We completed the first total synthesis of puerarin (1), an isoflavone C-glycoside. The key intermediate, β-d-glucopyranosyl-2,6-dimethoxybenzene (9), was obtained by coupling of a lithiated aromatic reagent (3) with pyranolactone (2) in 56% yield. Condensation of (16) with p-methoxybenzaldehyde gave the chalcone (17). The protected chalcone (18) was cyclized to (19) in the presence of Tl(NO3)3. Demethylation of (19) was accomplished by refluxing with TMSI in CH3CN to give puerarin (1).  相似文献

(Z)-5-(2-(1H-Indol-3-yl)-2-oxoethylidene)-3-phenyl-2-thioxothiazolidin-4-one (7a-q) derivatives have been synthesized by the condensation reaction of 3-phenyl-2-thioxothiazolidin-4-ones (3a-h) with suitably substituted 2-(1H-indol-3-yl)-2-oxoacetaldehyde (6a-d) under microwave condition. The thioxothiazolidine-4-ones were prepared from the corresponding aromatic amines (1a-e) and di-(carboxymethyl)-trithiocarbonyl (2). The aldehydes (6a-h) were synthesized from the corresponding acid chlorides (5a-d) using HSnBu3.  相似文献

Ligands (2-pyridyl-2-furylmethyl)imine, (L1), (2-pyridyl-2-thiophenemethyl)imine (L2), and (2-pyridyl-2-thiopheneethyl)imine (L3) were synthesized by condensation reactions and obtained in good yields. Reactions of L1-L3 with either [PdClMe(cod)] or [PdCl2(cod)] gave the corresponding monometallic palladium(II) complexes 1-5 in very good yields. Molecular structures of complexes 1, 4 and 5 indicated that the ligands are bidentate and coordinate to the palladium metal through the imine and pyridine nitrogen atoms. When complexes 3-5 were treated with NaBAr4, cationic species, 3a, 4a, and 5a were produced which catalyzed polymerization of ethylene though with very low activities. 1H NMR spectroscopy studies showed that these cationic species were very stable in solution. DFT calculations showed high ethylene coordination barriers to the cationic species 3a, 4a and 5a.  相似文献

A series of 2-imino-1,4-benzoxazines (8a-e) were prepared by the one-pot, three-component, condensation of salicylaldehyde (4), various ortho-aminophenols (5), and 2,6-dimethylphenylisonitrile (6). The structures of four of the crystalline benzoxazine derivatives (8b-e) were unambiguously established by X-ray analysis.  相似文献

Spironaamidine (1), a unique spiroquinone-containing alkaloid, was isolated from the marine sponge, Leucetta microraphis, along with two known imidazole alkaloids, naamidine H (2) and (9E)-clathridine 9-N-(2-sulfoethyl)imine (3). Novel condensation reaction of tropone with N-substituted and N,N′-disubstitued barbituric acids in Ac2O afforded 5-(cyclohepta-2′,4′,6′-trienylidene)pyrimidine-2(1H),4(3H),6(5H)-trione derivatives (8a-f) in moderate to good yields. The 13C NMR spectral study of 8a-f revealed that the contribution of zwitterionic resonance structures is less important as compared with that of 8,8-dicyanoheptafulvene. The rotational barriers (ΔG) around the exocyclic double bond of mono-substituted derivatives 8a-c were obtained to be 14.51-15.03 kcal mol−1 by the variable temperature 1H NMR measurements. The electrochemical properties of 8a-f were also studied by CV measurement. Upon treatment with DDQ, 8a-c underwent oxidative cyclization to give two products, 7 and 9-substituted cyclohepta[b]pyrimido[5,4-d]furan-8(7H),10(9H)-dionylium tetrafluoroborates (11a-c·BF4 and 12a-c·BF4) in various ratios, while that of disubstituted derivatives 8d-f afforded 7,9-disubstituted cyclohepta[b]pyrimido[5,4-d]furan-8(7H),10(9H)-dionylium tetrafluoroborate (11d-f·BF4) in good yields. Similarly, preparation of known 5-(1′-oxocycloheptatrien-2′-yl)-pyrimidine-2(1H),4(3H),6(5H)-trione derivatives (14a-d) and novel derivatives 14e,f was carried out. Treatment of 14a-c with aq. HBF4/Ac2O afforded two kinds of novel products 11a-c·BF4 and 12a,c·BF4 in various ratios, respectively, while that of 14d-f afforded 11d-f. The product ratios of 11a-c·BF4 and 12a-c·BF4 observed in two kinds of cyclization reactions were rationalized on the basis of MO calculations of model compounds 20a and 21a. The spectroscopic and electrochemical properties of 11a-f·BF4 and 12a-c·BF4 were studied, and structural characterization of 11c·BF4 based on the X-ray crystal analysis and MO calculation was also performed.  相似文献

The Pd(OAc)2-catalyzed oxyarylation of electron-rich (8 and 12) and electron-poor (10) olefins by ortho-iodophenols (3a-d) was studied using Ag2CO3 as the base, in acetone, and in the presence and absence of PPh3. The corresponding adducts of oxyarylation were obtained in moderate yields. The reaction mechanism was examined by electrospray ionization mass spectrometry (ESI-MS). Cationic arylpalladium intermediate (14), formed by the oxidative insertion of Pd(0) into 3a, and the cationic palladacycles (15), obtained by reaction of 14 with olefins 8 and 12, were intercepted by ESI-MS and characterized by ESI-MS/MS.  相似文献

A new indole alkaloid, 7β-hydroxy-7H-mitraciliatine (1) and a new oxindole alkaloid, isospeciofoleine (2) together with nine known alkaloids were isolated from Mitragyna speciosa and characterized by NMR, CD, and MS spectroscopic data analyses. The 1H and 13C NMR spectroscopic data of isospeciofoline (3), isorotundifoline (4), paynantheine (5), and 3-isopaynantheine (6) were also reported for the first time.  相似文献

1-Fluoroindan-1-carboxylic acid (FICA) (1) was designed and synthesized as its methyl ester (FICA Me ester) (4) in order to develop an efficient chiral derivatizing agent (CDA) which excels α-methoxy-α-(trifluoromethyl)phenylacetic acid (MTPA) in capability. FICA Me ester (4) was prepared by fluorination of methyl 1-hydroxyindan-1-carboxylate (3) with (diethylamino)sulfur trifluoride (DAST) and derived to the esters of racemic secondary alcohols by ester exchange reaction. The resulting ΔδF value was large in the case of 2-butyl ester of FICA (5a), whereas not detectable in the case of the corresponding MTPA ester (6a). The magnitude of the ΔδH values was similar to that of MTPA esters. The diastereomers of (R)-(−)-8-phenylmenthyl ester of FICA (5i) was separated and their 1H NMR analyses revealed that the concept of the modified Mosher's method was successfully applied to 5i.  相似文献

Three new aromatic sesquiterpenes (1, 2, and 4), one new dimeric sesquiterpene of the cyclolaurane-type (3), one sesquiterpene alcohol of bisabolene type (8) along with three previously reported metabolites (5-7), were isolated from the organic extracts of Laurencia microcladia, collected from the Chios island in the North Aegean Sea. The structures of the new natural products, as well as their relative stereochemistries, were established by means of spectral data analyses, including 2D experiments. The cytotoxicity of the isolated metabolites was evaluated against five human tumor cell lines.  相似文献

Reaction of acyl chlorides or acid anhydrides with isomeric 2:3 adducts derived from condensation of l-serine (1a), l-threonine (1b) and l-cysteine (1c) methyl esters with formaldehyde afforded N-acyl-pseudoprolines 7-19 in various yields. These 2:3 adducts can be considered as synthetic equivalents of oxaproline and thiaproline moieties. The present work revealed the versatile behaviour of the two 2:3 adducts as a consequence of the ring-chain tautomerism occurring in the five- and/or seven-membered rings.  相似文献

Starting from citraconic anhydride (13), a simple multistep (9-10 steps) synthesis of naturally occurring butyrolactones maculalactone A (3), maculalactone B (1), maculalactone C (2) and nostoclide I (4) have been described with good overall yields via dibenzylmaleic anhydride (20) and benzylisopropylmaleic anhydride (27). The two anhydrides 20 and 27 were prepared by SN2′ coupling reactions of appropriate Grignard reagents with dimethyl bromomethylfumarate (14), LiOH-induced hydrolysis of esters to acids, bromination of carbon-carbon double bond, in situ dehydration followed by dehydrobromination and chemoselective allylic substitution of bromoatom in disubstituted anhydrides 19 and 26 with appropriate Grignard reagents. The NaBH4 reduction of these anhydrides 20 and 27 furnished the desired lactones 21 and 29, respectively. The lactone 21 on Knoevenagel condensation with benzaldehyde, furnished maculalactone B (1), which on isomerization gave maculalactone C (2). Selective catalytic hydrogenation of 1 gave maculalactone A (3). The conversion of lactone 29 to nostoclide I (4) is known.  相似文献

An atom-efficient and environment-friendly approach for the synthesis of amidoalkyl naphthols (4a-x) via multicomponent one-pot reaction of 2-naphthol (1), aromatic aldehyde (2) and amide (3) catalyzed by P2O5 has been developed. Concomitant supplementation of a histone deacetylase inhibitor, suberoylanilide hydroxamic acid, and a DNA methyltransferase inhibitor, 5-azacytidine, to the culture medium of a plant endophytic fungus, Pestalotiopsis acaciae, dramatically altered its metabolic profiles. As a result, three novel aromatic compounds, 2′-hydroxy-6′-hydroxymethyl-4′-methylphenyl-2,6-dihydroxy-3-(2-isopentenyl)benzoate (1), 4,6-dihydroxy-7-hydroxymethyl-3-methylcoumarin (2) and 4,6-dihydroxy-3,7-dimethylcoumarin (3), were isolated, along with five known polyketides, endocrocin (4), pestalotiollide B (5), pestalotiopyrone G (6), scirpyrone A (7) and 7-hydroxy-2-(2-hydroxypropyl)-5-methylchromone (8).  相似文献

The synthesis and properties of a novel type of bis(heteroazulen-3-yl)methyl cations, bis(2-oxo-2H-cyclohepta[b]furan-3-yl)methyl cation salt and nitrogen analogues, (9a-c·PF6) and (9a-c·BF4), as well as bis(heteroazulen-3-yl)ketones (12a-d) are studied. The synthetic method was based on a TFA-catalyzed electrophilic aromatic substitution on the heteroazulenes (6a-d) with paraformaldehyde to afford the corresponding disubstituted methane derivatives 7a-d, followed by oxidative hydrogen abstraction with DDQ, and subsequent exchange of the counter-anion by using aq. HPF6 or aq. HBF4. In addition, the reaction of 7a-d with 2.2 equiv. amounts of DDQ afforded carbonyl compounds 12a-d. The delocalization of the positive charge of 9a-c was evaluated by the 1H and 13C NMR spectral data. The thermodynamic stability of cations 9a-c was evaluated to be in the order 9a<9b<9c on the basis of their reduction potentials measured by cyclic voltammetry (CV) and pKR+ values (2.6-10.3) obtained spectrophotometrically. The reduction waves of cations 9a-c were irreversible, suggesting the dimerization of the radical species generated by one-electron reduction. This was demonstrated by the reduction of 9a·BF4 with Zn powder to give dimerized product 14a. In addition, the quenching of 9a·BF4 with MeOH/NaHCO3 gives ether derivative 15a, which is proposed for the precursor for synthesizing tris(heteroazulene)-substituted methyl cations bearing two different heteroazulene-units.  相似文献

The complex [(η6-p-cymene)Ru(μ-Cl)Cl]21 reacts with pyrazole ligands (3a-g) in acetonitrile to afford the amidine derivatives of the type [(η6-p-cymene)Ru(L)(3,5-HRR′pz)](BF4)2 (4a-f), where L = {HNC(Me)3,5-RR′pz}; R, R′ = H (4a); H, CH3 (4b); C6H5 (4c); CH3, C6H5 (4d) OCH3 (4e); and OC2H5 (4f), respectively. The ligand L is generated in situ through the condensation of 3,5-HRR′pz with acetonitrile under the influence of [(η6-p-cymene)RuCl2]2. The complex [(η6-C6Me6)Ru(μ-Cl)Cl]22 reacts with pyrazole ligands in acetonitrile to yield bis-pyrazole derivatives such as [(η6-C6Me6)Ru (3,5-HRR′pz)2Cl](BF4) (5a-b), where R, R′ = H (5a); H, CH3 (5b), as well as dimeric complexes of pyrazole substituted chloro bridged derivatives [{(η6-C6Me6)Ru(μ-Cl) (3,5-HRR′pz)}2](BF4)2 (5c-g), where R, R′ = CH3 (5c); C6H5 (5d); CH3, C6H5 (5e); OCH3 (5f); and OC2H5 (5g), respectively. These complexes were characterized by FT-IR and FT-NMR spectroscopy as well as analytical data. The molecular structures1 of representative complexes [(η6-C6Me6)Ru{3(5)-Hmpz}2Cl]+5b, [(η6-C6Me6)Ru(μ-Cl)(3,5-Hdmpz)]22+5c and [(η6-C6Me6)Ru(μ-Cl){3(5)Me,5(3)Ph-Hpz}]22+5e were established by single crystal X-ray diffraction studies.  相似文献

Water-soluble p-sulfonatocalix[7]arene 1 has been synthesized in good yield through standard procedures and its conformational preferences have been investigated by Monte Carlo conformational searches. The acid-base properties of 1 were investigated by means of potentiometric titration, obtaining pKa values in agreement with those reported for other p-sulfonatocalix[n]arene homologs. The binding ability of 1 toward organic quaternary ammonium cations such as Diquat (2), Paraquat (3), and Chlormequat (4) was investigated by means of 1H NMR titrations in D2O at pD=7.3, DOSY NMR measurements, and 2D ROESY NMR spectroscopy. Spectrofluorimetry proved to be a useful method for the determination of trace amounts of 2 and 3 in aqueous solution by using Acridine Orange bound to 1 as a chemical indicator.  相似文献

Four new minor alkaloids, stryvomicine A (1), deoxy-isostrychnine-chloromethochloride (2), β-colubrine-chloromethochloride (3) and α-colubrine-chloromethochloride (4) were isolated from the seeds of Strychnos nux-vomica. Stryvomicin A possesses an unprecedented 6/5/7/8/6/7/6 heptacyclic ring system, which is the first example of the linkage of the C3–O–C26–N4 bond in strychnan-type alkaloids. Compound 5 induced the occurrence of autophagy and might have potential implications in autophagy related diseases.  相似文献

Three diruthenium(III) compounds Ru2(L)4Cl2, where L is mMeODMBA (N,N′-dimethyl-3-methoxybenzamidinate, 1a), DiMeODMBA (N,N′-dimethyl-3,5-dimethoxy benzamidinate, 1b), or DEBA (N,N′-diethylbenzamidinate, 1c), were prepared from the reactions between Ru2(OAc)4Cl and respective HL under reflux conditions. Metathesis reactions between 1 and LiC2Y resulted in bis-alkynyl derivatives Ru2(L)4(C2Y)2 [Y=Ph (2), SiMe3 (3), SiiPr3 (4) and C2SiMe3 (5)]. The parent compounds 1 are paramagnetic (S=1), while bis-alkynyl derivatives 2-5 are diamagnetic and display well-solved 1H- and 13C-NMR spectra. Molecular structures of compounds 1b, 1c, 2c, 3c and 4b were established through single crystal X-ray diffraction studies, which revealed RuRu bond lengths of ca. 2.32 Å for parent compounds 1 and 2.45 Å for bis-alkynyl derivatives. Cyclic voltammograms of all compounds feature three one-electron couples: an oxidation and two reductions, while the reversibility of observed couples depends on the nature of axial ligands.  相似文献
