Prostate-specific antigen (PSA) is a serum glycoprotein overproduced in prostate cancer, the total of which is comprised of two major forms, free and complexed. The common method for measuring of PSA is an Enzyme-linked immunosorbent assay (ELISA). Limits of detection using commercial PSA ELISA kits for free and total PSA were determined in our laboratory to be 1 and 10 ng/mL, respectively. A value of 0.10 was obtained for the free to total PSA ratio, a ratio of 0.25 being indicative of prostate cancer. A possible improvement in the sensitivity and detection limits of free and total PSA may be achieved by the ALYGNSA system (Clarizia et al., Anal Bioanal Chem 393:1531–1538, 2009). This fluorescent assay system employed a selective polymer biolinker system proven to enhance primary antibody orientation. Using this system, free and total PSA detection limits of 0.13 and 0.63 ng/mL, respectively, were realized. This amounted to an 8- and 15-fold improvement in detection limits for free and total PSA, respectively. A free to total PSA ratio of 0.20 was maintained in this study and may be useful for definitive diagnosis of prostate, as well as, other cancers.  相似文献

A simple and portable electrochemical immunosensor for the detection of total prostate specific antigen (t‐PSA) in human serum was developed using a double‐layer nanogold particles and dendrimer‐functionalized polyvinyl chloride (PVC) membrane as immunosensing interface. To fabricate such a multifunctional PVC electrode, an o‐phenylenediaminedoped PVC membrane was initially constructed, then nanogold particles and poly(amidoamine) G4‐dendrimer with a sandwich‐type format were assembled onto the PVC membrane surface, and then t‐PSA antibodies (anti‐PSA) were adsorbed on the nanogold surface. The detection principle of the immunosensor is based on the change in the electric potential before and after the antigen‐antibody interaction. The experimental conditions and the factors influencing the performance of the immunosensor were investigated. Under optimal conditions, the proposed immunosensor exhibits good electrochemical behavior in the dynamic range of 0.5–18 ng/mL relative to t‐PSA concentration with a relative low detection limit of 0.1 ng/mL (S/N=3). The precision, reproducibility, and stability of the immunosensor are acceptable. In addition, 43 serum specimens were assayed by the as‐prepared immunosensor, and consistent results were obtained in comparison with those obtained by the standard enzyme‐linked immunosorbent assay (ELISA). Compared with the conventional ELISAs, the developed immunoassay system was simple and rapid without labeling and separation steps. Importantly, the immobilization and detection methodologies could be extended for the immobilization and detection of other biomarkers.  相似文献

The BioCD is a spinning-disc interferometric biosensor on which antibodies are immobilized to capture target antigens from biological samples. In this work, BioCDs measured the interferometric response to prostate-specific antigen (PSA). The ideal detection limit for PSA was determined using a BioCD with 12,500 printed target antibody spots with a corresponding number of reference protein spots. Statistical analysis projects the detection limit of PSA as a function of the number of spots included in the average. When approximately 10,000 spot pairs were averaged, the 3σ detection limit was 60 pg/ml in a 2 mg/ml simple protein background. A standard format for BioCD immunoassays uses 96 wells with 32 target spots paired with reference spots. In serum, the detection limit for this format was 1 ng/ml in 3:1 diluted female human serum using a sandwich assay with a nonfluorescent mass tag.  相似文献

An enzyme and antibody dual labeled gold nanoparticles enhancing chemiluminescence strategy was developed for highly sensitive CE immunoassay (IA) of prostate‐specific antigen (PSA). In this work, gold nanoparticles were labeled with horseradish peroxidase and antiprostate specific antigen‐antibody, and used as the marker (Ab*). After PSA (antigen, Ag) was added into the system, a noncompetitive immune reaction was happen between Ab* and Ag to form an immune complex (Ag–Ab*). Subsequently, the obtained Ag–Ab* and unreacted Ab* were separated by CE, and the chemiluminescence intensity of Ag‐Ab* was used to estimate PSA concentration. The calibration curve showed a good linearity in the range of 0.25–10 ng/mL. Based on a S/N of 3, the detection limit for PAS was estimated to be 0.092 ng/mL. Proposed CE method was applied for PSA quantification in human serum samples from healthy volunteers and patients with prostate cancer. The obtained results demonstrated that the proposed CE method may serve as an alternative tool for clinical analysis of PSA.  相似文献

Cancer antigen 125 (CA-125) is a glycoprotein biomarker that denotes the presence of ovarian and reproductive cancers in women, with serum concentrations of CA-125 greater than 35 U/ml considered indicative of potential malignancies. A fluorescent immunoassay recently developed in our laboratory employing the ALYGNSA antibody-orientation system has been used to measure CA-125 levels. This system displayed significantly increased sensitivity with a detection limit of 1.5 U/ml compared to that of a commercial CA-125 enzyme-linked immunosorbent assay (15 U/ml) This tenfold lower level of detection of the ALYGNSA CA-125 assay should permit better identification and monitoring of ovarian cancer.  相似文献

Zearalenone, a mycotoxin biosynthesized by various Fusarium fungi, is widely found as a contaminant in grains and animal feeds. This study describes a rapid and sensitive LC/MS/MS assay method for the quantification of zearalenone in rat serum. The assay was validated to demonstrate the specificity, linearity, recovery, lower limit of quantification (LLOQ), accuracy and precision. The multiple reaction monitoring was based on the transition of m/z 317.0 → 130.9 for zearalenone and 319.0 → 204.8 for zearalanone (internal standard). The assay utilized a single liquid–liquid extraction with t‐butyl methyl ether and isocratic elution, and the LLOQ was 0.5 ng/mL using 0.1 mL rat serum. The assay was linear over a concentration range from 0.5 to 200 ng/mL, with correlation coefficients >0.9996. The mean intra‐ and inter‐day assay accuracy was 101.2–112.9 and 96.3–108.0%, respectively. The mean intra‐ and inter‐day precision was between 1.3–7.6 and 3.6–10.6%, respectively. Screening of Prostate-specific antigen (PSA) in human blood is the most common approach to diagnose prostate cancer. The joint application of biology and electrochemistry has shown a tremendous rise in research towards the development of electrochemical diagnostic tools for various diseases. The present study demonstrates the development of an effective immunosensing platform incorporating hydroquinone (HQ) immobilized, fullerene-C60 and copper nanoparticles (CuNPs) composite film on glassy carbon electrode (HQ@CuNPs-reduced-fullerene-C60/GCE) for the selective, quick and trace detection of PSA. In order to fabricate immunosensor sequential immobilization of primary antibody (Ab1), blocking agent (bovine serum albumin (BSA)), antigen (prostate-specific antigen (PSA)) and secondary antibody (Ab2) tagged with horseradish peroxide (HRP) was carried out on HQ@CuNPs-reduced-fullerene-C60/GCE. Electrochemical characterization and the signal response of immunosensor were tested using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS). Due to the synergetic effect of fullerene-C60 and CuNPs, the novel nanocomposite film exhibited excellent catalytic activity towards hydrogen peroxide (H2O2) reduction for greatly amplified immunosensing signals. HQ@CuNPs-fullerene-C60/GCE exhibited a well-defined redox peak and accelerated electrochemical reduction of H2O2 without any interference of dissolved oxygen and false-positive result in phosphate buffer solution (PBS) at pH 7.0. The parameters influencing the electrochemical response were optimized. Under the optimized conditions, wide linearity between PSA concentrations and current responses ranging from 0.005 ng/mL to 20 ng/mL with the lower detection limit of 0.002 ng/mL was obtained at the proposed immunosensor. The clinical applicability of the proposed immunosensor was successfully tested in serum and urine samples. Results revealed that the proposed immunosensor may create new boundaries in the identification of PSA in human blood samples.  相似文献

Influenza A virus, both seasonal and pandemic, has the potential to cause rampant devastating disease around the world. The most relied upon methods of viral detection require days, skilled workers, and laboratory settings to complete properly. Here, we report two methods for the detection of the nucleoprotein from inactivated influenza A (IFA-NP), a patented polymer–protein antibody orientation immuno-method, termed ALYNGSA, and a newly fabricated optical label-free Fabry–Perot interferometric immunosensor. The ALYGNSA assay for IFA-NP had a level of detection below 5 μg/mL of inactivated virus sample. The label-free detection through Fabry–Perot interferometry with the ALYGNSA orientation yielded an improved sensitivity to 1 μg/mL over the fluorescence sandwich assay alone. Characterization of the detection surface by fluorescence microscopy and non-contact AFM corroborated interferometry results. The resulting label-free detection method has the prospective for adaptation into a portable multi-chip sensor capable of real-time in situ detection of influenza A virus.  相似文献

A fluorescence polarization (FP) immunoassay, based on a monoclonal antibody and an ochratoxin A (OTA)-fluorescein tracer, has been developed for rapid screening of OTA in red wine. Wine samples were diluted with methanol and passed through aminopropyl solid-phase extraction columns prior to the FP assay. Average recoveries from samples spiked with OTA at levels of 2.0 and 5.0 ng/mL were 79% with RDS of 11% (n = 6). The limit of detection of the FP immunoassay was 0.7 ng/mL OTA, and the whole analysis was performed in less than 10 min. The assay was tested on 154 red wine samples (naturally contaminated or spiked at level ranging from 0.1 to 5.0 ng/mL) and compared with an high-performance liquid chromatography/immunoaffinity column clean-up method, showing a good correlation (r = 0.9222). Their compliance with the European regulation (2.0 ng/mL OTA maximum permitted level) was correctly assessed for 70% of the analyzed samples of red wine, whereas confirmatory analyses were required for the remaining ones with OTA levels close to the regulatory limit. No false-negative or positive results were observed using the FP immunoassay. The proposed FP assay is a useful screening method for OTA in red wines, when high throughput is required, that could also be used for white and rosé wines, which are known to contain less interfering compounds such as polyphenols.  相似文献

A novel electrochemical immunosensor using functionalized silica nanoparticles (Si NPs) as protein tracer has been developed for the detection of prostate specific antigen (PSA) in human serum. The immunosensor was carried out based on a heterogeneous sandwich procedure. The PSA capture antibody was immobilized on the gold electrode via glutaraldehyde crosslink. After reaction with the antigen in human serum, Si NPs colabeled with detection antibody and alkaline phosphatase (ALP) was sandwiched to form the immunocomplex on the gold electrode. ALP carried by Si NPs convert nonelectroactive substrate into the reducing agent and the latter, in turn, reduce metal ions to form electroactive metallic product on the electrode. Linear sweep voltammetry (LSV) was used to quantify the amount of the deposited silver and give the analytical signal for PSA. The parameters including the concentration of the ALP used to functionalize the Si NPs and the enzyme catalytic reaction time have been studied in detail and optimized. Under the optimum conditions of immunoreaction and electrochemical detection, the electrochemical immunosensor was able to realize a reliable determination of PSA in the range of 1–35 ng/mL with a detection limit of 0.76 ng/mL. For six human serum samples, the results performed with the electrochemical immunosensor were in good agreement with those obtained by chemiluminescent microparticle immunoassay (CMIA), indicating that the electrochemical immunosensor could satisfy the need of practical sample detection.  相似文献

We describe herein the combination of electrochemical immunosensors using single-wall carbon nanotube (SWNT) forest platforms with multi-label secondary antibody-nanotube bioconjugates for highly sensitive detection of a cancer biomarker in serum and tissue lysates. Greatly amplified sensitivity was attained by using bioconjugates featuring horseradish peroxidase (HRP) labels and secondary antibodies (Ab(2)) linked to carbon nanotubes (CNT) at high HRP/Ab(2) ratio. This approach provided a detection limit of 4 pg mL(-)(1) (100 amol mL(-)(1)), for prostate specific antigen (PSA) in 10 microL of undiluted calf serum, a mass detection limit of 40 fg. Accurate detection of PSA in human serum samples was demonstrated by comparison to standard ELISA assays. PSA was quantitatively measured in prostate tissue samples for which PSA could not be differentiated by the gold standard immunohistochemical staining method. These easily fabricated SWNT immunosensors show excellent promise for clinical screening of cancer biomarkers and point-of-care diagnostics.  相似文献

Ferruginol, a diterpene phenol, has recently received attention for its extensive pharmacological properties, including anti‐tumor, antibacterial, cardio‐protective and gastroprotective effects. In the present study, a high‐performance liquid chromatographic (HPLC) method was developed for determination of ferruginol in rat plasma and applied for the pharmacokinetics study. The HPLC assay was performed with a VP ODS‐C18 column. The mobile phaseconsisted of methanol and 1% acetic acid solution (90:10, v/v). The flow rate was 1.0 mL/min, and the wavelength was set at 270 nm. This method was linear over the studied range of 0.1–10.0 µg/mL for ferruginol. The correlation coefficient was 0.9998. The intra‐day and inter‐day precisions were better than 4 and 5%, respectively. The extraction recovery and accuracy were greater than 97 and 96%, respectively. The detection limit was 30 ng/mL. The mean maximum concentration of ferruginol in rat plasma was 3.14 µg/mL at 40 min after oral administration at a dose of 20 mg/kg. Ferruginol was absorbed quickly p.o. with t1/2ka = 14.86 min and had a high rate of elimination with t1/2 = 41.73 min. Electrochemical detection method allowing to detect prostate-specific antigen (PSA), a biomarker of prostate cancer (PCa), with PSA glycoprofiling was applied in an analysis of PCa serum samples for the first time. Electrochemical impedance spectroscopy (EIS) as a label-free method with immobilized anti-PSA was applied for PSA detection and lectins to glycoprofile captured PSA on the same surface. A proper choice of blocking agent providing high selectivity of biosensor detection with the immobilized anti-PSA antibody was done. The biosensor could detect PSA down to 100 ag/mL with a linear concentration working range from 100 ag/mL up to 1 μg/mL, i.e. 10 orders of concentration magnitude and the sensitivity of (5.5 ± 0.2)%/decade. The results showed that a commercial carbo-free blocking solution was the best one, reducing non-specific binding 55-fold when compared to the immunosensor surface without any blocking agent applied, while allowing to detect PSA. The biosensor response obtained after addition of lectin (i.e. proportional to the amount of a particular glycan on PSA) divided by the biosensor response obtained after incubation with a sample (i.e. proportional to the PSA level in the sample) was applied to distinguish serum samples of PCa patients from those of healthy individuals. The results showed that Maackia amurensis agglutinin (MAA) recognizing α-2,3-terminal sialic acid can be applied to distinguish between these two sets of samples since the MAA/PSA response obtained from the analysis of the PCa samples was significantly higher (5.3-fold) compared to the MAA/PSA response obtained by the analysis of samples from healthy individuals. Thus, combined analysis of serological PSA levels together with PSA glycoprofiling of aberrant glycosylation of PSA (i.e. increase in the level of α-2,3-terminal sialic acid) has a potential to improve detection of PCa.  相似文献

A simple and cost‐effective HPLC method was established for quantification of 5‐hydroxyeicosatetraenoic acid (5‐HETE) in human lung cancer tissues. 5‐HETE from 27 patients' lung cancer tissues were extracted by solid‐phase extraction and analyzed on a Waters Symmetry C18 column (4.6 × 250 mm, 5 µm) with a mobile phase consisting of methanol, 10 mm ammonium acetate, and 1 m acetic acid (70:30:0.1, v:v:v) at a flow rate of 1.0 mL/min. The UV detection wavelength was set at 240 nm. The calibration curve was linear within the concentration range from 10 to 1000 ng/mL (r2 > 0.999, n = 7), the limit of detection was 1.0 ng/mL and the limit of quantitation was 10.0 ng/mL for a 100 µL injection. The relative error (%) for intra‐day accuracy was from 93.14 to 112.50% and the RSD (%) for intra‐day precision was from 0.21 to 2.60% over the concentration range 10–1000 ng/mL. By applying this method, amounts of 5‐HETE were quantitated in human lung cancer tissues from 27 human subjects. The established HPLC method was validated to be a simple, reliable and cost‐effective procedure that can be applied to conduct translational characterization of 5‐HETE in human lung cancer tissues. Gold nanorods (AuNRs) integrated with ZnCdHgSe near-infrared quantum dots (AuNRs-ZnCdHgSe QDs) were successfully synthesized and characterized by transmission electron microscope, X-ray photoelectron spectroscopy, and X-ray diffraction. A glassy carbon electrode was decorated with the aforementioned AuNRs-ZnCdHgSe QDs nanocomposite, which provides a biocompatible interface for the subsequent immobilization of prostate specific antibody (anti-PSA). After being successively treated with glutaraldehyde vapor and bovine serum albumin solution, a photoelectrochemical immunosensing platform based on anti-PSA/AuNRs-ZnCdHgSe QDs/GCE was established. The photocurrent response of ZnCdHgSe QDs was tremendously improved by AuNRs due to the effect of resonance energy transfer which can be deduced from the dependence of the enhanced efficiency on the AuNRs with different length-to-diameter ratios and spectral absorption characteristics. A maximum photocurrent was obtained when the absorption spectrum of AuNRs matched well with the emission spectrum of ZnCdHgSe QDs. A photoelectrochemical immunosensor for prostate specific antigen (PSA) was achieved by monitoring the photocurrent variation. The photocurrent variation before and after being interacted with PSA solution exhibits a good linear relationship with the logarithm of its concentration (logcPSA) in the range from 1.0 pg mL−1 to 50.0 ng mL−1. The detection limit of this photoelectrochemical immunosensor is able to reach 0.1 pg mL−1 (S/N = 3). Determining PSA in clinical human serum was also demonstrated by using the developed anti-PSA(BSA)/AuNRs-ZnCdHgSe QDs/GCE electrode. The results were comparable with those obtained from an enzyme-linked immunosorbent assay method.  相似文献

A sensitive and selective high performance liquid chromatography-peroxyoxalate chemiluminescence (PO-CL) method has been developed for the simultaneous determination of chlorpheniramine (CPA) and monodesmethyl chlorpheniramine (MDCPA) in human serum. The method combines fluorescent labeling with 4-(4,5-diphenyl-1H-imidazole-2-yl)phenyl boronic acid using Suzuki coupling reaction with PO-CL detection. CPA and MDCPA were extracted from human serum by liquid–liquid extraction with n-hexane. Excess labeling reagent, which interfered with trace level determination of analytes, was removed by solid-phase extraction using a C18 cartridge. Separation of derivatives of both analytes was achieved isocratically on a silica column with a mixture of acetonitrile and 60 mM imidazole-HNO3 buffer (pH 7.2; 85:15, v/v) containing 0.015% triethylamine. The proposed method exhibited a good linearity with a correlation coefficient of 0.999 for CPA and MDCPA within the concentration range of 0.5–100 ng/mL. The limits of detection (S/N = 3) were 0.14 and 0.16 ng/mL for CPA and MDCPA, respectively. Using the proposed method, CPA could be selectively determined in human serum after oral administration.  相似文献

A novel method is proposed for the extraction-thermal lens quantification of cobalt with Nitroso-R-Salt based on the distribution of the colored complex in a two-phase aqueous system on the basis of poly-ethylene glycol (PEG) and an ammonium sulfate solution followed by its thermal lens detection in the extract. The limit of detection is 0.3 μM (20 ng/mL); the lower limit of the analytical range is 0.7 μM (40 ng/mL); the relative standard deviation for the concentrations 1–50 μM makes 1–3% (n = 6, P = 0.95). In the determination of cobalt by spectrophotometry under the same conditions, the detection limit is 10 μM (0.6 μg/mL) and the lower limit of the analytical range is 40 μM (2.5 μg/mL). The precision of thermal lens measurements in PEG solutions is higher in comparison to that in aqueous ones because of the weaker interference of convection in aqueous solutions of PEG.  相似文献

A simple, miniaturized microplate chemiluminescence assay for determination of methyl parathion (MP) was developed in 384-microwell plates. Zirconia (ZrO2) was added in microwell for adsorption of acetylcholinesterase (AChE). The developed assay is based on inhibition of AChE by MP. A good dynamic range 0.008–1,000 ng/mL was obtained for MP with limit of detection 0.008 ng/mL. Intrabatch and interbatch reproducibility for miniaturized assay was obtained with % RSD up to 3.07 and 5.66, respectively. In 384 well plate formats, 70 samples were simultaneously analyzed within 20 min with assay volume of 41.5 μL. The application of developed assay was extended for MP remediation. Column containing ZrO2 was utilized for remediation where MP was selectively adsorbed. Under optimized condition, adsorption of MP on ZrO2 was found to be 98–99% with 2-h contact time in real water samples. Adsorption of MP on ZrO2 column followed by quantification using developed bioassay provides a novel approach to monitor remediation. The applicability of assay was successfully extended for determination of MP in water samples after removal through ZrO2.  相似文献

Systematic evolution of ligands by exponential enrichment is a traditional approach to select aptamer, which has a great potential in biosensing field. However, chemical modifications of DNA library or targets before selection might block the real recognition and binding sites between aptamers and their targets. In this study, a label‐ and modification‐free‐based in situ selection strategy was developed to overcome this limitation. The strategy is an attempt to screen bovine serum albumin aptamers according to the principle of electrophoretic mobility shift assay, and allowed single‐stranded DNA sequence to be fully exposed to interact with bovine serum albumin which was mixed with the agarose gel beforehand. After eight rounds of selection, specific aptamer with low dissociation constant (Kd) value of 69.44 ± 7.60 nM was selected and used for subsequent establishment of fluorescence biosensor. After optimization, the optimal aptasensor exhibited a high sensitivity toward bovine serum albumin with a limit of detection of 0.24 ng/mL (linear range from 1 to 120 ng/mL). These results indicated that the label‐ and modification‐free‐based in situ selection strategy proposed in this work could effectively select specific aptamer to develop aptasensor for sensitive detection of bovine serum albumin or other targets in actual complicated samples.  相似文献

A highly sensitive ultra high pressure liquid chromatography (UHPLC‐MSMS) method for estimation of ropinirole in rat brain homogenate and plasma has been validated. The method was successfully used for the degradation kinetics in different stress condition and regulated temperature. The chromatographic separation was achieved using isocratic mobile phase, consisting of acetonitrile–2mM ammoniumacetate (28:72 v/v; 0.25 mL min?1). The mass spectrometer was operated in synapt mass spectrometry mode via positive electrospray ionization using the transitions m/z 260 → m/z 261 for ropinirole, and m/z 324.39 → m/z 262.161 as a parent ion of escitalopram (IS). The assay for ropinirole was linear over the range of 0.5–100 ng mL?1 (r2; 0.999). The intra‐ and inter day precisions were less than 11.2% in terms of relative standard deviation (R.S.D.), and the accuracy was within ±6.4% in terms of relative error (RE). The mean extraction‐efficiency of QC samples (MQC, 8 ng/mL) was ≥80%. The lower limit of quantification (LLOQ) was 0.049 ng/mL where as lower limit of detection (LLOD) was 0.016 ng/mL. All the peaks of degradation were well resolved. The degradation kinetics of ropinirole, showed highest stability (t1/2 256.66/h; t0.9, 39.11/h) in acidic medium, lower stability in alkaline environment (t1/2, 103.43/h; t0.9, 15.76/h) and highly susceptible in oxidative environment (t1/2, 21.58/h; t0.9, 3.28/h). The applicability of this assay was demonstrated and successfully applied for pharmacokinetic profiling of ropinirole in Wister rat brain homogenate after intranasal administration.  相似文献
