利用微波加热法,由5,10,15,20-四(4-甲氧羰基苯基)卟啉水解合成了四(4-羧基苯基)卟啉,并对反应条件进行了探讨。结果表明:以吡啶为溶剂,氢氧化钾为催化剂,以65W的微波功率辐射30rnin,可使5,10,15,20-四(4.甲氧羰基苯基)卟啉完全水解。反应时间是传统加热法用时的1/48。  相似文献

在“血卟啉衍生物”(HPD)[1]肿瘤光动力疗法取得令人瞩目成就的同时,化学家们证明了临床使用的HPD制剂并非原意义上的血卟啉衍生物,而是一种复杂的卟啉混合物[2],其中血卟啉(HP),羟乙基-乙烯基-次卟啉(HVD),原卟啉(PP)约占总量的80%,Dougherty等分离得到其余20%未知结构羧基卟啉,认为此为HPD的有效成分,经进一步分析测定其主要成分是二血卟啉醚(DHE)[3]。  相似文献

为合成白三烯A4的关键中间体, 本文试验了若干7-手性中心-6,5-双键庚酸衍生物的不对称环氧化方法, 发现从D-甘油醛得的顺式烯化合物可选择性地羟内酯化, 游离羟基甲磺酰化后甲醇纳处理可得所需的5S, 6R氧桥, 水解, 过碘酸钠断键后即可顺利得到目标化合物.  相似文献

合成了3-氨基-7,8-二甲氧基香豆素及其衍生物共11个化合物,其中3个化合物(2b、2d、2e)为新型香豆素芳酰胺类化合物。 通过猝灭1,1-二苯基-2-三硝基苯肼(DPPH)、2,2'-联氮二(3-乙基苯并噻唑-6-磺酸)二铵盐阳离子自由基(ABTS)和羟自由基实验考察了所合成化合物的抗氧化活性,结果表明化合物2b对DPPH自由基、羟自由基的清除能力超出或接近对照品维生素C,而衍生物2a、2b和2c的抗氧化活性优于母体。 故酰化可提高3-氨基-7,8-二甲氧基香豆素的抗氧化性能,尤其是普遍提高了羟基自由基的清除能力。  相似文献

1. 本文叙述了从α-苯甲醯-β-乙氧基丙烯酸乙酯开始经过四个步骤合成了制备氯霉素的有用中间体--dl-threo-3-氯代-3-苯基-2-氨基丙醇盐酸盐(Ⅳ)。2. 用 Reformatsky 反应合成了α-甲氧甲基-β-羟基-β-苯基丙酸甲酯(Ⅹ),虽然产量甚小,但说明了β位置含有甲氧基的α溴酯化合物可以应用于 Reformatsky 反应,而所得的产物为相应的β羟基酸酯而非不饱和酸酯。3. 进行了β羰基醯胺与甲醛的缩合,结果所得者为二分子β羰基醯胺与一分子甲醛缩合成戊二酸醯胺衍生物,而未取得羟甲基化合物。 4. 对硝基苯甲醯乙酸乙酯与过量的甲醛进行缩合,结果获得双羟甲基化合物,αα′-双-羟甲基-对硝基苯甲醯乙酸乙酯(ⅩⅧ)。  相似文献

Abstract— A first report on the biological evaluation of a series of isomerically pure benzoporphyrin derivatives ( cis- and frarcs-isomers) as methyl esters is described. In preliminary in vivo studies, the n- bexyl ether analogues of both cis- and trans -isomers of benzoporphyrin derivatives were found to be more active than the industrially prepared benzoporphyrin derivative, a mixture of monocarboxylic acids (BPDMA, Quadralogic Technologies, Vancouver). Further studies with 4-de-vinyl-4-(I-hexyloxyethyl) benzoporphyrin derivative showed that, like BPDMA, it had reduced residual skin phototoxicity compared in mice with Photofrin®. The uptake and clearance characteristics of BPDMA were also compared with the 4-(1-hexyloxyethyl)-derivative by in vivo reflection spectroscopy.  相似文献

The plasma distribution and biodistribution of benzoporphyrin derivative were examined. Two analogs of benzoporphyrin derivative were mixed with human plasma in vitro and recovered in the lipoprotein fractions upon separation by chromatography or ultracentrifugation. The majority of both analogs was recovered with high density lipoprotein. The effect of prebinding benzoporphyrin derivative to lipoproteins on the biodistribution of the drug in vivo was studied in tumor bearing DBA/2J mice. At 3, 8 and 24 h post-injection, tumor and tissue samples were excised and analyzed for benzoporphyrin derivative content. Precomplexing benzoporphyrin derivative with low density lipoprotein or high density lipoprotein led to significantly (P less than 0.05) greater tumor accumulation than in aqueous solution.  相似文献

Methods for the separation of photosensitizer isomers, such as benzoporphyrin derivative monoacid, benzoporphyrin ethyl monoacid, 2-[1-hexyloxyethyl]-2-devinylpyropheophorbide-a, diethyleneglycol diester benzoporphyrin derivative, tin ethyl etiopurpurin, and phthalocyanine tetrasulfonate, have been systematically developed by CE. Detection was accomplished by UV absorption at 214 nm or by LIF with excitation at 442/488 nm and emission at 690 nm. The effects of three major experimental parameters of buffer types, organic solvents, and surfactant additives are described. The optimized separation conditions were determined so as to provide satisfactory separation efficiency and analysis time. The methods are shown to be suitable for the separation and determination of porphyrin and phthalocyanines regioisomers, diastereoisomers, and enantiomers.  相似文献

The influence of lipoprotein association on in vitro tumor cell killing and in vivo tumor photosensitization with benzoporphyrin derivative (BPD) has been investigated in M-1 tumor bearing mice. The association of benzoporphyrin mono acid ring A with either low or high density lipoprotein increased tumor cell killing in an in vivo/in vitro cytotoxicity assay performed 3 h post intravenous drug administration. Eight hours following photosensitizer injection only low density lipoprotein (LDL) mixtures produced significant (P less than or equal to 0.005) increases in tumor cell killing compared to BPD in unfractionated plasma. The efficacy of in vivo photosensitization in the presence of lipoproteins correlated with the in vivo/in vitro cytotoxicity. Association of BPD with low or high density lipoproteins resulted in delayed tumor regrowth and higher cure rates when light exposure (125J/cm2) was performed 3 h post drug administration. When light exposure was performed 8 h post-injection only LDL-BPD mixtures led to enhanced tumor eradication compared to BPD administered in aqueous solution or unfractionated plasma.  相似文献

A novel divinyl ether was synthesized by a convenient method with high yield.Then the divinyl ether was combined with 2- hydroxyethyl methacrylate and acrylic acid,respectively,generating difunctional polymeric crosslinkers with(hemi)acetal structure that was labile in acid.The chemical structures of the divinyl ether and crosslinkers were confirmed by ~1H NMR and elemental analysis.The crosslinkers were employed in free-radical polymerization to prepare polymer gel and gel particles. Due to the(hemi)acetal structure in the crosslinking segment,the polymer gel and particles exhibited degradable ability in strong acid.  相似文献

Hydrophilic films based on blends of poly(acrylic acid) and poly(2‐hydroxyethyl vinyl ether) were prepared by casting. The characterization of the films was performed by differential scanning calorimetry (DSC), dynamic mechanical thermal analysis (DMTA), and scanning electron microscopy (SEM). It was shown that an increase of poly(2‐hydroxyethyl vinyl ether) content in the blends considerably decreases the glass transition temperature of the samples. The films containing 10 and 20 mol‐% of poly(2‐hydroxyethyl vinyl ether) show behavior of polymers in the glassy state, but a further increase of nonionic polymer content in the blend (30–50 mol‐%) provides the mechanical properties typical of a rubbery state. The content of water traces in the films has a significant effect on the mechanical properties of the materials.

10-Chloro-1,7-ditosyl-8-phenyl-4,1,7-benzoxadiazecine is obtained by reacting the sodium salt of N , N ′-ditosyl-2-amino-5-chlorobenzhydrylamine with the ditosylate of bis(hydroxyethyl) ether in DMF. The structure of the title compound was established by X-ray crystal structure analysis. Substituted 3-phenoxy-3-perfluoroalkylprop-2-enals 3a-s are synthesized in high yields starting from a gem-iodoacetoxy derivative 1 and phenoxides 2. Then efficient syntheses of push-pull derivatives 4, 5,8a,b, and nonconjugated analogues 6 and 7 illustrate the synthetic potentialities of 3. Stereochemical studies of these perfluoroalkyl-containing trisubstituted olefinic derivatives 3-8b revealed that the (4)J(CF) coupling constant observed in the (13)C NMR spectra was crucial in the determination of their configurations and conformations in solution. The solvent polarity effect on the stereochemistry of push-pull compounds 3-5 and 8a,b was studied. Unusual significant medium polarity effect on the stereochemistry of imino enol ether derivative 4 was observed.  相似文献

Treatment of aldosterone with 35% HCl in EtOH or in MeOH followed by the picolinyl derivatization gave the picolinyl derivative of aldosterone-ethyl ether, 8, or methyl ether, 9, as a single and well-shaped liquid chromatographic peak. Picolinyl derivatization of aldosterone produced 21-picolinyl derivative of 18,20-anhydro-hemiacetal derivatives, 6, with poor chromatographic peak with wide half-width. Further conversion of 6 to 8 required long reaction time (>4 h). Structure of each picolinyl or alkyl ether-picolinyl derivative, was carefully elucidated by nuclear magnetic resonance spectroscopy, electron ionization mass spectrometry and liquid chromatography-electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). Enhancement of sensitivity (approximately 10-fold) in positive-LC-ESI-MS/MS of aldosterone was confirmed by the use of the alkyl ether-picolinyl derivatization when compared to the underivatized molecule.  相似文献
