Electrospray ionization (ESI) Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR MS) combined with H/D exchange reactions was utilized to explore the existence of different b5+ and b4+ fragment ion conformers/isomers of hexapeptide WHWLQL in the gas phase. Distinct H/D exchange trends for protonated WHWLQL ([M + H]+) and its b5+ and b4+ fragment ions (with ND3) were observed. Isolated 12Call isotopomers of both b5+ and b4+ fragment ions yielded bimodal distributions of H/D exchanged product ions. The H/D exchange reaction kinetics also confirmed that b5+ and b4+ fragment ions exist as combination of slow-exchanging (“s”) and fast-exchanging (“f”) species. The calculated rate constant for the first labile hydrogen exchange of [M + H]+ (k[M + H] + = 3.80 ± 0.7 × 10−10 cm3 mol−1 s−1) was ∼30 and ∼5 times greater than those for the “s” and “f” species of b5+, respectively. Data from H/D exchange of isolated “s” species at longer ND3 reaction times confirmed the existence of different conformers or isomers for b5+ fragment ions. The sustained off-resonance irradiation collision-activated dissociation (SORI-CAD) of WHWLQL combined with the H/D exchange reactions indicate that “s” and “f” species of b5+ and b4+ fragment ions can be produced in the ICR cell as well as the ESI source. The significance of these observations for detailed understanding of protein sequencing and ion fragmentation pathways is discussed.  相似文献

Organophosphate esters (OPEs) are chemical compounds incorporated into materials as flame‐proof and/or plasticizing agents. In this work, 13 non‐halogenated and 5 halogenated OPEs were studied. Their mass spectra were interpreted and compared in terms of fragmentation patterns and dominant ions via various ionization techniques [electron ionization (EI) and chemical ionization (CI) under vacuum and corona discharge atmospheric pressure chemical ionization (APCI)] on gas chromatography coupled to mass spectrometry (GC‐MS). The novelty of this paper relies on the investigation of APCI technique for the analysis of OPEs via favored protonation mechanism, where the mass spectra were mostly dominated by the quasi‐molecular ion [M + H]+. The EI mass spectra were dominated by ions such as [H4PO4]+, [M–R]+, [M–Cl]+, and [M–Br]+, and for some non‐halogenated aryl OPEs, [M]+● was also observed. The CI mass spectra in positive mode were dominated by [M + H]+ and sometimes by [M–R]+, while in negative mode, [M–R] and more particularly [X] and [X2]‐● were mainly observed for the halogenated OPEs. Both EI and APCI techniques showed promising results for further development of instrumental method operating in selective reaction monitoring mode. Instrumental detection limits by using APCI mode were 2.5 to 25 times lower than using EI mode for the non‐brominated OPEs, while they were determined at 50‐100 times lower by the APCI mode than by the EI mode, for the two brominated OPEs. The method was applied to fish samples, and monitored transitions by using APCI mode showed higher specificity but lower stability compared with EI mode. As part of a mass spectrometric investigation of the binding properties of sulfonamide anion receptors, an atmospheric pressure chemical ionization mass spectrometric (APCI-MS) method involving direct infusion followed by thermal desorption was employed for identification of anionic supramolecular complexes in dichloromethane (CH2Cl2). Specifically, the dansylamide derivative of tris(2-aminoethyl)amine (tren) (1), the chiral 1,3-benzenesulfonamide derivatives of (1R,2S)-(+)-cis-1-amino-2-indanol (2), and (R)-(+)-bornylamine, (3), were shown to bind halide and nitrate ions in the presence of (n−Bu)4N+X (X = Cl, NO3, Br, I). Solutions of receptors and anions in CH2Cl2 were combined to form the anionic supramolecular complexes, which were subsequently introduced into the mass spectrometer via direct infusion followed by thermal desorption. The anionic supramolecular complexes [M+X], (M=13, X=Cl, NO3, Br, I) were observed in negative mode APCI-MS along with the deprotonated receptors [M−H]. Full ionization energy of the APCI corona pin (4.5 kV) was necessary for obtaining mass spectra with the best signal-to-noise ratios.  相似文献

Saturated and unsaturated, linear, branched, and cyclic hydrocarbons, as well as polyaromatic and heteroaromatic hydrocarbons, were successfully ionized by atmospheric pressure chemical ionization (APCI) using small hydrocarbons as reagents in a linear quadrupole ion trap (LQIT) mass spectrometer. Pentane was proved to be the best reagent among the hydrocarbon reagents studied. This ionization method generated different types of abundant ions (i.e., [M + H]+, M+•, [M – H]+ and [M – 2H]+ •), with little or no fragmentation. The radical cations can be differentiated from the even-electron ions by using dimethyl disulfide, thus facilitating molecular weight (MW) determination. While some steroids and lignin monomer model compounds, such as androsterone and 4-hydroxy-3-methoxybenzaldehyde, also formed abundant M+• and [M + H]+ ions, this was not true for all of them. Analysis of two known mixtures as well as a base oil sample demonstrated that each component of the known mixtures could be observed and that a correct MW distribution was obtained for the base oil. The feasibility of using this ionization method on the chromatographic time scale was demonstrated by using high-performance liquid chromatography (HPLC) with hexane as the mobile phase (and APCI reagent) to separate an artificial mixture prior to mass spectrometric analysis.  相似文献

The use of 5-aminosalicylic acid (5-ASA) as a new matrix for in-source decay (ISD) of peptides including mono- and di-phosphorylated peptides in matrix-assisted laser desorption/ionization (MALDI) mass spectrometry (MS) is described. The use of 5-ASA in MALDI-ISD has been evaluated from several standpoints: hydrogen-donating ability, the outstanding sharpness of molecular and fragment ion peaks, and the presence of interference peaks such as metastable peaks and multiply charged ions. The hydrogen-donating ability of several matrices such as α-cyano-4-hydroxycinnamic acid (CHCA), 2,5-dihydroxybenzoic acid (2,5-DHB), 1,5-diaminonaphthalene (1,5-DAN), sinapinic acid (SA), and 5-ASA was evaluated by using the peak abundance of a reduction product [M + 2H + H]+ to that of non-reduced protonated molecule [M + H]+ of the cyclic peptide vasopressin which contains a disulfide bond (S-S). The order of hydrogendonating ability was 1,5-DAN > 5-ASA > 2,5-DHB > SA = CHCA. The chemicals 1,5-DAN and 5-ASA in particular can be classified as reductive matrices. 5-ASA gave peaks with higher sharpness for protonated molecules and fragment ions than other matrices and did not give any interference peaks such as multiply-protonated ions and metastable ions in the ISD mass spectra of the peptides used. Particularly, 1,5-DAN and 5-ASA gave very little metastable peaks. This indicates that 1,5-DAN and 5-ASA are more “cool” than other matrices. The 1,5-DAN and 5-ASA can therefore be termed “reductive cool” matrix. Further, it was confirmed that ISD phenomena such as N-Cα bond cleavage and reduction of S-S bond is a single event in the ion source. The characteristic fragmentations, which form a− and (a + 2)-series ions, [M + H − 15]+, [M + H − 28]+, and [M + H − 44]+ ions in the MALDI-ISD are described.  相似文献

The dinuclear copper complex (α-cyano-4-hydroxycinnamic acid (CHCA) copper salt (CHCA)4Cu2), synthesized by reacting CHCA with copper oxide (CuO), yields increased abundances of [M + xCu − (x−1)H]+ (x = 1–6) ions when used as a matrix for matrix-assisted laser desorption ionization (355 nm Nd:YAG laser). The yield of [M + xCu − (x−1)H]+ (x = 1∼6) ion is much greater than that obtained by mixing peptides with copper salts or directly depositing peptides onto oxidized copper surfaces. The increased ion yields for [M + xCu − (x−1)H]+ facilitate studies of biologically important copper binding peptides. For example, using this matrix we have investigated site-specific copper binding of several peptides using fragmentation chemistry of [M + Cu]+ and [M + 2Cu − H]+ ions. The fragmentation studies reveal interesting insight on Cu binding preferences for basic amino acids. Most notable is the fact that the binding of a single Cu+ ion and two Cu+ ions are quite different, and these differences are explained in terms of intramolecular interactions of the peptide-Cu ionic complex.  相似文献

Upon CA, ESI generated [M + H]+ ions of chalcone (benzalacetophenone) and 3-phenyl-indanone both undergo losses of H2O, CO, and the elements of benzene. CA of the [M + H]+ ions of 2-methoxy and 2-hydroxychalcone, however, prompts instead a dominant loss of ketene. In addition, CA of the [M + H]+ ions of 2-methoxy-β-methylchalcone produces an analogous loss of methylketene instead. Furthermore, the [M + D]+ ion of 2-methoxychalcone upon CA eliminates only unlabeled ketene, and the resultant product, the [M + D − ketene]+ ion, yields only the benzyl-d 1 cation upon CA. We propose that the 2-methoxy and 2-hydroxy (ortho) substituents facilitate a Nazarov cyclization to the corresponding protonated 3-aryl-indanones by mediating a critical proton transfer. The resultant protonated indanones then undergo a second proton transport catalysis facilitated by the same ortho substituents producing intermediates that eliminate ketene to yield 2-methoxy- or 2-hydroxyphenyl-phenyl-methylcarbocations, respectively. The basicity of the ortho substituent is important; for example, replacement of the ortho function with a chloro substituent does not provide an efficient catalyst for the proton transports. The Nazarov cyclization must compete with an alternate cyclization, driven by the protonated carbonyl group of the chalcone that results in losses of H2O and CO. The assisted proton transfer mediated by the ortho substituent shifts the competition in favor of the Nazarov cyclization. The proposed mechanisms for cyclization and fragmentation are supported by high-mass resolving power data, tandem mass spectra, deuterium labeling, and molecular orbital calculations.  相似文献

The structures of the major adduct ions formed in ammonia chemical ionization of thirteen aliphatic and aromatic ketones have been studied by mass analysed ion kinetic energy spectrometry. The [M+NH3+H]+ ion is shown to have a protonated carbinolamine structure, [M+2NH3+H]+ to be a protonated carbinolamine with hydrogen-bonded ammonia and [2M+NH3+H]+ to be, at least in part, a protonated carbinolamine with hydrogen-bonded ketone. These structures may imply a nucleophilic addition of ammonia at the carbonyl of the ketone-ammonium ion complex. An unusual hydroxy migration is seen in the internal rearrangement of the [2M+NH3+H]+ ion leading to the formation of a protonated imine.  相似文献

Matrix assisted laser desorption/ionization (MALDI) time-of-flight (TOF) mass spectrometry (MS) and theoretical calculations [density functional theory (DFT)] were utilized to investigate the influence of cysteine side chain on Cu+ binding to peptides and how Cu+ ions competitively interact with cysteine (−SH/SO3H) versus arginine. Results from theoretical and experimental (fragmentation reactions) studies on [M+Cu]+ and [M+2Cu−H]+ ions suggest that cysteine side chains (−SH) and cysteic acid (−SO3H) are important Cu+ ligands. For example, we show that Cu+ ions are competitively coordinated to the −SH or SO3H groups; however, we also present evidence that the proton of the SH/SO3H group is mobile and can be transferred to the arginine guanidine group. For [M+2Cu−H]+ ions, deprotonation of the −SH/SO3H group is energetically more favorable than that of the carboxyl group, and the resulting thiolate/sulfonate group plays an important role in the coordination structure of [M+2Cu−H]+ ions, as well as the fragmentation patterns.  相似文献

The detection of anabolic androgenic steroids (AAS) is one of the most important topics in doping control analysis. Gas chromatography coupled to (tandem) mass spectrometry (GC–MS(/MS)) with electron ionization and liquid chromatography coupled to tandem mass spectrometry have been traditionally applied for this purpose. However, both approaches still have important limitations, and, therefore, detection of all AAS is currently afforded by the combination of these strategies. Alternative ionization techniques can minimize these drawbacks and help in the implementation of a single method for the detection of AAS. In the present work, a new atmospheric pressure chemical ionization (APCI) source commercialized for gas chromatography coupled to a quadrupole time‐of‐flight analyzer has been tested to evaluate the ionization of 60 model AAS. Underivatized and trimethylsylil (TMS)‐derivatized compounds have been investigated. The use of GC–APCI–MS allowed for the ionization of all AAS assayed irrespective of their structure. The presence of water in the source as modifier promoted the formation of protonated molecules ([M+H]+), becoming the base peak of the spectrum for the majority of studied compounds. Under these conditions, [M+H]+, [M+H‐H2O]+ and [M+H‐2·H2O]+ for underivatized AAS and [M+H]+, [M+H‐TMSOH]+ and [M+H‐2·TMSOH]+ for TMS‐derivatized AAS were observed as main ions in the spectra. The formed ions preserve the intact steroid skeleton, and, therefore, they might be used as specific precursors in MS/MS‐based methods. Additionally, a relationship between the relative abundance of these ions and the AAS structure has been established. Hydralazine has been widely employed in the development of drugs, derivatization reagents, and ligands. In the present work, we reported a new type of dehydrogenated ion [M ? H]+ that was produced from the hydralazine derivative of hexanal in electrospray ionization mass spectrometry (ESI‐MS). The formation of [M ? H]+ ions in the ESI‐MS was found to be independent on the mobile phase composition of the liquid chromatography and ESI source parameters. A series of hydralazine derivatives of aldehyde were investigated to confirm this phenomenon. The results showed that hydralazine derivatives of aldehydes that contained an sp3 hybridization carbon with a hydrogen at the α‐position of aldehydes could form the unexpected [M ? H]+ ions, whereas hydralazine derivative of acetone could only generate [M + H]+ ion in the ESI‐MS. We proposed the possible formation mechanism of [M ? H]+ ion for the hydralazine derivatives of aldehydes: the [M ? H]+ ion was possibly formed by the loss a hydrogen molecule (H2) from the protonated ion [M + H]+. The results obtained from density functional theory (DFT) calculations supported this proposed formation mechanism of [M ? H]+ ion.  相似文献

A new method is described for simultaneous determination of 3-methylindole (3MI) and indole in porcine adipose tissue. Sample preparation included liquid–liquid extraction with n-hexane and 75% aqueous acetonitrile. The acetonitrile extracts were analysed by liquid chromatography–mass spectrometry (LC–MS) using atmospheric pressure chemical ionization (APCI) with selective ion monitoring of protonated ions [M + H]+. This method showed excellent linearity over the concentration range tested (from 2 to 500 ng mL−1 for 3MI and from 1 to 500 ng mL−1 for indole) and good accuracy (recovery of 92 ± 10% for 3MI and 91 ± 10% for indole). This new LC–MS method was compared with traditional colorimetric method for 3MI equivalent. Additionally, the correlation between 3MI concentrations in adipose tissue and plasma was studied. The described LC–MS method can be used to quantify 3MI and indole in porcine adipose tissue in various endocrinological or meat science studies.  相似文献

A discharge adaptor, composed of a metal casing and platinum (Pt) wire needle, was directly attached to an electrospray ionization (ESI) probe tip, to transform the ionization into atmospheric pressure chemical ionization (APCI). Six generic drugs were analyzed with the developed discharge adaptor (DA) and two commercial interfaces. The DA interface produced more intense radical anions, [M].–, and less sodium adduct ions, [M + Na]+, than the ESI interface, whereas almost the same molecular ions were detected as the APCI interface. The effects of solvent and desolvation gas flow in the DA interface were similar to those in the ESI interface, but differed from those in the APCI interface. Better sensitivity of the tested drugs was obtained relative to the commercial APCI interface. For human plasma samples, the DA interface also demonstrated good tolerance to plasma matrices, linearity from 5 or 20 to 500 ng/mL (r2 > 0.99) and ruggedness. The ion-molecule reactions of dimethyl ether ions CH3OCH3 + and (CH3OCH3)H+, and four- to seven-membered ring lactams with methyl substituents in various positions were characterized by using a quadrupole ion trap mass spectrometer and a triple-quadrupole mass spectrometer. In both instruments, the lactams were protonated by dimethyl ether ions and formed various combinations of [M + 13] +, [M + 15] +, and [M + 45] + adduct ions, as well as unusual [M + 3] + and [M + 16] + adduct ions. An additional [M + 47] + adduct ion was formed in the conventional chemical ionization source of the triple-quadrupole mass spectrometer. The product ions were isolated and collisionally activated in the quadrupole ion trap to understand formation pathways, structures, and characteristic dissociation pathways. Sequential activation experiments were performed to elucidate fragment ion structures and stepwise dissociation sequences. Protonated lactams dissociate by loss of water, ammonia, or methylamine; ammonia and carbon monoxide; and water and ammonia or methylamine. The [M + 16] + products, which are identified as protonated lactone structures, are only formed by those lactams that do not have an N-methyl substituent. The ion-molecule reactions of dimethyl ether ions with lactams were compared with those of analogous amides and lactones.  相似文献

Low-energy CAD product-ion spectra of various molecular species of phosphatidylserine (PS) in the forms of [M−H] and [M−2H+Alk] in the negative-ion mode, as well as in the forms of [M+H]+, [M+Alk]+, [M−H+2Alk]+, and [M−2H+3Alk]+ (where Alk=Li, Na) in the positive-ion mode contain rich fragment ions that are applicable for structural determination. Following CAD, the [M−H] ion of PS undergoes dissociation to eliminate the serine moiety (loss of C3H5NO2) to give a [M−H−87] ion, which equals to the [M−H] ion of a phoshatidic acid (PA) and give rise to a MS3-spectrum that is identical to the MS2-spectrum of PA. The major fragmentation process for the [M−2H+Alk] ion of PS arises from primary loss of 87 to give rise to a [M−2H+Alk−87] ion, followed by loss of fatty acid substituents as acids (RxCO2H, x=1,2) or as alkali salts (e. g., RxCO2Li, x=1,2). These fragmentations result in a greater abundance of [M−2H+Alk−87−R2CO2H] than [M−2H+Alk−87−R1CO2H] and a greater abundance of [M−2H+Alk−87−R2CO2Li] than [M−2H+Alk−87−R1CO2Li]; while further dissociation of the [M−2H+Alk−87−R2(or 1)CO2Li] ions gives a preferential formation of the carboxylate anion at sn-1 (R1CO2) over that at sn-2 (R2CO2). Other major fragmentation process arises from differential loss of the fatty acid substituents as ketenes (loss of Rx′CH=CO, x=1,2). This results in a more prominent [M−2H+Alk−R2′CH=CO] ion than [M−2H+Alk−R1′CH=CO] ion. Ions informative for structural characterization of PS are of low abundance in the MS2-spectra of both the [M+H]+ and the [M+Alk]+ ions, but are abundant in the MS3-spectra. The MS2-spectrum of the [M+Alk]+ ion contains a unique ion corresponding to internal loss of a phosphate group probably via the fragmentation processes involving rearrangement steps. The [M−H+2Alk]+ ion of PS yields a major [M−H+2Alk−87]+ ion, which is equivalent to an alkali adduct ion of a monoalkali salt of PA and gives rise to a greater abundance of [M−H+2Alk−87−R1CO2H]+ than [M−H+2Alk−87−R2CO2H]+. Similarly, the [M−2H+3Alk]+ ion of PS also yields a prominent [M−2H+3Alk−87]+ ion, which undergoes consecutive dissociation processes that involve differential losses of the two fatty acyl substituents. Because all of the above tandem mass spectra contain several sets of ion pairs involving differential losses of the fatty acid substituents as ketenes or as free fatty acids, the identities of the fatty acyl substituents and their positions on the glycerol backbone can be easily assigned by the drastic differences in the abundances of the ions in each pair.  相似文献

The mass spectrometric (MS) and tandem mass spectrometric (MS/MS) behavior of six nitrocatechol-type glucuronides using atmospheric pressure chemical ionization (APCI) and electrospray ionization (ESI) was systematically studied, and the effect of operation parameters on the fragmentations are presented. The positive ion APCI- and ESI-MS spectra showed an intense protonated molecule and the respective negative ion spectra a deprotonated molecule with minimal fragmentation. The main fragment ions in the MS/MS spectra of the protonated and deprotonated molecules were [M + H - Glu]+ and [M - H - Glu]-, respectively, formed by the loss of the glucuronide moiety. The measured limits of detection indicated that ESI is a significantly more efficient ionization method than APCI in the negative and positive ion modes for the compounds studied. MS/MS was found to be less sensitive, but more reliable and simple than MS due to the absence of chemical noise.  相似文献

The gas-phase structures of deprotonated, protonated, and sodium-cationized complexes of diethyl phosphate (DEP) including [DEP − H], [DEP + H]+, [DEP + Na]+, and [DEP − H + 2Na]+ are examined via infrared multiple photon dissociation (IRMPD) action spectroscopy using tunable IR radiation generated by a free electron laser, a Fourier transform ion cyclotron resonance mass spectrometer (FT-ICR MS) with an electrospray ionization (ESI) source, and theoretical electronic structure calculations. Measured IRMPD spectra are compared to linear IR spectra calculated at the B3LYP/6-31G(d,p) level of theory to identify the structures accessed in the experimental studies. For comparison, theoretical studies of neutral complexes are also performed. These experiments and calculations suggest that specific geometric changes occur upon the binding of protons and/or sodium cations, including changes correlating to nucleic acid backbone geometry, specifically P–O bond lengths and ∠OPO bond angles. Information from these observations may be used to gain insight into the structures of more complex systems, such as nucleotides and solvated nucleic acids.  相似文献

A combination of electrospray ionization (ESI), multistage, and high-resolution mass spectrometry experiments are used to examine the gas-phase fragmentation reactions of radical cations of cysteine containing di- and tripeptides. Two different chemical methods were used to form initial populations of radical cations in which the radical sites were located at different positions: (1) sulfur-centered cysteinyl radicals via bond homolysis of protonated S-nitrosocysteine containing peptides; and (2) α-carbon backbone-centered radicals via Siu’s sequence of reactions (J. Am. Chem. Soc. 2008, 130, 7862). Comparison of the fragmentation reactions of these regiospecifically generated radicals suggests that hydrogen atom transfer (HAT) between the α C-H of adjacent residues and the cysteinyl radical can occur. In addition, using accurate mass measurements, deuterium labeling, and comparison with an authentic sample, a novel loss of part of the N-terminal cysteine residue was shown to give rise to the protonated, truncated N-formyl peptide (an even-electron xn ion). DFT calculations were performed on the radical cation [GCG].+ to examine: the relative stabilities of isomers with different radical and protonation sites; the barriers associated with radical migration between four possible radical sites, [G.CG]+, [GC.G]+, [GCG.]+, and [GC(S.)G]+; and for dissociation from these sites to yield b2-type ions.  相似文献
