Reaction of 2-indanone with malonitrile and ethyl cyanoacetate provides a simple way of preparing substituted indeno[1,2-a]fluorenes. Tetracyanoethylene does not yield the desired 1-dicyano-methylene-2-indanone but gives a bisproduct (5), which with phenylhydrazine, yields 2,3-dihydro-5H-indeno-[2,1-c]pyridazine (6). In the presence of aromatic amines, 2-indanone reacts with triethyl orthoformate to give 1-anilinomethylene-2-indanones.  相似文献

Derivatives of the following six ring systems were synthesized:
  1. 3,10-Dihydro-[1]benzothieno[2,3-d]imidazo[1,5-a]-pyrimidine (I)
  2. 6H-[1]Benzothieno[2,3-d]pyrazino[1,2-a]pyrimidine (II)
  3. 1,5-Dihydro-[1]benzothieno[2,3-d]imidazo[1,2-a]-pyrimidine (III)
  4. 6H-[1]Benzothieno[2,3-d]pyrimido[1,2-a]pyrimidine (IV)
  5. 1,5-Dihydro-imidazo[1,2-a]thieno[2,3-d]pyrimidine (V)
  6. 4H-Pyrimido[1,2-a]thieno[2,3-d]pyrimidine (VI)
The first four types are new heterocyclic systems. 2-Aminomethyl-5,6,7,8-tetrahydro-[1]benzothieno[2,3-d]pyrimidin-4(3H)-one (5), which was used as intermediate for typesI andII, was synthesized by various methods. TypesIII andIV were prepared from 2-methylthio-5,6,7,8-tetrahydro-[1]-benzothieno[2,3-d]pyrimidin-4(3H)-one via the corresponding 2-benzylamino derivatives, followed by ring closure.  相似文献

The discovery of a novel synthesis of new fused bicyclic isoxazoles, for example, N-methyl-3-phenyl-5,6-dihydro-4H-isoxazolo[3,4-c]azepin-8-amine (2a), N-methyl-3-phenyl-4,5-dihydroisoxazolo[3,4-c]pyridin-7-amine (2b) and N-methyl-3-phenyl-4H-pyrrolo[3,4-c]isoxazol-6-amine (2d) in high yield is reported. We speculate that the reaction proceeds via acid-mediated intramolecular 1,3-dipolar cycloaddition from 2-nitro-1,1-ethenediamines 1a,b,d.  相似文献

The behavior of methyl 3,3,3-trifluoro-2-(thiazol-2-ylimino)propionate (1) in cycloaddition and cyclocondensation reactions was studied. Cycloaddition of dimethylcyanamide to imine 1 gave a thiazolo[3,2-a][1,3,5]triazine derivative. Cyclocondensations of imine 1 with 2-aminothiazoline, N-cyclohexylbenzamidine, methyl (Z)-3-aminobut-2-enoate, and 6-amino-1-benzyluracil yielded dihydroimidazo[2,1-b]thiazol-5(6H)-one, 4,5-dihydroimidazol-5-one, 4,5-dihydro-1H-pyrrol-5-one, and dihydro-1H-pyrrolo[2,3-d]pyrimidine-2,4,6-trione derivatives, respectively.  相似文献

New 6,8-dihydro-5H-benzo[f]pyrazolo[3,4-b]quinolines 6 have been obtained in a novel solvent-free three-component reaction involving β-tetralone along with 5-aminopyrazoles 1 and benzaldehydes 2. The isomeric 6,10-dihydro-5H-benzo[h]pyrazolo[3,4-b]quinolines 9 could not be prepared in a similar fashion directly from α-tetralone, but were obtained by the reaction of amines 1 with benzylidene-derivative 10 of α-tetralone in similar conditions. The yields of quinolines obtained via this novel protocol were good and the reaction times varied from few minutes to just few seconds.  相似文献

Derivatives of 5H-thiazolo[3.2?a]thieno[2.3?d]pyrimidine(A), 5H-[1]benzothieno[2.3?d]thiazolo[3.2?a]pyrimidine (B), 4H-thiazolo[3.2?a]thieno[3.2?e]pyrimidine (C) and 5H-[1]benzo-thieno[3.2?e]thiazolo[3.2?a]pyrimidine (D) were synthesized by various methods. Similar reactions are leading to derivatives of thieno[2′.3′∶4.5]pyrimido[2.1?b][1.3]thiazine (E) and [1]benzothieno[2′.3′∶4.5]pyrimido[2.1?b][1.3]thiazine (F).C, D, E, andF are new heterocyclic ring systems. An innovative synthesis of aryl tethered 1,3-dimethylimidazo[4,5-b]pyrazin-2-ones 4 and 6 has been delineated through base catalyzed ring transformation of 6-aryl-4-(piperidin-1-yl)-2H-pyran-2-one-3-carbonitriles 1 and methyl 6-aryl-4-methylsulfanyl-2H-pyran-2-one-3-carboxylates 5 with 7-acetyl-1,3-dimethyllumazine 2 with subsequent ring contraction of the fused pyrimidine to an imidazole ring. An additional product, methyl [6-(1,3-dimethyl-2-oxo-2,3-dihydro-1H-imidazo[4,5-b]pyrazin-5-yl)-4-thiophen-2-ylpyran-2-ylidene]acetate 8b, was also isolated from the reaction of 5 and 2, as a minor constituent.  相似文献

β-Acetylvinyl-triphenylphosphonium bromide1 reacts with CMP to form the 3,N4-etheno-derivative {[6-(5′-phosphoribofuranosyl)-2-methyl-5-oxo-imidazo [1.2-c]pyrimidin-3-yl]-methyl}triphenyl-phosphonium bromide (2). Guanine affords mainly the lin. condensation product [(6-methyl-9-oxo-imidazo[1.2-a]-purin-7-yl)-methyl]triphenylphosphonium bromide (3) and the angular tricyclic product [(6-methyl-9-oxo-imidazo[2.1-b]purin-5-yl)-methyl]-triphenylphosphonium bromide (4). For comparison we synthesized the angular condensed heterocycle5, (6.8-dimethyl-9-oxo-imidazo[2.1-b]purin-5-yl)-methyl]triphenylphosphonium bromide, by reaction of 1-methylguanine with1, and the corresponding linear derivative6 [(4.6-dimethyl-9-oxo-imidazo[1.2-a]purin-7-yl)-methyl]-triphenylphosphoniumbromide from 3-methylguanine and1. AHofmann-type degradation of3 with the anion of diethyl malonate led to7, diethyl (6-methyl-9-oxo-imidazo[1.2-a]purin-7-yl)-methylmalonate, a compound whose structure resembles some Y-bases in t-RNA.Wittig reaction of the silylated nucleoside derivative8 a {[2-methyl-5-oxo-6-(2′.3′.5′-tris-trimethylsilyl)-ribofuranosyl-imidazo[1.2-c]pyrimidin-3-yl]methyl}-triphenylphosphonium bromide, with C6H5CHO resulted in the 2-methyl-3(ω-styryl)-6[2′.3′.5′-tris-(trimethylsilyl)]ribofuranosyl-imidazo[1.2-c] pyrimidin-5-one (9).  相似文献

5,6-Dihydro-4-hydroxy-6,6-dimethyl-2H-thiopyrane-2-thione (1 I) and its tautomer 2-mercapto-4H-thiopyrane-4-one (1 II) react with aliphatic aldehydes under different reaction conditions to yield mainly 5R-7,8-dihydro-2H,5H,6H-thiopyrano[2,3—b:6,5—b′]-bisthiopyran-4,6(3H)-diones2 and 2′R,4′R-5,6,6′,7′-tetrahydro-2-thioxo-spiro(4H-thiopyran-3(2H), 3′(4′H)-2′H,5′H-thiopyrano-[2,3—b]-thiopyran)-4,5′-diones3. The mechanisms of formation of the condensates2 and3 and their stereochemistry are discussed. The reaction yielding2 is analogous to the condensation of dimedone with subsequent anhydride formation.3 might be generated byDiels-Adler reaction of intermediately formed 2-thioxo-3-alkylidenethiopyranones4. An X-ray crystal structure analysis was carried out on3 b to establish its configuration and conformation.  相似文献

Three unique propeller-shaped helicenyl amines compounds: N,N-diphenyl-N-naphtho[2,1-b]thieno[2,3-b:3′,2′-d]dithiophene-5-yl-amine (1), N-phenyl-N,N-di(naphtho[2,1-b]thieno[2,3-b:3′,2′-d]dithiophene-5-yl)amine (2), and N,N,N-tri(naphtho[2,1-b]thieno[2,3-b:3′,2′-d]dithiophene-5-yl)amine (3) were efficiently synthesized by Wittig reaction and oxidative photocyclization. The crystal structures of 1, 2 and molecular configuration optimization (DFT-B3LYP/6-31+G(d)) of 3 reveal that the steric hindrance from the moiety of trithia[5]helicene effectively forces the nitrogen atom and the three bonded carbon atoms to coplanar and the interplanar angles of the facing terminal thiophene ring and benzene ring becoming larger when the helical arm increased from 1 to 3. Electrochemical properties and UV–vis absorption behaviors of 1, 2, 3 were primarily determined by the moiety of trithia[5]helicene.  相似文献

The key 3-(2-oxo-2H-chromen-3-yl)-2-oxo-2H,5H-pyrano[3,2-c]chromen-5-yl acetates 3 were synthesized in high yields by cyclocondensation of 4-oxo-4H-chromen-3-carbaldehydes 1 with coumarin-3-acetic acids 2 under mild conditions. The reaction pathway involves aldol condensation and subsequent intramolecular lactonization to afford 2-oxo-2H,5H-pyrano[3,2-c]chromene skeleton 3. Further treatment of acetates 3 with alcohols, water or nitrogen containing compounds led to 5-alkoxy-, 5-hydroxy- or 5-acylamino-2H,5H-pyrano[3,2-c]chromen-2-ones 4-6 via nucleophilic substitution of acetyloxy group at C-5. Acetates and hydroxyl derivatives 3 and 5 undergo facile rearrangement in an acid medium yielding 5-hydroxypyrano[2,3-b]chromen-2(10aH)-ones 7. Twelve prepared compounds were evaluated on their antineoplastic activities on 60 human tumour cell line panels in NCI USA. The obtained biological results confirmed that 3-(2-oxo-2H-chromen-3-yl)-2H,5H-pyrano[3,2-c]chromen-2-one represents a new leading skeleton suitable for further antitumour activity study.  相似文献

Treatment of N-substituted 2-methyl-1H-indoles 1 with S2Cl2 and DABCO in chloroform gave the corresponding [1,2]dithiolo[4,3-b]indole-3(4H)-thiones 5 by the addition of triethylamine in high yield. 1,2-Dithiole-3-thiones 5 underwent cycloaddition with one or two DMAD equivalents to afford either 2-(3-thioxo-1,3-dihydro-2H-indol-2-ylidene)-1,3-dithioles 10 or fused 4,5-dihydrothiopyrano[3,2-b]indoles 9.  相似文献

3-(6-Phenylimidazo[2,1-b]thiazol-5-yl)quinoxalin-2(1H)-ones (qunoxalinone) (6a-q) have been synthesized by the reaction of ethyl 2-oxo-2-(6-phenylimidazo[2,1-b]thiazol-5-yl)acetates (4a-e) with suitably substituted o-phenylenediamines (5a-f) under microwave heating. The ethyl 2-oxo-2-(6-phenylimidazo[2,1-b]thiazol-5-yl)acetates (4a-e) were prepared by the reaction of 6-phenylimidazo[2,1-b]thiazoles (3a-e) with ethyl chlorooxoacetate in refluxing 1,4-dioxane whereas the thiazoles (3a-e) were synthesized by the reaction of 2-bromo-1-phenylethanones (2a-e) with thiazol-2-amine in refluxing acetone.  相似文献

Alkylation of 11-benzyl-3,11-dihydro-4H-pyrimido[5′,4′:5,6]pyrido[3,2-b]indol-4-one with methyl iodide and methyl bromoacetate in DMF gave 3-alkylpyrimidopyridoindolones as the corresponding salts. The reaction in acetone in the presence of K2CO3 yielded 3,6-disubstitution products. Alkylation with DMF dimethyl acetal gave a mixture of the 3- and 6-alkylpyrimidopyridoindol-4-one bases. The structure of 4-oxo-4,6-dihydro-3H-pyrimido-[5′,4′:5,6]pyrido[3,2-b]indol-11-ium chloride (3b) was proved by X-ray diffraction analysis.  相似文献

A novel, two-step, facile route for the synthesis of pyrrolo[2,3-b]quinoxalines via 2,3-dioxopyrroles, enhanced by microwave irradiation, is presented. The newly synthesized 2,3-dioxo-5-halophenyl pyrrolo precursors 4ac as well as the non-aromatized ethyl 2-(4-halophenyl)-1-methyl-2,4-dihydro-1H-pyrrolo[2,3-b]quinoxaline-3-carboxylates 6ac and the aromatized ethyl 2-(4-halophenyl)-1-methyl-1H-pyrrolo[2,3-b]quinoxaline-3-carboxylates 7ac were evaluated for their antioxidant, cytostatic, and antiviral properties. Most of them proved to be potent hydroxyl radical scavengers and inhibited in vitro lipid peroxidation. The compounds showed moderate antiproliferative activity, while 6a inhibited vaccinia virus at an EC50 value of 2 μM, and 4c and 6c inhibited Sindbis virus at EC50 values of 4 μM.  相似文献

Nitration of 2,3-dihydro-1H-imidazo[4,5-b]pyridin-2-one gave its 5-nitro derivative which was subjected to alkylation with dimethyl sulfate, diethyl sulfate, and benzyl(dimethyl)phenylammonium chloride. The resulting 1,3-dimethyl-, 1,3-diethyl-, and 1,3-dibenzyl-5-nitro-2,3-dihydro-1H-imidazo[4,5-b]pyridin-2-ones were reduced to the corresponding 1,3-dialkyl-5-amino-2,3-dihydro-1H-imidazo[4,5-b]pyridin-2-ones, and the latter reacted with itaconic acid to produce 1-(1,3-dialkyl-2-oxo-2,3-dihydro-1H-imidazo[4,5-b]pyridin-5-yl)-5-oxopyrrolidine-3-carboxylic acids. 1-(2-Oxo-2,3-dihydro-1H-imidazo[4,5-b]pyridin-5-yl)-5-oxopyrrolidine-3-carboxylic acid was obtained by analogous reaction with 5-amino-2,3-dihydro-1H-imidazo[4,5-b]-pyridin-2-one.  相似文献

Some chemical transformations of 3,4-bis(4-aminofurazan-3-yl)furoxan (1) and 3,4-bis-(4-nitrofurazan-3-yl)furoxan (2) were considered. Compounds 1 and 2 are valuable synthons for the preparation of linear and cyclic compounds containing the 3,4-bis(furazan-3-yl)furoxan fragment. The reaction of compound 2 with a series of N- and O-nucleophiles afforded novel heterocyclic systems: 7-R-7H-difurazano[3,4-b:3′,4′-f]furoxano[3″,4″-d]azepine and difurazano[3,4-b:3′,4′-f]furoxano[3″,4″-d]oxepin.  相似文献
