This paper describes selected modification and structure-activity relationship of the small molecule HIV-1 inhibitor, 4-benzoyl-1-[(4-methoxy-1H-pyrrolo[2,3-b]pyridin-3-yl)oxoacetyl]-2-(R)-methylpiperazine (BMS-378806). The results revealed: i) that both the presence and configuration (R vs. S) of the 3-methyl group on the piperazine moiety are important for the antiviral activity, with the 3-(R)-methyl derivatives showing the highest activity; ii) that the electronegativity of the C-4 substituent on the indole or azaindole ring seems to be important for the activity, with a small, electron-donating group such as a fluoro or a methoxy group showing enhanced activity, while a nitro group diminishes the activity; iii) that the N-1 position of the indole ring is not eligible for modification without losing activity; and iv) that bulky groups around the C-4 position of the indole or azaindole ring diminish the activity, probably due to steric hindrance in the binding. We found that a synthetic bivalent compound with two BMS-378806 moieties being tethered by a spacer demonstrated about 5-fold enhanced activity in an nM range against HIV-1 infection than the corresponding monomeric inhibitor. But the polyacrylamide-based polyvalent compounds did not show inhibitory activity at up to 200 nM.  相似文献

A quartz crystal microbalance has been proposed for studies on the temperature dependence of the linear rate of a reaction interface advance in topochemical reactions of the thermal decomposition of solids. A quartz crystal microbalance has been used in investigations of the CuSO4 · 5H2O dehydration. The data agree fairly well with those available in the literature. Advantages and disadvantages of the method proposed are discussed.  相似文献

An investigation into the evaporation of sessile droplets of latex and clay particle suspensions is presented in this work. The quartz crystal microbalance (QCM) has been used to study the interfacial phenomena during the drying process of these droplets. Characteristic changes of the crystal oscillating frequency and crystal resistance (damping of the oscillating energy) have been observed and related to the different stages of the evaporation process. Measurements have been made for latex particle sizes from 1.9 to 10 microm and for rough and polished crystals using drops from 0.3 to 1.5 microL. The behavior of the QCM is shown to depend strongly on the size of particles present and on the morphology of the crystal surface. One of the most striking features is a drastic damping of the oscillation energy and corresponding rise in frequency observed during the final stages of evaporation, particularly for the clay suspensions.  相似文献

In this paper we report a method for the determination of 4-aminophenol (4-AP) in solution using a quartz crystal microbalance (QCM) sensor. 4-AP reacts with (para-unsubstituted) phenols to form hydrophobic indophenol dye species that precipitate out and adsorb to the surface of the crystal to produce a shift in the crystal resonant frequency. This frequency change, due to in-situ indophenol mass adsorption, can be related to the initial 4-AP concentration. A range of phenols (namely o-cresol, 1-naphthol, resorcinol, catechol and 8-hydroxyquinoline) and their reaction with 4-AP were tested. Ammonium persulfate (APS) and potassium periodate were used as initiators to improve the speed of the reaction and the rate of formation of the precipitate. APS elicited improved signal in terms of response times and frequency shifts compared with KIO4. Of the phenols studied, resorcinol gave the best response time of 6 min for 4-AP determination. The reaction of resorcinol with 4-AP gave extended response times and signal size with decreasing concentration of 4-AP (in the range 2-5 mM).  相似文献

The selective proteolytic activation of the HIV-1 envelope glycoprotein gp160 by furin and other precursor convertases (PCs) occurs at the carboxyl side of the sequence Arg508-Glu-Lys-Arg511 (site 1), in spite of the presence of another consensus sequence: Lys500-Ala-Lys-Arg503 (site 2). We report on the solution structural analysis of a 19-residue synthetic peptide, p498, which spans the two gp160-processing sites 1 and 2, and is properly digested by furin at site 1. A molecular model is obtained for p498, by means of molecular dynamics simulations, from NMR data collected in trifluoroethanol/water. The peptide N-terminal side presents a 9-residue helical segment, enclosing the processing site 2; the C-terminal segment can be described as a loop exposing the processing site 1. A hypothesis for the docking of p498 onto the catalytic domain of human furin, modeled by homology and fitting previous site-directed mutagenesis studies, is also presented. p498 site 1 is shown to have easy access to the furin catalytic site, unlike the nonphysiological site 2. Finally, on the basis of available data, we suggest a possible structural motif required for the gp160-PCs recognition.  相似文献

Protein imprinting leading to enhanced rebinding of ferritin to ternary lipid monolayers is demonstrated using a quartz crystal microbalance. Monolayers consisting of cationic dioctadecyldimethylammonium bromide, non-ionic methyl stearate, and poly(ethylene glycol) bearing phospholipids were imprinted with ferritin at the air/water interface of a Langmuir-Blodgett trough and transferred hydrated to hydrophobic substrates for study. This immobilization was shown by fluorescence correlation spectroscopy to significantly hinder any further diffusion of lipids, while rebinding studies demonstrated up to a six-fold increase in ferritin adsorption to imprinted versus control monolayers. A diminished rebinding of ferritin to its imprint was observed through pH reduction to below the protein isoelectric point, demonstrating the electrostatic nature of the interaction. Rebinding to films where imprint pockets remained occupied by the template protein was also minimal. Studies with a smaller acidic protein revealed the importance of the steric influence of poly(ethylene glycol) in forming the protein binding pockets, as albumin-imprinted monolayers showed low binding of ferritin, while ferritin-imprinted monolayers readily accommodated albumin. The controllable structure-function relationship and limitations of this system are discussed with respect to the application of protein imprinting in sensor development as well as fundamental studies of proteins at dynamic interfaces.  相似文献

Recent reports using particle image velocimetry and cone-and-plate rheometers have suggested that a simple Newtonian liquid flowing across a superhydrophobic surface demonstrates a finite slip length. Slippage on a superhydrophobic surface indicates that the combination of topography and hydrophobicity may have consequences for the coupling at the solid--liquid interface observed using the high-frequency shear-mode oscillation of a quartz crystal microbalance (QCM). In this work, we report on the response of a 5 MHz QCM possessing a superhydrophobic surface to immersion in water--glycerol mixtures. QCM surfaces were prepared with a layer of SU-8 photoresist and lithographically patterned to produce square arrays of 5 mum diameter circular cross-section posts spaced 10 microm center-to-center and with heights of 5, 10, 15, and 18 microm. Non-patterned layers were also created for comparison, and both non-hydrophobized and chemically hydrophobized surfaces were investigated. Contact angle measurements confirmed that the hydrophobized post surfaces were superhydrophobic. QCM measurements in water before and after applying pressure to force a Cassie-Baxter (non-penetrating) to Wenzel (penetrating) conversion of state showed a larger frequency decrease and higher dissipation in the Wenzel state. QCM resonance spectra were fitted to a Butterworth-van Dyke model for the full range of water-glycerol mixtures from pure water to (nominally) pure glycerol, thus providing data on both energy storage and dissipation. The data obtained for the post surfaces show a variety of types of behavior, indicating the importance of the surface chemistry in determining the response of the quartz crystal resonance, particularly on topographically structured surfaces; data for hydrophobized post surfaces imply a decoupling of the surface oscillation from the mixtures. In the case of the 15 microm tall hydrophobized post surfaces, crystal resonance spectra become narrower as the viscosity-density product increases, which is contrary to the usual behavior. In the most extreme case of the 18 microm tall hydrophobized post surfaces, both the frequency decrease and bandwidth increase of the resonance spectra are significantly lower than that predicted by the Kanazawa and Gordon model, thus implying a decoupling of the oscillating surface from the liquid, which can be interpreted as interfacial slip.  相似文献

The electrochemical quartz crystal microbalance (EQCMB) method has been used to evaluate the processes which occur in/on the palladium electrode in basic solutions. Hydrogen electrosorption in palladium is accompanied by an additional frequency shift that can be attributed to the stresses generated inside the Pd metal. A non-linear dependence between the mass change and the charge consumed during hydrogen oxidation in the Pd electrode has been found for hydrogen absorbed in the α- and β-phases. This effect precludes the objective estimation of the amount of hydrogen absorbed inside the Pd electrode. The EQCMB method has been used, however, for studying the surface electrode processes on the Pd electrode, i.e. specific anion adsorption, surface oxidation and dissolution. Also, the structure of the palladium oxide formed on the Pd surface during electrochemical oxidation is discussed in this paper and the effect of the anodic limiting potential on the oxide structure is reported. The electrochemical quartz crystal microbalance (EQCMB) method has been used to study the processes of hydrogen absorption/desorption in Pd-Ni alloy electrodes. It was found that hydrogen electrosorption is accompanied by an additional frequency shift, attributed to the stresses generated inside the alloy. The influence of stresses on the EQCMB response depends on the amount of absorbed hydrogen and the alloy composition. From the comparison of the EQCMB results with Pd-Ni alloy absorption capabilities data, it was concluded that the decrease of the hydrogen sorption capacity at a Ni content of ca. 25–30 at% is due to an excessive generation of stresses in the alloy lattice. The electrochemical preparation of copper layers was investigated using a quartz crystal microbalance (QCM) with damping monitoring. The damping increase during the deposition could be separated into two contributions arising from an internal and an external friction process. External friction comes from the coupling of the shear motion of the rough metal surface to the viscous liquid. An application of the oscillating quartz crystal microbalance (QCM) in the measurements of sorption rates and equilibria in polymer—vapor systems is presented. The sorption isotherms of methanol, ethanol and n-heptane in cellulose acetate and poly(hexamethylene adipamide) were measured with the QCM at 298K. The results agree reasonably with the Flory model except at low absorbate activities where appreciable deviations due to localized absorption were observed, as verified with IR spectroscopy. The diffusion coefficient of methanol in cellulose acetate was also calculated from the sorption rate data measured with the QCM. The values obtained at different methanol concentrations correlate well with the published diffusion coefficients.  相似文献

Acquired immune deficiency syndrome (AIDS) has prevailed over the last 30 years. Although highly active antiretroviral therapy (HAART) has decreased mortality and efficiently controlled the progression of disease, no vaccine or curative drugs have been approved until now. A viral inactivator is expected to inactivate cell-free virions in the absence of target cells. Previously, we identified a gp120-binding protein, mD1.22, which can inactivate laboratory-adapted HIV-1. In this study, we have found that the gp41 N-terminal heptad repeat (NHR)-binding antibody D5 single-chain variable fragment (scFv) alone cannot inactivate HIV-1 at the high concentration tested. However, D5 scFv in the combination could enhance inactivation activity of mD1.22 against divergent HIV-1 strains, including HIV-1 laboratory-adapted strains, primary HIV-1 isolates, T20- and AZT-resistant strains, and LRA-reactivated virions. Combining mD1.22 and D5 scFv exhibited synergistic effect on inhibition of infection by divergent HIV-1 strains. These results suggest good potential to develop the strategy of combining a gp120-binding protein and a gp41-binding antibody for the treatment of HIV-1 infection.  相似文献

The quartz crystal microbalance (QCM) was used to study the variability of acoustic properties of living cells on the sub-second time scale. A confluent cell layer of rat cardiac myocytes was grown onto the electrode of quartz crystal resonator. The cell layer performed periodic, synchronous contractions at a rate of about 1.5 Hz. In order to monitor these rather fast changes in the state of the cells, the QCM was operated in a "fast mode", which allows sampling of the shift of the resonance frequency and energy dissipation with a rate of up to 100 Hz. The contractions were clearly reflected in periodic variations of the resonance frequency and the bandwidth. The rate of the contractions, in particular, could be easily detected in this way. Building on the rate of contraction, the setup can be used to monitor the response of the cell layer to heart stimulating drugs like isoproterenol. Depending on the concentration of isoproterenol, the beat rate was found to increase by up to a factor of two.  相似文献

The preparation of a nonporous solid surface for native chemical ligation is described. A cysteine residue is covalently attached to the surface by means of a series of reactions. In a reaction analogous to that used for native chemical ligation, the surface‐attached cysteine residue reacts with a thioester to form an amide linkage. All of the reaction steps except the derivatization of the nonporous solid surface with amino‐ended silane are conducted within the flow cell of a quartz crystal microbalance with dissipation monitoring. This sensitive instrument allows each reaction step to be followed in real time, with simultaneous quantification of the mass added and removed in different steps. The number of protected cysteine residues attached per square nanometer is consistent with the number of protecting groups removed in each deprotection step and also with the number of thioesters reacting with the deprotected cysteine. The surface processes at carbon and platinum electrodes have been studied using the electrochemical quartz crystal microbalance technique in organic electrolyte solutions for lithium ion batteries. The changes in resonance frequency were analyzed as a function of the electrode potential, indicating that the process depended not only on the electrode material but also on the cathode potential. In the solution containing LiBF4 as the electrolyte, the main product at the platinum surface was Li2CO3 and LiF, whereas formation of lithium alkylcarbonates was the primary process at the platinum and carbon electrodes in LiPF6 solution.  相似文献

Concerning the redeposition of particulate soils in the detergent process, particle deposition onto substrates in aqueous solutions was investigated by the application of the quartz crystal microbalance (QCM) technique, and the effects of the kinds of the particle and substrate and the addition of ethanol were discussed by the extended DLVO theory. The film of polyethylene, Nylon 6 or cellulose acetate as a substrate was prepared on the gold electrode of the QCM by a spin-coating method. The electrode with or without the polymer film was perpendicularly immersed in the aqueous dispersion of spherical polyethylene or nylon particles. The total mass of particles deposited onto the electrode was determined, in situ, from frequency change of the QCM. The deposited mass was also determined from the difference in frequency measured in air before and after the immersion in the dispersion. In both cases, the particle deposition increased with immersion time and attained apparent equilibrium after 30–60 min. Apparent equilibrium deposition was large for the polyethylene particle compared with the Nylon 12 particle. For either particle, a considerable difference in the deposited amount was observed among the substrates. In all systems, the particle deposition drastically decreased by addition of ethanol to the aqueous dispersion. The results were discussed in terms of the electrical double layer, the Lifshitz–van der Waals and acid–base interactions between the particle and the substrate, which were calculated using the experimentally determined surface free energy components and electrokinetic potentials.  相似文献

A new quartz crystal microbalance sensor is developed to determine formic acid at low concentrations. Four previously selected polymers with acid-base characteristics were tested as possible coatings. Polyoxyethylene bis [amine] presented the best results. The sensor is rapid, sensitive [0.67 Hz/(mg/m3)], and reversible at low concentrations. The detection limit for formic acid (7.2 mg/m3) is comparable with the short term exposure limit and the threshold limit values. It presents a fast mechanical response to pressure changes, so that it can be quickly used in different environments and situations. The sensor also shows a good stability in a temperature range typical of work atmospheres (16-36 degrees C). It has a wide linear range (7.2-911.2 mg/m3) and a long useful time. It is also applicable to other low molecular mass carboxylic acids such as acetic acid.  相似文献

A new sensor based on oxidative combustion of nitroaromatics to NO2 and its detection with a quartz crystal microbalance (QCM) coated with copper phthalocyanine (CuPc) was developed for determination of nitroaromatics in landfill gas. An alternative method based on gas chromatography–mass spectrometry (GC–MS) was also used in order to assess the performance of the proposed method. The results show that the analytical apparatus based on the QCM is less expensive than the GC–MS, and that the analytical error is 0.8% for both methods.  相似文献

The effect of the immersion angle on the overtone mode of the quartz crystal ‘microbalance’ (QCM, piezoelectric quartz crystal) was investigated in a Newtonian liquid. The measurement using the impedance analyzer revealed that the resonant frequency shift of the Nth overtone mode of the QCM was depend on the immersion angle, and, at each immersion angle, was times larger than that of the fundamental mode. These tendencies were also observed in the resistance of the Butterworth-Van Dyke equivalent circuit. Furthermore, on the basis of these results, we have discussed the immersion angle dependence of the resonant frequency shift and the resistance.  相似文献
