ABSTRACT

A new method for mutagenicity testing of carcinogens is proposed in this paper. Mutagenic activity is examined through monitoring of the growth situation of Salmonella typhimurium TA98 strain with a series piezoelectric quartz crystal (SPQC) sensor. This method needs no immobilization and preincubation of microorganism, and is simple and rapid. It can be applied to detect mutagen directly. When it was used to study the mutagenicity of dimethyl sulfate, the test time was 4 h at 37°C. There was a good linear relationship between frequency shift and the dose of mutagen in the range 2.5-20 μg/mL and the regression equation was ∠F = 53.82 + 3.81C. Some experimental conditions are also discussed in detail.  相似文献

用溶胶-凝胶与接枝共聚物组成的有机-无机杂化材料固定皮状丝孢酵母制备BOD生物传感器,在研制的在线BOD监测仪上考察其性能。结果表明,该传感器线性高达85.0mg/L BOD,稳定性良好、使用寿命长,可连续使用90d以上,保存14个月后仍可保持70%以上的活性。本方法用于实际样品分析,得到了与传统的5d法较一致的结果,可用于污水的在线监测。  相似文献

ABSTRACT

A rapid and simple method for monitoring the growth of Thiobacillus ferrooxidans is put forward by using a series piezoelectric quartz crystal (SPQC). The SPQC was applied to continuously determine the variation of frequency shift during the culture of Thiobacillus ferrooxidans, with a conductive electrode as the probe. The frequency shift as a function of time agrees with a typical 'S - shape' model, a piezoelectric sensor responding well to the growth of Thiobacillus ferrooxidans. The fundamentals for determination of the bacterial growth activity using the SPQC is also described; the frequency shift is only dependent on the electrical conductivity of the solution and the dielectric constant.  相似文献

Abstract

Sulfur dioxide was detected and determined in air by a piezoelectric quartz crystal sensor coated with 4-aminoantipyrine 1-hydroxyetil-2-heptadecenyl imidazol (amine 220) solution (1:1 v/v in chloroform). The analytical response curve is linear over the concentration range from 0.70 to 5.0 ppm of SO2. Good linearities (r = 0.9990, 0.9995 and 0.9968) and sensitivities (18.0, 33.4 and 50.7 Hz/ppm) were found, respectively for exposure times of 30, 60 and 90 seconds. The sensor can be used for more than six months without loss in sensitivity and presented good reversibility and reproducibility. Among some possible interferents tested, only nitrogen dioxide and moisture caused major frequency changes.  相似文献

本文通过对几种微生物的筛选,找到活性较高的河流污泥微生物制成生化需氧量传感器,重点进行了培养基成分及测试条件等方面的优化,实验结果表明,电极对BOD标准物质线性响应范围为10-60mg/L响应时间为15min,连续稳定地工作寿命在30天以上,用于实际污水测定时,与BOD5有良好的相关性。  相似文献

采用混合菌BODseed制备微生物传感器快速监测生化需氧量(BOD),研究了矫正溶液选择、样品自降解及恒温装置对测量准确度的影响。 结果表明,采用葡萄糖 谷氨酸矫正溶液测得污水处理厂污水结果较BOD5方法偏低50%以上,而以污水本身制作矫正曲线取得了与BOD5一致的测试结果;样品在保存6 h后有机物自降解会使测量结果偏低40%;快速BOD在线监测时不仅需要对微生物传感器恒温,同时也需要对样品及缓冲溶液恒温,这有利于样品快速接近缓冲溶液温度,减少空气饱和时间,从而提高监测效率,提升测试准确度。  相似文献

研究了以间苯二酚杯[4]芳烃为敏感涂层的石英压电晶体(PQC)传感器,在中 性磷酸盐缓冲液体系中对神经传递质多巴胺和抗坏血酸的响应,发现间苯二酚杯 [4]芳烃对多能上能下胺有很好的响应选择性,这归因于间苯二酚杯[4]芳烃与多巴 胺分子结构相匹配,形成主客体超分子体系。以间苯二酚杯[4]芳烃为涂层的POC传 感器对液相中多巴胺具有响应快、重现性好、灵敏度高的特点,线性响应范围为3. 5-500 μg/g。  相似文献

生化需氧量(BOD)是一种表征水体有机污染程度的综合指标,广泛应用于水体监测和废水处理厂的运行控制中。由于BOD的标准测定方法需时5d,不能及时地反映水质状况和反馈处理信息,因此快速测定BOD的方法和仪器化研究近年来得到广泛的重视。其中利用生物传感器测定BOD是一种有效地快速测定废水中可生化降解有机物的方法。文章介绍了生物传感器的工作原理及其生物敏感材料,讨论了BOD快速测定值(BODst)与标准BOD5值的一致性问题。对目前市场上常见的几种BOD测定仪的性能进行了比较。  相似文献

Abstract

The frequency shift of a piezoelectric crystal sensor showed a linear dependence on the concentration for drugs which can be hydrolyzed in base aqueous solutions, at a given reacting time after the addition of the base, when the base concentration is basically constant during the entire measurement period. The sensitivity of the analysis method was improved, if crystal responses at different reacting times were recorded and utilized to estimate component concentration, when used for determining atropine sulfate in a pharmaceutical compound. The analytical results indicated that the selective and sensitive analytical procedure could be easily applied in true low concentration pharmaceutical analysis.  相似文献

Glomalin is a soil protein abundantly occurring in the soil. In the current time, knowledge about glomalin is limited and there are also missing simple test for the determination of glomalin in the environment. This work is devoted to construction of a biosensor which is expected to be a simple device for the determination of glomalin in extracts from soil samples. The biosensor was constructed using an antibody against glomalin and piezoelectric quartz crystal microbalance (QCM) sensor platform allowing label free assay. Electrodes of QCM were activated using cysteamine and glutaraldehyde and finally, an antibody against glomalin was immobilized. Glomalin was acquired from various soil samples by extraction in an autoclave and its content was determined by a standard spectrophotometric test. Time necessary to bind sufficient amount of glomalin was discovered for the biosensor and four hours incubation interval corresponded with maximal efficacy. Limit of detection for the biosensor based assay was found to be equal to 3.40 μg/g which is enough to cover all the tested soil samples containing glomalin in a concentration from 291 μg/g to 3.47 mg/g. The assay also fully correlated with the standard tests. In a conclusion, the piezoelectric biosensor seems to be a suitable platform for the determination of glomalin in samples of environment origin. The method represents an improvement of the current analytical platforms that are based on measurement of total protein content in soil extract.  相似文献

A glucose-sensing system was developed in which an electrodeless piezoelectric quartz crystal was used to measure mass changes on the surface of a quartz plate during the reduction of Ag(NH3)+2 by glucose. A satisfactory correlation was obtained between the frequency shift and the glucose concentration in the range 1.0–25 mM, and the total reducing sugar in urine was determined. Treatment with 8 M nitric acid after each measurement was effective for cleaning the quartz surface, and the electrodeless piezoelectric quartz crystal sensor possessed excellent reproducibility and reusability during repeated use over 400 times. For the determination of total reducing sugar in urine (or blood), the present method can avoid the interferences occurring in a colorimetric method such as the color and turbidity of clinical specimens.  相似文献

Albumin is a vital plasma protein in the control of osmotic pressure of human blood and is used as a screening marker for renal disease and Type I and Type II diabetic mellitus. A mass-sensitive immunosensor based on quartz crystal microbalance was used to determine microalbumin in urine on the antihuman albumin layer which was bonded to the protein A layer on the surface of a silver electrode. The analytical conditions were optimized for protein A and antihuman albumin on the electrode, and a calibration curve and the correlation between the microalbumin measured by the immunosensor and by a conventional method were evaluated. The results demonstrated that the optimal conditions for protein A coating on the silver electrode were 1?mg/ml and 2?h incubation at room temperature. The optimal conditions for antihuman albumin on the protein A-modified surface were 0.5?mg/ml at 2?h. The dose-response curve was linear from 0.0001 to 0.1?mg/ml. The comparison for the determination of microalbumin in urine by this method and a conventional approach showed good correlation with R2?=?0.999 (P?<?0.01). The receiver operating characteristic curve analysis showed 98.7% sensitivity, 100% specificity, and 1.00 for the area under the curve for the 0.02?mg/ml cutoff value. This system was label free and easy to use with high sensitivity and specificity. Therefore, the system may be used in place of commercial methods in the clinical laboratory.  相似文献

利用氧荧光猝灭原理研制了台式生化需氧量(BOD)传感检测仪器。仪器的工作系统由检测系统和软件系统两部分组成。该检测仪操作简便,检测快速,可进行海水BOD含量的现场检测。仪器的性能考察结果表明,在0.5~50mg/L浓度范围内BOD响应的线性相关系数为0.9978。实际海水样品的检测结果显示,仪器对海水样品的平行测定结果相对标准偏差小于±5%,并且与标准五日生化需氧量(BOD5)方法所获得的数值的相关性为0.99。仪器具有较长的使用寿命和良好的稳定性。  相似文献

A new method was proposed for monitoring antibiotics and other pollutants based on the assessment of the growth of Staphylococcus aureus (S. aureus) with a piezoelectric impedance analysis technique. During the growth process of S. aureus, the motional resistance variation ( jR1) increases and the frequency shift ( jf) of the piezoelectric quartz crystal (PQC) decreases correspondingly. The two responses can conveniently be used in process analysis. Under the given experimental conditions, a linear relationship between the resistance detection time (RDT) and logarithm value of the concentration of penicillin was obtained in the range of 0.01-10 µg/ml with a detection limit of 4 2 10-3µg/ml. Other antibiotics and pollutants also have the similar linear relationships. The proposed method is simple, convenient and sensitive. Particularly, it does not need the immobilization of the microorganism. This method was used for monitoring of wastewater from a hospital.  相似文献

In this study, we developed a simple strategy to prepare a biofilm reactor (BFR) sensor for the universal biochemical oxygen demand (BOD) determination. The microorganisms in fresh water were domesticated by artificial seawater with different salinity gradients successively to prepare the BFR sensor. The prepared BFR sensor exhibits an efficient ability to degrade a variety of organic substances. The linear range of BOD determination by the BFR sensor is 1.0–10.0 mg/L−1 with a correlation coefficient of 0.9951. The detection limit is 0.30 mg/L according to three times of signal-to-noise ratio. What is more, the BFR sensor displayed excellent performances for the BOD determination of different water samples, including both fresh water and seawater. The 16S-rRNA gene sequencing technology was used to analyze the microbial species before and after the domestication. The results show that it is a general approach for the rapid BOD determination in different water samples.  相似文献

An unexpected frequency response for a piezoelectric quartz crystal (PQC) sensor to liquid density and viscosity was reported. For a PQC oscillating in a liquid phase, the frequency shifts (△f) show a wave-shape response to liquid density (p) and viscosity 03) in fine structure, ifthe longitudinal wave effect was not eliminated. This result is different from the well-knownlinear relationship between of Af and (ρη)1/2. An oscillating frequency-temperature curve of thesensor was observed and explained.  相似文献
