Optical rubbery ormosils sensor for the detection of ammonia   总被引:1,自引:0,他引:1  
Rubbery ormosil films with immobilized aminofluorescein (AF) were investigated to develop an optochemical sensor for the determination of ammonia in water. The gel precursors with tetramethoxysilane (TMOS) and dimethyldimethoxysilane (DiMeDMOS) were deposited on glass supports, and characterized in terms of response to pH, and to dissolved ammonia at constant pH. After preconditioning the sensing film was stable for 6 months. The detection limit for ammonia in water was 0.2 μg mL–1 (S/N 2), the response being linearly dependent on concentration in the range of 0.5 to 80 μg mL–1 ammonia. The response time was less than 5 min. The effects of sodium chloride concentration, temperature, and coexisting metal ions and compounds were investigated. Summary A capillary zone electrophoretic method for the analysis of phenolic acids in soil and plant extracts was developed with direct UV detection using a phosphate electrolyte solution. The electrophoretic separation required the phenolic acids to be charged at a pH above their pKa in order to achieve their migration towards the anode. Electroosmotic flow (EOF) was reversed in direction by adding tetradecyltrimethylammonium bromide (TTAB). Factors affecting the separation selectivity, including the buffer pH and EOF modifiers, were investigated systematically. Eight phenolic acids were separated and detected in 10 min using an electrolyte containing 25 mM phosphate, 0.5 mM TTAB and 15% acetonitrile (v/v) at pH of 7.20. Linear plots for the test phenolic acids were obtained in a concentration range of 0.01–1 mM with detection limits in the range of 1.0–7.0 μM. The recoveries ranged from 92.8 to 102.3% in soil and plant tissues samples spiked at 100 μM and the relative standard deviation based on the peak area were ranged 2.0 to 4.5%. The proposed method was used for the determination of phenolic acids in plant tissue and soil extracts with direct injection.  相似文献

Sol–gel encapsulation has been used as the basis for detecting cortisol by an immunoassay approach. Previous research showed that antibodies immobilized in the pores of a sol–gel derived silica were able to bind cortisol and be used as an immunosensor. However, this approach was not effective when measuring cortisol levels in human serum because of interference from other fluorescence sources. The present paper describes a protocol which overcomes these limitations and enables sol–gel immunoassays to effectively measure cortisol in human serum over the physiological range of cortisol blood concentrations in an adult (2–28 μg/dL). The method involves a standard additions approach in which various amounts of cortisol are added to the serum. The cortisol concentration values obtained with our sol–gel immunoassay were typically within 10% of the values obtained by traditional analytical methods. The protocol presented here represents a significant contribution to sol–gel sensing and immunoassays in particular, because of the ability to detect an analyte in human serum. In addition, this work reports the first comparison between results from a sol–gel immunosensor and an alternative immuno-binding method for analyte detection.  相似文献

Summary HPTLC densitometry and HPLC are considered for the simultaneous determination of the degradation products of piroxicam (2-aminopyridine, DP-I and DP-II). The substances were separated on silica gel with fluorescence indicator in ethylacetate — toluene — diethylamine (10∶10∶5) and toluene — absolute ethanol — glacial acetic acid (8∶1.2∶0.5) systems. The measuring absorbance (detection of reflectance) of the separated substances was carried out “in situ” at 296 nm using 4-level calibration (external standard, nonlinear regresson function) in the concentration range 600–1200 ng 2-aminopyridine/spot and 300–600 ng DP-I and DP-II/spot. The HPLC method was carried out using RP-8 stationary phase and methanol + phosphate-citrate buffer, pH 3 mobile phase with addition of sodium pentanesulfonate (40+60, v/v). 2-aminopyridine wass detected at 300 nm, DP-I at 280 nm and DP-II at 248 nm. The concentration range for 2-aminopyridine is 2–40 μg/ml, for DP-I and DP-II 2–20 μg/ml (for an injection volume of 10 μl). The results were evaluated by linear regression analysis.  相似文献

The method integrates the transitory retention and fluorescence detection of both compounds on Sephadex QAE A-25 resin packed into a conventional flow-through cell. A monochannel manifold with two alternative carriers is used. At pH 2.0 (first carrier) salicylic acid is selectively retained on the solid support and after developing the analytical signal it is desorbed. At pH 11.0 (second carrier) both salicylic acid and salicylamide are simultaneously and transitorily retained on the solid, the analytical signal now corresponding to both analytes. The monochromators were tuned at 260 (excitation) and 415 (emission) nm, respectively. The calibration graph for salicylamide is linear over the range 0.01 to 0.32 μg mL–1 and for salicylic acid from 0.04 to 1.0 μg mL–1 in the presence of each other. The relative standard deviation and the sampling frequency for the determination of salicylamide (0.20 μg mL–1) and salicylic acid (0.50 μg mL–1) were 1.1% and 35 h–1, and 0.9% and 45 h–1, respectively. Good results on application to individual determination or mixture resolution in pharmaceutical samples testify to the usefulness of the proposed sensor. We demonstrate in this work that 2-μm-sized Ag (μAg) powders can be used as a core material for constructing biomolecular sensing/recognition units operating via surface-enhanced resonance Raman scattering (SERRS). This is possible because μAg powders are very efficient substrates for both the diffuse reflectance IR and the surface-enhanced Raman scattering–SERRS spectroscopic characterization of molecular adsorbates prepared in a similar manner on silver surfaces. Besides, the agglomeration of μAg particles in a buffer solution can be prevented by the layer-by-layer deposition of cationic and anionic polyelectrolytes such as poly(allylamine hydrochloride) (PAH) and poly(acrylic acid) (PAA). In this particular study, we used rhodamine B isothiocyanate (RhBITC) as a SERRS marker molecule, and μAg powders adsorbed consecutively with RhBITC and PAH–PAA bilayers were finally derivatized with biotinylated poly(l-lysine). On the basis of the nature of the SERRS peaks of RhBITC, those μAg powders were confirmed to selectively recognize streptavidin molecules down to concentrations of 10−10 g mL−1. Since a number of different molecules can be used as SERS–SERRS marker molecules, the present method proves to be an invaluable tool for multiplex biomolecular sensing/recognition via SERS and SERRS.  相似文献

In pH 4.5 Britton-Robinson(BR)buffer solution,erythrosin(ET)can react with diphenhydramine(DP)to form a 1:1 ion-association complex,which not only results in the change of the absorption spectra,but also results in the great enhancement of resonance Rayleigh scattering(RRS)and the quenching of fluorescence.Furthermore,a new RRS spectrum will appear,and the maximum RRS wavelength was located at about 580 nm.In this work,the spectral characteristics of the absorption,fluorescence and RRS,the optimum conditions of the reaction and the properties of an analytical chemistry were inves- tigated.A sensitive,simple and new method for the determination of DP by using erythrosin as a probe has been developed.The detection limits for DP were 0.0020μg/mL for RRS method,0.088μg/mL for absorption method and 0.094μg/mL for fluorophotometry.There was a linear relationship between the absorbance,RRS and fluorescence intensities and the drug concentration in the range of 0.0067-2.0, 0.29-6.4 and 0.31-3.2μg/mL,respectively.The effects of the interaction of diphenhydramine and erythrosin on the absorption,fluorescence and resonance Rayleigh scattering spectra were discussed. In light polarization experiment,the polarization of RRS at maximum wavelength was measured to be P =0.9779,and it revealed that the RRS spectrum of DP-ET complex consists mostly of resonance scat- tering and few resonance fluorescence.In this study,enthalpy of formation and mean polarizability were calculated by AM1 quantum chemistry method.In addition,the reaction mechanism and the rea- sons for the enhancement of scattering spectra and the energy transfer between absorption,fluores- cence and RRS were discussed.  相似文献

Errors in the determination of low concentrations by spectrophotometry are investigated with the uranium-thiocyanate system as an example. The reagent blank has significant absorption and measurements are made at 375 nm instead of the λmax. The error in the intercept of the calibration curve is an important factor in such measurements and the errors involved in the estimation of 1 μg/ml (normal working range 4–40μg/ml) are studied. It is shown that both random and systematic errors associated with the intercept are responsible for observed errors. The two types of errors are resolved by ANOVA (analysis of variance). The error in the measurement of a single value is estimated and compared with measured values for different calibration ranges. It is seen that two factors predominantly influence the error in the measured concentration — the variance from regression and the closeness of the measured value to the mean of the calibration range.  相似文献

Bovine serum albumin (BSA) is a soft globular protein that undergoes conformational changes through several identified transition steps in the pH range 2–13.5. The ability to change conformation makes BSA capable of complexing different ligands from fatty acids to cations or drugs and carries them in the bloodstream. Microcalorimetric titration of BSA with NaOH solution was performed to measure the enthalpy of conformational changes. Two exothermic enthalpy changes were found in the course of the titration between pH 3 and 9.5, which can be identified with the E–F, and the F–N transitions. The enthalpy change at pH 3.5 (transition from the E to the F form of BSA, folding of intra-domain helices in domain I) is independent of the protein concentration. The second transition (F–N, folding of domain III) was observed at pH 4.8 for the 0.1% BSA solution, but it shifted to higher pH values as the protein concentration increased to 0.2% and 0.3%. The tightening of the protein structure with increasing pH was verified measuring intrinsic fluorescence of tryptophan residues. At even higher pH value, pH 10.5, fluorescence measurements revealed protein expansion. The BSA conformational changes were also measured by dynamic light scattering. The hydrodynamic diameter was smaller at the i.e.p. of BSA (5–7 nm at pH ~5) and larger at the two ends of the pH range (17.5 nm at pH 2 and 8.3 nm at pH 10).  相似文献

Two spectrophotometric methods were applied to the simultaneous assay of chlorhexidine hydrochloride (CHL) and lidocaine hydrochloride (LIH) in pharmaceutical formulations. Using derivative spectrophotometry, CHL was determined by measurement of its first derivative signal at 290 nm (peak to zero amplitude) in the concentration range 5–9 μg/mL, and LIH was analysed by measurement of its second derivative signals at 272 and 276 nm (peak to peak amplitude) in the concentration range 160–480 μg/mL. With the partial least-squares (PLS-2), the experimental calibration matrix was constructed using 9 samples. The concentration ranges considered were 5–7 μg/mL for CHL and 220, 240, 260 μg/mL for LIH. The absorbances were recorded between 240 and 310 nm at every 5 nm.  相似文献

A simple, rapid and reproducible HPLC method was developed and validated for the simultaneous determination of olmesartan (OLM) medoxomil and hydrochlorothiazide (HCT) in combined tablets. Chromatography was carried out on a 4.6 mm I.D × 200 mm, 5 μm cyano column with methanol–10 mM phosphoric acid containing 0.1% triethylamine (pH 2.5, 50:50 v/v) at a flow rate of 1.0 mL min−1 and UV detector was set at 260 nm. Valsartan (VAL) was used as internal standard (IS). A linear response was observed in the range of 0.2–6 μg mL−1 (r 2 = 0.9998) for OLM and 0.1–4 μg mL−1 (r 2 = 0.9999) for HCT, respectively. The method showed good recoveries (99.56% for OLM and 99.48% for HCT) and the relative standard deviation (RSD) values for intra- and inter-day precision were 0.70–1.59 and 0.80–2.00% for OLM and 1.20–1.37 and 1.63–1.93% for HCT, respectively. The developed method was applied successfully for quality control assay of OLM and HCT in combined tablets and in vitro dissolution studies.  相似文献

Cardiolipin (CL) plays an essential role as a marker for cell apoptosis. Quantitative detection of phospholipids (PLs) by UV absorbance is problematic due to the presence of few double bonds in the structure. Although 10-N-nonyl acridine orange (NAO) has been utilized for fluorescent visualization of liposomes and mitochondria through its interaction with CL, in this work, we have developed a specific fluorescent method for CL in solution using NAO. The interaction of sodium n-dodecyl sulfate (SDS), used to treat cells prior to lipid extraction, and other PLs found in cell membranes such as phosphatidic acid (PA), phosphatidylcholine (PC), phosphatidylethanolamine (PE), phosphatidylglycerol (PG), phosphatidiylserine (PS), and sphingomyelin (SM) with NAO is investigated. The fluorescence intensity of the 0.5 μM NAO signal is strongly quenched by SDS below 25% methanol in water but with a methanol content above 50%, no quenching of NAO by SDS is observed. No fluorescence quenching of NAO with a 50% methanol/50% water solvent by the previously mentioned PLs or 4–20 μM cholesterol with the exception of PG at above 8 μM is noted. Using this 50% methanol/50% water solvent, the fluorescence signal due to the NAO–CL interaction is quite stable from 3 to at least 15 min. With excitation and emission wavelengths set at 518 and 530 nm, respectively, 20 μM NAO provides an inverse linear fluorescence response at 0.2–10 μM CL with a correlation coefficient of 0.9929. The detection limit is 0.2 μM and the limit of quantification is 0.6 μM. Structurally analogous acridine orange and phenosafranin dyes are less effective as fluorescent probes for CL. The CL in the whole cell and membrane samples is quantitatively determined by standard addition to range from 0.2 to 1.5 μM. The level of CL in cell membrane samples, previously subjected to staurosporine which initiates cell apoptosis, is increased but not significantly through use of the t-test.  相似文献

Summary The fluorogenic reagent 2-naphthyloxycarbonyl chloride (NOC-Cl) has been used for the automated precolumn derivatization of biogenic amines (BAs) at ambient followed by liquid-chromatographic separation of the derivatives formed. For optimized derivatization samples in 0.5 M borate buffer (pH 9.0) were derivatized with 5 mM NOC-Cl in acetonitrile (MeCN) for 3 minutes. Excess of reagent was scavenged by addition of 20 mM glycine in water. For HPLC a Superspher? RP-18e column and gradient elution using 0.1 M sodium acetate buffer (pH 4.4) and MeCN were used. The NOC-derivatives were detected by fluorescence at an emission wavelength of 335 nm at an excitation wavelength of 274 nm. This method allows the detection of BAs (2-phenylethylamine, putrescine, histamine, cadaverine, tyramine, spermidine, spermine) found in food and beverages (fruit juices, wines, various vinegars, fermented cabbage juice, and salmon). Detection limit of BAs are approximately 49–113 μg kg−1 with the exception of histamine (747 μg kg−1) (injected amounts: 18–41 pg histamine 267 pg), at a signalto-noise ratio of 3:1. The limits of determination are approximately 82–189 μg kg−1 (histamine 1245 μg kg−1) at a signal-to-noise ratio of 5:1. The correlation coefficients of linearity are 0.9910–0.9976.  A column preconcentration method has been established for the spectrophotometric determination of traces of nitrite using diazotization and coupling on an naphthalene-tetradecyldimethylbenzylammonium (TDBA)-iodide (I) adsorbent. Nitrite ion reacts with sulfanilic acid (SA) in the pH range 1.8–3.0 for the SA-1-naphthol system and in the pH range 2.3–3.2 for the SA-1-naphthylamine-4-sulfonate system (SA-NAS system) in hydrochloric acid medium to form water-soluble colourless diazonium cations. These cations were coupled with 1-naphthol in the pH range 1.6–4.6 and with NAS in the pH range 2.6–5.0 to be retained on naphthalene-TDBA-I packed in a column. The solid mass was dissolved from the column with 5 mL of dimethylformamide (DMF) and the absorbance measured at 418 nm for the SA-1-naphthol system and at 485 nm for the SA-NAS system. The calibration curve was linear over the concentration range 0.02–0.87 mg/L for SA-1-naphthol and 0.02–0.80 mg/L in the sample for SA-NAS. The molar absorptivity was calculated to be 1.70×104 L mol-1 cm-1 for SA-1-naphthol and 1.66×104 L mol-1 cm-1 for SA-NAS. The detection limits were found to be 0.014 and 0.016 mg/L for SA-1-naphthol and SA-NAS, respectively. The preconcentration factors were 8 and 6 for SA-1-naphthol and SA-NAS, respectively. Replicate determinations of seven sample solutions containing 6.6 μg of nitrite for SA-1-naphthol and 5.3 μg of nitrite for SA-NAS gave mean absorbances of 0.486 and 0.382 with relative standard deviations of 0.49 and 0.58%, respectively. Interferences due to various foreign ions have been studied and the method has been applied to the determination of 27–65 μg/L levels of nitrite in natural waters. The thermal stability of the recombinant green fluorescent protein (GFPuv) expressed by Escherichia coli cells and isolated by three-phase partitioning extraction with hydrophobic interaction chromatography was studied. The GFPuv (3.5–9.0 μg of GFPuv/mL) was exposed to various pH conditions (4.91–9.03) and temperatures (75–95°C) in the 10 mM buffers: acetate (pH 5.0–7.0), phosphate (pH 5.5–8.0), and Tris-HCl (pH 7.0–9.0). The extent of protein denaturation (loss of fluorescence intensity) was expressed in decimal reduction time (D-value), the time exposure required to reduce 90% of the initial fluorescence intensity of GFPuv. For pH 7.0 to 8.0, the thermostability of GFPuv was slightly greater in phosphate buffer than in Tris-HCl. At 85°C, the D-values (pH 7.1–7.5) ranged from 7.24 (Tris-HCl) to 13.88 min (phosphate) The stability of GFPuv in Tris-HCl (pH>8.0) was constant at 90 and 95°C, and the D-values were 7.93 (pH 8.38–8.92) and 6.0 min (pH 8.05–8.97), respectively. The thermostability of GFPuv provides the basis for its potential utility as a fluorescent biologic indicator to assay the efficacy of moist-heat treatments at temperatures lower than 100°C.  相似文献

A new method was developed for the simultaneous determination of lidocaine, proline and lomefloxacin in human urine by capillary electrophoresis-electrochemiluminescence detection with Ru(bpy)3 2+. Conditions of the separation and detection were investigated and optimized. It was proved that 20 mM phosphate buffer at pH 6.7 could achieve the most favorable resolution, and the high sensitivity of detection was obtained by using the detection potential at 1.15 V and 5 mM Ru(bpy)3 2+–60 mM phosphate buffer at pH 7.6 in the detection reservoir. The detection limits were 0.02 μg mL−1 for lidocaine, 0.03 μg mL−1 for proline and 0.06 μg mL−1 for lomefloxacin. Relative standard deviations of the ECL intensity and the migration time were 3.5 and 1.1% for 6 μg mL−1 lidocaine, 3.2 and 1.0% for 6 μg mL−1 proline and 3.7 and 1.2% for 6 μg mL−1 lomefloxacin, respectively. A baseline separation for lidocaine, proline and lomefloxacin was achieved within 360 s. The developed method was successfully applied to determine the amounts of lidocaine, proline and lomefloxacin in human urine. The recovery and RSD were in the range of 93.3–97.2 and 3.8–4.9%, respectively.  相似文献

An anthracycline drug, adriamycin, was chemically conjugated onto the backbone of glycol chitosan via an acid-labile cis-aconityl linkage. The physicochemical characteristics of the glycol chitosan–adriamycin (GC–ADR) conjugates were investigated by dynamic light scattering, atomic force microscopy, and fluorescence spectroscopy. The GC–ADR conjugates were capable of forming nano-sized self-aggregates in an aqueous medium, when the adriamycin content in the conjugate was in the range of 2.0–5.0 wt.%. The self-aggregates were spherical in shape, and had mean diameters of 238–304 nm, depending on the adriamycin content. The critical aggregation concentrations of the conjugates, estimated by the fluorescence quenching method, were as low as 1.0–2.5×10−2 mg/ml. The size of self-aggregates was not affected by the polymer concentration in the range from 50 to 2,000 μg/ml, and was maintained up to 8 days in phosphate-buffered saline (pH 7.4), indicating high colloidal stability. The release of adriamycin from self-aggregates was significantly dependent on the pH of the medium due to the cis-aconityl linkage; e.g., the amount of adriamycin released for 4 days was 7.3±0.3% at pH 7, whereas it was 29.3±1.9% at pH 4. The cell viability results demonstrated that free adriamycin shows more potent cytotoxicity than the conjugates, primarily attributed to the sustained release of adriamycin from self-aggregates. In conclusion, the self-aggregates, formed by GC–ADR conjugates, might be useful for the site-specific delivery of adriamycin in a sustained manner.  相似文献

A simple and fast flow injection fluorescence quenching method for the determination of iron in water has been developed. Fluorimetric determination is based on the measurement of the quenching effect of iron on salicylic acid fluorescence. An emission peak of salicylic acid in aqueous solution occurs at 409 nm with excitation at 299 nm. The carrier solution used was 2 × 10−6 mol L−1 salicylic acid in 0.1 mol L−1 NH4+/NH3 buffer solution at pH 8.5. Linear calibration was obtained for 5–100 μg L−1 iron(III) and the relative standard deviation was 1.25 % (n = 5) for a 20 μL injection volume iron(III). The limit of detection was 0.3 μg L−1 and the sampling rate was 60 h−1. The effect of interferences from various metals and anions commonly present in water was also studied. The method was successfully applied to the determination of low levels of iron in real samples (river, sea, and spring waters).  相似文献

Olive cake as low-cost abundantly available sorbent has been characterized by N2 at 77 K adsorption, porosity analysis, elemental analysis and IR spectra and has been used for preconcentrating of uranium(VI) and thorium(IV) ions prior to their determination spectrophotometrically. The optimum pH values for quantitative sorption of U(VI) and Th(IV) are 4–7 and 3–7, respectively. The enrichment factor for the preconcentration of U(VI) and Th(IV) were found to be 125 and 75 in the given order. The sorption capacity of olive cake is in the range of 2,260–15,000 μg g−1 for Th(IV) and in the range of 1,090–17,000 μg g−1 for U(VI) at pH 3–7. The sorbent exhibits good reusability and the uptake and stripping of the studied ions were fairly rapid. The elution of U(VI) and Th(IV) was performed with 0.3–1 M HCl/1–2 M HNO3 and 0.3–0.8 M HCl/1 M HNO3, respectively. The precision of the method was 1.8 RSD% for U(VI) and 2.5 RSD% for Th(IV) in a concentration of 1.00 μg mL−1 for 10 replicate analysis. The influence of some electrolytes and cations as interferents was discussed. Separation of U(VI) and Th(IV) from other metal ions in synthetic solution was achieved.  相似文献
