The first total synthesis of sphingolipid (2S,3R,4E)‐N2‐octadecanoyl‐4‐tetradecasphingenine ( 1a ), a natural sphingolipid isolated from Bombycis Corpus 101A, and of its styryl analogue 1b was achieved in good overall yield (Schemes 1 and 2). The key step involved the installation with (E) stereoselectivity of a long lipophilic chain or phenyl group on allyl alcohol derivative 3 via a cross‐metathesis reaction (→ 5a or 5b ). The N‐Boc protected 3 was easily accessible from (S)‐Garner aldehyde.  相似文献

(S)‐N‐(3,5‐dinitrobenzoyl)leucine‐N‐phenyl‐N‐propylamine‐bonded silica was used as a chiral stationary phase for separation of a set of racemic π‐acidic and π‐basic α‐amino acid amides in electrolyteless ACN‐water eluents by CEC in the RP and polar organic (PO) modes. The effect of the amount of water in the ACN‐water eluent on chiral separation was examined. As water is added to ACN, retention was shortened but resolution and selectivity deteriorated severely. Retention, enantioselectivity, and resolution factors obtained in 100% ACN were compared with those in an n‐hexane‐isopropanol eluent with a small amount of water by normal phase (NP) CEC. Much shorter retention times with comparable enantioselectivities were observed with 100% ACN, demonstrating the advantage of separation on (S)‐N‐(DNB)leucine‐N‐phenyl‐N‐propylamine‐bonded silica in PO‐CEC over NP‐CEC.  相似文献

A convenient diastereoselective synthesis of diisopropyl (2R,3R)‐3‐{{{(R/S)‐aryl[(diethoxyphosphinyl)amino]methyl}hydroxyphosphinyl}oxy}‐2‐hydroxybutanedioate through Mannich‐type reactions is reported. The reactions take place under mild conditions in good yields, and this makes it possible to introduce various substituents at the α‐position to the P‐atom of α‐aminophosphonates. The chiral diisopropyl (4R,5R)‐2‐chloro‐1,3,2‐dioxaphospholane‐4,5‐dicarboxylate ( 3 ) was found to be a good phosphonylating agent in this stereoselective reaction.  相似文献

A novel Cu(OAc)2·H2O catalyzed coupling reaction of N‐substituted‐2‐iodobenzamides with malononitrile to afford N‐substituted‐3‐amino‐4‐cyano‐isoquinoline‐1(2H)‐ones is described. The reaction proceeded in DMSO at 90°C for 5 h in nitrogen without external ligands.  相似文献

A number of novel chiral diamines 3 , (1R,2R)‐N‐monoalkylcyclohexane‐1,2‐diamines, were designed and synthesized from trans‐cyclohexane‐1,2‐diamine and applied to the catalytic asymmetric Henry reaction of benzaldehyde and nitromethane to provide β‐nitroalcohol in high yield (up to 99%) and good enantiomeric excess (up to 89%). By using ligand (1R,2R)‐N1‐(4‐methylpentan‐2‐yl)cyclohexane‐1,2‐diamine ( 3g ), the reaction was optimized in terms of the metal ion, temperature, solvent and base. Further experiments indicated that the complex, 3g –Cu(OAc)2, was an efficient catalyst in the asymmetric Henry reaction between different aldehydes and nitromethane, and the desired products have been obtained with high chemical yields (up to 99%) and high enantiomeric excess (up to 93%). The optimized catalyst promoted the diastereoselective Henry reaction of various aldehyde substrates and nitroalkane, which gave the corresponding anti‐selective adduct with up to 99% yield and 83:17 anti/syn selectivity. Upon scaling up to gram quantities, the β‐nitroalcohol was obtained in good yield (96%) with excellent selectivities (93% ee). The chiral induction mechanism was tentatively explained on the basis of a previously proposed transition‐state model. Copyright © 2014 John Wiley & Sons, Ltd.  相似文献

A series of Zn (II), Pd (II) and Cd (II) complexes, [(L) n MX 2 ] m (L = L‐a–L‐c; M = Zn, Pd; X = Cl; M = Cd; X = Br; n, m = 1 or 2), containing 4‐methoxy‐N‐(pyridin‐2‐ylmethylene) aniline ( L‐a ), 4‐methoxy‐N‐(pyridin‐2‐ylmethyl) aniline ( L‐b ) and 4‐methoxy‐N‐methyl‐N‐(pyridin‐2‐ylmethyl) aniline ( L‐c ) have been synthesized and characterized. The X‐ray crystal structures of Pd (II) complexes [L 1 PdCl 2 ] (L = L‐b and L‐c) revealed distorted square planar geometries obtained via coordinative interaction of the nitrogen atoms of pyridine and amine moieties and two chloro ligands. The geometry around Zn (II) center in [(L‐a)ZnCl 2 ] and [(L‐c)ZnCl 2 ] can be best described as distorted tetrahedral, whereas [(L‐b) 2 ZnCl 2 ] and [(L‐b) 2 CdBr 2 ] achieved 6‐coordinated octahedral geometries around Zn and Cd centers through 2‐equivalent ligands, respectively. In addition, a dimeric [(L‐c)Cd(μ ‐ Br)Br] 2 complex exhibited typical 5‐coordinated trigonal bipyramidal geometry around Cd center. The polymerization of methyl methacrylate in the presence of modified methylaluminoxane was evaluated by all the synthesized complexes at 60°C. Among these complexes, [(L‐b)PdCl 2 ] showed the highest catalytic activity [3.80 × 104 g poly (methyl methacrylate) (PMMA)/mol Pd hr?1], yielding high molecular weight (9.12 × 105 g mol?1) PMMA. Syndio‐enriched PMMA (characterized using 1H‐NMR spectroscopy) of about 0.68 was obtained with Tg in the range 120–128°C. Unlike imine and amine moieties, the introduction of N‐methyl moiety has an adverse effect on the catalytic activity, but the syndiotacticity remained unaffected.  相似文献

The thermal behavior and kinetic parameters of the exothermic decomposition reaction of N‐N‐bis[N‐(2,2,2‐tri‐nitroethyl)‐N‐nitro]ethylenediamine in a temperature‐programmed mode have been investigated by means of differential scanning calorimetry (DSC). The results show that kinetic model function in differential form, apparent activation energy Ea and pre‐exponential factor A of this reaction are 3(1 ‐α)2/3, 203.67 kJ·mol?1 and 1020.61s?1, respectively. The critical temperature of thermal explosion of the compound is 182.2 °C. The values of ΔS ΔH and ΔG of this reaction are 143.3 J·mol?1·K?1, 199.5 kJ·mol?1 and 135.5 kJ·mol?1, respectively.  相似文献

N-甲基-2-单芳基苯并噻唑啉 (1) 在固体状态时可以稳定地储藏在空气中。但是,它们在不同溶剂中却表现出了不同的行为。在醇溶液或二甲亚砜-水的体系中,1相对稳定;而在其它有机溶剂如:丙酮,氯仿,二氯甲烷和乙酸乙酯等溶剂中,会发生自发偶联反应生成相应的邻-(N-芳酰基-N-甲氨基苯)-二硫化合物 (2)。对这些新的化合物 (1) 和 (2) 进行了体外人乳腺癌细胞 (MDA-MB-231) 血管内皮生长因子靶点筛选,大部分化合物都表现出了抑制活性。实验结果表明这两类化合物 (1) 和 (2) 作为血管内皮生长因子抑制剂值得进行深入研究。  相似文献

A predominantly trans‐1,2‐disubstituted ethane system – N,N,N‐trimethyl‐(3,3‐dimethylbutyl)ammonium iodide – is of particular interest for conformational analysis, because it contains both an organic and a highly polar substituent, making it soluble and thus applicable to study in a large variety of solvents. The fraction of the trans conformer of this molecule in a wide range of protic and aprotic solvents was determined by the nuclear magnetic resonance proton couplings to be approximately 90%, in contrast to the previously assumed 100%. The consistently strong preference of the trans conformation should establish N,N,N‐trimethyl‐(3,3‐dimethylbutyl)ammonium iodide as a possibly useful ‘trans‐standard’ in conformational analysis, much more so than 1,2‐ditert‐butylethane, which has a poor solubility in many solvents. In a search for new insect growth regulators with unusual biological properties and different activity spectrum, we thought that the preservation of the bioactive unit and the introduction of 2‐methyl‐3‐(triphenylgermanyl)propoxycarbonyl in Ntert‐butyl‐N,N′‐dibenzoylhydrazine would enhance their larvicidal activities to a significant degree. Therefore, we designed and synthesized N′‐tert‐butyl‐N′‐[2‐methyl‐3‐(triphenylgermanyl)propoxycarbonyl]‐N‐benzoylhydrazine and analogs by two procedures. These novel compounds were characterized by elemental analyses, IR, and 1H NMR. At the same time, Ntert‐butyl‐N‐substitutedbenzoylhydrazines were prepared by a new method, and some reactions involved were studied. The preliminary results indicate that some compounds have inhibitory effects against plant pathogenetic bacteria such as early blight of tomato. Copyright © 2002 John Wiley & Sons, Ltd.  相似文献

We have developed a one‐pot procedure for the preparation of N,N‐disubstituted (Z)‐4‐(halomethylidene)‐4H‐3,1‐benzothiazin‐2‐amines 3 from 2‐(2,2‐dihaloethenyl)phenyl isothiocyanates 1 , easily accessible from known 2‐(2,2‐dihaloethenyl)benzenamines by a three‐step sequence, and secondary amines. Thus, the isothiocyanates 1 react with secondary amines to afford the corresponding thiourea derivatives, of which the treatment with NaH provides the desired products.  相似文献

An efficient stereoselective total synthesis of (3R,5R)‐1‐(4‐hydroxyphenyl)‐7‐phenylheptane‐3,5‐diol ( 1 ) is reported based on the Mukaiyama aldol reaction. The total synthesis of compound 1 was accomplished with 30% overall yield in simple eight steps from commercially available trans‐cinnamaldehyde.  相似文献

The synthesis and the X‐ray structure of the three new N‐(arylcarbonyl)‐substituted derivatives 2a – 2c of (2R)‐bornane‐10,2‐sultam are presented and discussed. Direct comparison of the solid‐state analyses shows that the dipole‐directed SO2/C?O anti‐/syn‐conformations may be very sensitive to weak electronic/electrostatic repulsions of the heteroatom lone pairs. The optimum interactions are reached when the lone pair of the β‐positioned heteroatom is oriented in the O(3)?C(11)? N(1) plane. Such rare syn‐conformations may be observed with at least up to 1.8 kcal/mol higher energy as compared to their ground states. Additionally, these anti/syn‐conformations are also very sensitive to external influences such as, for example, the crystal‐packing forces.  相似文献

Starting from (S)‐serine, a new method was developed for the synthesis of the β‐amino acid part of sitagliptin in ten steps and with an overall yield of 30%. The crucial step of the synthesis was the ring opening of N‐ and O‐protected (R)‐aziridin‐2‐methanol with (2,4,5‐trifluorophenyl)magnesium bromide to give N‐ and O‐protected (R)‐2‐amino‐3‐(2,4,5‐trifluorophenyl)propan‐1‐ol.  相似文献
