血小板聚集在血栓形成中起着先导而关键的作用,而严重威胁人类生命健康的心脑血管疾病多数与血栓形成和血栓栓塞有着密切的联系[1]。二氢哒嗪酮类化合物对抑制血小板聚集有较强的药理活性,对二磷酸腺苷(ADP)、花生四烯酸(AA)、血小板活化因子(PAF)和胶原(Collagen)引起的血小板聚集均有抑制作用。深入研究二氢哒嗪酮类化合物的构效关系,以期寻找活性更强的抗血小板聚集药物,是开发新型心脑血管系统疾病药物的重要研究方向之一。本文设计合成了6-(4-取代苯基)-5-甲基-4,5二氢-3(2H)哒嗪酮类化合物[2,3,4],并进行了体外药理实验。1实验部…  相似文献

The reaction of 2-amino-4-methyl-6-(2-pyridyl)-7,8-dihydroindazolo[4,5-d]thiazole, obtained by treating 3-methyl-4-oxo-1-(2-pyridyl)-4,5,6,7-tetrahydroindazole with pyridinium bromide perbromide and then with thiourea, and 2-amino-4-methyl-6-phenyl-7,8-dihydroindazolo[4,5-d]thiazole with 4-bromo-, 4-fluoro-, 4-dimethylamino-, 4-methoxy-, 3,4-dimethoxy-, and 3,4-methylenedioxybenzaldehydes, furfural, pyridinecarbaldehyde, and thiophenecarbaldehyde gave the corresponding Schiff bases. The products of the condensation of these aminothiazoles with cinnamaldehyde, 1-(2-pyridyl)- and 4-chloro-1-(2,4-difluorophenyl)-5-formyl-3-methyl-6,7-dihydroindazoles, 2-formyl-dimedone, and 2-formyl-1,3-indanedione were also obtained.  相似文献

设计了一条工艺简单、环境友好、成本较低的新型二氧戊环类化合物合成路线. 通过缩醛、氨解和酰胺还原三步反应得到目标产物,产率提高,平均收率达到52.1%,高于文献报道的5步合成反应收率22.4%. 通过新设计的合成路线合成了5个新化合物,并经质谱、核磁和元素分析测试技术确证了其结构.  相似文献

The nitrosation of N-aryl-3-oxobutanethioamides afforded 1-(6-R-benzothiasol-2-yl)-1-hydroxyimino-2-propanones that at oximation with hydroxylamine were converted into 1-(6-R-benzothiazol-2-yl)-1,2-dihydroxyiminopropanes. The title compound, (4R,5R)-1,3-dioxolance-4,5-bis(2-pyridineacrboxylic acid), has been synthesized and characterized by single-crystal X-ray diffraction, IR, NMR, and MS analyses. Crystal structure of the title compound was grown from ETOH by slow diffusion at room temperature. The title compound crystallizes in monoclinic, space group C2 with a = 17.805(3), b = 11.459(3), c = 11.1656(17) , β = 113.066(4)°, V = 2095.9(6) 3, Z = 4, F(000) = 880, Dc = 1.332 Mg/m-3, C23H20N2O6, Mr = 420.41 and μ = 0.10 mm-1  相似文献

Alkylation of the sodium salt and the trimethyl silylated derivatives of 6-methyl-1,2,4-triazin-3(4H)-one 1-oxide with chloromethoxyethyl acetate, n-hexyl chloride and benzyl bromide gave the 4-substituted products. However, attempts to achieve the ring closure of N4-(2-acetoxyethoxymethyl)thiosemicarbazide with bicarbonyl compounds to the corresponding as-triazines under different reaction conditions was not possible without disruption of the acetoxyethoxymethyl moiety. Although the as-triazine nucleoside analog II did not show antileukemic activity, this and other 4-alkylated as-triazine 1-oxides revealed good growth inhibitory effects against a representative spectrum of microorganisms.  相似文献

(R)-(+)-2-Methyl-2-ethyl-3-propiothiolactone was synthesized by debenzylation and cyclization of (?)-2-methyl-2-ethyl-3-benzylmercaptopropionyl chloride under the conditions of Friedel-Crafts synthesis, and by dehydration of (R)-(+)-2-methyl-2-ethyl-3-mercaptopropionic acid with dicyclohexyl carbodiimide. The configuration of the (+)-propiothiolactone was determined by chemical interconversion with (?)-2-methyl-2-ethylsuccinic acid, the absolute configuration of which is known to be (R). The polymerization of (R)-(+)-2-methyl-2-ethyl-3-propiothiolactone was performed in bulk with tetrabutylammonium versatate as catalyst. The specific rotation of the polymer ([α]D +151.7°) compared with the rotation of the low molecular weight model compound (R)-(+)-2-methyl-2-ethyl-3-acetylmercapto-thiolpropionic acid methyl ester ([α]D +55.0°) shows a significant enhancement, thus suggesting the possibility of the presence of rigid conformations in polymer chain.  相似文献

The C-glucopyranosyl nucleosides (1-4) containing the N-acetyl glucosaminyl and uridine units have been synthesized as nonhydrolyzable substrate analogues of UDP-GlcNAc aimed to inhibit the chitin synthases. The key intermediate, 4-(2'-(N-acetylamino)-3', 4',6'-tri-O-benzyl-2'-deoxy-alpha-D-glucopyranosyl)but-2-enoic acid (5), was prepared from the perbenzylated (N-acetylamino)-alpha-C-allylglucoside (7), by successive oxidative cleavage, Wittig olefination, and ester deprotection. The coupling of the acid 5 with the hydroxyl or amine function of the uridine derivatives (6a or 6b) afforded, respectively, the ester 12 and amide 14. The dihydroxylation of the conjugated double bond in ester 12 or amide 14 was better achieved with osmium tetraoxide/barium chlorate, leading to the expected diols 13 and 15 as a mixture of two diastereoisomers. The desired compounds 1-4 were obtained after catalytic hydrogenation of compounds 12-15.  相似文献
