Summary Two commercially available liquid crystals, 4-cyano-4′-n-pentyl-1,1′-bipheny and 4-cyano-4′-n-pentoxy-1,1′-bipheny, are bonded to a silica hydride surface via hydrosilation in the presence of a free radical iniator, t-butyl peroxide. Elemental analysis, diffuse reflectance Fourier trans-form infrared spectroscopy, and13C and29Si CP-MAS NMR spectroscopy are used to confirm the success of the bonding reaction. The13C CP-MAS spectra suggest a difference in the bonded phase morphology of the two materials. Static hydrolytic stability tests indicate these materials do not degrade significantly in both acidic and basic solutions. Chromatographic tests confirm that these two bonded phase behave differently with respect to their retention of PAHs, alkyl-substituted benzenes and benzodiazepines.  相似文献

Summary A propyldimethylphenylsilane stationary phase was prepared by a hydrosilylation reaction on the double bond of an allyl bonded phase intermediate. The carbon load on the silica was between 8.8–9.0%. Evaluation of the material by FTIR indicated high conversion of the double bond by the hydrosilylation reaction. The bonded material displayed reversedphase properties as determined by the retention behavior of alkylarylketones but it was less hydrophobic than either C-8 or C-18. The stability at both low and high pH was excellent. Separations of pharmaceutical compounds, a mixture of anilines, and a mixture of dopamine and epinephrine were satisfactory.  相似文献

Grignard and trichlorosilane coupling methods have been applied to the synthesis of surface-modified silicas carrying alkyl chains of varying length, terminally substituted by a hydroxyl group. Summary The versatility of the allyl phase is exploited by using it as an intermediate in the production of a weak anion exchange material that would be difficult to synthesize directly using organochlorosilane reagents. The stability of the allyl phase under the severe reaction conditions (170° and 140 atm of CO) of aminomethylation also is demonstrated. The product N-butylpyrrolidine phase is characterized spectroscopically by ESCA and DRIFT which also indicate a high conversion (greater than 90 %) of the allyl phase. In addition, the carbon-nitrogen ratio of the product phase supports a high yield. Chromatographic characterization is done by the retention indices of the alkylarylketone homologous series and by capacity factor (k′) determinations for inorganic anions and amino acids.  相似文献

The retention of a solute in RP chromatography is a very complex process which depends on many factors. Therefore, the study of the influence of a mobile phase modifier concentration on the retention in different reversed phase chromatographic systems is very important for understanding the rules governing retention and mechanisms of substance separation in a chromatographic process. Composition changes and the nature of mobile phases enable tuning of the separated analytes' retention over a wide range of retention parameters and optimization of the chromatographic process as well. Optimization of the chromatographic process can be achieved by several different methods; one of them is the so-called interpretative strategy. The key approach adopted in this strategy is the implementation of adequate retention models that couple the retention of solute with the composition of a mixed mobile phase. The use of chemically bonded stationary phases composed of partially non-bonded silica matrix and organic ligands bonded to its surface in everyday chromatography practice leads to questions of the correct definition of the retention model and the dominant retention mechanism in such chromatographic systems. The retention model for an accurate prediction of retention factor as a function of modifier concentration and the heterogeneity of the adsorbent surface should be taken into consideration. In this work the influence of mobile-phase composition on the retention of sixteen model substances such as phenols, quinolines, and anilines used as test analytes in different RP-TLC systems with CN-, NH2-, and Diol-silica polar bonded stationary phases has been studied. The aim of this study is to compare the performance of three valuable retention models assumed as the partition, adsorption/partition, and adsorption mechanism of retention. All the models were verified for different RP-TLC systems by three statistical criteria. The results of investigations presented in this work demonstrate that the best agreement between the experimental and calculated Rf values was obtained by the use of new-generation retention models, which assume heterogeneity of adsorbent surface. The results reported here show that heterogeneity of the adsorbent surface may be important in analysis of the elution process in liquid chromatography. Consideration of the goodness of fit for the experimental data to the examined retention models is in conformity with the adsorption mechanism of retention on all polar bonded stationary phases in most eluent systems for most investigated compounds.  相似文献

Summary The allyl bonded phase is used as an intermediate in the synthesis of a weak cation exchange material. The double bond is first converted to an aldehyde by an ozonation reaction and then subsequent oxidation to the carboxylate group is accomplished with dichromate. Diffuse reflectance Fourier transform infrared spectroscopy (DRIFT) is used to monitor the conversion process. The presence of the aldehyde functional group is confirmed by reaction with 2,4-dinitrophenylhydrazine. ESCA spectra reveal no residual chromium on the silica surface. The product is tested chromatographically by the retention of alkylarylketones, the ion-exchange behavior of both monovalent and divalent cations, the retention of nucleosides and bases, and the separation of a mixture of hemoglobins.  相似文献

以3-巯丙基三甲氧基硅烷为偶联剂,将聚N-异丙基丙烯酰胺(PNIPAM)键合到硅胶上,制得了键合固定相(SI-PNIPAM)填料,并用元素分析、红外光谱等对其进行了表征。以甲醇-水为二元流动相,用多环芳烃、碱性物质对该固定相进行了色谱评价,并考察了该固定相的适用pH范围及水解稳定性。结果表明:该固定相具有较好的色谱性能与温敏特性,并且在pH 2.5~7.5时稳定性良好。  相似文献

The effects of stationary and mobile phase on retention of 18 beta-adrenolytic drugs (beta-blockers) have been studied. Four 'deactivated surface' stationary phases (polar-embedded or end-capped) were examined. Special attention was drawn to the cholesterolic (SG-CHOL) and alkylamide (SG-AP) stationary phases, and their application for analysis of the compounds. The retention of analyzed substances was also examined in terms of mobile phase composition. Sixteen different configurations of mobile phases were prepared, all based on methanol and acetonitrile with ammonium acetate and ammonium formate. The difference in retention between ammonium formate and acetate water solutions, and peak shape changes related to the addition of triethylamine (TEA), were investigated. Principal component analysis was used to find the similarities between stationary phases. Polar-embedded phases synthesized on the same sorbent possess very similar properties. All phases based on silica gel compared with the monolithic column also showed similarities in retention of beta-blockers. The addition of TEA to the mobile phase did not influence strongly the retention, and analysis of asymmetry factors showed only a little peak broadening for a few compounds on the monolithic column.  相似文献

The properties of four cholesteric bonded stationary phases differing in the nature of the spacer and the end-capping were assessed using simple chromatographic tests based on the retention of nonpolar compounds and of planar or nonplanar probe solutes. All cholesteric columns showed a hydrophobicity close to that of conventional octadecyldimethylsilyl (ODS) materials. Non-end-capped cholesteric bonded phases showed greater selectivity than ODS ones and both end-capped cholesteric bonded phases exhibit behavior intermediate between that of the non-end-capped original material and that of the ODS bonded phase.  相似文献

A rapid and stereospecific HPLC micromethod to quantify flurbiprofen enantiomers was developed. Both flurbiprofen enantiomers and indomethacin, used as internal standard, were extracted with methylene chloride from 100 microL of acidified plasma. The resolution of the R- and S-forms was performed on a bonded vancomycin chiral stationary phase (Chirobiotic V) with 20% of tetrahydrofuran in ammonium nitrate (100 mM, pH 5) as mobile phase. Calibration curves were linear in the range 0.5-10 microg/mL for both enantiomers. A good accuracy (< or = 5%) was obtained for all quality controls, with intra-day and inter-day variation coefficients equal or less than 7.7%. Recovery of both enantiomers was found in the range 77.4-86.3%. The lower limit of quantitation was 0.25 microg/mL for both enantiomers, without interference of endogenous components. This validated micromethod has been successfully applied for quantifying R- flurbiprofen and S- flurbiprofen in rat plasma.  相似文献

以聚乙二醇(PEG)为致孔剂,四甲氧基硅烷(TMOS)和乙烯基三甲氧基硅烷(VTMS)为杂化硅胶前驱体,在乙酸催化作用下使硅烷发生水解,在尿素加热分解提供的碱性环境下水解的硅烷进一步缩聚得到杂化硅胶整体材料。将此整体材料用球磨机研磨,然后用三羟甲基氨基甲烷处理,并洗涤干燥得到粒径为3 μm左右的硅胶颗粒。探索了不同反应条件对硅胶颗粒的大小、比表面积和孔径、表面形貌和分散性的影响;当TMOS和VTMS体积比为3:1时可以得到孔径为7.5 nm和比表面积为245 m2/g的硅胶颗粒。通过对所制得的硅胶颗粒表面进行C18(十八烷基二甲基氯硅烷)键合修饰和巯基-烯点击反应,得到混合型高效液相色谱固定相。对此固定相的测试结果表明以上硅胶色谱填料的制备方法具有一定的实用性。  相似文献

Summary A chromatographic and thermodynamic study of the compound [4-(allyloxy)benzoyl]-4-methoxyphenyl (ABMP) as a model of a chemically bonded liquid crystal stationary phase for HPLC was undertaken. A number of polycyclic aromatic hydrocarbons (PAHs) and two small solutes, carvone and pulegone, were studied under varying solvent and temperature conditions. Plots of log k vs. % organic in the mobile phase were not completely linear in all cases. The van't Hoff plots revealed at least one phase transition. The enthalpies of solute transfer from the mobile phase to the ABMP phase were determined for several PAHs. All tests indicate that ABMP possess liquid crystal properties when bonded to particulate silica.  相似文献

以ODS为参比,研究了5种水溶性维生素和5种核苷在姜黄素键合硅胶固定相(CCSP)上的色谱行为,考察了甲醇、流动相pH和离子强度对CCSP分离这类极性化合物的影响,探讨了该新固定相的色谱保留机理。结果表明,在V(甲醇)∶V(0.01 mol/L NaH2PO4)=40∶60(pH 3.5)流动相体系中,5种水溶性维生素在CCSP上实现较好分离;核苷的分离则是在以纯水为流动相的条件下实现的。与ODS柱相比,在相同条件下,CCSP对5种水溶性维生素具有较高的选择性,洗脱顺序与ODS具有显著差异,且保留明显比ODS强(B6除外);与此相反,相同色谱条件下,CCSP对核苷的保留比ODS弱,CCSP可实现核苷的简便有效分离,同时胸苷与黄苷的洗脱顺序也与ODS不同。以上说明不同保留机理的存在,CCSP具有典型的反相色谱特征,但疏水性比ODS弱;极性的配体使CCSP在对溶质的分离分析中除疏水作用外,还存在n-π和π-π作用、氢键作用、偶极-偶极等作用;协同作用的结果使CCSP在水溶性极性化合物的分离分析中显示出优势。  相似文献
