an enzyme biosensor. The surface topographies of the SiO2/BSA/Au composite nanoparticles were investigated by using scanning electronic microscopy. The stepwise self-assemble procedure of the biosensor was further characterized by means of cyclic voltammetry and chronoamperometry. The enzyme biosensor showed high sensitivity, good stability and selectivity, a wide linear response to hydrogen peroxide (H2O2) in the range of 8.0 × 10-6 ∼ 3.72 × 10-3 mol/L, with a detection limit of 2.0 × 10-6 mol/L. The Michaelies-Menten constant KMapp K_M^{app} value was estimated to be 2.3 mM.  相似文献

In the presence of carbonate and uranine, the chemiluminescent intensity from the reaction of luminol with hydrogen peroxide was dramatically enhanced in a basic medium. Based on this fact and coupled with the technique of flow-injection analysis, a highly sensitive method was developed for the determination of carbonate with a wide linear range. The method provided the determination of carbonate with a wide linear range of 1.0 × 10−10–5.0 × 10−6 mol L−1 and a low detection limit (S/N = 3) of carbonate of 1.2 × 10−11 mol L−1. The average relative standard deviation for 1.0 × 10−9–9.0 × 10−7 mol L−1 of carbonate was 3.7% (n = 11). Combined with the wet oxidation of potassium persulfate, the method was applied to the simultaneous determination of total inorganic carbon (TIC) and total organic carbon (TOC) in water. The linear ranges for TIC and TOC were 1.2 × 10−6–6.0 × 10−2 mg L−1 and 0.08–30 mg L−1 carbon, respectively. Recoveries of 97.4–106.4% for TIC and 96.0–98.5% for TOC were obtained by adding 5 or 50 mg L−1 of carbon to the water samples. The relative standard deviations (RSDs) were 2.6–4.8% for TIC and 4.6–6.6% for TOC (n = 5). The mechanism of the chemiluminescent reaction was also explored and a reasonable explanation about chemical energy transfer from luminol to uranine was proposed. A carbon composite amperometric hydrogen peroxide sensor has been developed using a sol-gel technique. Toluidine blue (TB), which acts as the redox mediator, was covalently immobilized via glutaraldehyde crosslinking with an organically modified silane, namely 3-aminopropyltrimethoxysilane (APTMOS). Methyltrimethoxysilane (MTMOS) was used as the additional monomer; this controls the hydrophobicity of the electrode surface, thus limiting the wettability. The immobilization of TB within the sol-gel matrix was confirmed with FTIR studies. The sol-gel mixture containing TB immobilized in APTMOS and MTMOS was mixed with graphite powder in order to prepare the carbon composite electrode. The electrode was characterized using voltammetric techniques and its electrocatalytic activity for the reduction of hydrogen peroxide was also studied. The carbon composite electrode has the advantage of sensing H2O2 at a lower potential and with a higher sensitivity, and interferences due to ascorbic acid, uric acid and acetaminophen were greatly minimized. The linear range for the determination of H2O2 extends from 5.37 × 10−6 to 6.15 × 10−3 M, with a correlation coefficient of 0.9981. The detection limit was found to be 2.15 × 10−6 M. The covalent immobilization of TB effectively prevents the leakage of the water-soluble mediator during measurements. The modified electrode, aside from electrocatalyzing the reduction of H2O2, exhibits distinct advantages in terms of surface renewal in the event of surface fouling, as well as simple preparation, good chemical and mechanical stability, and good reproducibility. A tubular membrane made of Zr0.8Y0.2O1.9 (60 vol%) and was prepared by a standard ceramic process. Oxygen permeation through the membrane tube was examined by exposing its outer shell to air and sweeping its inner wall with pure helium or CO balanced with helium. An oxygen flux of 9.2×10−9 mol cm−2 s−1 was measured at 950°C under air/He gradient, and a larger flux of 3.2×10−8 mol cm−2 s−1 at 930°C under air/CO gradient. The membrane tube was found to exhibit excellent stability under highly reducing atmosphere and elevated temperatures. The oxygen permeation rate is likely to be increased through the modification of the surface and reduction of the membrane thickness.  相似文献

A hybrid film is fabricated by casting hemoglobin (Hb)–carbon nanoparticles (CNPs)–polyvinyl alcohol (PVA) suspension on glassy carbon electrode (GCE). The resulting film shows a three-dimensional nanoporous structure. In the hybrid film, the ultraviolet visible (UV–Vis) absorption spectra of Hb keep almost unchanged. The organic–inorganic hybrid material can promote the direct electron transfer of Hb. A pair of well-defined and quasireversible peaks with a formal potential of −0.348 V (vs saturated calomel electrode) is obtained, which is caused by the electrochemical reaction of the Fe(III)/Fe(II) couple of Hb. The electron transfer rate constant (k s) is estimated to be 3.9 s−1. The immobilized Hb exhibits high stability and excellent electrochemical catalysis to the reduction of oxygen (O2), hydrogen peroxide (H2O2), and nitrite (). The catalytic currents are linear to the concentrations of H2O2 and from 1.96 to 112 μM and from 0.2 to 1.8 mM, respectively. Therefore, the hybrid film may be a good matrix for protein immobilization and biosensor fabrication.  相似文献

A fast and sensitive approach that can be used to detect norfloxacin in human urine using capillary electrophoresis with end-column electrochemiluminescence (ECL) detection of is described. The separation column was a 75-μm i.d. capillary. The running buffer was 15 mmol L−1 sodium phosphate (pH 8.2). The solution in the detection cell was 50 mmol L−1 sodium phosphate (pH 8.0) and 5 mmol L−1 The ECL intensity varied linearly with norfloxacin concentration from 0.05 to 10 μmol L−1. The detection limit (S/N=3) was 0.0048 μmol L−1, and the relative standard deviations of the ECL intensity and the migration time for eleven consecutive injections of 1.0 μmol L−1 norfloxacin (n=11) were 2.6% and 0.8%, respectively. The method was successfully applied to the determination of norfloxacin spiked in human urine without sample pretreatment. The recoveries were 92.7–97.9%.   相似文献

A microfluidic system incorporating chemiluminescence detection is reported as a new tool for measuring antioxidant capacity. The detection is based on a peroxyoxalate chemiluminescence (PO-CL) assay with 9,10-bis-(phenylethynyl)anthracene (BPEA) as the fluorescent probe and hydrogen peroxide as the oxidant. Antioxidant plugs injected into the hydrogen peroxide stream result in inhibition of the CL emission which can be quantified and correlated with antioxidant capacity. The PO-CL assay is performed in 800-μm-wide and 800-μm-deep microchannels on a poly(dimethylsiloxane) (PDMS) microchip. Controlled injection of the antioxidant plugs is performed through an injection valve. Of the plant-food based antioxidants tested, β-carotene was found to be the most efficient hydrogen peroxide scavenger (SA HP of 3.27 × 10−3 μmol−1 L), followed by α-tocopherol (SA HP of 2.36 × 10−3 μmol−1 L) and quercetin (SA HP of 0.31 × 10−3 μmol−1 L). Although the method is inherently simple and rapid, excellent analytical performance is afforded in terms of sensitivity, dynamic range, and precision, with RSD values typically below 1.5%. We expect our microfluidic devices to be used for in-the-field antioxidant capacity screening of plant-sourced food and pharmaceutical supplements. Pathogen detection is important for health and safety reasons. Several outbreaks all over the world have shown the need for rapid, qualitative, quantitative, and, particularly, multianalyte detection systems. Hence, a multichannel flow-through chemiluminescence microarray chip for parallel detection of pathogenic bacteria was developed. The disposable chip made of acrylonitrile–butadiene–styrene (ABS) copolymer was devised as a support for a multiplexed sandwich immunoassay. Calibration and measurement was possible in one experiment, because the developed chip contains six parallel flow-through microchannels. Polyclonal antibodies against the pathogenic bacteria Escherichia coli O157:H7, Salmonella typhimurium, and Legionella pneumophila were immobilized on the chip by microcontact printing in order to use them as specific receptors. Detection of the captured bacteria was carried out by use of specific detection antibodies labelled with biotin and horseradish peroxidase (HRP)–streptavidine conjugates. The enzyme HRP generates chemiluminescence after adding luminol and hydrogen peroxide. This signal was observed by use of a sensitive CCD camera. The limits of detection are 1.8 × 104 cells mL−1 for E. coli O157:H7, 7.9 × 104 cells mL−1 for L. pneumophila, and 2.0 × 107 cells mL−1 for S. typhimurium. The overall assay time for measurement and calibration is 18 min, enabling very fast analysis.   相似文献

The electrocatalytic oxidation of glutathione (GSH) has been studied at the surface of ferrocene-modified carbon paste electrode (FMCPE). Cyclic voltammetry (CV), double potential step chronoamperometry, and differential pulse voltammetry (DPV) techniques were used to investigate the suitability of incorporation of ferrocene into FMCPE as a mediator for the electrocatalytic oxidation of GSH in buffered aqueous solution. Results showed that pH 7.00 is the most suitable for this purpose. In the optimum condition (pH 7.00), the electrocatalytic ability of about 480 mV can be found and the heterogeneous rate constant of catalytic reaction was calculated as . Also, the diffusion coefficient of glutathione, D, was found to be 3.61 × 10–5 cm2 s−1. The electrocatalytic oxidation peak current of glutathione at the surface of this modified electrode was linearly dependent on the GSH concentration and the linear analytical curves were obtained in the ranges of 3.2 × 10–5 M–1.6 × 10–3 M and 2.2 × 10–6 M–3.5 × 10–3 M with cyclic voltammetry and differential pulse voltammetry methods, respectively. The detection limits (3σ) were determined as 1.8 × 10–5 M and 2.1 × 10–6 M using CV and DPV, respectively. Finally, the electrocatalytic oxidation of GSH at the surface of this modified electrode can be employed as a new method for the voltammetric determination of glutathione in real samples such as human plasma.  相似文献
