Abstract— Hypocotyl elongation in etiolated mustard ( Sinapis alba L.) seedlings is known to be controlled by phytochrome (Pfr) through a threshold mechanism. The Pfr threshold value required to suppress hypocotyl growth was low (3 times 10−2% Pfr, based on total phytochrome in the hypocotyl at 36 h after sowing = 100%). In the present study the question was addressed whether the threshold control by Pfr of hypocotyl elongation also operates in light-pretreated, partly de-etiolated seedlings after transfer to darkness. The experimental results show that this is the case. Calculation of the threshold level in far-red light pretreated seedlings led to a very low value (3 times 10−7%) compared to etiolated seedlings (3 times 10−2%). In red light pretreated seedlings the threshold level was calculated to be 9 times 10−7%. Since the light pretreatment affected the rate of degradation of phytochrome strongly (half-life of Ptot in continuous red light was found to be 35 min in far-red pretreated instead of 47 min in etiolated material), the difference in threshold level between far-red and red pretreated material cannot be interpreted unambiguously. However, the conclusion can be drawn that light nretreatment strongly increases the degradation rate of Pfr and decreases the threshold level.  相似文献

Abstract— In mustard ( Sinapis alba L.) cotyledons appearance of 'potential capacity' to phosphorylate is controlled by phytochrome (Pfr) through a threshold, all-or-none reaction. Phytochrome operates only if the hypocotyl hook is connected to the cotyledons for at least 2.5 min after the end of a 1 min inductive light pulse. Partial irradiations of hook and cotyledons show that in the case of induction of 'potential capacity' rapid transmission of the Pfr signal occurs from the hypocotyl hook to the cotyledons. Phytochrome within the cotyledons is totally ineffective in this regard. A 3 min red light pulse suffices for full transmission of the signal. It seems that an explanation of the facts we have observed requires a revision of the present views about communication between the organs of a plant.  相似文献

Abstract —Synthesis* of the enzyme lipoxygenase (LOG)? in the cotyledons of the mustard seedling (Sinapis alba L.) is controlled by phytochrome (Pfr) through a threshold (all-or-none) mechanism. The data of the present paper confirm the previous assumption (Oelze-Karow and Mohr, 1973) that the primary reaction of Pfr (Pfr+ X → PfrX ? PfrX‘) is the site of the highly cooperative threshold reaction. Suppression of LOG synthesis depends on the presence of PfrX’. However, PfrX‘ is only stable above the threshold level of Pfr. If the level of Pfr is below the threshold, PfrX is stable, and no suppression of LOG synthesis occurs. As long as the level of Pfr remains below the threshold, no destruction of Pfr takes place. Destruction of Pfr occurs only as long as [Pfr]?is above the threshold level. Thus the simplest formulation of the actual threshold reaction in the LOG response is PfrX?frX’ state at [Pfr] below threshold no Pfr destruction LOG synthesis suppressed state at [Pfr] above threshold Pfr destruction(1kd LOG synthesis unimpaired The reversible threshold reaction is thus an integral part of the “primary reaction” of Pfr occurring at the “matrix” specific for the LOG response. The data and conclusions on the LOG response are consistent with an “open phytochrome-receptor model” recently advanced by E. Schäfer (1975). The data are not consistent with the concept that a rapid dark reversion (Pfr→Pr) exists in dicotyledonous seedlings and that the degree of Pfr dark reversion strongly depends on the initial photostationary state, φ?, established by a saturating light pulse.  相似文献

Abstract— Six hours of irradiation with white or far-red light strongly stimulates the 'end of day' inhibition of hypocotyl elongation in dark grown Sinapis alba L. Measurements of both reversion kinetics of the inductive light pulse and 'null point' experiments (Hillman, 1965. 1972) indicate that this response is controlled by stable phytochrome. The extent of the reversible response (i. e. length after far-red light minus length after red light) showed rhythmic oscillations after a 6, 48 and 54 h white light pretreatment. The rhythm is started by the transition from light to dark and neither phase nor amplitude is influenced by the level of P at this transition.  相似文献

Abstract— The relationship between phototropism and axis extension was examined in light-grown mustard (Sinapis alba L.) seedlings using the low pressure sodium lamp (SOX)? technique to eliminate growth responses due to phytochrome. Addition of blue light caused no net inhibition of hypocotyl elongation, but plants showed a phototropic response. Curvature was caused by a simultaneous inhibition of growth on the illuminated side of the hypocotyl and an acceleration on the shaded side. Phototropism thus occurs independently of axis elongation and suggests that they are two separate processes. The results are inconsistent with the Blaauw theory of phototropism.  相似文献

Abstract— Hypocotyl straight growth in mustard (Sinapis alba L.) responds very strongly and in precisely the same way to low fluence rate red (RL) and white light (WL). The effect of weak light can be attributed fully to light absorption by phytochrome. Only with increasing fluence rate an effect of blue light (BL) comes into play which cannot be explained by the action of phytochrome. However, this specific action of BL can be demonstrated in hypocotyl growth of mustard seedlings only up to 5 days after sowing (25°C). With older seedlings control of hypocotyl growth seems to be exerted exclusively via phytochrome. Regarding the far-red light dependent “high irradiance reaction” (FR-HIR) it was found that it plays a dominant role in growth of mustard only during a relatively short period. It tends to disappear in favor of a RL-HIR between 3 and 4 days after sowing. It is concluded that the seedling exhibits a largely endogenous temporal pattern of responsiveness to light. Phototropism of the mustard seedling can be elicited by low fluence rates (< 1 mW m?2) of unilateral BL. This same light has no effect on straight growth. It is concluded that BL-dependent phototropic growth response of a hypocotyl and the effect of BL on longitudinal growth of the hypocotyl are unrelated phenomena.  相似文献

Abstract— The development of plastids in the palisade parenchyma cells of the cotyledons of mustard seedlings ( Sinapis alba L.) was studied by electron microscopy. In darkness the etioplasts undergo a sequence of morphogenic changes previously recognized in principle in bean and barley leaves, as summarized by Rosinski, J. and W. G. Rosen (1972) Quart. Rev. Biol. 47 , 160–190. From 12 to 36 h after sowing, an increase in the percentage of etioplast profiles with paracrystalline prolamellar bodies can be observed. Thereafter, the degree of organization and size of the prolamellar bodies decrease. 60 h after sowing, the etioplasts show only remnants of prolamellar bodies with irregularly spaced tubules. Continuous far-red light, which is considered to operate via phytochrome, counteracts the decay of organization of the prolamellar body and strongly increases the size of the plastids. The effect of continuous far-red light (onset of light 36 h after sowing) can be substituted by 12 h of far-red light given between 36 and 48 h after sowing. It is shown with red and far-red light pulses that the morphogenic effect of long-term far-red light on plastid size and appearance of the prolamellar body is exclusively due to phytochrome (Pfr). Changes by light in the amounts of protochlorophyll(ide) or chlorophyll(ide) do not affect these results. The action of Pfr on the structure of the prolamellar body is a relatively fast process, occurring within 3 h. Formation of thylakoids does not seem to be under phytochrome control. Rather, this response seems to be related to the protochlorophyll(ide)→ chlorophyll(ide) a transformation.  相似文献

Abstract. Continuous far-red light operating through phytochrome reduces the rate of hypocotyl lengthening by a factor of 8. Consonant with this was a reduction of up to 12% in the proportion of total cytoplasmic ribosomes present as polysomes. A 2% reduction was elicited by brief red light. Evidence is presented which argues against these changes arising from a change in extractability or nucleolytic activity. The speed of the response (1.5 h) is much slower than that for inhibition of lengthening (15 min). The data therefore indicate that phytochrome is unlikely to act by controlling the general rate of translation in this instance.  相似文献

Abstract— Mustard ( Sinapis alba L.) seedlings were grown on moistened vermiculite without added nutrients for 17 days in medium white light (4.6 lx). During this period cotyledons as well as primary leaves expressed signs of senescence as defined by the loss of chlorophyll(ide) a ('degreening'). While degreening was detectable 9 days after sowing in the cotyledons it was not measurable before 15 days after sowing in the primary leaves. Phytochrome retards the loss of chlorophyll(ide) a in both organs; however, it does not change the temporal pattern of greening and degreening.  相似文献

Abstract— Etiolated turions of Spirodela polyrhiza are positively photoblastic and show a phytochrome-mediated low fiuence germination response. The far-red light (FR) reversibility decreased with the delay of FR irradiation (lag phase 1.06 ± 0.03 days after red light irradiation; half-maximal response 1.9 days). The action of the far-red-absorbing form of phytochrome (Pfr) was only realized by a germination response if exogenously applied Ca2+ was present. Calcium step-down (from 1 mM to 0.9 μ M Ca2+) and Ca2+ step-up (from 0.9 μ M to 1 m M Ca2+) experiments were carried out to determine the Ca2+-sensitive phase. There was no time gap between the two phases determined by the step-down and step-up experiments but a clear coincidence of both curves. Pulse treatments (24 h) with Ca2+ (1 m M ) showed the upper part of this common curve to represent the most Ca2+-sensitive phase. The Ca2+-sensitive phase was within the Pfr-requiring phase. After reversion of Pfr by FR pulses there was only a negligible response to the high Ca2+-concentration, independent of the delay between the red light (R) and FR pulses. These results are compatible with the assumption of Ca2+ acting as a second messenger of Pfr. However, the Ca2+-insensitivity in the first 12 h after the R pulse points against this hypothesis.  相似文献

Abstract— A rapid transient increase in the growth rate of Cucumis sativus L. seedlings is found to occur after a transition from fluorescent white light to darkness. The post-illumination burst of growth starts after about 20 min of darkness and lasts for 30 to 50 min. The response occurs after long periods of continuous white light, in cycles of white light and darkness, whether the cotyledons are covered or exposed. The post-illumination burst is an expression of the loss of photocontrol of hypocotyl inhibition via the specific blue light photoreceptor because (a) its kinetics resemble those of recovery from blue-light inhibition, and (b) it only occurs on transition from a blue-rich to a blue-poor light environment.  相似文献

Abstract— In sunflower ( Helianthus annuus L.) it has been found that continuous white-light during the flowering stage causes loss of protandry so that filaments do not elongate. Experiments have been carried out to test the nature of the pigment involved in the photoinhibition of elongation. The results suggest that the elongation of the filament is controlled by the phytochrome system and that a low photoequilibrium is required to obtain the response. Abstract— The effects of phytochrome status on extracellular peroxidase activity were investigated in Sinapis alba L. seedlings grown for 12 days under continuous white light and transferred to darkness after a red light or a far-red light pulse. The rates of extension growth and dry matter accumulation in the first internode were increased by the far-red light pulse. Extracellular proteins, obtained by low speed centrifugation of intact internodes infiltrated with CaCl2, were separated by isoelectrofocusing, and four extracellular acidic peroxidases were resolved, the most active being A3 and A4 (both ˜60 kD). The activity of A4 was reduced by the far-red light pulse perceived by phytochrome, while the activity of A3 was unaffected. The promotion of internode extension growth caused by far-red light is biphasic [Casal and Smith (1989) Plant, Cell Environ. 12 ,511–50]. Changes in peroxidase activity were detected prior to the second, but not to the first phase of the internode growth promotion. The effects on both growth and peroxidase activity were virtually restricted to the upper half of the internode and, once established, did not subsequently increase in magnitude.
In contrast to the effects mediated by phytochrome, blue light pretreatments affected growth but not extracellular peroxidase activity. Wounding the internode reduced extension growth, increased the activity of A3, but caused no significant effects on A4.
Other extracellular proteins, separated in sodium dodecyl sulphate polyacrylamide gels and stained with Coomassie blue, showed no significant differences. The concentration of extracellular proteins was higher in the upper than the lower half of the internode.
Results are discussed in terms of phytochrome effects in light grown plants, peroxidase activitv-arowth relationships, and extracellular peroxidase isoform functions.  相似文献

A series of polycondensed aromatic N-heterocycles (acridine, benzo-f-quinoline 1,2,7,8-dibenzacridine and 3,4,5,6-dibenzacridine) were adsorbed from the gas phase and from liquid solution on highly dispersed silica gels with very different specific surface areas and pore sizes. The translational mobility of the adsorbed species was quantified by the triplet decay and the delayed fluorescence following bimolecular triplet-triplet annihilation after pulsed laser irradiation. The decay kinetics were analyzed with conventional second order rate equations and with the fractal approach. The first method is reliable without limitations on adsorbents with large pore diameters. It yields second order annihilation constants of 4 times 1012-6 times 1011 dm2 mol?1 s?1 depending on the masses and sterical requirements of the adsorbates. For the second method a spectral dimension ds= 4/3 was used. This method is quantitatively applicable to all heterocycles adsorbed on silica gel 60 that have very small pore sizes. An activation energy of 4.9 ± 0.5 kcal mol?1 was obtained for the translational diffusion of acridine on hydroxylated silica gel.  相似文献

Abstract— Action spectra for growth reduction within the 260 nm to 305 nm waveband were measured for hypocotyls and roots of young etiolated cress ( Lepidium sativum ) seedlings. The action spectra show increasing photon effectiveness with decreasing wavelength and resemble those due to DNA damage. Using short term irradiations, photoreceptors absorbing in the visible range were found to be without influence. As no photoreactivation could be found and the seedlings showed no outward signs of damage, this growth effect may be due to a UV photoreceptor. A modeling calculation was carried out to estimate the effectiveness of solar UV-B on this response both under present conditions and under reduced ozone levels. Even under present conditions, solar UV-B could be involved in regulating growth during the period immediately after germination.  相似文献

The plastid glyceraldehyde-phosphate dehydrogenase and phosphoribulokinase of mustard cotyledon extracts were activated by preincubation with ATP and with ATP and dithiothreitol respectively. By in vitro activation prior to assay, it was possible to determine the potential activities, which appear to have been directly proportional to the amount of each enzyme protein present. In this way it was possible to deduce the net synthesis of these two enzymes. The induction of synthesis of glyceraldehyde-phosphate dehydrogenase and phosphoribulokinase, by continuous far red or white light were similar hut net synthesis in continuous far red continued longer for glyceraldehyde-phosphate dehydrogenase than for phosphoribulokinase. The kinetics of the development of the glyceraldehyde-phosphate dehydrogenase potential activity were very close to those reported by Bruning et al. (1975) for this enzyme. The data do not permit elimination of either the single switch or multiple switches hypotheses for the action of phytochrome. Continuous white illumination gave results similar to those for continous far red for the net synthesis of the two enzymes but it was more effective than far-red in bringing about enzyme activation in vivo.  相似文献

Abstract— Rapid inhibition of hypocotyl extension of de-etiolated Cucumis sativus L. by blue light is described, and compared with responses to white fluorescent light. Rapid inhibition of hypocotyl growth by blue light via the specific blue light photoreceptor requires a minimum Buence rate. Above this minimum value rapid modulations of growth rate accompany changes in blue light fluence rate. An initial response to blue light, or to a step-up in blue fluence rate takes less than 5 min. A drop from high to low fluence rate blue light (or to darkness) is followed by a recovery of the growth rate after about 20 min. A change from a low fluence rate to darkness elicits a more rapid recovery of growth rate (within 7 min). Similar responses were obtained in seedlings de-etiolated for a few hours and for several days.  相似文献

Hypocotyl growth in etiolated seedlings of wild-type and an aurea mutant of tomato (Lycopersicon esculenturn Mill.), that appears to be deficient in labile phytochrome, is strongly inhibited by UV radiation in the region of 300–400 nm. The role of phytochrome in the UV-mediated inhibition of hypocotyl growth was studied using different experimental approaches: (1) by comparing the effectiveness of treatments of increasing duration of exposure to 692 nm and UV radiation; (2) by modifying the UV spectral range with specific cut-off filters. The experimental results suggest that the UV-induced inhibition of growth in wild-type tomato is mediated to a large extent by the longer wavelengths of the UV-A region and is mediated mainly by phytochrome. In contrast, at wavelengths < 305 nm a strong UV-B effect was found in the aurea mutant, suggesting a preeminent action of a specific UV-B absorbing photoreceptor that displays less action in the wild-type.  相似文献
