We report a novel 1:1 cocrystal of β‐alanine with dl ‐tartaric acid, C3H7NO2·C4H6O6, (II), and three new molecular salts of dl ‐tartaric acid with β‐alanine {3‐azaniumylpropanoic acid–3‐azaniumylpropanoate dl ‐tartaric acid–dl ‐tartrate, [H(C3H7NO2)2]+·[H(C4H5O6)2], (III)}, γ‐aminobutyric acid [3‐carboxypropanaminium dl ‐tartrate, C4H10NO2+·C4H5O6, (IV)] and dl ‐α‐aminobutyric acid {dl ‐2‐azaniumylbutanoic acid–dl ‐2‐azaniumylbutanoate dl ‐tartaric acid–dl ‐tartrate, [H(C4H9NO2)2]+·[H(C4H5O6)2], (V)}. The crystal structures of binary crystals of dl ‐tartaric acid with glycine, (I), β‐alanine, (II) and (III), GABA, (IV), and dl ‐AABA, (V), have similar molecular packing and crystallographic motifs. The shortest amino acid (i.e. glycine) forms a cocrystal, (I), with dl ‐tartaric acid, whereas the larger amino acids form molecular salts, viz. (IV) and (V). β‐Alanine is the only amino acid capable of forming both a cocrystal [i.e. (II)] and a molecular salt [i.e. (III)] with dl ‐tartaric acid. The cocrystals of glycine and β‐alanine with dl ‐tartaric acid, i.e. (I) and (II), respectively, contain chains of amino acid zwitterions, similar to the structure of pure glycine. In the structures of the molecular salts of amino acids, the amino acid cations form isolated dimers [of β‐alanine in (III), GABA in (IV) and dl ‐AABA in (V)], which are linked by strong O—H…O hydrogen bonds. Moreover, the three crystal structures comprise different types of dimeric cations, i.e. (AA)+ in (III) and (V), and A+A+ in (IV). Molecular salts (IV) and (V) are the first examples of molecular salts of GABA and dl ‐AABA that contain dimers of amino acid cations. The geometry of each investigated amino acid (except dl ‐AABA) correlates with the melting point of its mixed crystal.  相似文献

Monofunctional polylactones were prepared by Bu2Sn(OMe)2‐initiated ring‐opening polymerization of ε‐caprolactone (εCL) followed by acylation with bromoacetylbromide. Telechelic polylactones and polylactides were prepared via ring‐expansion polymerization with 2,2‐dibutyl‐2‐stanna‐1,3‐dioxepane (DSDOP) or 2,2‐dibutyl‐2‐stanna‐pentaoxacyclotridecane (Bu2SnTEG) as cyclic initiator. In situ combination of the polymerization with condensation by means of bromoacetylbromide yielded polylactones having bromoacetate endgroups. These endgroups were subjected to nucleophilic substitution with 3‐mercaptopropyl trimethoxysilane (3‐MPTMS). Analogous experiments were conducted with dl‐lactide. The telechelic trimethoxysilyl‐endcapped polylactones were characterized by viscosity, 1H and 13C NMR‐spectroscopy, and MALDI‐TOF mass spectrometry. α‐Methyl glutamic acid (L ‐L )‐, (L ‐D )‐, (D ‐L )‐, and (D ‐D )‐γ‐dimers were synthesized from L ‐ and D ‐glutamic acids, and the obtained dimers were subjected to polycondensation with 1‐(3‐dimethylaminopropyl)‐3‐ethylcarbodiimide hydrochloride and 1‐hydroxybenzotriazole hydrate as condensation reagents. Poly‐γ‐glutamic acid (γ‐PGA) methyl ester with the number‐average molecular weights of 5000∼20,000 were obtained by polycondensation in N,N‐dimethylformamide in 44∼91% yields. The polycondensation of (L ‐L )‐ and (D ‐D )‐dimers afforded the polymers with much larger |[α]D | compared with the corresponding dimers. The polymer could be transformed into γ‐PGA by alkaline hydrolysis or transesterification into α‐benzyl ester followed by hydrogenation. © 2001 John Wiley & Sons, Inc. A series of activated urethane‐type derivatives of γ‐benzyl‐L ‐glutamate were synthesized, and their potential as monomers for polypeptide synthesis was investigated. The derivatives of the focus of this work were a series of N‐aryloxycarbonyl‐γ‐benzyl‐L ‐glutamate 1 , of which aryl groups were phenyl, 4‐chlorophenyl, and 4‐nitrophenyl. These urethanes 1 were reactive in polar solvents such as dimethylsulfoxide, N,N‐dimethylformamide (DMF), and N,N‐dimethylacetamide (DMAc), and were efficiently converted into poly(γ‐benzyl‐L ‐glutamate) (poly(BLG)) under mild conditions; at 60 °C without addition of any catalyst. Among the three urethanes, that having 4‐nitrophenoxycarbonyl group 1c was the most reactive to give poly(BLG) efficiently, as was expected from the highly electron deficient nature of the nitrophenoxycarbonyl group. On the other hand, the urethane 1a having phenoxycarbonyl group was also efficiently converted into poly(BLG), in spite of the intrinsically less electrophilicity of the phenoxycarbonyl group. In addition, the successful formation of poly(BLG) by the reaction of 1a favored its diluted concentration (0.1 M) much more than 2.0 M, the optimum initial concentration for 1c . 1H NMR spectroscopic analyses of the reactions in situ revealed that the predominant pathway from 1 to poly(BLG) involved the intramolecular cyclization of 1 into the corresponding N‐carboxyanhydride, with release of phenol and its successive ring‐opening polymerization with release of carbon dioxide. © 2008 Wiley Periodicals, Inc. This work reports a modular and rapid approach to the stereoselective synthesis of a variety of α‐ and β‐(1→2)‐linked C‐disaccharides. The key step is a Ni‐catalyzed cross‐coupling reaction of D ‐glucal pinacol boronate with alkyl halide glycoside easily prepared from commercially available D ‐glucal. The products of this sp2–sp3 cross‐coupling reaction can be converted to glucopyranosyl, mannopyranosyl, or 2‐deoxy‐glucopyranosyl C‐mannopyranosides by one‐ or two‐step stereoselective oxidative–reductive transformations. To the best of our knowledge, we demonstrated the first synthetic application of a challenging sp2–sp3 Suzuki‐Miyaura cross‐coupling reaction in carbohydrate chemistry.  相似文献

A gene of α‐L ‐arabinofuranosidase (Abf) from Trichoderma koningii G‐39 was successfully expressed in Pichia pastoris. The recombinant enzyme was purified to > 90% homogeneity by a cation‐exchanged chromatography. The purified enzyme exhibits both α‐L ‐arabinofuranosidase and β‐D ‐xylosidase (Xyl) activities with p‐nitrophenyl‐α‐L ‐arabionfuranoside (pNPAF) and 2,4‐dinitrophenyl‐β‐D ‐xylopyanoside (2,4‐DNPX) as substrate, respectively. The stability and the catalytic feature of the bifunctional enzyme were characterized. The enzyme was stable for at least 2 h at pH values between 2 and 8.3 at room temperature when assayed for Abf and Xyl activities. Enzyme activity decreased dramatically when the pH exceeded 9.5 or dropped below 1.5. The enzyme lost 35% of Abf activity after incubation at 55 °C for 2 h, but retained 95% of Xyl activity, with 2,4‐DNXP as substrate, under the same conditions. Further investigation of the active site topology of both enzymatic functions was performed with the inhibition study of enzyme activities. The results revealed that methyl‐α‐L ‐arabinofuranoside inhibition is noncompetitive towards 2,4‐DNPX as substrate but competitive towards pNPAF. Based on the thermal stability and the inhibition studies, we suggest that the enzymatic reactions of Abf and Xyl are performed at distinct catalytic sites. The recombinant enzyme possesses both the retaining transarabinofuranosyl and transxylopyranosyl activities, indicating both enzymatic reactions proceed through a two‐step, double displacement mechanism.  相似文献

Semihydrogenation of α,β‐unsaturated ynoates and ‐ynones bearing a γ‐alkoxy group can be performed using triphenylphosphine and water. α,β‐Unsaturated ynoates were reduced to a mixture of cis and trans α,β‐unsaturated enoates, whereas, ynones were reduced to trans α,β‐unsaturated enones as the only products.  相似文献

Disconnections between in vitro responses and those observed in whole cells confound many attempts to design drugs in areas of serious medical need. A method based on 1D 1H NMR spectroscopy is reported that affords the ability to monitor the hydrolytic decomposition of the carbapenem antibiotic meropenem inside Escherichia coli cells expressing New Delhi metallo‐β‐lactamase subclass 1 (NDM‐1), an emerging antibiotic‐resistance threat. Cell‐based NMR studies demonstrated that two known NDM‐1 inhibitors, L ‐captopril and ethylenediaminetetraacetic acid (EDTA), inhibit the hydrolysis of meropenem in vivo. NDM‐1 activity in cells was also shown to be inhibited by spermine, a porin inhibitor, although in an in vitro assay, the influence of spermine on the activity of isolated NDM‐1 protein is minimal. This new approach may have generic utility for monitoring reactions involving diffusible metabolites in other complex biological matrices and whole‐cell settings, including mammalian cells.  相似文献

A silk‐like extensible poly(α,L ‐amino acid) fiber is created by self‐assembly of poly(α,L ‐lysine) and poly(α,L ‐glutamic acid) at their aqueous solutions' interface. Distinguishing features of the PLL/PLG fiber are the high extensibility and good stretch. Stretching after spinning changes this fiber to a rigid and strong one. The concept and the poly(α,L ‐amino acid) fibers themselves open doors for the production of new protein fibers which are more silk‐ and wool‐like.  相似文献

The crystal structure of methyl α‐d ‐mannopyranosyl‐(1→3)‐2‐O‐acetyl‐β‐d ‐mannopyranoside monohydrate, C15H26O12·H2O, ( II ), has been determined and the structural parameters for its constituent α‐d ‐mannopyranosyl residue compared with those for methyl α‐d ‐mannopyranoside. Mono‐O‐acetylation appears to promote the crystallization of ( II ), inferred from the difficulty in crystallizing methyl α‐d ‐mannopyranosyl‐(1→3)‐β‐d ‐mannopyranoside despite repeated attempts. The conformational properties of the O‐acetyl side chain in ( II ) are similar to those observed in recent studies of peracetylated mannose‐containing oligosaccharides, having a preferred geometry in which the C2—H2 bond eclipses the C=O bond of the acetyl group. The C2—O2 bond in ( II ) elongates by ~0.02 Å upon O‐acetylation. The phi (?) and psi (ψ) torsion angles that dictate the conformation of the internal O‐glycosidic linkage in ( II ) are similar to those determined recently in aqueous solution by NMR spectroscopy for unacetylated ( II ) using the statistical program MA′AT, with a greater disparity found for ψ (Δ = ~16°) than for ? α‐l ‐Fucosidases (EC 3.2.1.51) are exo‐glycosidases. On the basis of the multi‐alignment of amino acid sequence, α‐l ‐fucosidases were classified into two families of glycoside hydrolases, GH‐29 and GH‐95. They are responsible for the removal of l ‐fucosyl residues from the non‐reducing end of glycoconjugates. Deficiency of α‐l ‐fucosidase results in Fucosidosis due to the accumulation of fucose‐containing glycolipids, glycoproteins and oligosaccharides in various tissues. Recent studies discovered that the fucosylation levels are increased on the membrane surfaces of many carcinomas, indicating the biological function of α‐l ‐fucosidases may relate to this abnormal cell physiology. Although the gene of human α‐l ‐fucosidase (h‐fuc) was cloned, the recombinant enzyme has rarely been overexpressed as a soluble and active from. We report herein that, with carefully control on the growing condition, an active human α‐l ‐fucosidases (h‐Fuc) was successfully expressed in Escherichia coli for the first time. After a series steps of ion‐exchange and gel‐filtration chromatographic purification, the recombinant h‐Fuc with 95% homogeneity was obtained. The molecular weight of the enzyme was analyzed by SDS‐PAGE (~50 kDa) and confirmed by ESI mass (50895 Da). The recombinant h‐Fuc was stable up to 55 °C with incubation at pH 6.8 for 2 h; the optimum temperature for h‐Fuc is approximately 55 °C. The enzyme was stable at pH 2.5–7.0 for 2 h; the enzyme activity decreased greatly for pH greater than 8.0 or less than 2.0. The Km and kcat values of the recombinant h‐Fuc (at pH 6.8) were determined to be 0.28 mM and 17.1 s?1, respectively. The study of pH‐dependent activity showed that the recombinant enzyme exhibited optimum activity at two regions near at pH 4.5 and pH 6.5. These features of the recombinant h‐Fuc are comparable to the native enzyme purified directly from human liver. Studies on the transfucosylation and common intermediate of the enzymatic reaction by NMR support that h‐Fuc functions as a retaining enzyme catalyzing the hydrolysis of substrate via a two‐step, double displacement mechanism.  相似文献

Two kinds of new glycopolymers, (P(VB‐1‐GlcaH‐co‐AAm), 9 ) and (P(VB‐1‐Glco‐co‐AAm), 10 ), were synthesized through the radical copolymerization of styrene derivatives bearing pendant D ‐glucaric and D ‐gluconic moieties, N‐(p‐vinylbenzyl)‐1‐D ‐glucaramide (VB‐1‐GlcaH, 7 ), and N‐(p‐vinylbenzyl)‐D ‐gluconamide (VB‐1‐Glco, 8 ), with acrylamide (AAm). Glycopolymer 9 bearing the pendant glucaric moiety at the first position inhibited the hydrolysis of a model compound for xenobiotics‐β‐glucuronide conjugates, p‐nitrophenyl β‐D ‐glucuronide, uncompetitively, in contrast to the competitive inhibition in the presence of the corresponding isomeric glycopolymer bearing the pendant D ‐glucaric unit at the sixth position (P(VB‐6‐GlcaH‐co‐AAm), 3 ) reported in our previous article. On the other hand, another copolymer 10 bearing the gluconic moiety was found not to inhibit the hydrolysis as well as the corresponding copolymer bearing pendant gulonic unit (P(VB‐6‐Glco‐co‐AAm), 4 ). These results indicate that the hydrolysis is influenced not only by existence of pendant carboxyl units but also by the direction on the linkage of the glyco‐units to the polymer frame. Therefore the configurational position of hydroxy groups in pendant glyco‐units in macromolecular inhibitors may be essential for the interaction with β‐glucuronidase. © 2006 Wiley Periodicals, Inc. Poly(ethylene‐bε‐caprolactone) (PE‐b‐PCL) diblock copolymers were synthesized by ring‐opening polymerization (ROP) of ε‐caprolactone (CL) with α‐hydroxyl‐ω‐methyl polyethylene (PE‐OH) as a macroinitiator and ammonium decamolybdate (NH4)8[Mo10O34] as a catalyst. Polymerization was conducted in bulk (130–150°C) with high yield (87–97%). Block copolymers with different compositions were obtained and characterized by 1H and 13C NMR, MALDI‐TOF, SAXS, and DSC. End‐group analysis by NMR and MALDI‐TOF indicates the formation of α‐hydroxyl‐ω‐methyl PE‐b‐PCL. The PE‐b‐PCL degradation was studied using thermogravimetric analysis (TGA) and alkaline hydrolysis. Novel thermoplastic elastomers based on multi‐block copolymers of poly(l ‐lysine) (PLL), poly(N‐ε‐carbobenzyloxyl‐l ‐lysine) (PZLL), poly(ε‐caprolactone) (PCL), and poly(ethylene glycol) (PEG) were synthesized by combination of ring‐opening polymerization (ROP) and chain extension via l ‐lysine diisocyanate (LDI). SEC and 1H NMR were used to characterize the multi‐block copolymers, with number‐average molecular weights between 38,900 and 73,400 g/mol. Multi‐block copolymers were proved to be good thermoplastic elastomers with Young's modulus between 5 and 60 MPa and tensile strain up to 1300%. The PLL‐containing multi‐block copolymers were electrospun into non‐woven mats that exhibited high surface hydrophilicity and wettability. Differing from the moisture‐sensitive α‐amino acid N‐carboxyanhydrides (AA‐NCAs) monomers, N‐phenoxycarbonyl α‐amino acids (AA‐NPCs) can be prepared and stored in open air. In this contribution, we report that the controlled polymerizations of AA‐NPC monomers of Otert‐butyl‐dl ‐serine (BRS‐NPC), Nε‐benzyloxycarbonyl‐l ‐lysine (ZLL‐NPC) and Nε‐trifluoroacetyl‐l ‐lysine (FLL‐NPC) initiated by amines are surprisingly able to tolerate common nucleophilic impurities such as water and alcohols at a level of monomer concentration. The structures of polypeptides synthesized in the presence of water or alcohols agree well with the designed ones in the case of repeated chain extensions. Detailed mechanism study and density functional theory calculation reveal that the low concentration of AA‐NCA and the high activity of amines are the key factors to the controllability of AA‐NPC polymerizations. The water‐ and alcohol‐tolerant property in polymerizations of AA‐NPCs encourages the following studies on unprotected (phenolic) hydroxyl groups containing AA‐NPCs. The controllable polymerizations of N‐phenoxycarbonyl l ‐tyrosine (LT‐NPC) and N‐phenoxycarbonyl S‐(3‐hydroxypropyl)‐l ‐cysteine (HLC‐NPC) initiated by amines are confirmed and reported for the first time, which extends the library of AA‐NPCs and polypeptides as well. All the universality of library, the convenience of monomer preparation, and the controllability and water‐ and alcohol‐tolerant property of polymerization of AA‐NPCs significantly enhance the feasibility of polypeptide synthesis, making AA‐NPC approach a promising synthetic method of polypeptides. © 2019 Wiley Periodicals, Inc. The article describes the use of a fullerene (C60)‐β‐cyclodextrin conjugate, synthesized via 1,3dipolar cycloaddition, for the ultrasensitive electrochemical detection of p‐nitrophenol. This conjugate was successfully immobilized on the surface of a glassy carbon electrode and the developed device showed high activity towards p‐nitrophenol due to the synergetic effect of C60, the latter becoming highly conductive upon reduction. The determination of p‐nitrophenol was performed by using square wave voltammetry over a concentration range from 2.8×10?9 mol L?1 to 4.2×10?7 mol L?1 and the detection limit was calculated to be 1.2×10?9 mol L?1.  相似文献

The stereoselective synthesis of the non‐lactonic part of the natural G2 checkpoint inhibitor, (Z)‐cryptofolione, has been accomplished. Butane‐1,4‐diol was used as the starting material, and the stereogenic centers were generated through L ‐proline‐catalyzed α‐aminoxylation and Maruoka asymmetric allylation. We attempted to convert this non‐lactonic moiety to (Z)‐cryptofolione via olefin cross‐metathesis reaction, but by this approach another naturally occurring lactonic compound, goniothalamin, was obtained.  相似文献

A series of efficient zinc catalysts supported by sterically bulky benzotriazole phenoxide ( BTP ) ligands are synthesized and structurally characterized. The reactions of diethyl zinc (ZnEt2) with CMe2PhBTP ‐H, t‐BuBTP ‐H, and TMClBTP ‐H yield monoadduct [(μ‐ BTP )ZnEt]2 ( 1 – 3 ), respectively. Bisadduct complex [( t‐BuBTP )2Zn] ( 4 ) results from treatment of ZnEt2 with t‐BuBTP ‐H (2 equiv.) in toluene, but treatment of TMClBTP ‐H with ZnEt2 in the same stoichiometric proportion in Et2O produces five‐coordinated monomeric complex [( TMClBTP )2Zn(Et2O)] ( 5 ). The molecular structures of compounds 1 , 4 , and 5 are characterized by X‐ray crystal structure determinations. All complexes 1 – 5 are efficient catalysts for the ring‐opening polymerization of ε‐caprolactone (ε‐CL) in the presence of 9‐anthracenemethanol. Experimental results indicate that complex 3 exhibits the greatest activity with well‐controlled character among these complexes. The polymerizations of ε‐CL and β‐butyrolactone catalyzed by 3 are demonstrated in a “living” character with narrow polydispersity indices (monomer‐to‐initiator ratio in the range of 25–200, PDIs ≤ 1.10). The “immortal” character of 3 provides a way to synthesize as much as 16‐fold polymer chains of poly(ε‐CL) (PCL) with narrow PDI in the presence of a catalyst in a small proportion. This contribution describes the synthesis and ring‐opening (co)polymerization of several L ‐lysine N‐carboxyanhydrides (NCAs) that contain labile protective groups at the ?‐NH2 position. Four of the following L ‐lysine NCAs were investigated: N?‐trifluoroacetyl‐L ‐lysine N‐carboxyanhydride, N?‐(tert‐butoxycarbonyl)‐L ‐lysine N‐carboxyanhydride, N?‐(9‐fluorenylmethoxycarbonyl)‐L ‐lysine N‐carboxyanhydride, and N?‐(6‐nitroveratryloxycarbonyl)‐L ‐lysine N‐carboxyanhydride. In contrast to the harsh conditions that are required for acidolysis of benzyl carbamate moieties, which are usually used to protect the ?‐NH2 position of L ‐lysine during NCA polymerization, the protective groups of the L ‐lysine NCAs presented here can be removed under mildly acidic or basic conditions or by photolysis. As a consequence, these monomers may allow access to novel peptide hybrid materials that cannot be prepared from ?‐benzyloxycarbonyl‐L ‐lysine N‐carboxyanhydride (Z‐Lys NCA) because of side reactions that accompany the removal of the Z groups. By copolymerization of these L ‐lysine NCAs with labile protective groups, either with each other or with γ‐benzyl‐L ‐glutamate N‐carboxyanhydride or Z‐Lys NCA, orthogonally side‐chain‐protected copolypeptides with number‐average degrees of polymerization ≤20 were obtained. Such copolypeptides, which contain different side‐chain protective groups that can be removed independently, are interesting for the synthesis of complex polypeptide architectures or can be used as scaffolds for the preparation of synthetic antigens or protein mimetics. © 2003 Wiley Periodicals, Inc. Natural protein fibers, such as silk, having high‐performance characteristics have been important materials in biopolymer research. This article reports the development of a silk‐like extensible poly(α,L ‐amino acid) fiber inspired by self‐assembly of polypeptides in living systems. Electrostatic interaction was employed as the driving force for building the fiber, and we succeeded in spinning the fiber from an aqueous solution interface between poly(α,L ‐lysine) (PLL) and poly(α,L ‐glutamic acid) (PLG). When the PLL/PLG fiber was formed, the conformations of PLL and PLG were changed from random to β‐structures. A remarkable feature of the PLL/PLG fiber is the high extensibility. Mechanical stretching of the PLL/PLG fiber resulted in a change from an extensible fiber to a rigid and strong fiber. These features depend on the molecular conformation and the deviation in the amino acid composition of the PLL/PLG fibers. This concept and the poly(α,L ‐amino acid) fibers themselves allow the production of new protein fibers and aid the development of the science of protein folding as well as giving insight into the noncovalent interactions involved in self‐assembly.
