用纳米金增效以提高微重量法检测核酸的灵敏度。实验研究了纳米金粒径大小对核酸探针固定的表面密度的影响以及探针密度对靶核酸杂交效率的影响。研究表明,核酸检测灵敏度与纳米金的粒径大小有密切关系。与粒径为5、15 nm的纳米金相比,用粒径25 nm的纳米金可以获得较高的杂交效率,检测的灵敏度也较高。本实验方法检测核酸的线性范围为0.05~1.2×10-6mol/L;检出限为1.0×10-8mol/L。  相似文献

 A novel ion-selective sensor for procaine hydrochloride has been proposed and applied successfully to analysis of pharmaceutical preparations. The sensor is based on modification of the Ag-electrode of an AT-cut piezoelectric quartz crystal with a PVC membrane containing procaine tetraphenylborate. A linear relation between the logarithm of the response frequency and the logarithm of the procaine hydrochloride concentration was found experimentally and could be explained by combination of a modified Sauerbrey equation and the Freundlich isotherm. The detection range was from 8.3 × 10−8 to 5.0 × 10−3 M with a detection limit of 8.3 × 10−8 M. The selectivity of the sensor and the influence of experimental parameters such as solution pH and detection time were investigated. Received February 10, 1999. Revision October 10, 1999.  相似文献

Abstract

Sulfur dioxide was detected and determined in air by a piezoelectric quartz crystal sensor coated with 4-aminoantipyrine 1-hydroxyetil-2-heptadecenyl imidazol (amine 220) solution (1:1 v/v in chloroform). The analytical response curve is linear over the concentration range from 0.70 to 5.0 ppm of SO2. Good linearities (r = 0.9990, 0.9995 and 0.9968) and sensitivities (18.0, 33.4 and 50.7 Hz/ppm) were found, respectively for exposure times of 30, 60 and 90 seconds. The sensor can be used for more than six months without loss in sensitivity and presented good reversibility and reproducibility. Among some possible interferents tested, only nitrogen dioxide and moisture caused major frequency changes.  相似文献

ABSTRACT

A new method for mutagenicity testing of carcinogens is proposed in this paper. Mutagenic activity is examined through monitoring of the growth situation of Salmonella typhimurium TA98 strain with a series piezoelectric quartz crystal (SPQC) sensor. This method needs no immobilization and preincubation of microorganism, and is simple and rapid. It can be applied to detect mutagen directly. When it was used to study the mutagenicity of dimethyl sulfate, the test time was 4 h at 37°C. There was a good linear relationship between frequency shift and the dose of mutagen in the range 2.5-20 μg/mL and the regression equation was ∠F = 53.82 + 3.81C. Some experimental conditions are also discussed in detail.  相似文献

采用简单的溶剂热法制备金属有机框架化合物MIL-125(Ti)和NH_2-MIL-125(Ti)材料,并通过X射线衍射仪、扫描电子显微镜、傅氏转换红外线光谱分析仪和比表面积测试仪对所得样品进行表征。将2种材料修饰构建为石英晶体微天平传感器,测试其在11%~97%RH相对湿度范围内的湿度感测性能。实验结果表明,基于MIL-125(Ti)和NH_2-MIL-125(Ti)构建的传感器对湿度具有灵敏度高、重复性好、响应/恢复快等特点。相对于没有氨基修饰的MIL-125(Ti),NH_2-MIL-125(Ti)材料修饰的传感器对湿度表现出更高的响应性能。在环境湿度约52%时,NH_2-MIL-125(Ti)传感器对11%RH湿度响应值比MIL-125(Ti)湿度传感器的大57 Hz,说明氨基功能化对MIL-125(Ti)的湿敏性能有显著的增强作用。此外,通过Materials Studio模拟计算获得了水分子与MIL-125(Ti)及NH_2-MIL-125(Ti)作用的吸附焓,也证明氨基功能化对MIL-125(Ti)的湿度敏感性能具有增强作用。  相似文献

CRISPR/Cas是大多数细菌及古细菌基于RNA的后天免疫系统。由CRISPR/Cas系统改造而成的CRISPR/Cas技术已成为一种强大的基因编辑工具,广泛应用于基因功能研究和基因修饰与治疗。除了作为基因编辑工具,Ⅱ类Cas蛋白具有的"附属切割"特性,已被开发成一种快速、低成本且高灵敏的核酸检测工具,在核酸分子诊断领域具有重要的应用潜力。该文总结了3种Ⅱ类Cas蛋白在核酸检测领域的代表性研究进展,并对CRISPR/Cas系统在该领域的应用前景进行了展望。  相似文献

Abstract

A novel piezoelectric quartz crystal microbalance (QCM) device with gas permeable membrane is proposed for the detection of microorganisms producing hydrogen sulfide (H2S). The detection theory is based on the adsorption of hydrogen sulfide onto the silver electrode of the piezoelectric crystal sensor, which causes a dramatic decrease in the resonant frequency of QCM. A 100 Hz frequency shift is chosen as the criteria value to judge the presence of microorganisms producing H2S. Factors affecting detection were investigated. Desiccant is of great practical importance in sensor response. This new biosensor can be a potential candidate for detecting bacteria which produce hydrogen sulfide.  相似文献

Thin, phenylboronic acid‐containing polymer coatings are potentially attractive sensory layers for a range of glucose monitoring systems. This contribution presents the synthesis and properties of glucose‐sensitive polymer brushes obtained via surface RAFT polymerization of 3‐methacrylamido phenylboronic acid (MAPBA). This synthetic strategy is attractive since it allows the controlled growth of PMAPBA brushes with film thicknesses of up to 20 nm via direct polymerization of MAPBA without the need for additional post‐polymerization modification or deprotection steps. QCM‐D sensor chips modified with a PMAPBA layer respond with a linear change in the shift of the fundamental resonance frequency over a range of physiologically relevant glucose concentrations and are insensitive toward the presence of fructose, thus validating the potential of these polymer brush films as glucose sensory thin coatings.

报道了用戊二醛将氨基修饰的寡核苷酸探针固定于氨基磁珠表面制成核酸传感器,传感器与生物素标记的DNA杂交,再利用亲和素-生物素偶合系统,偶联上亲和素标记的碱性磷酸酶。碱性磷酸酶催化底物α-萘酚磷酸酯(α-NATP)水解形成具有电化学活性的α-萘酚,用示差脉冲伏安法测定杂交结果,本法测得短链DNA片断的浓度线性范围为10-1000μg/L;最低检测限为3μg/L。作者优化了相关的检测条件,并成功测定了乙型肝炎病毒HBV-DNA的片段。  相似文献

An unexpected frequency response for a piezoelectric quartz crystal (PQC) sensor to liquid density and viscosity was reported. For a PQC oscillating in a liquid phase, the frequency shifts (△f) show a wave-shape response to liquid density (p) and viscosity 03) in fine structure, ifthe longitudinal wave effect was not eliminated. This result is different from the well-knownlinear relationship between of Af and (ρη)1/2. An oscillating frequency-temperature curve of thesensor was observed and explained.  相似文献
