Abstract— The sensitivity of phototactic orientation of pseudoplasmodia (slugs) of the cellular slime mold Dictyosrelium discoideum has been measured for white light and monochromatic light using computer aided directional statistics. The zero threshold for white light was found at about 10-5 Ix. An action spectrum for positive phototaxis has been calculated from fluence rate-response curves; it shows two major maxima at about 420 and 440 nm and secondary peaks at 560 and 610 nm. This action spectrum is significantly different from the one for phototactic orientation in Dictyostelium amoebae.  相似文献

The properties of a newly isolated anionic tobacco peroxidase from transgenic tobacco plants overexpressing the enzyme have been studied with respect to the chemiluminescent reaction of luminol oxidation. These were compared to the properties of horseradish peroxidase in the cooxidation of luminol and p -iodophenol, the enhanced chemiluminescence reaction. The pH, luminol and hydrogen peroxide concentrations were optimized for maximum sensitivity using the tobacco enzyme. The detection limit for the latter under the optimal conditions (2.5 m M luminol, 2 m M hydrogen peroxide, 100 m M Naborate buffer, pH 9.3) was about 0.1 p M , which is at least five times lower than that for horseradish peroxidase in enhanced chemiluminescence with p -iodophenol. The rate constants for the elementary steps of the enzyme-catalyzed reaction have been determined: k 1= 4.9 × 106 M −1 s1, k 2= 7.3 × 106 M −1 s−1, k 3= 2.1 × 106 M −1 s−1 (pH 9.3). The similarity of these rate constants is unusual for plant peroxidases. The high catalytic activity of tobacco peroxidase in the luminescent reaction is explained by the high reactivity of its Compound II toward luminol and the high stability of the holoenzyme with respect to heme dissociation. This seems to be a unique property of this particular enzyme among other plant peroxidases.  相似文献

Abstract— Chlamydomonas reinhardtii responds to a blue light stimulus by an oriented swimming (phototaxis) toward or away from the stimulus source. In this study it is established that the sign and strength of the phototactic response are a complex function of extracellular [Ca2+], stimulus fluence rate, time of analysis after onset of stimulation and light pretreatment. At very low extracellular [Ca2+] the response is weak and usually negative. At [Ca2+] close to the preconditioning level, phototactic response becomes stronger and positive. As [Ca2+] is raised further, the initial (2 s) response remains positive but the long term (20 s) becomes negative and very strong. At extremely high [Ca2+] the cells become immobile. This bimodal behavior suggests that two different mechanisms determine the direction of the turn. Data cannot be explained in terms of a simple model. The model which accounts for most of the details of the behavior is that of Kamiya and Witman (1984), which proposes that positive response is triggered by a transient increase in intracellular [Ca2+] and negative response by a decrease below unstimulated level of Ca2+, at least in the range of 10-9-10-6 M [Ca2+]. The strong negative orientation which follows an initial positive response above this level of [Ca2+], in these experiments, is best explained by an adaptation of the cells due to an increased (on average) intracellular [Ca2+].  相似文献

Abstract A photobiological study was camed out on the bacterium Escherichia coli in order to determine whether stimulation of growth occurred after irradiation of an inoculum with coherent red light. No enhancement or inhibition of growth was observed for cultures of the bacterium following irradiation of inocula with a Helium-neon laser (continuous wave, λ= 632.8 nm) at irradiances of 7.7 × 1015 and 1.8 × 1016 photons cm−2 s−1 using fluences of 4.5 × 10−-1 and 4.5 J cm−2 at each irradiance. Bacterial growth in irradiated and control cultures was monitored during a growth period of ca 2 h using a viable count technique after inocula in the early exponential phase had been diluted with fresh growth medium. These results do not provide support for the work of Kam et al . (1983, Nuov. Cim . 2D, 1138–1144), and Tiphlova and Karu (1988, Photochem. Photobiol . 48 , 467–471), which appear to show substantial enhancement of E. coli growth under these conditions.  相似文献

Abstract— N,N'-bis(2-ethyl-1,3-dioxolane)-kryptocyanine (EDKC), a lipophilic dye with a delocalized positive charge, photosensitizes cells to visible irradiation. In phosphate-buffered saline (PBS), EDKC absorbs maximally at 700 nm (ε= 1.2 × 105 M−1 cm−1) and in methanol, the absorption maximum is at 706 nm (ε= 2.3 × 105 M−1 cm−1). EDKC partitions from PBS into small unilamellar liposomes prepared from saturated phospholipids and into membranes prepared from red blood cells (RBC) and binds to human serum albumin (HSA). The EDKC fluorescence maximum red shifts from 713 nm in PBS to 720–725 nm in liposomes and RBC membranes and the fluorescence intensity is enhanced by factors of 14–35 compared to PBS (φ= 0.0046). EDKC is thermally unstable in PBS (T1/2= 2 h at 1.3 × 10−5 M EDKC), but stable in methanol. In liposomes and RBC membranes, EDKC is 10 times more stable than in PBS, indicating that it is only partially exposed to the aqueous phase. Quenching of EDKC fluorescence in liposomes and RBC membranes by trinitrobenzene sulfonate also indicates that EDKC is not buried within the membranes. Photodecomposition of EDKC was oxygen-dependent and occurred with a low quantum yield (6.4 × 10−4 in PBS). Singlet oxygen was not detected upon irradiation of EDKC in membranes or with HSA since the self-sensitized oxidation of EDKC occurred at the same rate in D2O as in H2O and was not quenched by sodium azide or histidine.  相似文献

Abstract
The shape of the fluence-response relationship for the phototropic response of the JK224 strain of Arabidopsis thaliana depends on the fluence rate and wavelength of the actinic light. At low fluence rate (0.1 μmol m-2s-1), the response to 450-nm light is characterized by a single maximum at about 9 μmol m-2. At higher fluence rate (0.4 μmol m-2s-1), the response shows two maxima, at 4.5 and 9 μmol m-2. The response to 510-nm light shows a single maximum at 4.5 μmol m-2. Unilateral preirradiation with high fluence rate (25 μmol m-2s-1) 510-nm light eliminates the maximum at 4.5 μmol m-2 in the fluence response curve to a subsequent unilateral 450-nm irradiation, while the second maximum at 9 μmol m-2 is unaffected. Based on these results, it is concluded that a single photoreceptor pigment has been altered in the JK224 strain of Arabidopsis thaliana.  相似文献

Abstract— Anionic polyelectrolytes functionalized with the 5-deazaflavin group (dFl) were synthesized. The lifetime of the triplet excited dFl in the polyelectrolytes with a 2-mol% dFl content (AdFl-2) was about 10 times longer than that of a low molecular weight analog (AdFl-M). 2-Mercaptoethanol (RSH) reduced the triplet dFl with the rate constant of k red= 2.01 × 108 M −1 s−1 for AdFl-M and k red= 4.4 × 107 M −1 s−1 for AdFl-2. A zwitterionic viologen (SPV) oxidized the triplet dFl with the rate constant of k red= 3.69 × 109 M −1 s−1 for AdFl-M and k ox= 7.4 × 108 M−1 s−1 for AdFl-2. The smaller rate constants for the polymer system were discussed in terms of the hindering effect of the macromolecular microenvironment. The back electron transfer was shown to be drastically slowed in the AdFl-2-SPV system as a result of the intensive electrostatic effect of the polyelectrolytes. The buildup of the viologen radicals was studied under the steady-state illumination of the three component systems including viologen and RSH. The dFl group was demonstrated to serve as a very efficient photosensitizer in the oxidative cycle in case back electron transfer was retarded. This is the case of the AdFl-2-SPV system which gave the quantum yield of about 0.4 for the SPV buildup. By comparison, the AdFl-2-MV2+ system resulted in a much slower buildup of MV +radicals.  相似文献

Abstract— Most sensitizers used for the photodynamic therapy (PDT) of tumors photobleach on illumination. Thus, it is of interest to examine the photobleaching behavior of new sensitizers proposed for use in PDT. This report surveys the quantum yields and kinetics of the photobleaching of mono- l -aspartyl chlorin e6 (NPe6), a hydrophilic chlorin that has many of the photoproperties desirable in a sensitizer for clinical PDT. It is a very effective sensitizer for the PDT of several types of model tumors in animals and is now in Phase I clinical trials. The quantum yield of NPe6 photobleaching in pH 7.4 phosphate buffer in air was 8.2 × 10−4; this is greater than the yields for typical porphyrin photosensitizers. For example, the yields for hematoporphyrin and uroporphyrin are 4.7 × 10 5 and 2.8 × 10−5, respectively. The yield decreased significantly in organic solvents of low dielectric constant. The Sn derivative of NPe6 was more light stable than NPe6 (yield = 5.7 × 10 −6), while the Zn derivative was more sensitive (yield = 1.9 × 10−2). Oxygen appeared to be necessary for the photobleaching of NPe6; however, bleaching was not inhibited by 100 mM azide, an efficient quencher of singlet oxygen. The photooxidizable substrates cysteine, dithiothreitol and furfuryl alcohol increased the quantum yield of photoblcaching two- to four-fold, while the electron acceptor, met-ronidazole, increased it almost six-fold. Abstract— The steady-state (254 nm) photolysis of 9–(β-d-erythropentofuranosyl)adenine (adenosine) in aqueous solution was studied. Photodestruction yields on the order of 1.3 × 10−3) were determined at room temperature by measuring the initial decrease in the absorption maximum as a function of irradiation time. The use of high performance liquid chromatography (HPLC) permitted a more exact determination of the yield (2.5 × 103). The formation of photoproducts was also studied using HPLC. In the photolysis of 50 μ M aqueous solutions of adenosine under anaerobic conditions at least 11 stable photoproducts are formed that absorb at 260 nm, the wavelength of maximum absorption of adenosine. The major photoproduct was also isolated and characterized as adenine; its formation yield was determined to be 4.5 × 104. This yield is affected by the presence of oxygen and by the initial concentration of adenosine employed. Fluorescence emission and excitation spectra were used to monitor the formation of highly fluorescent photoproducts that emit with maxima at 365, 398 , and 430 nm and absorb in the wavelength region of 240–380 nm.
The reactive species in the photodestruction mechanism were established using substrates that react selectively with the respective short-lived species. Photoionization is a primary photoprocess implied by these studies. The triplet state of adenosine also contributes to the photodestruction mechanism.  相似文献

Abstract— The ability of UV-irradiation (254 nm) to induce formation of RNA-protein crosslinks in tobacco mosaic virus (TMV) particles have been studied by Cs2SO4 density gradient centrifugation, analytical centrifugation, nitrocellulose filter binding and two-dimensional peptide mapping. RNA-protein crosslinks were found to be formed on UV-irradiation of TMV, but the parallel process of UV-induced RNA chain breakage complicated their quantitation. Using speciall devised equations, the quantum yield of RNA-protein crosslink formation was found to be 0.65 × 10−5 and that of RNA chain break formation 0.95 × 10−5.  相似文献

Abstract— Ferrideuteroporphyrin in benzene, water or micelle solutions containing primary or secondary alcohols as well as in pure or basic 2-propanol solutions is clearly reduced to the ferrous state by continuous light irradiation in the Soret region. Quantum yields range between 4 × 10−4 and 3 × 10−2 depending on the solvents used and on the coordination state of the ferric porphyrin. As inferred from laser pulse photolysis experiments, the primary chemical step appears to be the homolytic cleavage of the bond between the ferric ion and a coordinated alcoholate anion leading to the ferrous porphyrin and the alkoxy radical. This cleavage is found to occur within less than 50 ns. The alkoxy radical rearranges leading to the α-hydroxyalkyl radical which reacts with excess ferric porphyrin leading to further reduction. The reaction rate constant for the reaction of α-hydroxyisopropyl radicals is found to be k = (2.1 ± 0.3) × 108 M −1 s−1 in pure 2-propanol. As expected, this rate is greatly increased in basic 2-propanol where α-hydroxyisopropyl radicals deprotonate.  相似文献

Abstract— 5-Methylcytosine and 5-methyldeoxycytidylic acid are fluorescent in aqueous solution at room temperature and neutral pH. 5-Methylcytosine, 10-3M, pH 8.5, 25°C, has a quantum yield of 5 ×10-4, 5-Methyldeoxycitydylic acid, 10-4M, pH 7.5, 20°C, has a quantum yield of 8 × 10-4. Emission maxima are 2.91 and 2.80μ-1. At pH 14, the quantum yield of 5-methylcytosine is 1.6 × 10-2; the emission maximum is 2.82μ-1. At pH I, the quantum yield of both compounds is less than or equal to 10-4. Both compounds were chromatographically homogeneous, had absorption spectra which agreed with published data, and excitation spectra which agreed closely with absorption spectra.  相似文献

Abstract— The fluorescence decay profiles, relative quantum yield, and transmission of the phycoerythrin a subunit, isolated from the photosynthetic antenna system of Nostoc sp., were measured using single picosecond laser excitation. The fluorescence decay profiles were found to be intensity independent for the intensity range investigated (4 × 1013 and 4 × 1015 photons-cm-2 per pulse). The decay profiles were fitted to a model assuming both chromophores absorb and fluoresce. The inferred total deactivation rates for the two chromophores, in the absence of energy transfer and when the effects of the response time of the streak camera and the finite pulse width are properly included, are 1.0 × 1010s' and 1.0 × 109 s 1 for the s and f chromophores. respectively, whereas the transfer rate between the two fluorophorcs is estimated to be 1.0 × 1010 s−1 giving a s→ f transfer rate on the order of (100 ps)−1. Steady-atate polarization measurements were found to be equal to those calculated using the rate parameters inferred from the kinetic model fit to the fluorescence decays. The apparent decrease in the relative fluorescence quantum yield and increase of the relative transmission with increasing excitation intensity is suggestive of ground state depletion and upper excited state absorption. Evidence suggests that exciton annihilation is absent within isolated α subunits for the intensity range investigated (4 × 1013 to 4 × 1015 photons-cm 2 per pulse).  相似文献

Abstract— A broad-band UVA source that emits primarily350–400 nm radiation and no measurable radiation below 340 nm was used to test toxicity and mutagenicity at the thymidine kinase locus in L5178Y, subclone 3.7.2C (TK+/-) mouse lymphoma cells. Cells were exposed to a fluence of 0 to 80 × 104 J/m2. The relationship between UVA fluence and survival was found to have a shoulder region followed by an exponential decrease in survival at higher fluence levels. An exposure-dependent increase in mutation was observed with increasing fluences from 0 to about 60 × 104 J/m2. An approximately 3- to 4-fold increase in mutations (trifluorothymidine resistance) over unexposed, control cells was seen at a fluence that resulted in 90% cell killing. We conclude that UVA radiation is a mutagen in the L5178Y mouse lymphoma cells used in this study.  相似文献

Abstract— Irradiation of aqueous solutions of plasmid DNA (pUC18) at pH 7.6 with 193 nm laser light results in low yields of prompt single strand breakage (air-saturated sample φssb= [1.5 ± 0.1] ± 10−4, argon-saturated sample φssb= [0.9 ± 0.1] ± 10−4). Treatment of the irradiated DNA samples with Escherichia coli formamidopyrimi-dine-DNA glycosylase (Fpg) protein results in an approximate 20-fold increase in the yield of single strand breakage (air-saturated sample φfpg= [33.1 ± 3.1] ± 10−4, argon-saturated sample φfpg= [23.8 ± 2.6] × 10 4). This result indicates that 193 nm light induces other modification) (most likely of the purine moieties) that are 20 times more abundant than prompt strand breakage within the DNA matrix.  相似文献

Abstract— The effects of repeated UV exposure on the skin of the European crested newt, Triturus cristatus carnifex , have been investigated. The animals were irradiated 3 times per week with a Westing-house FS40T12 fluorescent sun lamp (wavelength spectrum 275–350 nm). Two groups of animals received the same total fluence of 1.3 × 105 J/m2 in single fluences of either 1570 J/m2 (group A) or 9430 J/m2 (group C), and one group received a total fluence of 2.6 × 105 J/m2 in single fluences of 4710 J/m2 (group B). All the animals were killed 7 months after the first UV exposure, but at different intervals after the last exposure. Striking epidermal hyperplasia was found in the newts irradiated at the lower fluence rate (group A). In the animals given the higher total fluence (group B), the most prominent skin changes were dermal fibrosis and irregular thinning and thickening of the epidermis. No significant skin changes were found in group C., in which if there had been UV lesions, they had been repaired during the 5 month interval between the last irradiation and the killing of the animals. No skin tumors developed in any experimental group.  相似文献

Blue light induces the formation of Neurospora crassa protoperithecia.This photoinduction is completed in less than 24 h. Its threshold is about 4.2 J/m2. Red light is ineffective. The Bunsen-Roscoe law is obeyed at the fluence of 12.6 J/m2 for fluence rates from 5.25 × 10 2 to 1.05 W/m2.  相似文献

Abstract— Light-induced accumulations of amoebae of Dictyostelium discoideum in 'light traps' have been observed. The greatest accumulations are obtained with cell densities of about 8 × 104 cells/mm2. Accumulations are observed at incident fluence rates over about one decade both for white and for monochromatic light; higher fluence rates cause dispersal from the 'light trap'. An action spectrum for the photoaccumulation, calculated from fluence-response curves using the zero thresholds, shows a major peak between 405 and 410 nm and extends through most of the visible spectrum. This action spectrum does not coincide with any of the pigments known to be present in D. discoideum . The cellular basis for the photoaccumulation has been studied. No light effects on cell divisions or cell aggregation are observed during the 2 h duration of an accumulation experiment. Microvideographic analysis of single amoebae is consistent with the hypothesis that the amoebae are positively phototactic and move toward the light scattered from cells in the 'light trap', thus accumulating in the trap.  相似文献

Abstract— We characterized the fluorescence intensity decays of Indo-1, which is commonly used as an emission wavelength-ratiometric calcium probe. The apparent lifetime of the long-wavelength side of the emission of Indo-1 is dependent on Ca2+. This long-wavelength emission displays the characteristics of an excited-state reaction, that is, a negative preexponential component in thc multiexponential analysis. The emission spectra and lifetime of Indo-1 appear to be identical for one-photon and two-photon excitation at 351 and 702 mn, respectively, suggesting that the relative one- and two-photon cross sections are similar for the calcium-free and calcium-bound forms of Indo-1. Also, the two-photon cross section of Indo-1 is relatively high, about 4 × 10−49 cm4 s/photon molecule at 690 nm for both the calcium-free and calcium-bound forms. Hence, Indo-1 can be used for calcium imaging based on one- or two-photon excitation, using either emission wavelength ratios or lifetime imaging methods.  相似文献

Abstract— A comparison of the transient absorption spectra from the photolysis of disulfides in solution suggests that C-S bond breakage is a common primary photolytic process. This process becomes more important as the resulting carbon centered radical is stabilized by increasing alkyl substitution or resonance interaction with an aromatic system. The perthiyl radical product is characterized by λmax∽380 nm,ε380∽1700 M −1 cm−1 and decays by second order kinetics with k 2∽3.7×108 M −1 s−1 in water.
In the presence of O2, the photolysis of disulfides which produce the thiyl radical give transient absorptions in the 500–600 nm region. Possible identities of these transients are discussed.  相似文献
