A flow injection method was developed aimed at the determination of ascorbic acid in biological fluids, particularly fruit juices. The enzyme ascorbic oxidoreductase (EC 1.10.3.3), extracted fromCucurbita maxima, was immobilized onto alkylamine glass beads using glutaral-dehyde as a bifunctional agent. The ascorbic acid concentration was related to oxygen saturation. Fall in oxygen concentration, as a result of ascorbic acid oxidation, was detected by a low cost, homemade oxygen electrode. The calibration graph was linear over the range 0.05 to 3.00 mM (RSD 1%), the maximum number of samples that could be analysed was 90/h. The immobilized enzyme retained its initial activity for 2 mo with more than 600 assays.  相似文献

An electroanalytical strategy for the simultaneous determination of ascorbic acid (AA) and dehydroascorbic acid (DHA), is described. A palladized Al electrode is used for hydrodynamic amperometry of AA. While the decrease of anodic stripping voltammetry current of the K2UO2[Fe(CN)6]‐Pd/Al electrode prepared in the presence of DHA was the principal of the DHA determination. The calibration graph for both methods was linear over the concentration range 1–50 μM. The detection limit was found to be 0.5 μM. Some fresh fruit juices and vegetables of trace level of AA and DHA were analyzed as the typical example of application.  相似文献

A cresol red modified glassy carbon electrode was prepared using an electrochemical method. The cyclic voltammograms of the modified electrode indicate the presence of a couple of well-defined redox peaks, and the formal potential shifts in the negative direction with increasing solution pH. The modified electrode exhibits high electrocatalytic activity toward ascorbic acid oxidation, with an overpotential of 300 mV less than that of bare glassy carbon electrodes, and drastic enhancement of the anodic currents. The calibration graph obtained by linear sweep voltammetry for ascorbic acid is linear in the range of 50∼500 µM. The electrode markedly enhances the current response of dopamine and can separate the electrochemical responses of ascorbic acid and dopamine. The separation between the anodic peak potentials of ascorbic acid and dopamine is 190 mV by cyclic voltammetry. The linear sweep voltammetric peak currents for dopamine in the presence of 2 mM ascorbic acid vary linearly with a concentration of between 10 and 100 µM.  相似文献

Direct-current cyclic voltammetry is used to investigate the suitability of some ferrocene derivatives such as ferrocenecarboxylic acid, ferroceneacetic acid and ferrocenemethanol as mediators for ascorbic acid oxidation in aqueous solutions with low pH. The ascorbic acid coupled catalytically to three ferrocene derivatives exhibiting homogeneous second-order rate constants k(s), in the range 7.36 x 10(5) - 1.23 x 10(7). The catalytic oxidation peak current was linearly dependent on the ascorbic acid concentration and the linearity range obtained in the presence of ferrocenecarboxilic acid, having the largest second-order rate constant, was 5 x 10(-5) - 1.5 x 10(-3) M. The catalytic effect of the ferrocene derivatives on the electrochemical oxidation of ascorbic acid reduced the oxidation potential of ascorbic acid, resulting in the separation of the overlapping voltammograms of ascorbic acid and dopamine at the glassy carbon electrode in a mixture. This allowed the determination of ascorbic acid in the presence of dopamine. The calibration graph obtained by linear sweep voltammetry for ascorbic acid in the presence of dopamine of fixed concentration is linear in the range 5 x 10(-5) - 1.5 x 10(-3) M. In a similar manner, dopamine is determined in the presence of a high concentration of ascorbic acid, up to 100 times that of dopamine, using ferroceneacetic acid as the most suitable mediator for this purpose.  相似文献

A carbon paste electrode (CPE) modified by a monolayer film of sodium dodecyl sulfate (SDS) was used for detection of dopamine (DA). Cyclic voltammetry demonstrated improved response of the DA sensor. This suggests the effectivity of surface modification of CPE by SDS. Impedance spectroscopy was used for the characterization of CPE surface properties. The effect of SDS concentration on the electrode quality also reveals that SDS formed a monolayer on CPE surface with a high density of negative-charged end directed outside the electrode. As a result, the carbon paste electrode modified with SDS (SDS/CPE) exerted discrimination against ascorbic acid in physiological circumstance. Thus, it can selectively determine dopamine even in the presence of 220-fold AA combined with differential pulse stripping voltammetry. In pH 7.40 phosphate buffer solution, the oxidation peak current on differential pulse voltammograms increases linearly with the concentration of DA in the range of 5.0 x 10(-7) to 8.0 x 10(-4) mol . L(-1) with a detection limit of 5.0 x 10(-8) mol . L(-1). Satisfying results are achieved when detecting the DA in injection and simulated biology sample.  相似文献

Automatic ascorbic acid (AA) voltammetry was established in 24-well microtiter plates. The assay used a movable assembly of a pencil rod working, an Ag/AgCl reference and a Pt counter electrode with differential pulse voltammetry (DPV) for concentration-dependent current generation. A computer was in command of electrode (z) and microtiter plate (x, y) positioning and timed potentiostat operation. Synchronization of these actions supported sequential approach of all wells and subsequent execution of electrode treatment procedures or AA voltammetry at defined intervals in a measuring cycle. DPV in well solutions offered a linear current/concentration range between 0.1 and 8.0 mM, a sensitivity of about 1 μA mM−1 AA, and a detection limit of 50 μM. When used with a calibration curve or standard addition, automated voltammetry of samples with added known amounts of AA demonstrated good recovery rates. Also, the assay achieved the accurate determination of the AA content of vitamin C tablets, a fruit juice and an herbal tea extract. Robotic AA voltammetry has the advantage of conveniently handling multiple samples in a single measuring run without the continuous attention of laboratory personnel. It is a good option when the goal is cost-effective AA screening of sample libraries and has potential for applications in health care and the food processing, cosmetic and pharmaceutical industries.  相似文献

A carbon-paste electrode spiked with 1-[4-(ferrocenyl ethynyl)phenyl]-1-ethanone was constructed by the incorporation of 1-[4-(ferrocenyl ethynyl)phenyl]-1-ethanone in a graphite powder silicon oil matrix. It shown by cyclic voltammetry and double potential-step chronoamperometry, which this ferrocene derivative modified a carbon-paste electrode, can catalyze the ascorbic acid oxidation in an aqueous buffered solution. It has been found that under the optimum conditions (pH 7.00), the oxidation of ascorbic acid at the surface of this carbon paste modified electrode occurs at a potential of about 260 mV less positive than that of an unmodified carbon-paste electrode. The catalytic oxidation peak current was linearly dependent on the ascorbic acid concentration, and a linear calibration curve was obtained in the range of 6 x 10(-5) M-7 x 10(-3) M of ascorbic acid with a correlation coefficient of 0.9997. The detection limit (2sigma) was determined to be 6.3 x 10(-5) M. This method was also used for the determination of ascorbic acid in some pharmaceutical samples, such as effervescent tablets, ampoules and multivitamin syrup, by using a standard addition method. The reliability of the method was established by a parallel determination against the official method.  相似文献

Water present in undiluted acetic acid can be monitored in situ with Pt and Au microelectrodes using differential pulse and square‐wave voltammetries. Both reduction and oxidation peaks can be obtained. The best quantitative analytical results were obtained for the anodic peak, the Pt microelectrode, and differential pulse voltammetry. The anodic water peak is located at +1.55 V vs. the quasi reference Pt electrode. The voltammetric peak obtained at Pt electrode is apparently not a diffusional one, however, the calibration plot obtained by employing this peak is linear over a wide concentration range, up to 4 mol dm?3. The detection limit has been estimated as 2.3 mM or 0.0043 v/v%. The developed method may be particularly useful, since the Karl Fischer method can not be used to determine water in glacial acetic acid due to the estrification reaction of acetic acid with methanol. A voltammetric wave of undiluted acetic acid could not be obtained in the positive and negative ranges of potential.  相似文献

Self-doped polyaniline (PAN) film on platinum electrode surface has been synthesized via electrochemical copolymerization of aniline with orthanilic acid (OAA). Fourier transform infrared, UV–Vis, and elemental analysis indicate the formation of the copolymer and that the copolymer has the structure of a head-to-tail coupling of aniline and OAA units. It was found that the internal doping of PAN with OAA can extend the electroactivity of PAN in neutral and even in alkaline media. The obtained self-doped PAN (PAN-OAA)-coated platinum electrode is shown to be a good surface for the electrooxidation of ascorbic acid (AA) in phosphate buffer solution of pH 7. The anode peak potential of AA shifts from 0.63 V at bare platinum electrode to 0.34 V at the PAN-OAA-modified platinum electrode with greatly enhanced current response. A linear calibration graph is obtained over the AA concentration range of 5–60 mM using cyclic voltammetry. Rotating disk electrode voltammetry and chronoamperometry have been employed to investigate the electrooxidation of AA. The PAN-OAA-modified platinum electrode shows good stability and reproducibility.  相似文献

The present study reports the simultaneous determination of ascorbic acid (AA), dopamine (DA) and uric acid (UA) in phosphate buffer solution (pH 7.0) using graphene paste electrode modified with functionalized graphene sheets (GPE‐MFGSs). The presence of FGS inhibited the adsorption of AA owing to the electrostatic repulsion, but was favorable for the affinity adsorption of DA and UA via the ion exchange and hydrogen bonding mechanisms, respectively. This led to the decrease in the oxidation potential of AA and the significantly enhanced oxidation peak currents of DA and UA at the GPE‐MFGSs. By cyclic voltammetry and differential pulse voltammetry, the oxidation potentials of AA, DA, and UA, at the GPE‐MFGSs in a ternary mixture were found to be well resolved so that their simultaneous determination could be achieved. Furthermore, the influence of some experimental variables such as graphene paste composition, working solution pH, scan rate and pulse amplitude was studied. In addition, by differential pulse voltammetry, the linear dependence of peak current on the concentration was obtained in the ranges of 0.05–9.0, 0.03–13, and 0.03–5.5 µM with the lowest detection limits of 0.02, 0.01, and 0.01 µM for AA, DAand UA, , respectively.  相似文献

A chemically modified carbon paste electrode with 2,7‐bis(ferrocenyl ethyl)fluoren‐9‐one (2,7‐BFEFMCPE) was employed to study the electrocatalytic oxidation of ascorbic acid in aqueous solution using cyclic voltammetry, differential pulse voltammetry and chronoamperometry. The diffusion coefficient (D=1.89×10?5 cm2 s?1), and the kinetic parameter such as the electron transfer coefficient, α (=0.42) of ascorbic acid oxidation at the surface of 2,7‐BFEFMCPE was determined using electrochemical approaches. It has been found that under an optimum condition (pH 7.00), the oxidation of ascorbic acid at the surface of such an electrode occurs at a potential about 300 mV less positive than that of an unmodified carbon paste electrode. The catalytic oxidation peak currents show a linear dependence on the ascorbic acid concentration and linear analytical curves were obtained in the ranges of 8.0×10?5 M–2.0×10?3 M and 3.1×10?5 M–3.3×10?3 M of ascorbic acid with correlation coefficients of 0.9980 and 0.9976 in cyclic voltammetry and differential pulse voltammetry, respectively. The detection limits (2δ) were determined to be 2.9×10?5 M and 9.0×10?6 M with cyclic voltammetry and differential pulse voltammetry, respectively. This method was also examined for determination of ascorbic acid in pharmaceutical preparations.  相似文献

A simple, sensitive and reliable electrochemical sensor has been developed based on CuO nanostructures modified glassy carbon electrode for simultaneous determination of hydroquinone (HQ) and ascorbic acid (AA). The CuO nano material was synthesized by aqueous chemical growth method using different sources of OH. The characterization of nano material was performed by Fourier transform infrared spectroscopy, X‐ray diffraction, field emission scanning electron microscopy and energy dispersive X‐ray spectroscopy. The glassy carbon electrode was modified by CuO nano material using drop cast method and studied by cyclic voltammetry. The CuO/GCE exhibited excellent electrocatalytic activity towards the oxidations of HQ and AA in borate buffer solution (pH 8.0) and the corresponding electrochemical signals have appeared as two well resolved oxidation peaks with significant peak potential differences of (0.21V vs. Ag/AgCl). Differential pulse voltammetry was used for simultaneous determination of HQ and AA using the CuO/GCE. At the optimum conditions, for simultaneous determination by synchronous change of the analyte concentrations, the linear response ranges were between 0.0003–0.355 mM for HQ and 0.0001–0.30 mM for AA respectively. Furthermore, CuO/GCE was successfully applied for the independent determination of AA in fruit juices as well as for the simultaneous determination of HQ and AA in cosmetic samples.  相似文献

A selective dopamine determination using a nafion-coated clinoptilolite-modified carbon paste electrode in the presence of ascorbic acid was studied. Both cyclic voltammetry (CV) and differential pulse anodic stripping voltammetry (DPASV) were used for measurements of dopamine. To improve the selectivity of the clinoptilolite-modified carbon paste electrode in presence of a high concentration of ascorbic acid, the electrode surface was coated with nafion membrane. Experimental parameters affecting the determination of dopamine, including the clinoptilolite ratio, nafion membrane thickness, preconcentration time, preconcentration solution pH, stripping solution pH and interferences are discussed. The developed sensor has a wide linear range, a low detection limit, and good stability and reproducibility. The sensor offers a good alternative to existing analytical methods for dopamine, permits a relatively short analysis time, and is simple, selective and inexpensive.  相似文献

A simple modified TiO(2) nanotubes electrode was fabricated by electrodeposition of Pd, Pt and Au nanoparticles. The TiO(2) nanotubes electrode was prepared using the anodizing method, followed by modifying Pd nanoparticles onto the tubes surface, offering a uniform conductive surface for electrodeposition of Pt and Au. The performance of the modified electrode was characterized by cyclic voltammetry and differential pulse voltammetry methods. The Au/Pt/Pd/TiO(2) NTs modified electrode represented a high sensitivity towards individual detection of dopamine as well as simultaneous detection of dopamine and uric acid using 0.1 M phosphate buffer solution (pH 7.00) as the base solution. In both case, electro-oxidation peak currents of dopamine were linearly related to accumulated concentration over a wide concentration range of 5.0 × 10(-8) to 3.0 × 10(-5) M. However in the same range of dopamine concentration, the sensitivity had a significant loss at Pt/Pd/TiO(2) NTs electrode, suggesting the necessity for Au nanoparticles in modified electrode. The limit of the detection was determined as 3 × 10(-8) M for dopamine at signal-to-noise ratio equal to 3. Furthermore, the Au/Pt/Pd/TiO(2) NTs modified electrode was able to distinguish the oxidation response of dopamine, uric acid and ascorbic acid in mixture solution of different acidity. It was shown that the modified electrode possessed a very good reproducibility and long-term stability. The method was also successfully applied for determination of DA in human urine samples with satisfactory results.  相似文献

Cyclic voltammetry and differential pulse voltammetry were used to investigate the electrochemical behavior of ascorbic acid (AA) on palladium coated nanoporous gold film (PdNPGF) electrode. The deposition of palladium was done through oxidation of copper UPD layer by palladium ions. This low Pd‐loading electrode behaved as the nanostructured Pd for electrocatalytic reaction. The PdNPGF electrode exhibits excellent electrocatalytic behavior by enhancing the AA oxidation peak current due to synergistic influence of the Pd film and NPGF. The kinetic parameters such as electron transfer coefficient, α, was 0.47 and the voltammetric responses of the PdNPGF electrode were linear against concentration of AA in the ranges of 2.50–33.75 mM and 0.10–0.50 mM with CV and DPV respectively.  相似文献

The enzyme catalase (EC: 1.11.1.6) has been covalently coupled onto the surface of glassy carbon (GC) powder matrix using a 16 atom spacer arm. The enzyme coupled powder was made into a paste electrode that was used to study the electrochemical properties. Standard electrochemical techniques like cyclic voltammetry, differential pulse voltammetry and flow injection analysis studies were carried out using this paste electrode. The cyclic voltammogram of the modified paste exhibited a clear increase in the reduction peak at −180 mV in the presence of hydrogen peroxide. The potential at which maximum Faradaic activity was observed was determined using differential pulse voltammetry, which showed a clear peak at −100 mV. This potential was used to monitor the response of the electrode to varying substrate concentrations using a home made setup for flow injection analysis. A linear increase in the current values in the range 0.1–1 mM hydrogen peroxide concentration was observed in our system.  相似文献

A chemically modified electrode based on a chitosan-multiwall carbon nanotube (MWNT) coated glassy carbon electrode (GCE) is described, which exhibits an attractive ability to determine dopamine (DA) and ascorbic acid (AA) simultaneously. The modified electrode exhibited a high differential pulse voltammetry (DPV) current response to DA at 0.144 V and AA at -0.029 V (vs. SCE) in a 0.1 mol l(-1) phosphate buffer solution (pH = 7.2). The properties and behaviors of the chitosan-multiwall carbon nanotube modified electrode (MC/GCE) were characterized using cyclic voltammetry (CV) and DPV methods. The mechanism for the discrimination of dopamine from ascorbic acid at MC/GCE is discussed. The linear calibration range for DA and AA were 5 x 10(-7) mol l(-1) to 1 x 10(-4) mol l(-1) (r = 0.997), and 5 x 10(-6) mol l(-1) to 1 x 10(-3) mol l(-1) (r = 0.996), respectively. The MC/GCE showed good sensitivity, selectivity and stability.  相似文献

A simple, selective and precise voltammetric method for the determination of ascorbic acid in pharmaceutical preparations and fresh fruit juices-complex matrices containing various reducing compounds-is described. The method is based on the electrocatalytic oxidation of ascorbic acid in homogeneous solution using electrogenerated ferriciniumcarboxylic acid as mediator. The pH and mediator concentration affecting the performance of the electrocatalytic oxidation of the analyte were optimized. The method was applied to determine vitamin C in deeply coloured, viscous and turbid fruit juice samples with ascorbic acid contents ranging from 15-45 mg per 100 ml, without further dilution, concentration or other pre-treatment of the samples. The amount of mediator used varied depending on the ascorbic acid concentration in the samples. The method was also used for pharmaceutical analysis using a calibration graph. For fruit juice samples the standard addition technique was adopted to prevent the matrix affecting the accuracy of the determination. The relative standard deviation for the analysis of vitamin C in fruit juices ranged from 1.5-5%. The reliability of the method was established by parallel determination against the official methods.  相似文献

The voltammetric behaviour of the antioxidant tert-butylhydroxyanisole (BHA), at a carbon paste electrode modified with the electron mediator nickel phthalocyanine, is described, and a method for the determination of this antioxidant, based on its oxidation on the modified electrode, is proposed. Cyclic voltammograms showed a well-defined oxidation peak for BHA slightly shifted towards less positive potentials with respect to that obtained at the plain carbon paste electrode. The peak current measured at the modified electrode is considerably higher than that obtained at the unmodified electrode. A modifier percentage of 2%, a methanol percentage of 2% and a 0.1 mol/HClO(4) medium were chosen as working conditions. The i(p)vs v1 2 plot obtained by linear sweep voltammetry showed a linear relationship over the whole scan rate range studied (5-2000 mV/sec) which is typical of a diffusion-controlled current. Using differential pulse voltammetry at DeltaE = 50 mV, linear calibration graphs were obtained in the concentration ranges 1.0-30.0, 0.10-1.0 and 0.02-0.10 mg/l BHA. The detection limit was 0.0036 mg/l (2.0 x 10(-8) mol/l). Interferences from other substances commonly present in commercial antioxidant mixtures were tested. The developed method was applied to the determination of BHA in spiked potato flakes.  相似文献

A mercury-sensitive chemically modified graphite paste electrode was constructed by incorporating modified silica gel into a conventional graphite paste electrode. The functional group attached to the (3-chloropropyl) silica gel surface was 2-mercaptoimidazole, giving a new product denoted by 3-(2-thioimidazolyl)propyl silica gel, which is able to complex mercury ions. Mercury was chemically adsorbed on the modified graphite paste electrode containing 3-(2-thioimidazolyl)propyl silica (TIPSG GPE) by immersion in a Hg(II) solution, and the resultant surface was characterized by cyclic and differential pulse anodic stripping voltammetry. One cathodic peak at 0.1 V and other anodic peak at 0.34 V were observed on scanning the potential from -0.1 to 0.8 V (0.01 M KNO3; v = 2.0 mV s(-1) vs. Ag/AgCl). The anodic peak at 0.34 V show an excellent sensitivity for Hg(II) ions in the presence of several foreign ions. A calibration graph covering the concentration range from 0.02 to 2 mg L(-1) was obtained. The detection limit was estimated to be 5 microg L(-1). The precision for six determinations of 0.05 and 0.26 mg L(-1) Hg(II) was 3.0 and 2.5% (relative standard deviation), respectively. The method can be used to determine the concentration of mercury(II) in natural waters contaminated by this metal.  相似文献
