Hydrazones 6a–6n were synthesized from different amino acids with various aldehydes under reflux in methanol/ethanol. The structures of synthesized compounds were ascertained by elemental analysis and spectroscopic techniques. A comparative study of the antimicrobial activity and cytotoxicity was carried out of the N‐protected amino acids, their esters, hydrazides, and the respective hydrazones, providing good results in cytotoxicity studies.  相似文献

α-Amino acids protected at nitrogen in quite different ways can be transformed without racemization into the corresponding α-amino aldehydes. Provided one chooses the right protecting groups (e.g., two benzyl residues on nitrogen) it is possible for the first time to carry out Grignard-like, aldol, and Me3SiCN additions, and hetero-Diels–Alder reactions with a high degree of nonchelation control. If the reactions of classical carbanions turn out to be nonselective, transmetalation, for example into organotitanium reagents, constitutes an important tool in controlling stereoselectivity. Diastereoselectivity can be reversed by specific variation of the protecting group and reagent. “Protecting-group tuning”, “metal tuning”, and “ligand tuning” are very useful in reactions of α-amino aldimines as well. The α-amino aldehydes can also be converted by Wittig reactions into electron-poor γ-amino olefins, which in turn undergo stereoselective cuprate, Michael, and cycloadditions.  相似文献

从20种天然氨基酸197个GETAWAY指数经主成分分析得出一种新3D氨基酸描述子——VSGETAWAY[vector of principal component scores for GETAWAY (geometry, topology and atom-weights assembly)]. 将其应用于48个苦味活性二肽、31个血管舒缓激肽促进剂和20个促凝血酶原激酶抑制剂结构表征并以偏最小二乘(PLS)对3个体系建立定量构效关系(QSAR)模型, 得复相关系数(Rcum2)与交互检验复相关系数(Qcum2)分别为0.887和0.753; 0.995和0.708; 0.999和0.802. 研究结果表明, VSGETAWAY描述子操作简便、结构表达能力强, 有望成为多肽药物QSAR研究中一种有效的结构表征方法.  相似文献

The Schiff base ligand 4-methyl-2-pentanone thiosemicarbazone (MPTSC) (HL) has been synthesized by the interaction of 4-methyl-2-pentanone (MP) and thiosemicarbazone (TSC). The Ni(II), Cu(II), and Fe(III) binary complexes of this ligand have been prepared. The ternary complexes of VO(IV) and Mn(II) ions with HL and glutamine (Glu) as a secondary ligand, in addition to VO(IV), Mn(II), and La(III) with HL and glycine (Gly) as a secondary ligand, have also been synthesized. The binary and ternary complexes have been characterized based on elemental analysis, IR, UV-VIS, molar conductance, mass spectra, magnetic moment, and ESR measurements. The magnetic moment, UV, and ESR studies suggest that Ni(II) and Cu(II) complexes are square planar, whereas Fe(III), Mn(II), and La(III) complexes have octahedral geometry, but VO(IV) ternary complexes have square pyramidal geometry. The analytical data indicate that the metal-to-ligand ratio in binary complexes is 1:1, except HL-Cu(II) chloride complex where the metal-to-ligand to secondary ligand ratio in ternary complexes is 1:1:1. The anticancer studies showed that the anticancer activity is in the decreasing order: ternary complexes > binary complexes > free ligand (HL).

从20种天然氨基酸的50个物化性质出发,按照疏水、立体和电性特征将其分类后分别进行主成分分析,并将产生的得分矢量即VHSE(principal component score vector of hydrophilicity,steric,and electronic properties)作为氨基酸结构描述子用于肽的定量构效关系研究。与已有方法相比,VHSE描述子具有物化意义明确、结果更易解释等特点。应用该描述子并结合逐步回归变量筛选和偏最小二乘建模方法,在对苦味二肽和血管舒缓激肽促进剂等体系的定量构效关系研究中,均取得了优于已有文献的结果。  相似文献

One-step synthesis of substituted hydantoins can be achieved by the palladium-catalyzed “ureidocarbonylation” of aldehydes with urea derivatives and carbon monoxide [Eq. (1)]. This surprisingly selective protocol converts substituted ureas into 1,5- and 1,3,5-substituted hydantoins in yields of up to 93 %.  相似文献

A triazene-based synthetic strategy for the construction of the complex biaryl ethers and a Suzuki coupling reaction were the key steps in the synthesis of precursor 1 of the aglycon of vancomycin, which already contains the complete skeleton of the target compound. The cleavage of the triazene unit from the D ring and the removal of the other protecting groups led to the aglycon of vancomycin. These strategies should be particularly valuable for the synthesis of other naturally occurring glycopeptide antibiotics and offer opportunities for the synthesis of combinatorial libraries of compounds of the vancomycin family for chemical biology studies.  相似文献

Two activations are better than one : The chiral bifunctional guanidine 1 , which features an amino amide backbone, catalyzes the asymmetric Michael addition of a range of substrates and gives products with excellent stereoselectivities. The method also allows the efficient synthesis of optically pure β‐amino acid esters. Both the guanidine group and the NH proton of the amide were important for the dual activation.

Densities and apparent molar volumes of aqueous 2-chloroethanol were determined at temperatures from 15.0 to 34.4°C using digital densimetry. The results of the volumetric measurements have been used to calculate the following thermodynamic quantites at 25°C: V 2 o = 55.05 ± 0.02 cm3-mol–1, (V 2 o/T)p = 0.01486 ± 0.00318 cm3-K–1-mol–1, and (2 V 2 o/T 2)p = 0.02972 ± 0.00318 cm3-K–2-mol–1. Partial molar volumes of transfer from water to 1 mol-dm–3 2-chloroethanol have also been determined for L-glycine, L-alanine, L-serine, L-glutamic acid, and L-aspartic acid at 35.0°C. The transfer results have been explained in terms of the nature of the interactions of the groups in the solute and solvent. Hydration numbers of L-glycine and L-alanine have also been calculated in aqueous 2-chloroethanol.  相似文献

从天然氨基酸的25个结构与拓扑变量中经主成分分析得到一种新的氨基酸描述子——VSTV (principal component scores vector of structural and topological variables).应用该描述子对以下3个体系,即血管紧张素转化酶抑制剂(2肽)、抗菌18肽和促凝血酶原激酶抑制剂(6~12肽)进行分子结构参数化表达,并在此基础上通过偏最小二乘回归(PLSR)建立定量构效关系(QSAR)模型,取得了优于文献的结果.模型的复相关系数(R2)和交互检验复相关系数(Q2)分别为0.789, 0.767; 0.996, 0.879; 0.981, 0.480.  相似文献

A concise approach for the transformation of various S-amino acids into the 5-alkyl-3-phenyl-2-thioxoimidazolidin-4-one heterocycles using phenylisothiocyanate is described. Syntheses of the benzyl or the trityl protected α- and β-anomers of vinyl sulfone-modified pent-2-enofuranosides have been initiated by the ring opening of the suitably masked methyl α-lyxofuranosyl-epoxide or methyl β-ribofuranosyl-epoxide or by the nucleophilic displacement of the leaving groups in benzyl protected 3-O-tosyl xylofuranoside and 3-O-mesyl ribofuranoside by p-thiocresol. In case of the latter set of starting materials, α- and β-methyl glycosides formed in almost equal ratio only from the derivatives of d-xylose. For the synthesis of α- and β-anomers of vinyl sulfone-modified hex-2-enopyranosides, a d-glucose derivative was selected over a d-allose derivative as the starting material because the former almost exclusively produced the required methyl pyranosides whereas the latter produced a mixture. All sulfides were converted to vinyl sulfone-modified carbohydrates by the sequential application of oxidation, mesylation and base induced elimination reactions.  相似文献

The synthesis of bolaamphiphiles from unusual β‐amino acids or an alcohol and C12 or C20 spacers is described. Unusual β‐amino acids such as a sugar amino acid, an AZT‐derived amino acid, a norbornene amino acid, and an AZT‐derived amino alcohol were coupled with spacers under standard conditions to get the novel bolaamphiphiles 5 – 8 (Scheme 1), 12 and 13 (Scheme 2), and 17 and 20 (Scheme 3). Some of these compounds, on precipitation from MeOH/H2O, self‐assembled into organized molecular structures.  相似文献

Various substituted homotaurines (=3‐aminopropane‐1‐sulfonic acids) 6 were readily synthesized in satisfactory to good yields via the Michael addition of thioacetic acid to alk‐2‐enamides 3 (→ 4 ), followed by LiAlH4 reduction (→ 5 ) and performic acid oxidation (Scheme 1). The configuration of ‘anti’‐disubstituted homotaurine ‘anti’‐ 6h was deduced from the 3‐(acetylthio)alkanamide (=S‐(3‐amino‐1,2‐dimethyl‐3‐oxopropyl) ethanethioate)‘anti’‐ 4h formed in the Michael addition, which was identified via the Karplus equation analysis, and confirmed by X‐ray diffraction analysis. The current route is an efficient method to synthesize diverse substituted homotaurines, including 1‐, 2‐, and N‐monosubstituted, as well as 1,2‐, 1,N‐, 2,N‐, and N,N‐disubstituted homotaurines (Table).  相似文献
