The chiral synthesis of maconelliol, [2,2-dimethyl-3-(1-methylethylidene)cyclobutyl] methanol, the alcohol moiety of the major sex pheromone component isolated from the pink hibiscus mealybug, Maconellicoccus hirsutus, is described. The compound was synthesized in six steps from alpha-pinene and the key step is dehydration of 5 to 7 through the intermediate 6. The absolute configuration of the naturally occurring maconelliol was determined as R.  相似文献

2,3-Dimethyl-2,3-diphenylbutanediol-1,4-ditosylate (7) was synthesized starting from 2-phenylpropionic acid (1). The acid chloride was brominated and transformed into methyl 2-phenyl-2-bromo-propionate (4) which was coupled with a zinc/copper couple to dimethyl 2,3-dimethyl-2,3-diphenylsuccinate (5). Reduction with lithium aluminum hydride to 2,3-dimethyl-2,3-diphenylbutanediol-1,4 (6) was followed by tosylation. The tosylate 7 a mixture of the meso and racemic compounds, could be separated into the pure isomers,a m. p. 170 °C andb m. p. 121 °C. The mixture of each individual pure compound, when treated with tetraalkyl-ammonium bromide, did not give the expected 2,3-dimethyl-2,3-diphenyl-1,4-dibromobutane (9) but rather 1,4-diphenyl-2,3-dimethylbutadiene-1,3 (8). The identity of the compound was established by independent unequivocal synthesis, the comparison of spectral characteristics, and mixed melting point.  相似文献

4,4-Dimethyl-6-methoxy-4-sila-1-tetralone (2) was prepared by a modified literature procedure and converted to 3-methoxy-6,6-dimethyl-6-silaestra-1,3,5(10),8,14-pentaen-17β-yl acetate (5c). Catalytic hydrogenation of 5c gave 3-methoxy-6,6-dimethyl-6-silaestra-1,3,5(10),8-tetraen-17β-yl acetate (6b), and its 14-iso- and Δ1,3,5(10),8(14) isomers, the proportions varying with the catalyst and solvent. Reduction of 6b with lithium-liquid ammonia, and O-demethylation, gave 6,6-dimethyl-6-silaestradiol (8b). Reduction of the 3-methyl ether of 8b with lithium-liquid ammonia-t-butanol and hydrolysis afforded 3-keto-6,6-dimethyl-6-silaestr-1(10)-en-17β-ol (15), which was catalytically reduced to its 1,10α-dihydro derivative 17. The 5,6 SiC bond of 8b, 15 and their derivatives was cleaved by boron tribromide, aq. ethanolic hydrogen fluoride, and other reagents, providing a series of 5,6-seco-6,6-dimethyl-6-silasteroids. X-ray crystallographic analysis of 17 and the 17α-ethynyl derivative of 15 confirmed the stereochemical assignments. None of the compounds which were subjected to uterotropic, anti-uterotropic, or post-coital assays, showed significant activity. A partially completed synthesis of 6-silaestradiol (21a) is described.  相似文献

A series of 1-(2,4-dichlorophenoxyacetoxy)alkylphosphonic acid dimethyl esters 5 and its corresponding phosphonate monosalts 6 were synthesized as potential herbicide. The phosphonate monosalts can be prepared from 1-(2,4-dichlorophenoxyacetoxy)alkylphosphonic acid dimethyl esters 5, which were synthesized by the condensation of O,O-dimethyl-1-hydroxyalkylphosphonates with dichlorophenoxyacetic chloride. This method provides a simple and efficient procedure for the synthesis of phosphonate derivatives containing sensitive groups to acid, base or water such as carboxylate ester bond; and the herbicidal activity of title compounds was evaluated in a set of experiments in greenhouse. Most of the compounds exhibited notable herbicidal activity.  相似文献

The cysteine protease, falcipain-2 is an important drug target in human malaria parasite Plasmodium falciparum. A new series of 2-(4-(substituted benzoyl)-1,4-diazepan-1-yl)-N-phenylacetamide derivatives 5(at) were designed as per pharmacophoric requirements of falcipain-2 inhibitors using ligand-based approach. The target compounds were synthesized from the key intermediate, 2-(1,4-Diazepan-1-yl)-N-phenylacetamide, by coupling it with appropriate carboxylic acids using carbodiimide chemistry. Structural features of target compounds were characterized by spectral data (1H NMR, and mass) and elemental analyses. The purity of the final compounds was confirmed by HPLC. The compounds were tested for their in vitro falcipain-2 inhibitor activity on recombinant falcipain-2 enzyme. Five compounds 5b, 5g, 5h, 5j, 5k showed good inhibitory activity (>60%), against falcipain-2 at 10 μM concentration, and fifteen compounds showed weak to moderate inhibitor activity. Compound 5g, the most potent compound from this series showed 72% inhibition at 10 μM concentrations.  相似文献

Synthesis of the heteroaromatic systems 6-thia-7H-benzo [b] thiophene (3) and 1,3-dimethyl-5-thia-4H-benzo [c] thiopbene (5) have been achieved by reduction and dehydration of 1,6-dithiaindan-4-one (4) and 1,3-dimethyl-2,5-dithiaindan-7-one (8) respectively. A similar attempt to synthesise 5-thia-4H-benzo [b] thiophene (4) by dehydration of 7-hydroxy-1,5-dithiaindane (16) resulted in the formation of 7,7′-bis (1,5-dithiaindanyl) ether.  相似文献

Several new 8-phosphabicyclo[3.2.1]oct-6-enes and -6-en-3-yl acetates were prepared by the alkyl (or aryl)dihalophosphane addition to cyclohepta -1,3-diene and 1-acetoxy cyclohepta-3,5-diene. The P-configuration in two P-epimer pairs (3, 4 and 5, 6) and three other compounds which were each obtained as a single isomer (1, 2 and 7) was determined by investigation of the NMR spectra, i.e. by complexation of the compounds with Eu(dpm)3 as well as from the various phosphorus-hydrogen coupling constants. Compounds possessing the same P-substituent were correlated among themselves and in the case of the PPh bearing compounds (1 and 7) further correlation to a known compound (8a) was established. The chemical behaviour of the C6C7 double bond is discussed and compared with the corresponding behaviour in trop-6-enes. Great steric hindrance was found for this bond, for which the following sequence is suggested:

3-Methoxy-7α-phenyl-1,5-dimethyl-3-borabicyclo[3.3.1]nonane 5 in solution at room temperature exists in the double chair conformation, as shown by NMR studies. Increasing the temperature leads to an increase in the population of the chair–boat conformation. At decreased temperature hindered rotation around the B---O bond is observed for 5. Dissolving 5 in deuteropyridine leads to the reversible formation of complex 6, which exists in the chair–boat conformation. The chair–boat conformation is also the most stable one for chelate compound 7 with a tetracoordinated boron atom.  相似文献

The cycloadditions of arylazides, benzonitrile oxides and diphenylnitrile imine to norbotnadiene derivatives 1a–c showed varying degree of site- and stereo-selectivity. With dipolarophile 1a arylazides and benzonitrile oxides attack, preferentially, the electron-poor tetrasubstituted double bond, while in the case of 1b and 1c is the substituted double bond which enters the cycloaddition. By contrast, 2-diazopropane and C-pnenyl-N-methyl-nitrone react with the sole tetrasubstituted double bond of 1a–c in stereo- and site-specific cycloadditions. The quantitative evaluation of the two possible reaction paths was performed by glc analysis. The compounds detected were those arising from Diels-Alder cycloreversions of the thermally labile intermediate adducts 2 and 3 (Scheme 1). The results were rationalized on the basis of a qualitative perturbation treatment which considers frontier orbital interactions only.  相似文献

In interactions of β-dicarbonyl compounds (2) with azodicarboxamidine (1), the primary reaction step consists of the addition of the active methylene group in 2 to the NN double bond in 1. A new heterocyclic system, 1,2,9-triaza-8-oxabicyclo-[5.3.0]-3,5,9-decatriene 4, has been synthesized by regiospecific 1,3-dipolar cycloaddition of nitrile oxides to the imine double bond of 1,2-diazepines. The quinazolinocarboline alkaloids including rutaecarpine (1a), euxylophoricine A (1b), and euxylophoricine C (1c) have been synthesized efficiently from the ring opened β-carboline derivative as key intermediate by a one-pot reductive-cyclization reaction. Rearrangement of β-ionone epoxide with aqueous formic acid gave 1,2-dihydro-1,1,6-trimethylnaphthalen (2), 1-(1,2,2-trimethylcyclopent-1-yl)-2-penten-1,4-dione (3), 4-(5,5-dimethyl-2-acetyl-1-cyclopenten-1-yl)-2-butanone (5), 4-(1,3,3-trimethyl-2-cyclohexanon-1-yl)-3-buten-2-one (6), 4-(2,3,6-trimethylphenyl)-2-butanone (7) and 6,6-dimethyl-2,5-10-undecatrione (8). NMR assignments are made for each compound while a computer assisted analysis of the A2B2 portion of 7 has been completed. A novel series of conjugates of benzosuberone and 1,2,3-triazole i.e. 3-(4-phenyl-1H-1,2,3-triazol-1-yl)propyl-9-chloro-2,3-dimethyl-6,7-dihydro-5H-benzo[7]annulene-8-carboxylic acids (8a-j) were synthesized in good to excellent yields catalysed by CuSO4 under milder reaction conditions and evaluated for their anti-proliferative activity. The structural elucidation of the prepared compounds was carried out using IR, 1H NMR, 13C NMR and Mass spectral analysis. The newly synthesized derivatives (8a-j) were evaluated for their anti-proliferative activity against four human cell lines and the novel derivatives showed moderate to excellent activity. The obtained results suggest that these compounds can be considered as new hits for anti-proliferative drug development programme and further SAR studies can help obtain better anticancer agents.  相似文献

A simple, highly useful synthesis of 1-(2-(4,5-dimethyl-2-phenyl-1H-imidazol-1-yl)ethyl)piperazine derivatives is achieved by a four component cyclo condensation of diacetyl, aromatic aldehyde, 2-(piperazin-1-yl)ethanamine and ammonium acetate using SO42−/Y2O3 as a catalyst in ethanol. The synthesized compounds were characterized by IR, 1H and 13C NMR and mass spectral studies. All the synthesized compounds were screened for their in vitro antimicrobial studies. Among the newly synthesized compounds 5d, 5e, 5h and 5m showed excellent antibacterial and antifungal activities when compared to the standard drugs.  相似文献

When acted upon by (CH3)2SO4 compounds 2, 3 were converted into corresponding fluorine-containing 1-methyl-1,2-dihydro-1,3-diazafluorenes 6, 7. 4a-Chloro-5,6,7,8,9,9-hexafluoro-2,2-dimethyl-4-trifluoromethyl-2,4a-dihydro-1,3-diazafluorene (8) has been synthesized by the interaction of compound 2 with SOCl2. Solution of compound 2 as well as 8 in CF3SO3H-CD2Cl2 generated 5,6,7,8,9,9-hexafluoro-2,2-dimethyl-4-trifluoromethyl-1,2,3,4-tetrahydro-1,3-diazafluorene-4-yl cation (2c). The structures of compounds 2, 3, 6-8 have been determined by single crystal X-ray diffraction.  相似文献

Synthetic approaches to anthracycline antibiotics were studied through the use of Claisen rearrangements on 1-methallyloxy-5-methoxyantraquinone (9) which required reducing conditions to proceed through a hydroquinone intermediate in situ. 1-(2′-Methylene-4′-pentenoxy)-5-methoxyanthraquinone (13) underwent a similar reductive rearrangement but also produced a spiro compound 16 as a result of an ene reaction between the phenol and side chain double bond. 1-Hydroxy-2-methally-5-methoxyanthraquinone (11) could not be oxidized to quinizarin 17. 1-Hydroxy-2-methally-5,9,10-trimethoxyanthracene (21) was oxidatively coupled to the dimer at C-2. 1H-Siladigermirene R4SiGe2 (2a) and 1H-trigermirene R4Ge3 (2b) (R = SiMetBu2) with a GeGe double bond were synthesized by the reaction of tetrachlorodigermane RGeCl2–GeCl2R with dilithiosilane R2SiLi2 and dilithiogermane R2GeLi2, respectively. The skeletal GeGe double bond of 2a is trans-bent (51.0(2)°) with a bond distance of 2.2429(6) Å. The reaction of both 2a and 2b with CH2Cl2 resulted in the formation of unusual four-membered ring compounds 5a and 5b as a result of a ring expansion reaction. 1H-Trisilirene 7a and 3H-disilagermirene 7b with an SiSi double bond also smoothly reacted with CH2Cl2 to yield the four-membered ring systems 8a and 8b, respectively.  相似文献
