A liquid chromatography/mass spectrometry (LC/MS) method for amnesic shellfish poisoning toxins in shellfish was developed and validated. Tissue homogenate (4 g) was extracted with 16 mL methanol-water (1 + 1, v/v). Dilution into acetonitrile-water (1 + 9, v/v) was followed by C18 solid-phase extraction cleanup. Domoic acid (DA) and epi-domoic acid were determined by LC/MS/MS with electrospray ionization and multiple reaction monitoring. External calibration was performed with dilutions of a certified reference standard. Advantages of this method include speed, lower detection limits, and a very high degree of specificity. The LC/MS response was highly linear, and there were no significant interferences to the determination of DA. Formal method validation was performed on 4 shellfish species. Fortification studies gave recoveries (mean +/- SD; n = 24) of 93 +/- 14% at 1 mg/kg, and 93.3 +/- 7.6% at 20 mg/kg over all the species. Analysis of a mussel certified reference material showed the bias as < 5%. The limits of detection and quantitation were 0.15 and 0.5 mg/kg, respectively. Routine application of the method over 4 months gave a recovery for the QC sample (1 mg/kg fortified blank mussel homogenate) run with each batch of 88.9 +/- 5.5% (mean +/- SD; n = 37). The total uncertainty of measurement results were estimated as 0.12 (12%) at 0.25-5 mg/kg and 0.079 (7.9%) at 5-50 mg/kg. The major contribution to the uncertainty was the repeatability of the LC/MS determination, probably arising from subtle matrix effects.  相似文献

A new high-performance liquid chromatography method was developed and validated for the quantitation of gliclazide and repaglinide in pharmaceutical formulations. Determination was performed using a LiChroCART RP-18 column, a mobile phase containing acetonitrile-phosphate buffer (pH 2.1; 60 + 40, v/v), and ultraviolet (UV) detection at 225 nm. Repaglinide was used as an internal standard for gliclazide determination and gliclazide for repaglinide assay. The method was validated with respect to linearity, precision, robustness, ruggedness, accuracy, and specificity. The calibration graphs ranged from 0.015 to 0.09 mg/mL for gliclazide and 0.06 to 0.36 mg/mL for repaglinide. Intra- and interday relative standard deviation values for the standard solutions were 0.70 and 1.01% for gliclazide and 0.78 and 0.93% for repaglinide, respectively. Total recoveries of gliclazide and repaglinide from the laboratory-prepared mixtures were 99.82 +/- 0.58 and 101.50 +/- 0.46% for gliclazide and repaglinide, respectively [mean +/- standard deviation (SD)]. In forced degradation studies, the effect of acid, base, oxidation, UV light, and temperature on both drugs was also investigated. Finally, the method was applied for the quality control of commercial gliclazide and repaglinide tablets. Total recovery was 100.40 +/- 0.35 and 104.46 +/- 0.23% for gliclazide and repaglinide, respectively (mean +/- SD).  相似文献

建立了测定化妆品中硝基甲烷的固相萃取-气相色谱-质谱分析方法。膏霜、水剂、散粉、香波、唇膏等不同类型的化妆品样品加入甲醇或无水乙醇超声提取后,提取液高速离心处理,浓缩上清液,采用Sep-Pak Silica固相萃取柱净化,收集80%甲醇-20%二氯甲烷洗脱液,浓缩后经无水硫酸钠脱水,进行气相色谱-质谱定性及定量分析。选用HP-INNOWax石英毛细管柱(30m×0.25mm×0.25μm),程序升温,流速1.0mL/min;采用电子轰击电离源,选择监测离子(m/z30、46、61,其中61为定量离子),外标法定量。方法的平均回收率为83.0%~96.4%,相对标准偏差(RSD)为1.0%~5.3%,方法对硝基甲烷的检出限为0.2mg/kg。该方法准确、灵敏度高,可用于化妆品中硝基甲烷的测定。  相似文献

建立了快速测定铅笔涂层中18种光引发剂的气相色谱-质谱联用法(GC/MS)。样品以二氯甲烷为提取溶剂,在30℃温度下,超声提取30 min后,萃取液用100 mg N-丙基乙二胺(PSA)分散固相萃取净化,以气相色谱-质谱法(GC-MS)测定,外标法定量。18种光引发剂在0.2~4.0 mg/L(或0.1~4.0 mg/L)范围内线性良好,相关系数R2均大于0.997,方法检出限(LODs)为0.4~1.0 mg/kg,定量限(LQDs)为1.0~2.0 mg/kg。3个加标水平(2.0或4.0、10.0和40.0 mg/kg)的回收率在75.2%~115.2%之间,相对标准偏差(RSDs)为1.0%~9.3%(n=6)。该方法可用于铅笔涂层中18种光引发剂含量的高通量检测。  相似文献

建立了微波萃取/GC-MS法检测电子电气产品中多氯萘的方法。将破碎后的样品以甲苯为溶剂进行微波萃取,萃取液经硅胶小柱净化、平行蒸发定量浓缩后采用气相色谱-质谱法测定多氯萘的含量,外标法定量。优化了样品前处理条件,包括前处理方式、微波萃取溶剂、萃取时间,以及净化小柱的选择。对于8种含不同氯原子数的多氯萘,在1~50 mg/L范围内,线性关系良好,相关系数均大于0.995;方法定量下限为0.2~1.0 mg/kg,相对标准偏差为2.7%~7.2%,回收率为77%~112%。该法操作简便、分析迅速、结果准确,可以满足对电子电气产品中多氯萘的定性确证和定量分析。  相似文献

A multi-residue method has been developed for determining 65 pesticide residues in greasy vegetable matrices such as avocado. Conventional organic solvent extraction assisted by a high-speed homogenizer was compared to pressurized liquid extraction (PLE) as extraction techniques. Following this, the lipophilic extract was purified using gel permeation chromatography (GPC). Alternative clean-up methods were also evaluated, as solid-phase extraction cartridges individually used and downstream coupled, but less effective lipophilic separation was archived. The pesticide residue determination was carried out using low-pressure gas chromatography coupled to tandem mass spectrometry (LP-GC-MS-MS), showing the applicability of this type of GC columns for the analysis of fat vegetable matrices. The proposed methodology was validated in avocado matrix. The recoveries were in the range 70-110%, with RSD values lower than 19%, at 12 and 50 microg/kg spiking levels. The limits of quantitation (LOQs) were in the range 0.04-8.33 microg/kg and the limits of detection (LODs) were between 0.01 and 2.50 microg/kg. All of them were lower than the maximum residue levels (MRLs) set by the European Union (EU) in avocado. The proposed method was evaluated analyzing pesticide residues in real avocado samples.  相似文献

A method was developed to determine 4-hexylresorcinol in shrimp meat. The procedure is based on extraction of test portions with methanol followed by liquid chromatographic analysis of the extracts, using a reversed-phase column and fluorimetric detection (excitation: 280 nm, and emission: 310 nm). The confidence interval of the recovery in working range of 1.5-2.5 mg/kg was 81.6 +/- 0.8%. The relative standard deviation in the working range was 2.1%. Limits of quantitation and detection were 6.59 and 1.98 ng/mL extract, respectively, corresponding to 0.26 and 0.08 mg/kg in shrimp.  相似文献

A method has been developed for the quantitation of imazaquin residues in soil. The herbicide was extracted from soil with methanol-water (2 + 1, v/v) and cleaned up by strong anion-exchange solid-phase extraction cartridges. Analysis was performed by using high-performance liquid chromatography with ultraviolet detection. Average recoveries through the method ranged from 90.7 to 100.6%, with relative standard deviation equal to or lower than 6.6%. The limit of detection was estimated to be 0.0015 mg/kg, and the minimum quantitation concentration of imazaquin in soil was 0.005 mg/kg. This method was successfully applied to evaluate imazaquin residue levels in soil and its dissipation rates in a soybean field in the Xisanqi District of Beijing, People's Republic of China. The dissipation study showed that the half life of imazaquin in soil was 10.37 +/- 0.0135 days at 3 different application rates.  相似文献

An extraction method for the quantitative analysis of polybrominated diphenyl ethers (PBDEs) in aqueous samples has been evaluated. The analytical methodology includes the sample filtration through glass fiber filter and subsequent extraction of dissolved phase compounds by C18 solid-phase disk extraction. Dependence of extraction efficiency on factors such as pollutant concentrations, sample volume, and stability during storage has been investigated. Mean extraction efficiencies of 97% for total PBDEs (13 tri- to heptabrominated congeners at spiking levels in the range of 15-90pg) with a RSD between 9 and 20% were achieved. Higher recoveries were observed for the more volatile PBDEs (112%) in relation to more brominated congeners (88%). The developed methodology was successfully applied to the analysis of these compounds in atmospheric deposition and snow samples from remote sites in Europe with method detection and quantitation limits in the range of 2.1-10pgL(-1) for almost all congeners, which allow the determination of PBDEs in remote areas with levels in the range of low to medium pgL(-1) for SigmaPBDEs.  相似文献

We developed a high-throughput method based on on-line solid-phase extraction liquid chromatography tandem mass spectrometry (SPE-LC-MS/MS) to determine N-terminal thymosin-β fragment peptide (N-acetyl-seryl-aspartyl-lysyl-proline, Ac-SDKP) in human plasma samples. Quantification of Ac-SDKP was performed using direct injection for on-line SPE based on C(18), reversed-phase LC separation and stable isotope dilution electrospray ionization-MS/MS in multiple reaction-monitoring (MRM) mode. The Ac-SDKP-(13)C(6), (15)N(2) (m/z 496 → 137) was synthesized for the internal standard. The MRM ion for Ac-SDKP was m/z 488 → 129 (quantitative ion)/226. The limit of detection and lower limit of quantitation were 0.05 and 0.1 ng/mL in standard solution, respectively. Recovery values were 98.3-100.4% with inter-day (relative standard deviation, RSD, 0.4-14.1%) and intra-day (RSD, 0.8-19.7%) assays. This method was applied to the measurement of Ac-SDKP levels in plasma from hemodialyzed subjects. Concentrations were 0.59 ± 0.23 ng/mL (pre-hemodialyzed subjects, n = 9) and 0.44 ± 0.19 ng/mL (post-hemodialyzed subjects, n = 9). All plasma Ac-SDKP levels were decreased by dialysis. Thus, plasma Ac-SDKP was decreased through dialysis in chronic kidney disease. The findings in this study will be useful for the treatment of anemia in chronic kidney disease with dialysis.  相似文献

This paper describes a sensitive and selective liquid chromatography with tandem mass spectrometry (LC-MS/MS) method for determination of the novel survivin suppressant YM155, 1-(2-methoxyethyl)-2-methyl-4,9-dioxo-3-(pyrazin-2-ylmethyl)-4,9-dihydro-1H-naphtho[2,3-d]imidazolium, which is developed for the treatment of solid tumors. This method uses a liquid-liquid extraction from 0.25 mL of dog plasma. LC separation was carried out on a Genesis Silica column (50 mm x 3.0 mm i.d.) at a flow-rate of 0.5 mL/min. Compounds were eluted using a mobile phase of 5 mm ammonium acetate and 0.1% formic acid in water-0.1% formic acid in acetonitrile, 17:83 (v/v). MS/MS detection was carried out with an MDS-Sciex API3000 triple quadrupole mass spectrometer in positive electrospray ionization mode. The standard curve was linear from 0.05 to 50 ng/mL (r > or = 0.9968). The lower limit of quantitation was 0.05 ng/mL. Good intra- and inter-day assay precision (within 7.4% RSD) and accuracy (within +/-12.3%) were obtained. The extraction recovery was 66.2%. The method was successfully applied to preclinical pharmacokinetic studies in dogs.  相似文献

A very fast chromatographic separation of isoflavonoids genistein, daidzein, formononetin and biochanin A was developed on a C18 high-speed column under isocratic conditions. The method was validated in terms of detection limits, quantitation limits (LOQs), linearity and precision. LOQs in 0.04-0.2 microg/g range were calculated, making feasible the determination of these compounds of nutritional concern at trace levels. Good linearity was demonstrated over three concentration orders of magnitude for each analyte (r2 0.990-1.000). The intra-day and inter-day repeatability was evaluated in terms of relative standard deviation (RSD%) at two concentration levels for each analyte (RSD% <9%). An optimization strategy was adopted to find the best conditions for the extraction of isoflavonoid aglycones from yellow soybeans using microwave-assisted extraction. The most relevant parameters resulted to be the microwave power, the extraction time and the acid concentration, optimal values being 600 W, 1 min and 12 M, respectively. When performing sample treatment on a fortified soybean sample, high recovery percentage was obtained for both compounds (94+/-8% for daidzein and 97+/-5% (n = 4) for genistein). The concentration level at which daidzein and genistein were found in the soybean sample were 1.21+/-0.15 mg/g and 2.38+/-0.09 mg/g (n=4), respectively.  相似文献

This article describes and critically evaluates a complete method for the quantitation of PCB in sediment samples. The extraction and clean-up procedure is described. Packed column and capillary column results are compared. Capillary column quantitation yields lower values for total PCB loading. Preferences for capillary column quantitation are discussed and explained. Capillary quantitation is based on the combination of an external Aroclor standard and an internal decachlorobiphenyl standard for normalizing data. In light of the recent report of synthesis of all 209 PCB congeners a suggestion is made to use these as absolute standards to establish a bank of primary standard Aroclors.  相似文献

A procedure for the enzymatic determination of alpha-glycerophosphate (alpha-GP) has been developed, using an automated in-house FIA system, with immobilized glycerol-3-phosphate oxidase (GPO) on non-porous glass beads, following optimization of the immobilization and analytical parameters. Fabricated single bead string reactors (SBSR) were used in connection with the FIA system, following optimization of its parameters. The half-life of GPO-SBSR regarding reduction of the enzyme activity was found to be 110 days for its use in 20 triplicate measurements daily and storage at 4 degrees C in the appropriate buffer. The regression equation of the calibration graph for the determination of alpha-GP was: A(max)=(10+/-2)x10(-4)+(22 134+/-12)x10(-4) (mmol l(-1)alpha-GP). The lower limit of quantitation was 0.74 mumol l(-1)alpha-GP and the RSD of the method 0.05% (r=0.9999). The same FIA system and procedure can be also used for the determination of the GPO activity, with the alpha-GP as substrate. The regression equation for this calibration graph was: A(max)=(23+/-18)x10(-4)+(190+/-1)x10(-4) (mug ml(-1) GPO), the lower limit of quantitation was 0.782x10(-3) mg ml(-1) (0.782 ppm) GPO and the RSD of the method 0.53% (r=0.9999). Serum samples obtained from hospitalized patients were deproteinized by gel filtration and analyzed under pseudo-first order conditions, at various concentrations of alpha-GP. A kinetic study of the reduction of alpha-GP in serum versus time is given and an observed reaction rate constant k(ob)=106.5x10(-4) min(-1) was determined.  相似文献

This paper describes the validation of a micellar electrokinetic capillary chromatography method for the direct determination of the 3-O-glucuronides of entacapone and its (Z)-isomer, the main urinary metabolites of entacapone in humans. Entacapone is a novel drug which, as a potent inhibitor of catechol-O-methyltransferase (COMT), is used as an adjunct in the standard therapy of Parkinson's disease. The 3-O-glucuronide of another COMT inhibitor, nitecapone, was used as internal standard (I.S.). The validation experiments were performed by using spiked urine samples that were extracted with Sep-Pak C18 cartridges before analysis. Determinations were carried out in a buffer of pH 7.0 containing 25 mM of phosphate, 50 mM of borate and 20 mM of sodium dodecyl sulfate, and by applying 15 kV over a 67 cm (60 cm to the detector) x 75 microns fused-silica capillary. UV detection was at 335 nm. The validity of the method was assessed by investigating the identity of the analytes, selectivity, limit of quantitation, linearity, within-day precision, extraction recovery, between-day precision and accuracy, electroosmotic flow stability and analyte stability. The method proved to be reproducible, sufficiently selective and accurate. Extraction recoveries of the analytes were > 94%. The limit of quantitation (LOQ) was 2 micrograms/ml and the assay was linear in the range 2-150 micrograms/ml with correlation coefficients better than 0.999 for both glucuronides. The repeatability of the method, expressed as the ratio of corrected peak area of the analytes to that of I.S., gave RSD values of < 5% even at the LOQ. Between-day precision (RSD) was < 7.5% for both glucuronides at 7.5 micrograms/ml. Determination of the glucuronide concentrations in urine samples of 34 patients treated with entacapone either orally (200 mg) or intravenously (25 mg) showed the method to be suitable for monitoring the concentrations of the glucuronide of entacapone after both oral and intravenous administration and those of the glucuronide of its (Z)-isomer after oral administration. The limited long term stability of the system requires, however, frequent recalibration in applications involving long sample series.  相似文献

Fourteen extraction methods commonly cited in the literature were evaluated for the quantitation of methionine (Met) and selenomethionine (SeMet) in a yeast candidate certified reference material (CRM). Species specific isotope dilution (ID) gas chromatography-mass spectrometry (GC-MS) was utilized to effectively compensate for potential errors, such as losses during derivatization and clean up steps. Despite different extraction methods, the same derivatization procedure using methyl chloroformate was applied with a single exception, which was based on digestion with cyanogen bromide with 2% SnCl2 in 0.1 M HCl. Significant differences in measured Met and SeMet concentrations were obtained when different extraction methods were used. A 4 M methanesulfonic acid reflux digestion was found to be the most efficient for both analytes. Digestion with CNBr with 2% SnCl2 in 0.1 M HCl for the determination of SeMet showed the second highest extraction efficiency. Despite frequent use of enzymatic hydrolysis for the extraction of SeMet from yeast, very low extraction efficiencies for both analytes were obtained for four of eight tested methods. Among these, the highest extraction efficiencies for both analytes were obtained using 20mg pronase and 10mg lipase with incubation at 37 degrees C for 24 h. However, recoveries remained nearly 30 and 50% lower for Met and SeMet, respectively, compared to extraction with methanesulfonic acid. Lowest extraction efficiencies for both analytes were obtained when HCl or tetramethylammonium hydroxide (TMAH) digestions were used. Efficient extraction was also achieved using 200 mg (or 400 mg) of protease XIV with incubation at 37 degrees C for 72 h (or 24 h). Concentrations of 3331+/-45 and 3334+/-39 microg g(-1) (mean and one standard deviation, n = 4) for SeMet were obtained using 200 mg (72 h incubation) and 400 mg (24 h incubation) of protease XIV, respectively, in agreement with a value of 3404+/-38 microg g(-1) obtained using a methanesulfonic acid reflux.  相似文献

A method was developed for the direct determination of free methionine in soy-based infant formula, with analyte separation and quantitation by reversed-phase liquid chromatography (LC), and UV absorbance at 214 nm, respectively. Sample preparation required only dilution with mobile phase and syringe filtration. Using a 0.02M KH2PO4 mobile phase (pH adjusted to 2.9 with 85% o-phosphoric acid) and 0.7 mL/min flow rate, methionine eluted at approximately 8 min, and total run time was 14 min after column regeneration with acetonitrile-water. System linearity was demonstrated as peak area versus analyte concentration, ranging from 80 to 120% of the formula specification for free methionine (r > 0.999, and all residuals < 0.45%). Intermediate precision relative standard deviation values were < 1.5% for ready-to-feed and reconstituted powder samples, and recoveries ranged from 98.0 to 103.5% for inter-method comparison with an amino acid analyzer method. The limit of quantitation was 3 mg methionine/L in the "as fed" infant formula. Despite the relatively weak UV absorptivity of methionine, the 214 nm signal was sufficiently intense in the 30-65 mg/L (201-436 microM) range to afford quantitation by peak area proportionation versus a 2-point external standard calibration. This direct UV detection after reversed-phase LC separation provides a simple and accurate method for determining free methionine without derivatization.  相似文献
