Unsubstituted imidazo[4,5-b]pyridine adds methyl iodide to the pyridine N atom. Treatment of the resulting iodide with base forms 4-methyl-4H-imidazo[4,5-b]pyridine. 5-Chloro-7-[1,3]oxazolo[4,5-b]pyridin-2-ylquinolin-8-ol was synthesized by a Pd-catalyzed arylation, which proceeds efficiently with 2 equivalents of benzylated clioquinol and 1 equivalent of oxazolo[4,5-b]pyridine. Trifluoro or pentafluoroacylation of heteroaryl-enamines 2a,b gave the corresponding perfluoroacylated heteroaryl-enamines 3a-c . Heating the latter compounds with diethyl iminomalonate gave 2-amino-3-pyridinecarboxylates 4a-c . Hydrolysis to the free acids 5a-c , and reaction with diphenylphosphoryl azide afforded the desired 1,3-dihydro-6-heteroaryl-5-perfluoroalkyl-2H-imidazo[4,5-b]pyridin-2-ones 6a-c .  相似文献

[formula: see text] Condensation of 3-phenylsulfonyl-1,3-isobenzofuranone (3) with benzocyclohept-6,7-en-5-ones such as 4 and 15 provides a straightforward, general method for synthesis of functionalized benzo[4,5]cyclohepta[1,2-b]naphthalenes (e.g., 5 and 16). This finding was used to achieve a brief and efficient preparation of 6,7-benzo-3,4-(1,4-dimethoxy-2,3-naphtho)-1,5-dioxosuberane (2), an established intermediate to the naturally occurring red pigment radermachol (1).  相似文献

Two novel 2,3-dihydro-1H-imidazo[1,2-b]pyrazole derivatives 7 and 8 have been prepared by hydrazinolysis with 2,4-dinitrophenylhydrazine of 1-(benzylideneamino)-2-(2-ethoxycarbonyl-2-nitromethylidene)-imidazolidine 4 . The precursor 4 was conveniently prepared from ethyl nitroacetate and 1-(benzylidene-amino)-2-(methylthio)imidazoline 3 . Two examples are presented in which ethyl aceto(and trifluoroaceto)acetate 2-nitrophenyl-hyrazone, 9 and 10 , both of which also feature the β-hydrazinoacrylate arrangement, are refluxed in ethanol containing hydrochloric acid and thereby converted into pyrazolones 11 and 12 , pyrazoles 13 and 14 , and ketone 2-nitrophenylhydrazones 15 and 16 , respectively.  相似文献

As part of a preliminary study on novel 5-HT3 ligands, the synthesis of a series of 1H-imidazo[1,2-b]-pyrazole derivatives is described. The bicyclic heteroaromatic nucleus was functionalized at positions 1, 6 and 7 to give the series of tropanyl derivatives 4a-g, 12a, 12d (Table 1). Different synthetic approaches were utilized to obtain the desired molecules: endo and exo 6-amides 4a, 12a and 6-ester 4b required two independent schemes due to the opposite behavior of the intermediate imidazolide 3 towards tropine and tropanamine. The 7-congeners, ester 4c , its tropinium salt 4e , the endo and exo amides 4d and 12d were prepared from the known common precursor 8 [1], while derivatives 4f-g , originated by functionalizing position 1, were obtained from 1H-imidazo[1,2-b]pyrazole by direct N-acylation. Since the structural features of these molecules seemed to meet the main rules of the S.A.R. studies published so far [2-5], they were evaluated “in vitro” for 5-HT3 receptor affinity (Table 2). The biochemical data show significant activity for derivatives 4a-e, 4g. These results are encouraging and justify further investigational work on this class of molecules.  相似文献

The splitting of the dihydropyridine ring of N-methyl-substituted 4-phenyl-5-oxo-4,5-dihydroindeno[1,2-b]pyridine in an acid medium takes place at the C-N bond.
