For the direct measurement of electrophoretic mobility, multiple-point (Shah function) detected, time-domain detector signals were converted into frequency-domain plots by means of Fourier transformation. Multiple sample plugs (up to a maximum of three) were introduced into the separation channel and the resultant time-domain signals were then Fourier-transformed. The multiple-sample injection technique has been successfully demonstrated for a one-component system and a separation. Though the number of fluorescing zones flowing through the illuminated length of the channel is greater than the number of analytes in the solution, Shah convolution Fourier transform detection (SCOFT) is able to identify the number of fluorescent species in the solution based on their migration velocities. The height of the fundamental peak increases as the number of injected sample plugs is increased. More importantly, the signal-to-noise ratio (S/N) is found to be proportional to the number of injected sample plugs. With these findings, the multiple-sample injection technique certainly has got many potential applications in trace analysis. The technique would be equally applicable to other separation techniques (e.g., high-performance liquid chromatography) and detection methods (e.g., absorption, refractive index).  相似文献

The analysis of biological samples (e.g., blood, urine, saliva, tissue homogenates) by capillary electrophoresis (CE) requires efficient sample preparation (i.e., concentration and clean-up) procedures to remove interfering solutes (endogenous/exogenous and/or low-/high-molecular-mass), (in)organic salts and particulate matter. The sample preparation modules can be coupled with CE either off-line (manual), at-line (robotic interface), on-line (coupling via a transfer line) or in-line (complete integration between sample preparation and separation system). Sample preparation systems reported in the literature are based on chromatographic, electrophoretic or membrane-based procedures. The combination of automated sample preparation and CE is especially useful if complex samples have to be analyzed and helps to improve both selectivity and sensitivity. In this review, the different modes of solid-phase (micro-) extraction will be discussed and an overview of the potential of chromatographic, electrophoretic (e.g., isotachophoresis, sample stacking) and membrane-based procedures will be given.  相似文献

A butyl methacrylate-co-ethylene dimethacrylate (BuMA-co-EDMA) monolith was synthesized by UV initiated polymerization at the inlet end of a 75 microm I.D. fused silica capillary that had been previously coated with a protein compatible polymer, poly(vinyl)alcohol. The monolith was used for on-line preconcentration of proteins followed by capillary electrophoresis (CE) separation. For the analysis of standard proteins (cytochrome c, lysozyme and trypsinogen A) this system proved reproducible. The run-to-run %RSD values for migration time and corrected peak area were less than 5%, which is typical of CE. As measured by frontal analysis using lysozyme as solute, saturation of a 1cm monolith was reached after loading 48 ng of protein. Finally, the BuMA-co-EDMA monolithic preconcentrator was coupled to a protein G monolithic column via a zero dead volume union. The coupled system was used for on-line removal of IgG, preconcentration of standard proteins and CE separation. This system could be a valuable sample preparation tool for the analysis of low abundance proteins in complex samples such as human serum, in which high abundance proteins, e.g., human serum albumin (HSA) and immunoglobulin G (IgG), hinder identification and quantification of low abundance proteins.  相似文献

On-line gas diffusion has been coupled to a capillary electrophoresis system (CE) via a specially designed interface. The sample is merged with a modifying solution, e.g., a strong acid, in a flow system to transform the analytes of interest into their respective gaseous forms. These transformed, gaseous analytes permeate through a PTFE membrane into an acceptor stream comprising of a tris-buffer. The continuously flowing acceptor stream is led into an injector forming an integrated part of a flow injection analysis (FIA) system. The sample receiving carrier stream in the FIA system, a chromate buffer, brings the sample, 50 mul, to the FIA-CE interface into which one end of a separation capillary has been inserted. A small portion of the injected sample enters the capillary (electrokinetic injection) and separation takes place. A UV detector is placed at the other capillary end and a run potential of 25 kV is applied to two platinum electrodes positioned in the flow system. Multiple sample injections can be performed in one uninterrupted electrophoretic run. A typical sampling frequency is 15 h(-1); each run may result in quantitation of at least five anions. The overall repeatability is in the range 1.8-3.6% (RSD). The technique has been applied to the analysis of real samples such as soft drinks, vinegar and wine. Selective discrimination of anions which are unable to form volatile species is accomplished. No off-line sample pre-treatment is needed.  相似文献

Capillary electrophoresis (CE) with a water-soluble ion-exchange polymer in the background electrolyte is very efficient for the separation of organic and inorganic anions because the ion-exchange selectivity, as well as differences in electrophoretic mobility, can be used for separating sample ions. Poly(diallyldimethylammonium chloride) (PDDAC) was employed for this purpose. A very stable electroosmotic flow was obtained between pH 2.3 and 8.5 due to the strong adsorption of PDDAC onto the capillary wall. The effect of ion exchange on the migration of sample anions and their separation was controlled by varying the concentration of PDDAC, the concentration and the type of salt used in the CE background electrolyte. Addition of organic solvent (e.g., acetonitrile) could also modify the sample migration and the separation. Baseline separations were obtained for anions with very similar mobilities, such as bromide and iodide, naphthalenesulfonates, and bi- and tricarboxylic acids. Typical separation efficiencies were between 195,000 and 429,000 theoretical plates per meter. Ten replicate separations gave an average RSD of 1.0% for migration times of the sample anions studied. Excellent separations were obtained for a variety of samples, including a separation of 17 inorganic and organic anions in less than 6 min.  相似文献

The present review highlights some modern aspects of biosensor revelation, a detection method which has already found a large number of applications in healthcare, food industry and environmental analysis. First, the concept of bio-recognition, which is at the heart of biosensor technology, is discussed, with emphasis on host-guest-like recognition mechanisms. This detection device has been successfully coupled, in its first applications, to chromatographic columns, which allow a high resolution of complex mixtures of analytes prior to interaction with the biosensing unit. The properties of the transducing elements, which should generate a signal (e.g., electrochemical, thermal, acoustic, optical) of proper intensity and of relative fast rise, are additionally evaluated and discussed. The review then focuses on potential applications of biosensing units in capillary electrophoresis (CE) devices. CE appears to be an excellent separation methodology to be coupled to biosensor detection, since it is based on miniaturized electrophoretic chambers, fast analysis times, complete automation in sample handling and data treatment and requires extremely small sample volumes. Although only a few applications of CE-based biosensors have been described up to the present, it is anticipated that this hyphenated technique could have a considerable expansion in the coming years.  相似文献

Analyses of cytokinins are very important in both plant physiological and biomedical research as they are implicated in many biological processes. Reliable, sensitive, selective and inexpensive methods that are flexible and designed for automation are required for these analyses. This review addresses the advances made in the separation and determination of cytokinins by CE as well as the other applications of CE (i.e., determination of dissociation constants and complexation constants of cytokinins). The various CE modes used to separate the compounds and the quantification strategies are examined. Special attention is also focused on those aspects that improve on the sensitivity and/or selectivity, such as sample extraction and preconcentration, on-line preconcentration techniques (stacking), and/or specific detectors (e.g., MS). With the coupling to the preconcentration techniques and certain detection systems, numerous CE methods can potentially be adapted for the analysis of cytokinins in complex biological samples. Therefore, we would anticipate wider applications of CE methods in the near future for cytokinin analyses, which should facilitate a decrease in analysis cost and should help to improve analysis efficiency.  相似文献

Fast capillary electrophoresis (CE) hyphenated to time-of-flight mass spectrometry (TOF-MS) of four organoarsenic species (glycerol oxoarsenosugar, sulfate oxoarsenosugar, arsenobetaine, arsenocholine) are presented using short length CE capillaries under high electric field strengths of up to 1.3 kV cm(-1) with small inner diameter (ID). The separation of arsenosugars by CE is demonstrated for the first time. An aqueous formic acid solution was employed as the background electrolyte (BGE) for the separation. Various acid concentrations were evaluated for their influence on migration times, separation efficiency as well as with regard to controlling the charge of the arsenic species. A 0.1 M formic acid/ammonium formate buffer (pH 2.8) proved to be suitable for the separation of the four species. A non-aqueous BGE was tested as an alternative buffer system for fast speciation analysis. Separation of arsenobetaine and arsenocholine could even be achieved within 10 s by pressure-assisted CE. Application of the optimized method for the analysis of extracts of a seagrass and a Wakame algae sample as well as the brown algae homogenate reference material IAEA-140/TM revealed a clear signal for the glycerol arsenosugar.  相似文献

Dual opposite injection capillary electrophoresis (DOI-CE) is a family of CE techniques in which the sample is introduced into both ends of the capillary. For the analysis of compounds with widely varying pKa values using a voltage-driven separation scheme, DOI-CE is superior to conventional CE with sample introduction at only one end of the capillary due to DOI-CE's broader elution window. To enhance the DOI-CE separation, a running buffer with a microemulsion system was developed. Since DOI-CE works best under conditions of low electroosmotic flow (EOF), the suppression of EOF via the addition of a multiply charged cation (e.g., Zn2+) to the buffer was investigated, and was found to suppress the EOF effectively at moderate concentrations (2.5-10 mM). Three different dual opposite injection modes were studied: simultaneous electrokinetic injection, sequential electrokinetic injection, and sequential hydrodynamic injection. The injection bias in the first two electrokinetic injection modes was compared with the sequential hydrodynamic injection. Corrections in the bias of the electrokinetic injections were discussed, and an improved approach was suggested. Finally, the effect of the relative concentration of the multiply charged cation in the sample plug and running buffer on the peak shape of co-electroosmotic and counter-electroosmotic ions was examined, and found to be much more influential on the latter.  相似文献

This paper aimed to build up a sensitive CE method for the analysis of tetracyclines (TCs) antibiotics (including tetracycline, chlorotetracycline, oxytetracycline, and doxycycline) with conventional UV detection. Here, the large volume sample stacking was applied to achieve in capillary preconcentration of the targets. To achieve large volume sample stacking, the essential step was a large volume of sample (around 83.3% of total capillary length from inlet to detection window) hydrodynamically loaded. Then, the reserved voltage was added in order to push the sample matrix out of the capillary. Due to different pH between sample solution (pH 4.6) and BGE (pH 11.0), the cationic TCs would turn into negatively charged while the sample matrix was removing from the capillary. Finally, the anionic TCs were stacked at the inlet for the subsequent separation. Although the loss of sample existed during their charge transformation, the LODs could be improved around 40 times than that obtained by normal hydrodynamic injection CE method. Here, the LODs were in the range of 8.1–14.5 μg/L, around 10 ppb that close to the level by electrochemiluminescence or laser‐induced fluorescence detection of TCs by CE. The precision was characterized by RSDs of migration times and peak areas, which were in the range of 0.19–0.24% and 0.97–2.54%, respectively. The recoveries of the developed method were in the range of 95–112% by spiking TCs in the tap water. The proposed inline preconcentration CE method could be a simple, speed, and sensitive method for the quantitative analysis of TCs.  相似文献

The Hadamard transform (HT) technique, which permits the S/N in CE to be improved, was applied to MEKC. Multiple sample injection of fluorescent analytes according to a Hadamard code sequence was performed using an optically gated sample injection technique, in which a sample plug was produced based on photodegradation by irradiation with an intense laser beam. The capillary and reservoirs were filled with a sample solution containing buffer components and SDS as a pseudostationary phase. A preliminary study confirmed that fluorescein ion could be photobleached in the presence of SDS. The optically gated sample injection technique was then applied to multiple sample injection, based on a Hadamard matrix. The S/N in the electropherogram obtained by HT-MEKC was improved substantially compared to that obtained by a single injection method. When the technique was applied to the separation of several amino acids labeled with FITC, the S/N ratio for each amino acid was enhanced, without any evidence of degradation in separation resolution. Moreover, HT-MEKC was applied to the analysis of amino acids contained in a Japanese beverage, resulting in improved S/Ns for the amino acids.  相似文献

A system of microchip capillary electrophoresis/electrospray ionization mass spectrometry (microchip-CE/ESI-MS) for rapid characterization of proteins has been developed. Capillary electrophoresis (CE) enables rapid analysis of a sample present in very small quantity, such as at femtomole levels, at high resolution. Faster CE/MS analysis is expected by downsizing the normal capillary to the microchip (microchip) capillary. Although rapidity and high resolution are advantages of CE separation, electroosmotic flow (EOF) instability caused by the interaction between proteins and the microchannel surface results in low reproducibility in the analysis of basic proteins under neutral pH conditions. By coating the microchannel surface with a basic polymer, polyE-323, basic proteins, which have pI values of over 7.5, could be separated and detected by microchip-CE/MS on quadrupole (Q) and time-of-flight (TOF) hybrid instruments. By increasing the cone and collision voltages during the analysis by microchip-CE/ESI-MS of a small protein, some product ions, which contain the sequence information, could also be obtained, i.e., 'top-down' analysis of the protein could be accomplished with this microchip-CE/MS system. To our knowledge, this is the first report of 'top-down' analysis of a protein by microchip-CE/MS. Since it requires a much shorter time and a smaller sample amount for analysis than the conventional liquid chromatography (LC)/ESI-MS method, microchip-CE/MS promises to be suitable for the high-throughput characterization of proteins.  相似文献

In this study we report an improved protocol that combines simplified sample preparation and micro-scale separation for mass spectrometric analysis of neuropeptides from individual neuroendocrine organs of crab Cancer borealis. A simple, one-step extraction method with commonly used matrix-assisted laser desorption/ionization (MALDI) matrix, 2,5-dihydroxybenzoic acid (DHB), in saturated aqueous solution, is employed for improved extraction of neuropeptides. Furthermore, a novel use of DHB as background electrolyte for capillary electrophoresis (CE) separation in the off-line coupling of CE to MALDI-Fourier transform mass spectrometric (FT-MS) detection is also explored. The new CE electrolyte exhibits full compatibility with MALDI-MS analysis of neuropeptides in that both the peptide extraction process and MALDI detection utilize DHB. In addition, enhanced resolving power and improved sensitivity are also observed for CE-MALDI-MS of peptide mixture analysis. Collectively, the use of DHB has simplified the extraction and reduced the sample loss by elimination of homogenizing, drying, and desalting processes. In the mean time, the concurrent use of DHB as CE separation buffer and subsequent MALDI matrix offers improved spectral quality by eliminating the interferences from typical CE electrolyte in MALDI detection.  相似文献

In this work, the use of capillary electrophoresis (CE) to analyze synthetic polymers is reviewed including works published till February 2004. The revised works have been classified depending on the CE mode (e.g., free solution capillary electrophoresis, capillary gel electrophoresis, etc.) and type of buffer (i.e., nonaqueous, aqueous and hydro-organic background electrolytes) employed to separate synthetic macromolecules. Advantages and drawbacks of these different separation procedures for polymer analysis are discussed. Also, physicochemical studies of complex polymer systems by CE are reviewed, including drug release studies, synthetic polyampholytes, dendrimers, fullerenes, carbon nanotubes and associative copolymers.  相似文献

A simple automated nanoliter scale injection device which allows for reproducible 5 nL sample injections from samples with a volume of <1 microL is successfully used for conventional capillary electrophoresis (CE) and Hadamard transform (HT) CE detection. Two standard fused silica capillaries are assembled axially through the device to function as an injection and a separation capillary. Sample solution is supplied to the injection capillary using pressure controlled with a solenoid valve. Buffer solution flows gravimetrically by the junction of the injection and separation capillaries and is also gated with a solenoid valve. Plugs of sample are pushed into the space between the injection and separation capillaries for electrokinectic injection. To evaluate the performance of the injection device, several optimizations are performed including the influence of flow rates, the injected sample volume and the control of the buffer transverse flow on the overall sensitivity. The system was then applied to HT-CE-UV detection for the signal-to-noise ratio (S/N) improvement of the nitric oxide (NO) metabolites, nitrite and nitrate. In addition, signal averaging was performed to explore the possibility of greater sensitivity enhancements compared to single injections.  相似文献

We developed a method for the analysis of multiplexed double-stranded DNA (dsDNA) samples complexed to various intercalating dyes using entangled polymer solution. A commercial single-column capillary electrophoresis (CE) instrument with diode array detection was used for multiplexed detection of DNA samples by addition of intercalating fluorescent molecules. A Phi X174HinfI and a pGEM DNA ladder (1 mg/mL) were used for the electrophoretic separation of dsDNA fragments ranging in size from 24 to 726 and 36 to 2645 bp, respectively. The results suggested that simultaneous electrophoretic separation of different DNA ladders multiplexed with different dyes could be performed in the same capillary yielding fast DNA sizing separations. CE analysis, which is often overpowered by slab gel in sample throughput, could now overcome this disadvantage by allowing multiplexed sample analysis in a fraction of the time needed for slab gel analysis. The separation efficiency of stained DNA molecules with both dyes were dramatically improved with buffers containing a large cation such as tetrapentylammonium ion (Npe(4) (+)) as the only cation in the buffer.  相似文献

Advantageous features (enhanced sensitivity and low baseline noise) of the multiplex measuring procedure, the Hadamard transform capillary electrophoresis (HT-CE), are experimentally demonstrated and critically discussed. To perform a perfect multiplex experiment in CE, sample species need to be dissolved in the background electrolyte medium and have very low concentration. The mismatch of electric conductivity resulting from a sample dissolved in water or in a separation buffer diluted with water will lead to sample stacking and corrupting the anticipated outcome. The multiplex measurements were carried out with benzyltriethylammonium bromide, resorcinol and 3,5-dihydroxybenzoic acid in the phosphate buffer, 511 sample/buffer injections were performed into the capillary according to the pseudorandom binary sequence. The averaged electropherogram of the single injection was calculated from the detection signal with the aid of the Fourier transform. The results illustrate the detrimental effect of sample matrix dilution with water and the effect of increased initial sample concentration on the multiplex measurement in CE. Multiplex advantage, in theory possible in the HT-CE, can be obtained at low concentration levels feasible with laser induced fluorescence and optically gated sampling. To achieve successful multiplex measurements with the UV detector, the single injection signal should be approximately at the baseline noise level.  相似文献

Ion migration in electrophoresis always leads to the formation of ionic boundaries. While some types of the ionic boundaries can be formed intentionally, e.g., to improve separation efficiency or sensitivity of the analysis, other, naturally formed boundaries, may cause unexpected effects during the electrophoretic experiment. Some of the boundaries often go unnoticed in practice; however, many effects of the ionic boundaries formed by the sample matrix and background electrolyte components are frequently observed in capillary electrophoresis (CE). The interpretation of these effects may not be trivial and different explanations of the same phenomena may be found in the literature. This critical article attempts to review some of the important effects of the ionic boundaries observed or utilized during the CE of biologically important samples.  相似文献

A review of recent developments in theoretical as well as application studies concerning the use of organic solvents, either as purely nonaqueous solvents, hydro-organic mixtures, or a combination of an organic solvent with another organic modifier(s), in the sample matrix and/or separation buffer for effecting sample pretreatment and/or improving separation performances in capillary electrophoresis (CE) is presented. In particular, recent advances made in furthering the basic understanding of selectivity changes that occur in capillary zone electrophoresis due the presence of organic solvents in the separation medium, based on in-depth studies of fundamental processes, such as acid-base chemistry, ion-ion and ion-solvent interactions, were discussed in detail. The utilization of organic solvents for improving the resolution of highly challenging and important separations, i.e., those involving the separation of positional and optical isomers, was also critically reviewed. Furthermore, a comprehensive survey of the use of organic solvents for on-line sample pretreatment, e.g., minimizing aggregation and maximizing solubilization of hydrophobic analytes, improving concentration detection sensitivity for analytes via the use of sample stacking, was presented and discussed. Moreover, recent applications involving the use of organic solvents for improving the CE separations of a variety of molecular species with significance in various disciplines, including biological, environmental and pharmaceutical areas, were summarized and tabulated.  相似文献

The cellulases CBH 58 from the fungus Phanerochaete chrysosporium and CBH I from the fungus Trichoderma reesei were compared as chiral selectors in capillary electrophoresis (CE) applying the partial filling technique. Amines, e.g., norephedrine, two bambuterol analogs, as well as acids, e.g., di-p-toluoyl tartaric acid and dibenzoyl tartaric acid, which could not be enantioseparated in the liquid chromatographic use of the selectors, could be separated in the corresponding CE experiments. Due to the very high enantioselectivities, terbutaline, alprenolol and propranolol could be completely enantioresolved with selector plugs shorter than the sample plugs. The affinity of propranolol to CBH 58 was so high at pH 7.0 that neither of the enantiomers reached the detector; therefore, a plug of the displacing disaccharide cellobiose was injected after the sample to elute the propranolol enantiomers. The enantiomers could also be made to leave the capillary at opposite ends, thereby causing an infinite enantioresolution. A new preconcentration technique was introduced, which takes advantage of the very high affinity of propranolol to CBH 58 and the eluting ability of cellobiose. A 12.5 cm long plug of rac-propranolol could be preconcentrated and enantioseparated in a single procedure.  相似文献
