Abstract— The triplet state spectra of anaerobically grown cells of Rhodopseudomonas sphaeroides GIC and R. sphaeroides wild type and aerobically grown cells of R. sphaeroides wild type were examined under reducing and non-reducing conditions at 100 K. by electron paramagnetic resonance (EPR). Large differences in the zero field splitting parameters, |D| and |E| of the triplets are seen to arise in the different preparations. The spectra are assigned to the triplet states of carotenoids, some of which are closely associated with the reaction center and others are contained in the light harvesting complex. Taken together with the triplet state data on Rhodospirillum rubrum wild type previously published the trends in the zero field splitting parameters can be understood in terms of the extent of electron derealization in carotenoids of varying length.  相似文献

The photolysis of lumichrome, riboflavin, flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD) was studied in air-saturated aqueous solution at room temperature in the presence of appropriate electron donors: ascorbic acid, aromatic amino acids or amines, e.g. ethylenediaminetetraacetate (EDTA). The overall reaction is conversion of oxygen via the hydroperoxyl/superoxide radical into hydrogen peroxide. The quantum yield of oxygen uptake increases with the donor concentration, e.g. up to 0.3 for riboflavin, FMN or FAD in the presence of EDTA or ascorbic acid (0.3-10mM). The formation of H(2)O(2) is initiated by quenching of the acceptor triplet state by the electron donor and subsequent reaction of the semiquinone radical with oxygen. Specific properties of flavins are discussed including the radicals involved and the pH and concentration dependences. The quantum yield of photodegradation is low under air, but substantial under argon, where the major product absorbing in the visible spectral range is the corresponding hydroquinone.  相似文献

The pH dependent behavior of two flavin cofactors, flavin-adenine dinucleotide (FAD) and flavin mononucleotide (FMN), has been characterized using femtosecond transient absorption spectroscopy for the first time. The flavin excited state was characterized in three states of protonation (Fl(-), Fl, and FlH(+)). We found that Fl and Fl(-) exhibit the same excited state absorption but that the lifetime of Fl(-) is much shorter than that of Fl. The transient absorption spectrum of FlH(+) is significantly different from Fl and Fl(-), suggesting that the electronic properties of the flavin chromophore become appreciably modified by protonation. We further studied the excited state protonation of the flavin and found that the protonation sites of the flavin in the ground and excited state are not equivalent. In the case of FAD, its excited state dynamics are controlled by the two conformations it adopts. At low and high pH, FAD adopts an "open" conformation and behaves the same as FMN. In a neutral pH range, FAD undergoes a fast excited state deactivation due to the "stacked" conformer. The transition from stacked to open conformer occurs at pH ~ 3 (because of adenine protonation) and pH ~ 10 (because of flavin deprotonation).  相似文献

The high resolution X-band electron para magnetic resonance (EPR) spectrum of quintet pyridyl-2,6-dinitrene was recorded after the photolysis of 4-amino-2,6-diazido-3,5-dichloropyridine in solid argon matrix at 15 K. This spectrum represents a new type of powder EPR spectra that are characteristic for quintet spin states with zero-field splitting parameters |E(q)/D(q)| approximately 1/4. All EPR lines of the quintet dinitrene were unambiguously assigned based on the eigenfield calculations of the Zeeman energy levels and angular dependencies of resonance magnetic fields. Owing to the high resolution of the experimental EPR spectrum, zero-field splitting parameters of the quintet dinitrene were determined with a high accuracy: D(q)=0.2100+/-0.0005 cm(-1) and E(q)=-0.0560+/-0.0002 cm(-1). These parameters provide correct information regarding the molecular angle Theta and distance r between two triplet sites in the molecule of quintet dinitrene. The measured molecular angle Theta=114.2 degrees+/-0.2 degrees is in excellent agreement with results of the density functional theory calculations. The analysis of the magnetic parameters shows that the spin population on the nitrene units in the quintet dinitrene is greater than that on the nitrene unit in the triplet nitrene.  相似文献

6,9-Di(tert-butyl)-1-methyltetrazolo[1,5-a]perimidine (1) has been synthesized from naphthalene in seven steps. The EPR spectra, recorded after irradiation of 1 in a butyronitrile matrix at 77 K (lambda = 351 nm) and in Ar and Xe matrixes at 4.6 K (lambda > or = 345 nm), showed a six-line, high-field signal (Delta m(S) = +/- 1), centered at 3350 G in butyronitrile, along with a half-field signal (Delta m(S) = +/- 2), which is characteristic for triplets. Simulation of the observed EPR spectra gave values for the zero-field splitting parameters of |D/hc|/cm(-1) = 0.0105, |E/hc|/cm(-1) = 0.0014 in butyronitrile and |D/hc|/cm(-1) = 0.0107, |E/hc|/cm(-1) = 0.0016 in Ar. These EPR parameters are consistent with the diradical 5,8-di(tert-butyl)-2-(N-methylimino)perimidine-1,3-diyl ((3)2) as source of the EPR spectra. Linearity of the Curie-Weiss plot and UB3LYP and (14/14)CASPT2 calculations of the singlet-triplet energy difference (DeltaE(ST) approximately 8-10 kcal/mol) indicate that the triplet is the ground state of 2, as predicted for such a nondisjoint diradical.  相似文献

Abstract— A continuation of studies is presented on the excited triplet state of flavins using EPR techniques. Detailed experiments are reported on the triplet state of flavin-mono-nucleotide (FMN) and flavin-adenine-dinucleotide (FAD). Action spectra of triplet yield are explained in terms of the optical absorption for FMN and FAD. Effects of light saturation, concentration quenching and oxygen on the triplet state are discussed. It is suggested that the rate constant k 3 for the intersystem crossover from the excited singlet to the triplet state is increased by oxygen and quenchers such as KI. Detailed kinetic studies are presented on the formation of the triplet state.  相似文献

Abstract— The triplet state of flavin derivatives and d-amino acid oxidase was observed by electron paramagnetic resonance at 77°K.
Flavin triplets (Δ m =± 2) originate from the isoalloxazine ring and are resonant at 1560 guass.The half-life of the FMN triplet in 1 N HCl is 15 nisec.This life-time is prolonged indirectly by the presence of paramagnetic species, such as oxygen or free radicals.
The flavin triplet state is pH dependent.In neutral solution the nlaximum triplet yield is obtained and the longest life-time is observed.The triplet state is affected by intra-and inter-molecular complex formation, FAD is partially quenched by indirectly substituted adenine.Tryptophan quenches completely the FMN triplet.The FAD triplet of d-amino acid oxidase is enhanced but the life-time is shortened relative to a pure FAD solution.  相似文献

Time-resolved and steady-state electron paramagnetic resonance (EPR) spectra have been observed for the lowest excited triplet (T(1)) states of phenazine (Phz) and its singly protonated cation (phenazinium) in sulfuric acid-ethanol mixtures at 77K. The single protonation appears to have little effect on the anisotropic sublevel populating rates of the T(1) state of phenazine. However, the zero-field splitting (ZFS) parameter D decreases on the protonation, reflecting the increase of delocalization of the two unpaired electrons. The sublevel preferentially populated by intersystem crossing (ISC) is T(y) in both phenazine and phenazinium (the y-axis is parallel to the in-plane long axis). From the analysis of the observed anisotropy in the ISC rates and the semi empirical molecular orbital calculations of the ZFS parameters, we concluded that the T(1) state of phenazinium is the (3)A(1)(pipi*) state.  相似文献

Abstract— The effect of flavin structure variation upon the binding process of flavin to hen egg white riboflavin was studied using fluorescence methods for formylmethylflavin (FMF), riboflavin (RF) and flavomononucleotide (FMN).
Measurements of flavin fluorescence intensities (steady state and phase-sensitive) and lifetimes were performed in a variety of RBP concentrations and temperatures (4 to 40°C). No fluorescence of flavoproteins was detected, while the fluorescence of flavins was found to be quenched by RBP. The overall quenching process is dominated by the static quenching (about 88%) due to the flavoprotein complex formation in the ground state, presumably a charge transfer complex.  相似文献

The blue-light sensitive photoreceptor, phototropin, is a flavoprotein which regulates the phototropism response of higher plants. The photoinduced triplet state and the photoreactivity of the flavin-mononucleotide (FMN) cofactor in two LOV domains of Avena sativa, Adiantum capillus-veneris, and Chlamydomonas reinhardtii phototropin have been studied by time-resolved electron paramagnetic resonance (EPR) and UV-vis spectroscopy at low temperatures (T < or = 80 K). Differences in the electronic structure of the FMN as reflected by altered zero-field splitting parameters of the triplet state could be correlated with changes in the amino acid composition of the binding pocket in wild-type LOV1 and LOV2 as well as in mutant LOV domains. Even at cryogenic temperatures, time-resolved EPR experiments indicate photoreactivity of the wild-type LOV domains, which was further characterized by UV-vis spectroscopy. Wild-type LOV1 and LOV2 were found to form an adduct between the FMN cofactor and the functional cysteine with a yield of 22% and 68%, respectively. The absorption maximum of the low-temperature photoproduct of wild-type LOV2 is red-shifted by about 15 nm as compared with the FMN C(4a)-cysteinyl adduct formed at room temperature. In light of these observations, we discuss a radical-pair reaction mechanism for the primary photoreaction in LOV domains.  相似文献

A method is described for the rapid determination of flavins in sea water, based on solid-phase extraction followed by ion-pair high-performance liquid chromatography (HPLC) with fluorescence detection. Riboflavin, flavin mononucleotide (FMN), and flavin adenine dinucleotide (FAD) and their photochemical breakdown products, lumiflavin, formylmethylflavin, and lumichrome can be determined with subpicomolar detection limits. The method was used at sea in the analysis of coastal and open ocean waters. In both environments, riboflavin, lumiflavin and lumichrome were routinely observed at concentrations in the picomolar range; lumiflavin and lumichrome were generally confined to the photic zone while riboflavin was present throughout the water column. Formylmethylflavin, FMN, and FAD were only occasionally observed; when present, these flavins were observed at consistently higher concentrations than riboflavin, lumiflavin and lumichrome.  相似文献

Abstract— Since purine free base in aqueous solution exists in different ionic forms at different pHs and most of its photoreactions are likely to involve the triplet excited states of these different ionic forms, electron paramagnetic resonance studies (EPR) have been performed in order to determine intersystem crossing quantum yields and other characteristic parameters of the excited triplet state of these forms. Intersystem crossing yields decrease with a decrease in pH, being 0.62, 0.37 and 0.10 in 8 M NaOH, 8 M NaCIO4 and 6M H3P04 glasses, respectively. Differences in triplet decay lifetimes 3.4, 2.5 and 3.1 s, as well as in root mean-square zero-field splitting (ZFS) parameter, D*, (0,1304, 0.1512 and 0.1353 cm-1) are also observed for the anionic, neutral and cationic species of purine free base. The EPR signals of the triplet state of the neutral and anionic forms have been observed simultaneously in the pH range of7–10.  相似文献

The excited state processes of riboflavin, flavin mononucleotide and flavin adenine dinucleotide in argon-saturated aqueous solution were studied in the presence of lysozyme or bovine serum albumin (BSA). UV–Vis absorption and fluorescence spectroscopy indicates that the noncovalent flavin-protein binding is relatively weak. Quenching of the flavin triplet state by BSA, observed by time-resolved photolysis, is less efficient than by lysozyme. Light-induced oxidation of the two proteins and reduction of the three flavins were studied. The quantum yields of the former and latter in the absence of oxygen are up to 0.1 and 0.04, respectively. The effects of pH and sensitizer and protein concentrations were examined in greater detail. The proposed reaction is electron transfer from the tryptophan moiety to the flavin triplet rather than excited singlet state.  相似文献

Abstract— Quenching of the excited states of lumiflavin and 3-methyl-5-deazalumiflavin by methyl-and methoxy-substituted benzenes and naphthalenes in methanol was investigated. The observed difference in the reactivity of acid and neutral lumiflavin triplets is explained thermodynamically by applying the Michaelis cycle, as being due to the higher reduction potential of the acid triplet. In this connection the p K values of lumiflavin triplet (p K M= 6.5) and semiquinone (p K M= 11.3) have also been determined in methanol. The difference in the reactivity between the singlet and triplet states of lumiflavin is found to be greater as predicted by the difference in excitation energy. The reactivities of the excited states of flavin and 5-deazaflavin differ only slightly in contrast to the marked difference in the ground state reactivities of electron transfer reactions. This is explained in terms of the model of Rehm and Weller. The pH dependence of the electron transfer quenching of 5-deazaflavin triplet was investigated in water, yielding a triplet p K of 2.5. In contrast to the flavin, this triplet p K does not significantly differ from the p K of the 5-deazaflavin ground state. From this, different sites of protonation are deduced for the photoexcited triplet states of flavin and 5-deazaflavin.  相似文献

The flavin adenine dinucleotide (FAD) cofactor of Aspergillus niger glucose oxidase (GO) in its anionic (FAD*-) and neutral (FADH*) radical form was investigated by electron paramagnetic resonance (EPR) at high microwave frequencies (93.9 and 360 GHz) and correspondingly high magnetic fields and by pulsed electron-nuclear double resonance (ENDOR) spectroscopy at 9.7 GHz. Because of the high spectral resolution of the frozen-solution continuous-wave EPR spectrum recorded at 360 GHz, the anisotropy of the g-tensor of FAD*- could be fully resolved. By least-squares fittings of spectral simulations to experimental data, the principal values of g have been established with high precision: gX=2.00429(3), gY=2.00389(3), gZ=2.00216(3) (X, Y, and Z are the principal axes of g) yielding giso=2.00345(3). The gY-component of FAD*- from GO is moderately shifted upon deprotonation of FADH*, rendering the g-tensor of FAD*- slightly more axially symmetric as compared to that of FADH*. In contrast, significantly altered proton hyperfine couplings were observed by ENDOR upon transforming the neutral FADH* radical into the anionic FAD*- radical by pH titration of GO. That the g-principal values of both protonation forms remain largely identical demonstrates the robustness of g against local changes in the electron-spin density distribution of flavins. Thus, in flavins, the g-tensor reflects more global changes in the electronic structure and, therefore, appears to be ideally suited to identify chemically different flavin radicals.  相似文献

Cultured mammalian cells essential are model systems in basic biology research, production platforms of proteins for medical use, and testbeds in synthetic biology. Flavin cofactors, in particular flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD), are critical for cellular redox reactions and sense light in naturally occurring photoreceptors and optogenetic tools. Here, we quantified flavin contents of commonly used mammalian cell lines. We first compared three procedures for extraction of free and noncovalently protein‐bound flavins and verified extraction using fluorescence spectroscopy. For separation, two CE methods with different BGEs were established, and detection was performed by LED‐induced fluorescence with limit of detections (LODs 0.5–3.8 nM). We found that riboflavin (RF), FMN, and FAD contents varied significantly between cell lines. RF (3.1–14 amol/cell) and FAD (2.2–17.0 amol/cell) were the predominant flavins, while FMN (0.46–3.4 amol/cell) was found at markedly lower levels. Observed flavin contents agree with those previously extracted from mammalian tissues, yet reduced forms of RF were detected that were not described previously. Quantification of flavins in mammalian cell lines will allow a better understanding of cellular redox reactions and optogenetic tools.  相似文献

2,3,5,6-Tetrafluorophenylnitren-4-yl (5) was synthesized in argon at 4 K via the photolysis of 2,3,5,6-tetrafluoro-4-iodo-phenyl azide (6). Electron paramagnetic resonance (EPR) spectroscopy allows us to observe triradical 5 in its quartet state with the zero-field splitting (ZFS) parameters |D/hc| = 0.285 and |E/hc| = 0.043 cm-1. The quartet ground state of 5 is in accordance with our previous infrared (IR) spectroscopic investigation, in which the high-spin quartet state, but no low-spin doublet state, of 5 was observed in solid argon at 4 K [Wenk, H. H.; Sander, W. Angew. Chem., Int. Ed. 2002, 41, 2742-2745]. Because annealing of the matrix at temperatures of >10 K results in the rapid recombination of the highly reactive species 5 with I atoms produced during the photolysis of 6, the Curie-Weiss behavior could not be investigated. However, the absence of low-spin states in the IR investigations, as well as the results of ab initio and density functional theory (DFT) calculations, strongly suggest that 5 has a robust quartet ground state that is best-described as an unprecedented sigma,sigma,pi-triradical. The ZFS of 5 has been successfully reproduced by DFT calculations, which furthermore provide qualitative insight into the origin of the observed EPR parameters.  相似文献

Blue-light sensitive photoreceptory BLUF domains are flavoproteins, which regulate various, mostly stress-related processes in bacteria and eukaryotes. The photoreactivity of the flavin adenine dinucleotide (FAD) cofactor in three BLUF domains from Rhodobacter sphaeroides, Synechocystis sp. PCC 6803 and Escherichia coli have been studied at low temperature using time-resolved electron paramagnetic resonance. Photoinduced flavin triplet states and radical-pair species have been detected on a microsecond time scale. Differences in the electronic structures of the FAD cofactors as reflected by altered zero-field splitting parameters of the triplet states could be correlated with changes in the amino-acid composition of the various BLUF domains' cofactor binding pockets. For the generation of the light-induced, spin-correlated radical-pair species in the BLUF domain from Synechocystis sp. PCC 6803, a tyrosine residue near the flavin's isoalloxazine moiety plays a critical role.  相似文献

Continuous-wave time-resolved EPR (cw-TREPR) and pulsed electron nuclear double resonance (ENDOR) studies have been carried out to clarify the electronic structure of the lowest excited triplet (Tl) state of 5,12-naphthacenequinone (5,12-NpQ) as well as 1,4-anthraquinone (1,4-AQ) and 6,13-pentacenequinone (6,13-PeQ). The Tl energy level and the D value of the zero-field splitting (ZFS) parameters only slightly decreased with the increasing pi-conjugated system. The Tl states of these linear para-acenequinones were assigned to the pi pi* character. In triplet 5,12-naphthacenequinone, more than 80% of the unpaired electron spins are localized on the naphthalene aromatic sub-system.  相似文献

Abstract— The photoreduction of riboflavin, FMN, and lumiflavin in a series of hydroxylic solvents has been examined using flash photolysis. A comparison of the relative quantum yields for the photoxidation of several alcohols and esters by lumiflavin demonstrated that the hydroxyl hydrogen of the alcohol is abstracted approximately twice as readily as is the alpha hydrogen. The use of glycerol as both solvent and reductant provided direct evidence that the initial reaction proceeds by a one-electron reduction to form the flavin semi-quinone. In the case of riboflavin (and FMN) the kinetic results are consistent with an initial intramolecular hydrogen atom transfer, analogous to photobleaching in aqueous solution, followed by a reaction of the semi-quinone with the reductant which prevents degradation of the ribityl side-chain. Quenching by iodide indicates that all the reactions proceed via the flavin triplet state, as is the case in aqueous systems.  相似文献
