In this paper, N‐isopropylacrylamide (NIPA) was synthesized by acrylonitrile and isopropanol. Poly(N‐isopropylacrylamide) (PNIPA) was prepared by a chemical method. The dependence of its swelling behavior on temperature was studied. Results showed that PNIPA hydrogel was a temperature‐sensitive gel. Its LCST (lower critical solution temperature) was about 32 °C, and its swelling ratio (at 20 °C) was about 17–18. Sodium acrylate (SA) and sodium methylacrylate (SMA) were copolymerized with NIPA respectively. Equilibrium swelling ratios of the copolymer hydrogels at lower temperature were two to three times that of PNIPA. The LCST of the copolymer hydrogels could be controlled between 32 and 45 °C by adjusting the content of SA or SMA. Kinetics of P(NIPA‐co‐SA) hydrogels, whether swelling or shrinking processes, were in good agreement with apparent second order kinetic equations. Several experiments were designed to separate aqueous bovine serum albumin solution using the hydrogels prepared above. The separation efficiency was about 80%. The dextran‐allyl isocyanate/poly(N‐isopropylacrylamide) (Dex‐AI/PNIPAAm) hydrogel was designed and prepared by copolymerization of the modified dextran with N‐isopropylacrylamide (NIPAAm). This novel Dex‐AI/PNIPAAm hydrogel is biodegradable and intelligent due to its biodegradable dextran linkage and thermosensitive PNIPAAm moiety. With an increase in dextran content, it exhibits the increased lower critical solution temperature (LCST) and decreased porous microstructure. Also, the thermosensitivity of this hydrogel is also controllable and adjustable depending on the different compositions.

Fast responsive temperature‐ and pH‐sensitive hydrogels of poly(N,N‐diethylacrylamide‐co‐acrylic acid) (P(DEA‐co‐AA)) have been synthesized successfully by a two‐step procedure, in which the initial polymerization was conducted at constant temperature for 15 min, followed by further polymerization at ?30°C for 12 hr. Swelling studies showed that hydrogels thus prepared had almost the same temperature and pH sensitivity compared with the conventional ones (polymerized at 24°C for 12 hr). However, hydrogels thus prepared had faster swelling/deswelling rates in distilled water than the conventional ones, and their deswelling rates in low pH buffer solutions were also faster than the conventional ones. These improved properties were attributed to the porous network structure, which was confirmed by the results of scanning electron microscopy. Copyright © 2009 John Wiley & Sons, Ltd.  相似文献

Chitosan (CS), CS‐poly(N‐isopropylacrylamide)(PNIPAM) and their dyed (pyrene) hydrogels were prepared using glutaraldehyde (Glu) as a crosslinker. The gelation rate, swelling behaviors in ethanol/water mixtures, electricity‐induced contraction and thermoresponse of the gels were investigated using fluorescence probe technique. Results showed that CS/Glu, and PNIPAM‐containing CS/Glu gels exhibited similar properties in all aspects examined, except that the transparence of the CS‐PNIPAM/Glu gel is very dependent upon the temperature. The CS‐PNIPAM/Glu gel is transparent below 30°C, whereas opaque above 32°C. It is expected that this observation may be useful for the design and preparation of new kinds of hydrogel devices. © 2000 John Wiley & Sons, Inc. In this paper, we report on a novel electrophoretic separation and analysis method for membrane pore‐forming proteins in multilayer lipid membranes (MLMs) in order to overcome the problems related to current separation and analysis methods of membrane proteins, and to obtain a high‐performance separation method on the basis of specific properties of the lipid membranes. We constructed MLMs, and subsequently characterized membrane pore‐forming protein behavior in MLMs. Through the use of these MLMs, we were able to successfully separate and analyze membrane pore‐forming proteins in MLMs. To the best of our knowledge, this research is the first example of membrane pore‐forming protein separation in lipid membranes. Our method can be expected to be applied for the separation and analysis of other membrane proteins including intrinsic membrane proteins and to result in high‐performance by utilizing the specific properties of lipid membranes.  相似文献

Adopting inorganic clay (hectorite) and MBA as physical and chemical cross‐linking agents, respectively, PNIPAM microgels were synthesized by SFEP. The chemical structure and morphology of the microgels were confirmed by FTIR, WXRD, and SEM. The temperature‐sensitivity of the microgels was investigated by DLS and UV spectrophotometers. The results inferred that clay platelets dispersed in an aqueous medium were fully exfoliated and could act as a kind of multifunctional cross‐linking agent and significantly reduced the hydrodynamic diameters of the microgels. In fact, the hydrodynamic diameters of the PNIPAM microgels with clay as cross‐linker ranged from 154 to 322 nm which was much smaller than that using MBA as chemical cross‐linker, the later was in the range of 284–808 nm on heating from 5 to 50 °C.

In this study, the effect of the level of crosslinking on the properties of poly(N‐isopropylacrylamide) (PNIPAAm) hydrogels was investigated in terms of their lower critical solution temperature (LCST), interior morphology, equilibrium swelling, and deswelling and swelling kinetics. The thermal analysis showed that PNIPAAm hydrogels, having a wide range of crosslinking levels, exhibited almost the same LCSTs, and this was different from what the conventional theory would have predicted. Scanning electron micrographs revealed that the interior network structure of the PNIPAAm matrix became more porous with an increase in the level of crosslinking. This more porous matrix provided numerous water channels for water diffusion in or out of the matrix and, therefore, an improved response rate to the external temperature changes during the deswelling process and the swelling process. © 2003 Wiley Periodicals, Inc. Dual temperature‐ and pH‐sensitive hydrogels composed of N‐isopropylacrylamide (NIPAM) and 2‐acrylamido‐2‐methyl‐propanosulfonic acid (AMPS) were prepared by free‐radical crosslinking copolymerization in aqueous solution at 22 °C. The mole percent of AMPS in the comonomer feed was varied between 0.0 and 7.5, while the crosslinker ratio was fixed at 5.0/100. The effect of AMPS content on thermo‐ and pH‐ induced phase transitions as well as equilibrium swelling/deswelling, interior morphology and network structure was investigated. The volume phase transition temperature (VPT‐T) was determined by both swelling/deswelling measurements and differential scanning calorimetry (DSC) technique. In addition, the volume phase transition pH (VPT‐pH) was detected from the derivative of the curves of the swelling ratio (dQv/dpH) versus pH. The polymer‐solvent interaction parameter (χ) and the average molecular mass between crosslinks ( ) of hydrogels were calculated from swelling ratios in buffer solutions at various pHs. The enthalpy (ΔH) and entropy (ΔS) changes appearing in the χ parameter of hydrogels were also determined by using the modified Flory–Rehner equation. The negative values for ΔH and ΔS indicated that the hydrogels had a negative temperature‐sensitive property in water, that is, swelling at a lower temperature and shrinking at a higher temperature. It was observed that the experimental swelling data of hydrogels at different temperature agreed with the modified Flory‐Rehner approach based on the affine network model. © 2008 Wiley Periodicals, Inc. Thermosensitive Poly(N‐isopropylacrylamide) (PNIPA) hydrogels were synthesized by a free radical solution polymerization in three different ways. Normal hydrogels were prepared at room temperature and normal cryogels were prepared at subzero temperature. A cation surfactant dodecyl dimethyl benzyl ammonium bromide (DDBAB) was used during preparation of novel cryogels in freezing state. The response rates of normal hydrogels were very slow, whereas the rates of both normal and novel cryogels were very fast because of the macroporous structure of the cryogels. Mixed solvents which were composed of pure water and 1,4‐dioxane at various concentrations were used instead of pure water during the polymerization. The effects of the mixed solvent on morphology, swelling ratio, and deswelling/reswelling kinetics of the three kinds of hydrogels were investigated. For normal hydrogels and normal cryogels, there was no remarkable difference no matter the mixed solvent or pure water was used. However, the properties of the resulted novel cryogels were much different with the concentration of dioxane. Finally, the resulted hydrogels were used for concentrating emulsified paraffin. Linear and crosslinked polymers based on N‐isopropylacrylamide (NIPAAm) exhibit unusual thermal properties. Aqueous solutions of poly(N‐isopropylacrylamide) (PNIPAAm) phase‐separate upon heating above a lower critical solution temperature (LCST), whereas related hydrogels undergo a swelling–shrinking transition at an LCST. A linear copolymer made of NIPAAm/acryloxysuccinimide (98/2 mol/mol) and two hydrogels with different hydrophilicities were prepared. Fourier transform infrared (FTIR) spectroscopy was employed to determine the transition temperature and provide insights into the molecular details of the transition via probing of characteristic bands as a function of temperature. The FTIR spectroscopy method described here allowed the determination of the transition temperature for both the linear and crosslinked polymers. The transition temperatures for PNIPAAm and the gel resulting from the crosslinking with polylysine or N,N′‐methylenebisacrylamide (MBA) were in the same range, 30–35 °C. For the gels, the transition temperature increased with the hydrophilicity of the polymer matrix. The spectral changes observed at the LCST were similar for the free chains and the hydrogels, implying a similar molecular reorganization during the transition. The C H stretching region suggests that the N‐isopropyl groups and the backbone both underwent conformational changes and became more ordered upon heating above the LCST. An analysis of the amide I band suggests that the amide groups of the linear polymer were mainly involved in hydrogen bonding with water molecules below the LCST, the chain being flexible and disordered in a water solution. During the transition, around 20% of these intermolecular hydrogen bonds between the polymer and water were broken and replaced by intramolecular hydrogen bonds. A novel one‐step approach is reported to prepare thermosensitive hydrogels simply by using hydroxypropyl‐β‐cyclodextrin (HP‐β‐CD)/glycidyl methacrylate (GMA)/N‐isopropylacrylamide (NIPAM) system. From GMA and HP‐β‐CD, HP‐β‐CD/GMA inclusion complex was prepared and identified with NMR, FTIR, and UV‐vis spectroscopies. GMA in the form of HP‐β‐CD/GMA complex was copolymerized with NIPAM in water with K2S2O8 as initiator, yielding hydrogels designated as poly(NIPAM‐CD‐GMA). The inclusion of CD in the hydrogels was confirmed by FTIR spectroscopy. The contents of CD and GMA placed considerable influence on the swelling ratio and temperature‐sensitivity of the produced hydrogels. The hydrogels bearing CD moieties showed higher swelling ratio and temperature‐sensitivity when compared with that without CD. The porous structure of the hydrogels containing CD was observed in the SEM images. Relevant mechanism of the ring‐opening reaction of epoxide groups in GMA, the subsequent crosslinking reactions and the formation of hydrogels containing CD moieties were proposed. © 2008 Wiley Periodicals, Inc. The temperature‐sensitive poly(N‐isopropylacrylamide) hydrogels, prepared by γ and electron‐beam (EB) irradiation, were studied using positron annihilation lifetime spectroscopy (PALS). The effect of water content in the hydrogel on the ortho‐positronium (o‐Ps) lifetime and intensity was investigated. The observed positronium lifetime suggests microstructural differences between γ‐ and EB‐synthesized hydrogels. The distribution in positronium lifetime indicates nonhomogeneity in the distribution of free‐volume holes in EB‐synthesized hydrogels. © 2000 John Wiley & Sons, Inc. The use of polymeric materials with temperature‐dependent degrees of swelling (especially polymers that exhibit lower critical solution temperature (LCST) behaviour in aqueous solutions) in microsystems requires the preparation and patterning of layers in the µm range. Copolymers based on N‐isopropylacrylamide were modi‐fied with a stilbazolium salt chromophore to yield photocrosslinkable temperature‐sensitive polymers. The chromophore and the polymers were characterized by UV, IR, 1H‐NMR (nuclear magnetic resonance) and 13C‐NMR spectra. The resulting polymers showed LCST behaviour, which was measured by differential scanning calorimetry. The photocrosslinking properties were studied by UV irradiation of the thin films and measurement of the changes in the UV absorption spectra. By irradiation of thin films through a mask it was possible to obtain patterned networks in the µm range (20 µm space width and ≥50 µm line width). A functionalized cyclam was synthesized by the attachment of a polymerizable acryloyl group to one of the four nitrogens on the cyclam molecule. The polymerization of the functionalized cyclam was performed with N‐isopropylacrylamide and N,N′‐methylene bisacrylamide, and the gels obtained were studied in the presence of different transition‐metal‐ion solutions. There was a drastic difference in the phase‐transition temperature (Tc) of the poly(N‐isopropylacrylamide) (PNIPAAm)/cyclam gel in comparison with the pure PNIPAAm gel. For the described system, a Tc shift of 15 °C was obtained. The presence of functionalized cyclam increased the hydrophilicity and Tc of the aforementioned polymer gels in deionized water (at pH 6) because of the presence of protonated amino moieties. The PNIPAAm/cyclam gels showed a dependence of the swelling behavior on pH. Tc of the pure PNIPAAm gel was weakly influenced by the presence of any transition‐metal ions, such as Cu2+, Ni2+, Zn2+, and Mn2+. The addition of Cu2+ or Ni2+ to the PNIPAAm/cyclam gel reduced Tc of the polymer gel, and a shift of approximately 12 °C was observed. © 2003 Wiley Periodicals, Inc. An improved, simple, and efficient method for the synthesis of lactose‐containing monomer acrylamidolactamine (LAM) has been reported. Free radical copolymerization of this monomer with N‐isopropylacrylamide (NIPAM) in the presence of the crosslinking reagent N,N′‐methylenebisacrylamide (BisA) (1.2 mol %) proceeded smoothly in an aqueous solution using potassium persulfate (KPS) and N,N,N′,N′‐tetramethylethylenediamine (TMEDA) as the initiating system and gave transparent hydrogels. Reactivity ratios were estimated from copolymerization reactions carried out in solution without BisA crosslinker and at low conversion, by using both linearization and nonlinearization methods. They were found to be rLAM = 0.75 and rNIPAM = 1.22. The swelling behavior of the hydrogels was studied by immersion of the hydrogels in deionized water at different temperatures. Equilibrium water uptake was increased when the LAM content was higher than 47 mol %, and reached ≈ 44‐fold with 100 mol % LAM at room temperature. Depending on the composition, the gels showed sharp swelling transitions with small changes in temperature. Differential scanning calorimetry (DSC) was used to characterize the swelling transition and the organization of water in the copolymer hydrogels. The amounts of freezable water in these hydrogels ranged from 81 to 89%, and was not correlated to the content of the sugar monomer. Interpenetrating polymer network (IPN) strategy was developed to fabricate novel hydrogels composed of cellulose and poly(N‐isopropylacrylamide) (PNIPAAm) with high mechanical strength and adjustable thermosensitivity. Cellulose hydrogels were prepared by chemically cross‐linking cellulose in NaOH/urea aqueous solution, which were employed as the first network. The second network was subsequently obtained by in situ polymerization/cross‐linking of N‐isopropylacrylamide in the cellulose hydrogels. The results from FTIR and solid 13C NMR indicated that the two networks co‐existed in the IPN hydrogels, which exhibited uniform porous structure, as a result of good compatibility. The mechanical and swelling properties of IPN hydrogels were strongly dependent on the weight ratio of two networks. Their temperature‐sensitive behaviors and deswelling kinetics were also discussed. This work created double network hydrogels, which combined the advantages of natural polymer and synthesized PNIPAAm collectively in one system, leading to the controllable temperature response and improvement in the physical properties. Novel temperature-sensitive poly(N-isopropylacrylamide)/amine-terminated polyamidoamine dendrimer G6-NH2 hydrogels with fast responsive properties were synthesized by forming semi-interpenetrating polymeric networks. In contrast to the conventional PNIPA hydrogel, these new gels showed rapid shrinking rate at the temperature above lower critical solution temperature (LCST), and exhibited higher equilibrium swelling ratio at room temperature. All these properties might be attributed to the incorporation of polyamidoamine dendrimer G6-NH2, which forms water-releasing channels and increases the hydrophilicity of PNIPA network. The novel hydrogels have potential applications in drug and gene delivery.  相似文献

Dual responsive cyclotriphosphazene (CTP)‐based hydrogels have been synthesized for a controlled release of FU, a hydrophilic drugs. These hydrogels composed of mono (methacryloyl‐2‐ethoxy)‐pentakis(N1,N1‐dimethylpropane‐1,3‐diamino)‐cyclotriphosphazene (HEMA (DMPDA)5CP), acryl amide and pectin were synthesized by free radical polymerization method using methylenebisacrylamide cross linker. The CTP hydrogels were characterized to understand the structure, drug nature in the network and morphology by FTIR, DSC, XRD and SEM, respectively. In this paper, the swelling (dynamic and equilibrium) properties of cyclotriphosphazene hydrogels were investigated, showing dual (pH and thermo) responsiveness and large variation in the swelling capacity. Based on these results the structural parameters of the hydrogel networks such as the average molecular weight between cross‐links (Mc) and polymer–solvent interaction parameter (χ) were determined. The CTP hydrogels has high FU loading efficiency 65 ± 0.5. In‐vitro FU release of these hydrogels was controlled for about 24 hr also hydrogel showed a distinct initial burst. The CTP hydrogels are bearing both hydrophilic groups of pectin and hydrophobic groups of CTP exhibited dual responsive behaviors with pH and temperature. Poly(N‐isopropylacrylamide) (PIPAAm), which is a well‐known temperature‐responsive polymer, is modified on substrates by various methods. At 37 °C, PIPAAm modified surface is hydrophobic and allows cells to adhere to and proliferate on the surface. By reducing temperature below the lower critical solution temperature of PIPAAm, the surface turns to hydrophilic and allows cells to detach themselves from the surface spontaneously. With this technology, cell sheet engineering is established several years ago. This review focuses on the preparations and characteristics of PIPAAm‐modified surfaces, and discusses the effect of surface properties on cell adhesion and deadhesion. In addition, the recent improvement of PIPAAm‐modified surfaces for cell culture and the clinical applications of cell sheets harvested from the surfaces are also mentioned. © 2014 Wiley Periodicals, Inc. Macroporous, temperature‐sensitive poly(N‐isopropylacrylamide) (PNIPAAm) hydrogels were synthesized with poly(ethylene glycol)s (PEGs; molecular weight = 2000–6000) as the pore‐forming agents. The influence of the molecular weight and PEG content on the responsive kinetics of these macroporous hydrogels was investigated. The PEG‐modified PNIPAAm hydrogels were characterized by the swelling ratio, deswelling–reswelling kinetics, Fourier transform infrared, and differential scanning calorimetry. The morphology of these hydrogels was analyzed with scanning electron microscopy. The prepared macroporous hydrogels exhibited some unique properties in comparison with the gels with low molecular weight PEGs (molecular weight < 2000) as the pore‐forming agents. In addition, a preliminary study on the controlled release of bovine serum albumin from these macroporous hydrogels was carried out. © 2002 Wiley Periodicals, Inc.
