Ternary mixtures of fluoroquinolones, with a 7-piperazinyl substituted group have been simultaneously determined in human urine samples by application of a multivariate calibration partial least squares (PLS) model. The calibration set was designed with 15 urine samples containing different concentrations of the three fluoroquinolones and 16 blank urine samples. The concentration range for the fluoroquinolones were up to 25 ng ml−1 for norfloxacin (NOR), 80 ng ml−1 for ofloxacin (OFLO) and 300 ng ml−1 for enoxacin (ENO). The method is based on the native fluorescence emission of these compounds in sodium dodecyl sulfate (SDS) medium, at pH 4.0, when exciting at 277 nm. A selection of the emission wavelength range used for the analysis was made for each component. Intraday and interday precision values were determined. Figures of merit as selectivity, sensitivity, limit of detection (LOD) and analytical sensitivity were also calculated. Using the standard addition methodology, five urine samples from five different persons, fortified with three concentration levels of the fluoroquinolones, were analyzed. The limits of detection in urine were 10.0, 0.5 and 0.8 ng ml−1 for ENO, NOR and OFLO, respectively.  相似文献

For the first time, a solid sensing zone implemented with unsegmented flow analysis is described for the simultaneous determination of two pesticides, thiabendazole and warfarin. The system works as a simple and rapid spectrofluorimetric biparameter sensor. The sensor is based on the retention of the analytes on the sensing solid zone (octadecyl silane C18 gel) placed in the detection zone itself into a quartz flow-cell. A temporary sequentiation in the arrival of the analytes to the sensing zone is achieved by on line separation using a pre-column of the same gel placed just before the flow cell. Thiabendazole is determined the first (using methanol 30% (v/v) as carrier/elution solution) because it passes through the pre-column while warfarin is strongly retained in it. Then, warfarin is conveniently eluted from the pre-column (using methanol 50% (v/v) as carrier/elution solution) the intrinsic fluorescence peak height measured at an excitation wavelength of 309 nm and an emission wavelength of 368 nm is used as analytical signal. Using a low sample volume (40 μl), the analytical signal showed a very good linearity in the range 10-800 ng ml−1 and 2-40 μg ml−1 with detection limits of 2.35 ng ml−1 and 0.54 μg ml−1 for thiabendazole and warfarin, respectively. The sensor was satisfactorily applied to the determination of these two analytes in pesticides and pharmaceutical preparations.  相似文献

The aim of this work was to quantify five commonly used pesticides (propoxur, carbaryl, carbendazim, thiabendazole and fuberidazole) in real samples as: tomato, orange juice, grapefruit juice, lemon and tangerine. The method used for the determination of these analytes in the complex matrices was high-performance liquid chromatography with diode array detection. In order to work under isocratic conditions and to complete each run in less than 10 min, the analysis was carried out applying multivariate curve resolution coupled to alternating least-squares (MCR–ALS). The flexibility of this applied multivariate model allowed the prediction of the concentrations of the five analytes in complex samples including strongly coeluting analytes, elution time shifts, band shape changes and presence of uncalibrated interferents. The obtained limits of detection (in μg L−1) using the proposed methodology were 2.3 (carbendazim), 0.90 (thiabendazole), 12 (propoxur), 0.46 (fuberidazole) and 0.32 (carbaryl).  相似文献

Two highly sensitive and selective methods based on gas chromatography coupled to mass spectrometry (GC-MS) in the selected ion monitoring (SIM) mode have been developed for the quantification of 2,6-dichlorophenol (2,6-DCP), a sex pheromone of the tick females of Anocentor nitens. Standard addition method and calibration curve techniques using 5-bromine-4-hydroxy-3-methoxybenzaldehyde (5-BrV) as internal standard (IS) afforded detection limit of 0.1 ng ml−1. The calibration curve was linear over the concentration range from 0.5 to 500 ng ml−1 for 2,6-DCP. Results show that the concentration range of sex pheromone in the extracts samples was 1.08-10.35 ng ml−1. The methods developed provided reliable procedures to determine amounts of 2,6-DCP present in ticks.  相似文献

This paper presents the development of a new flow-injection system combined with solid-surface fluorescence detection for the determination of the widely used fungicide thiabendazole. Nylon powder was probed as a novel solid support for building the optosensor. The method is based on the on-line immobilization of thiabendazole onto nylon in a continuous flow system, followed by the measurement of its native fluorescence. Aqueous samples are directly injected in a water carrier, resulting in a very simple and economical method. The analytical figures of merit obtained using 1500 μL of sample and 75% methanol (v/v) as eluting solution were: linear calibration range from 8 to 120 ng mL−1 (the lowest value corresponds to the quantitation limit), relative standard deviation, 0.9% (n = 5) at a level of 64 ng mL−1, limit of detection calculated according to 1995 IUPAC recommendations is to 2.8 ng mL−1, and sampling rate of 14 samples h−1. The potential interference from other agrochemicals, metal ions and common anions, and the viability of determining thiabendazole in real water samples were also evaluated.  相似文献

This paper presents two easy and selective methods for determining the active principles nafronyl (NFL) and naproxen (NAP), using a flow-through fluorescence optosensor based on the on-line immobilization on a nonionic-exchanger (Silica Gel, Davisil™ and Amberlite XAD 7, respectively) solid support. The determination was performed in 5×10−3 M HAc/NaAc buffer solution at pH 5 for NFL and 15×10−3 M glycine/HCl buffer solution at pH 2.5 for NAP at a working temperature of 20 °C. The fluorescence intensities were measured at λex/em=294/336 nm and λex/em=332/354 nm for NFL and NAP, respectively. The response time for these optosensors were practically instant, obtaining a linear concentration range between 0 and 700.0 ng ml−1 with a detection limit of 20.8 ng ml−1, an analytical sensitivity of 10.1 ng ml−1 and a standard deviation of 1.27% at a 500 ng ml−1 concentration level for NFL and a linear concentration range between 0 and 200.0 ng ml−1 with the detection limit of 13.3 ng ml−1, an analytical sensitivity of 6.0 ng ml−1 and a standard deviation of 3.52% at a 100 ng ml−1 concentration level for NAP. The proposed methods were satisfactorily applied to real samples (three commercial formulations and urine samples). The effects of the possible interferences were evaluated in all cases.  相似文献

A sensitive procedure has been developed for selenium and tellurium determination in milk by hydride generation atomic fluorescence spectrometry (HG-AFS) after microwave-assisted sample digestion. The method provides sensitivity values of 1591 and 997 fluorescence units ng−1 ml−1 with detection limits of 0.005 and 0.015 ng ml−1 for Se and Te, respectively. The application of the developed methodology to the analysis of cow milk samples of the Spanish market evidenced the presence of concentration ranges from 11.1 to 26.0 ng ml−1 for Se, and from 1.04 to 9.7 ng ml−1 for Te having found a good comparability with data obtained after dry-ashing of samples.  相似文献

A simple, sensitive, low-cost and rapid, flow injection system for the on-line preconcentration of lead by sorption on a microcolumn packed with silica gel funtionalized with methylthiosalicylate (TS-gel) was developped. The metal is directly retained on the sorbent column and subsequently then eluted from it by EDTA. Five variables (sample flow rate, eluent flow rate, eluent concentration, pH and buffer concentration) were considered as factors in the optimization process. Interactions between analytical factors and their optimal levels were investigated using two level factorial and Box-Behnken designs. The optimum conditions established were applied to the determination of lead by flow injection inductively coupled plasma atomic emission spectrometry (FI-ICP-AES). The proposed method has a linear calibration range from 10 to at least 500 ng ml−1 of lead. At a sample frequency of 24 h−1 and a 120 s preconcentration time, the enrichment factor was 41, the detection limit was 15.3 ng ml−1 (S/N=3) and the precision, expressed as relative standard deviation, was 0.9% (at 100 ng ml−1). Validation of the developed method was carried out against electrothermal atomic absorption spectrometry analysis without statistically significant differences between the proposed method and the atomic absorption method.  相似文献

This is the first report of the determination of aniline with tetra-substituted amino aluminium phthalocyanine (TAAlPc) by a fluorimetric method. In KBr-HCl solution, nitrite ion diazotizes TAAlPc, thus, the fluorescence of TAAlPc is dramatically quenched. However, there is less quenching in the presence of aniline and the recovery in fluorescence intensity is linear with the concentration of aniline. Based on this, a novel method has been developed for the determination of aniline in aqueous solutions. Under the optimal conditions, the calibration graph for aniline is from 5 to 300 ng ml−1 with a 3σ limit of detection of 1.8 ng ml−1. The relative standard deviation for nine replicate measurements of 100 ng ml−1 aniline is 1.7%. The method was applied to the analysis of water samples with satisfactory results.  相似文献

A new biacridine compound, 10,10′-dimethyl-3,3′-disulfo-9,9′-biacridine (DMDSBA) was synthesized, and its chemiluminescent characteristics were investigated in detail. DMDSBA was used to label anti-CEA antibody. The labeling ratio was estimated to be 1.15-1.32, and the average labeling ratio was 1.25. The results show that there are no obvious changes in the immunoreactivity of the labeled anti-CEA antibody and the quantum efficiency of DMDSBA after attaching to the anti-CEA antibody. In addition, the conditions of labeling reaction, sandwich immunoassay and chemiluminescent reaction have been studied. A new sandwich chemiluminescent immunoassay method was firstly established, and used to determine CEA in human serum, the calibration range is 1.0-100 ng ml−1 and the minimal detectable concentration of CEA is 0.53 ng ml−1, the relative standard deviation is 6.5% for 20 ng ml−1 CEA. This method was well-matched with radio immunoassay.  相似文献

A supramolecular solvent made up of vesicles of decanoic acid in the nano- and microscale regimes dispersed in a continuous aqueous phase is proposed for the extraction/preconcentration of benzimidazolic fungicides (BFs) from river and underground water samples prior to their determination by liquid chromatography (LC)/fluorimetry. The solvent is produced from the coacervation of decanoic acid aqueous vesicles by the action of tetrabutylammonium (Bu4N+). Carbendazim (CB), thiabendazole (TB) and fuberidazole (FB) are extracted on the basis of hydrophobic and π-cation interactions and the formation of hydrogen bonds. The extraction provides high preconcentration factors (160 for CB and 190 for TB and FB), requires a short time (the procedure takes less than 20 min and several samples can be simultaneously processed) and a low sample volume (20 mL), and avoids the use of toxic organic solvents. Because of the absence of matrix interferences and the low viscosity of the extracts, these can be directly injected into the chromatographic system without the need of cleaning-up or diluting them. Recoveries are not influenced by the presence of salt concentrations up to 1 M. The proposed method provides detection limits for the determination of CB, TB and FB in natural waters of 32, 4 and 0.1 ng L−1, respectively, and a precision, expressed as relative standard deviation (n = 11) of 5.5% for CB (100 ng L−1), 4.0% for TB (80 ng L−1) and 2.5% for FB (30 ng L−1). Recoveries obtained by applying this approach to the analysis of river and underground water samples fortified at the ng L−1 level are in the intervals 75–83, 95–102 and 97–101% for CB, TB and FB, respectively.  相似文献

An electrolytic hydride generation system for determination of another hydride forming element, cadmium, by catholyte variation electrochemical hydride generation (EcHG) atomic absorption spectrometry is described. A laboratory-made electrolytic cell with lead-tin alloy as cathode material is designed as electrolytic generator of molecular hydride. The influences of several parameters on the analytical signal have been evaluated using a Plackett-Burman experimental design. The significant parameters such as cathode surface area, electrolytic current, carrier gas flow rate and catholyte concentration have been optimized using univariate method. The analytical figures of merit of procedure developed were determined. The calibration curve was linear up to 20 ng ml−1of cadmium. The concentration detection limit (3σ, n = 8) of 0.2 ng ml−1 and repeatability (relative standard deviation, n = 7) of 3.1% were achieved at 10.0 ng ml−1. It was shown that interferences from major constituents at high concentrations were significant. The accuracy of method was verified using a real sample (spiked tap water) by standard addition calibration technique. Recovery of 104% was achieved for Cd in the spiked tap water sample.  相似文献

Partitioned dispersive liquid-liquid microextraction (PDLLME), using THF as the dispersive solvent and dichloromethane as the extraction solvent, was utilized to isolate and concentrate phenylurea herbicides (PUHs) from aqueous samples. In PDLLME, a dispersive solvent should be able to partition in the organic extractant droplets to effectively extract the polar organic compounds from aqueous samples. The mixture of the water-immiscible extractant and the partitioned dispersive solvent was obtained by centrifugation, dried under low pressure, reconstituted in methanol-water mixture (1:1), and injected into a HPLC system for the determination of PUHs. The enrichment factors of the PUHs ranged from 68 to 126 under the optimal conditions. The linear range was 0.5-100 ng ml−1 for each analyte, the relative standard deviations of PUHs were in the range of 1.5-5.9% (n = 5), and the detection limits (signal-to-noise ratio of 3) ranged from 0.10 to 0.28 ng ml−1 for the herbicides. The range of intraday precision (n = 5) for PUHs at the levels of 0.5, 5, and 50 ng ml−1 were 3.0-5.9%, 1.8-3.3%, and 2.2-3.6%, respectively. The range of interday precision (n = 5) at 0.5, 5, and 50 ng ml−1 were 0.4-1.8%, 1.2-2.4%, and 0.9-2.3%, respectively. The recoveries of PUHs from three spiked river water samples, at a level of 10 ng ml−1, were 91.2-104.1%. Due to its rapidity, ease of operation, and high recovery, PDLLME can be utilized to isolate and concentrate organic environmental contaminants such as PUHs from aqueous samples.  相似文献

A highly sensitive and relatively interference-free spectrophotometric method for determination of calcium is described. The method is based on the reaction between calcium ions and carboxyazo-p-CH3 in aqueous citrate medium of pH 7, to form a blue complex with maximum absorption at 716 nm. The calibration is linear up to 0.12 μg ml−1 calcium with a repeatability (R.S.D.) of 1.0% at a concentration of 0.04 μg ml−1 (n=5). The molar absorptivity of the complex and Sandell’s sensitivity are 3.5×105 l mol−1 cm−1 and 0.11 ng cm−2, its 10σ limit of quantification and the 3σ limit of detection were found to be 0.3 ng ml−1 and 0.09 ng ml−1 respectively. The influence of reaction variables and the effect of interfering ions are studied; no interference was observed in clinical samples. The proposed method has been applied directly to the determination of calcium in clinical samples without the need for pre-concentration, masking metal ions and digesting samples.  相似文献

The simultaneous determination of carbendazim, fuberidazole and thiabendazole was accomplished by cross-section (CS) fluorimetry in combination with multivariate calibration algorithms. The total luminescence information of the compounds was used to optimise the linear trajectories of the CS. A comparison between principal component regression (PCR) and two partial least squares (PLS) algorithms, PLS-1 and PLS-2, with different pre-processing methodologies was made. The final model, which applied the PLS-1 method, built using pesticide standard and emission spectra, was successfully used for the determination of these compounds in synthetic mixtures. However, a different PLS-1 multivariate calibration model, based on CS through the total luminescence spectroscopic data, was necessary for determining the cited pesticides in water samples. Mean centring was the best pre-processing technique in both PLS-1 models. This later calibration model was built from ultra-pure water samples spiked with known carbendazim, fuberidazole and thiabendazole concentrations, after solid-phase extraction (SPE). The method, which had a precision better than 5%, was shown to be suitable for carbendazim, fuberidazole and thiabendazole monitoring in water samples at trace levels.  相似文献

A simple and sensitive kinetic method for the determination of traces of mercury (70-760 ng ml−1) based on its inhibitory effect on the addition reaction between methyl green and sulfite ion is proposed. The reaction was monitored spectrophotometrically by measuring the decrease in absorbance of methyl green at 596 nm between 2 and 4 min using a fixed time method. Artificial neural networks with back propagation algorithm coupled with an orthogonal array design were applied to the modeling of the proposed kinetic system and optimization of experimental conditions. An orthogonal design was utilized to design the experimental protocol, in which pH, concentration of sulfite, temperature, and concentration of methyl green were varied simultaneously. Optimum experimental conditions in term of sensitivity were generated by using ANNs. The rate of decrease in absorbance is inversely proportional to the concentration of Hg(II) over entire concentration range tested (100-550 ng ml−1) with a detection limit of 45 ng ml−1 and a relative standard deviation at 200-400 ng ml−1 Hg(II) of 3.2% (n=5). A simple preconcentration step improved the limit of detection and linear dynamic range of the method to about 8 and 12-760 ng ml−1, respectively, by about 10 times enrichment of mercury between 12 and 75 ng ml−1. The method was based on enrichment of Hg(II) from dilute samples on an anionic ion exchanger fixed on a plastic strip and was applied to the determination of Hg(II) in environmental samples with satisfactory results.  相似文献

This paper discusses the first analytical determination of the widely used fungicide thiabendazole by nylon-induced phosphorimetry. Nylon was investigated as a novel solid-matrix for inducing room-temperature phosphorescence of thiabendazole, which was enhanced under the effect of external heavy-atom salts. Among the investigated salts, lead(II) acetate was the most effective in yielding a high phosphorescence signal. An additional enhancement of the phosphorescence emission was attained when the measurements were carried out under a nitrogen atmosphere. There was only a moderate increase in the presence of cyclodextrins. The room-temperature phosphorescence lifetimes of the adsorbed thiabendazole were measured under different working conditions and, in all cases, two decaying components were detected. On the basis of the obtained results, a very simple and sensitive phosphorimetric method for the determination of thiabendazole was established. The analytical figures of merit obtained under the best experimental conditions were: linear calibration range from 0.031 to 0.26 μg ml−1 (the lowest value corresponds to the quantitation limit), relative standard deviation, 2.4% (n = 5) at a level of 0.096 μg ml−1, and limit of detection calculated according to 1995 IUPAC Recommendations equal to 0.010 μg ml−1 (0.03 ng/spot). The potential interference from common agrochemicals was also studied. The feasibility of determining thiabendazole in real samples was successfully evaluated through the analysis of spiked river, tap and mineral water samples.  相似文献

A sensitive and simple solid-phase preconcentration procedure for enrichment of cadmium prior to analysis by flame atomic absorption spectrometry (FAAS) is described. The method is based on the adsorption of cadmium as CdI42− on naphthalene-methyltrioctylammonium chloride adsorbent, elution by nitric acid and subsequent determination by FAAS. The effect of pH, iodide concentration, sample flow rate, volume of the sample and diverse ions on the recovery of the analyte was investigated and optimum conditions were established. A preconcentration factor of 40 was achieved using the optimum conditions. The calibration graph was linear in the range 1-100 ng ml−1 cadmium in the initial solution. The detection limit based on the 3Sb criterion was 0.6 ng ml−1 and the relative standard deviations (RSD) were 3.9 and 1.05% for 5 and 40 ng ml−1, respectively (n=8). The method was successfully applied to the determination of cadmium added to river, tap and Persian Gulf water samples.  相似文献

A new method based on near-infrared (near-IR) fluorescence recovery, employing a two-reagent system which is composed of an anionic heptamethylene cyanine (HMC) and a polycationic phthalocyanine dye, Alcian blue 8GX, is presented for the determination of nucleic acids. With a maximum excitation wavelength at 766 nm and a maximum emission wavelength at 796 nm, the fluorescence recovery is linear with the concentration of nucleic acids added. Factors including the acidity of the medium, the reaction time, the optimal ratio of the two reagents, as well as the influence of foreign substance were all investigated. Meanwhile, the mechanism of fluorescence recovery was also studied. Under the optimal conditions, the linear ranges of the calibration curves were 10-250 ng ml−1 for calf thymus DNA (CT DNA) and 10-200 ng ml−1 for yeast RNA. The detection limits were 6.8 ng ml−1 for CT DNA and 6.3 ng ml−1 for yeast RNA, respectively. The method has been applied to the analysis of practical samples and the recovery results were satisfactory.  相似文献

Pesticides residues in aquatic ecosystems are an environmental concern which requires efficient analytical methods. In this study, we proposed a generic method for the quantification of 13 pesticides (azoxystrobin, clomazone, diflufenican, dimethachlor, carbendazim, iprodion, isoproturon, mesosulfuron-methyl, metazachlor, napropamid, quizalofop and thifensulfuron-methyl) in three environmental matrices. Pesticides from water were extracted using a solid phase extraction system and a single solid-liquid extraction method was optimized for sediment and fish muscle, followed by a unique analysis by liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS). Limits of quantification were below 5 ng L−1 for water (except for fluroxypyr and iprodion) and ranged between 0.1 ng g−1 and 57.7 ng g−1 for sediments and regarding fish, were below 1 ng g−1 for 8 molecules and were determined between 5 and 49 ng g−1 for the 5 other compounds. This method was finally used as a new routine practice for environmental research.  相似文献
