We present the development of a dynamic model for predicting the trajectory of microparticles in microfluidic devices, employing dielectrophoresis, for Hyperlayer field‐flow fractionation. The electrode configuration is such that multiple finite‐sized electrodes are located on the top and bottom walls of the microchannel; the electrodes on the walls are aligned with each other. The electric potential inside the microchannel is described using the Laplace equation while the microparticles' trajectory is described using equations based on Newton's second law. All equations are solved using finite difference method. The equations of motion account for forces including inertia, buoyancy, drag, gravity, virtual mass, and dielectrophoresis. The model is used for parametric study; the geometric parameters analyzed include microparticle radius, microchannel depth, and electrode/spacing lengths while volumetric flow rate and actuation voltage are the two operating parameters considered in the study. The trajectory of microparticles is composed of transient and steady state phases; the trajectory is influenced by all parameters. Microparticle radius and volumetric flow rate, above the threshold, do not influence the steady state levitation height; microparticle levitation is not possible below the threshold of the volumetric flow rate. Microchannel depth, electrode/spacing lengths, and actuation voltage influence the steady‐state levitation height.  相似文献

In this study, a dielectrophoresis field‐flow fractionation device was analyzed using a numerical simulation method and the behaviors of a set of different cells were investigated. By reducing the alternating current frequency of the electrodes from the value used in the original setup configuration and increasing the number of exit channels, total discrimination in cell trajectories and subsequent separation of four cell types were achieved. Cells were differentiated based on their size and dielectric response that are represented in their real part of Clausius–Mossotti factor at different frequencies. A number of novel designs were also proposed based on the original setup configuration. It was seen that by reducing the length of the main channel and the number of electrodes at low frequencies and not changing the inlet flow velocities, cell separation was still achieved successfully, although with a slightly larger electrode voltage. The shorter main channel decreased the residence time for the cells on the chip and also reduced the overall size of the device—these were improvements over the original design. The obtained results can be used to analyze other cell types by knowing their size and dielectric properties to design geometries that can ensure separation.  相似文献

The purpose of this study was to investigate the influence of ionic strength of the carrier liquid, cross flow rate, focus flow rate, and sample load on the retention behavior of liposomes in asymmetric flow field‐flow fractionation (AF4). Two differently prepared samples of large unilamellar vesicles (LUV) were used. Experiments were performed varying the factors systematically and evaluating their effect on both retention behavior of the liposomes and on particle size as obtained from online coupled multi‐angle light scattering (MALS) analysis. The results showed that the focus flow rate had the least influence on the elution of liposomes. Elution of LUV is mainly governed by the chosen cross flow condition and ionic strength of the carrier liquid as well as its sample load. Optimal fractionation and size analysis were achieved using a sample load of about 10 μg, a cross flow gradient from 1.0 to 0.1 mL/min over 35 min and a carrier solution of NaNO3 with a concentration of 10 mM.  相似文献

Asymmetric flow field‐flow fractionation is a valuable tool for the characterization of protein aggregates in biotechnology owing to its broad size range and unique separation principle. However, in practice asymmetric flow field‐flow fractionation is non‐trivial to use due to the major deviations from theory and the influence on separation by various factors that are not fully understood. Here, we report methods to assess the non‐ideal effects that influence asymmetric flow field‐flow fractionation separation and for the first time identify experimentally the main factors that impact it. Furthermore, we propose new approaches to minimize such non‐ideal behavior, showing that by adjusting the mobile phase composition (pH and ionic strength) the resolution of asymmetric flow field‐flow fractionation separation can be drastically improved. Additionally, we propose a best practice method for new proteins.  相似文献

Hollow-fiber flow field-flow fractionation (HF FlFFF) was applied for the separation and size characterization of airborne particles which were collected in a municipal area and prefractionated into four different-diameter intervals >5.0, 2.5-5.0, 1.5-2.5, <1.5 microm) by continuous split-flow thin (SPLIIT) fractionation. Experiments demonstrated the possibility of utilizing a hollow-fiber module for the high-performance separation of supramicron-sized airborne particles at steric/hyperlayer operating mode of HF FlFFF. Eluting particles during HF FlFFF separation were collected at short time intervals (approximately 10 s) for the microscopic examination. It showed that particle size and size distributions of all SPLITT fractions of airborne particles can be readily obtained using a calibration and that HF FlFFF can be utilized for the size confirmation of the sorted particle fraction during SPLITT fractionation.  相似文献

The flow field-flow fractionation (FIFFF) technique is a promising method for separating and analysing particles and large size macromolecules from a few nanometers to approximately 50 μm. A new fractionation channel is described featuring well defined flow conditions even for low channel heights with convenient assembling and operations features. The application of the new flow field-flow fractionation channel is proved by the analysis of pigments and other small particles of technical interest in the submicrometer range. The applicability of different concentration detection methods for online quantification of liposomes upon asymmetric flow field‐flow fractionation was investigated. Filter‐extruded egg phosphatidylcholine liposomes of different size were used. Online quantification using a differential refractive index (dRI) detector was found feasible for relatively high sample loads in the magnitude of 100 μg lipid (under the chosen fractionation conditions). UV–Vis detection of the turbidity of liposomes was ruled out as online detection method because turbidity increases with particle size and the signal is not only concentration but also particle‐size dependent. Staining of liposomes by Rhodamine phosphatidylethanolamine or Sudan Red and subsequent online UV–Vis detection at the absorption maximum of the dye enabled quantification with much higher sensitivity than dRI detection. Furthermore analyte loss and carry‐over phenomena upon repeated injection of varying liposome sample loads were studied using regenerated cellulose (RC) membranes as accumulation wall. It could be shown that RC membranes are prone to adsorption in case of very small sample loads (0.5 μg). This effect may be overcome by pre‐saturation of the membrane with sample loads of at least 2 μg. For higher sample loads adsorptive losses play a minor role. Recovery from pre‐saturated membranes reached approximately 100% and carry‐over was found negligible.  相似文献

To accurately characterize branched polysaccharides with high molecular weights from medicinal and edible mushrooms and identify the limitations of size exclusion chromatography, molecular characteristics of polysaccharides from Tremella fuciformis were determined and compared by asymmetrical flow field‐flow fractionation coupled with multiangle laser light scattering and refractive index detection, and size exclusion chromatography coupled with multiangle laser light scattering and refractive index detection, respectively. Results showed that molecular weights of three batches of T. fuciformis polysaccharides were determined as 2.167 × 106 (TF1), 2.334 × 106 (TF2), and 2.435 × 106 Da (TF3) by size exclusion chromatography, and 3.432 × 106 (TF1), 3.739 × 106 (TF2), and 3.742 × 106 Da (TF3) by asymmetrical flow field‐flow fractionation, as well as 3.469 × 106 Da (TF1) by off‐line multiangle laser light scattering, respectively. Results suggested that size exclusion chromatography was unable to accurately characterize T. fuciformis polysaccharides, which may be due to its limitations such as shear degradation and abnormal coelution. Compared to size exclusion chromatography, asymmetrical flow field‐flow fractionation could be a better technique for the molecular characterization of branched polysaccharides with high molecular weights from medicinal and edible mushrooms, as well as from other natural resources.  相似文献

The behavior of nanometer or micrometer-sized particles, dispersed in liquid phase and exposed to temperature gradient, is a complex and not yet well understood phenomenon. Thermal field-flow fractionation (TFFF), using conventional-size channels, played an important role in the studies of this phenomenon. In addition to thermal diffusion (thermophoresis) and molecular diffusion or Brownian movement, several secondary effects such as particle–particle and/or particle–wall interactions, chemical equilibria with the components of the carrier liquid, buoyant and lift forces, etc., may contribute to the retention and complicate the understanding of the relations between the thermal diffusion and the characteristics of the retained particles. Microthermal FFF is a new high-performance technique allowing much easier manipulation and control of the operational parameters within an extended range of experimental conditions in comparison with conventional TFFF. Consequently, in combination with various other methods, it is well suited for a detailed investigation of the mentioned effects. In this work, some contradictory published results concerning the thermal diffusion of the colloidal particles, studied by TFFF but also by other methods, are analyzed and compared with our experimental findings.  相似文献

The separation and manipulation of microparticles in lab on a chip devices have importance in point of care diagnostic tools and analytical applications. The separation and sorting of particles from biological and clinical samples can be performed using active and passive techniques. In passive techniques, no external force is applied while in active techniques by applying external force (e.g. electrical), higher separation efficiency is obtained. In this article, passive (pinched flow fractionation) and active (insulator‐based dielectrophoresis) methods were combined to increase the separation efficiency at lower voltages. First by simulation, appropriate values of geometry and applied voltages for better focusing, separation, and lower Joule heating were obtained. Separation of 1.5 and 6 μm polystyrene microparticles was experimentally obtained at optimized geometry and low total applied voltage (25 V). Also, the trajectory of 1.5 μm microparticles was controlled by adjusting the total applied voltage.  相似文献

Effects of mobile phase composition can play an effective role in modulating the retention of particles in gravitational field-flow fractionation (GFFF), the simplest and cheapest among field-flow fractionation (FFF) techniques. In the framework of an optimized procedure for the GFFF characterization of particulate systems, an experimental approach to the effects of the mobile phase composition on the retention of silica particles retention is presented. The role of the ionic strength and the presence of surfactant are emphasized, with special regards to the shape of the particles. Moreover, the first experimental evidence of potential-barrier GFFF is reported.  相似文献

Various Be-containing micro-particle suspensions were equilibrated with simulated lung fluid (SLF) to examine their dissolution behavior as well as the potential generation of nanoparticles. The motivation for this study was to explore the relationship between dissolution/particle generation behaviors of Be-containing materials relevant to Be-ore processing, and their epidemiologically indicated inhalation toxicities. Limited data suggest that BeO is associated with higher rates of beryllium sensitization (BS) and chronic beryllium disease (CBD) relative to the other five relevant materials studied: bertrandite-containing ore, beryl-containing ore, frit (a processing intermediate), Be(OH)2 (a processing intermediate), and silica (control). These materials were equilibrated with SLF at two pH values (4.5 and 7.2) to reflect inter- and intra-cellular environments in lung tissue. Concentrations of Be, Al, and Si in SLF increased linearly during the first 20 days of equilibration, and then rose slowly, or in some cases reached a maximum, and subsequently decreased. Relative to the other materials, BeO produced relatively low Be concentration in solution at pH 7.2; and relatively high Be concentration in solution at pH 4.5 during the first 20 days of equilibration. For both pH values, however, the Be concentration in SLF normalized to Be content of the material was lowest for BeO, demonstrating that BeO was distinct among the four other Be-containing materials in terms of its persistence as a source of Be to the SLF solution. Following 149 days of equilibration, the SLF solutions were fractionated using flow-field flow fractionation (FlFFF) with detection via ICP-MS. For all materials, nanoparticles (which were formed during equilibration) were dominantly distributed in the 10–100 nm size range. Notably, BeO produced the least nanoparticle-associated Be mass (other than silica) at both pH values. Furthermore, BeO produced the highest Be concentrations in the size range corresponding to < 3 kDa (determined via centrifugal ultrafiltration), indicating that in addition to persistence, the BeO produced the highest concentrations of truly dissolved (potentially ionic) Be relative to the other materials. Mass balance analysis showed reasonable sample recoveries during FFF fractionation (50–100%), whereas recoveries during ICP-MS (relative to acidified standards) were much lower (5–10%), likely due to inefficiencies in nebulizing and ionizing the nanoparticles.  相似文献

We applied flow field flow fractionation in a ceramic tubular membrane to study the diffusional behavior of sodium polystyrene sulfonate (NaPSS) as a function of salt and polyelectrolyte concentrations in the dilute concentration regime. NaPSS standards were fractionated in an aqueous carrier liquid with salt concentrations of 1–100 mmol L?1 and injected amounts of 0.2–20 μg. A sharp increase in the apparent diffusion coefficients, due to peak overloading, occurred at a critical polyelectrolyte concentration at the accumulation wall. A model has been developed that compares repulsive electrostatic interactions, calculated from the Derjaguin–Landau–Verweij–Overbeek theory, to the thermal energy. It was found that the observed critical concentration approximated the concentration of a closely packed lattice of hard spheres within an order of magnitude. The critical concentration appeared to be dependent on the salt concentration, but the influence of the molecular mass was less clear. Expressions for the migration velocity in overloaded channels were derived and confirmed experimentally for the two lowest molecular masses. Deviations occurred for high molecular mass NaPSS, possibly because of mutual attraction. These experiments confirmed the observations made by small‐angle neutron scattering and dynamic light scattering that the ordering of polyelectrolytes already occurred in the dilute regime. © 2001 John Wiley & Sons, Inc. The downstream processing of enveloped virus‐like particles is very challenging because of the biophysical and structural similarity between correctly assembled particles and contaminating vesicular particles present in the feedstock. We used hydroxyl‐functionalized polymethacrylate monoliths, providing hydrophobic and electrostatic binding contributions, for the purification of HIV‐1 gag virus‐like particles. The clarified culture supernatant was conditioned with ammonium sulfate and after membrane filtration loaded onto a 1 mL monolith. The binding capacity was 2 × 1012/mL monolith and was only limited by the pressure drop. By applying either a linear or a step gradient elution, to decrease the ammonium sulfate concentration, the majority of double‐stranded DNA (88–90%) and host cell protein impurities (39–61%) could be removed while the particles could be separated into two fractions. Proteomic analysis and evaluation of the p24 concentration showed that one fraction contained majority of the HIV‐1 gag and the other fraction was less contaminated with proteins originated from intracellular compartments. We were able to process up to 92 bed volumes of conditioned loading material within 3 h and eluted in average 7.3 × 1011 particles per particle fraction, which is equivalent to 730 vaccination doses of 1 × 109 particles.  相似文献

A new online concentration method, namely pressure‐assisted field‐enhanced sample injection (PA‐FESI), was developed and compared with FESI for the analysis of water‐soluble vitamins by CZE with UV detection. In PA‐FESI, negative voltage and positive pressure were simultaneously applied to initialize PA‐FESI. PA‐FESI uses the hydrodynamic flow generated by the positive pressure to counterbalance the reverse EOF in the capillary column during electrokinetic sample injection, which allowed a longer injection time than usual FESI mode without compromising the separation efficiency. Using the PA‐FESI method, the LODs of the vitamins were at ng/mL level based on the S/N of 3 and the RSDs of migration time and peak area for each vitamin (1 μg/mL) were less than 5.1%. The developed method was applied to the analysis of water‐soluble vitamins in corns.  相似文献

On the basis of previous studies, the particles in a dilute (volume fractions φ < 4 × 10–3) suspension in combined Poiseuille and electroosmotic “counterflow” at flow Reynolds numbers Re ≤ 1 accumulate, then assemble into structures called “bands,” within ∼6 μm of the channel wall. The experimental studies presented here use a small fraction of tracer particles labeled with a different fluorophore from the majority “bulk” particles to visualize the dynamics of individual particles in a φ = 1.7 × 10–3 suspension. The results at two different near-wall shear rates and three electric field magnitudes E show that the near-wall particles are concentrated about 150-fold when the bands start to form, and are then concentrated about 200-fold to a maximum near-wall volume fraction of ∼0.34. The growth in the near-wall particles during this accumulation stage appears to be exponential. This near-wall particle accumulation is presumably driven by a wall-normal “lift” force. The observations of how the particles accumulate near the wall are compared with recent analyses that predict that suspended particles subject to shear flow and a dc electric field at small particle Reynolds numbers experience such a lift force. A simple model that assumes that the particles are subject to this lift force and Stokes drag suggests that the force driving particles toward the wall, of O(10–17 N), is consistent with the time scales for particle accumulation observed in the experiments.  相似文献

Electrokinetic supercharging is one of the most powerful sample‐stacking methods that combines field amplified sample injection and transient ITP. In counter‐flow electrokinetic supercharging, a constant counter pressure is applied during sample injection in order to counterbalance the movement of the injected sample zone. As a result, there will be a pronounced increase in the amount of sample injected and the portion of the capillary available for electrophoresis. In this report, counter‐flow electrokinetic supercharging optimization factors such as the electric field application in the constant voltage and constant current modes, the magnitude of counter pressure, and the terminating electrolyte concentrations were investigated. The enrichments obtained with a 30 min injection of 10 nM catecholamines in 5 mM terminating electrolyte solution in the constant voltage mode applying a counter pressure of 1.3 psi were 41000‐fold for dopamine, 50 000‐fold for norepinephrine, and 32 000‐fold for epinephrine, yielding detection limits of 1.3, 1.4, and 1.2 nM, respectively, with absorbance detection at 200 nm.  相似文献

Amphiphilic diblock copolymers consisting of a hydrophobic core containing a polymerized ionic liquid and an outer shell composed of poly(N‐isoprolylacrylamide) were investigated by capillary electrophoresis and asymmetrical flow‐field flow fractionation. The polymerized ionic liquid comprised poly(2‐(1‐butylimidazolium‐3‐yl)ethyl methacrylate tetrafluoroborate) with a constant block length (n = 24), while the length of the poly(N‐isoprolylacrylamide) block varied (n = 14; 26; 59; 88). Possible adsorption of the block copolymer on the fused silica capillary, due to alterations in the polymeric conformation upon a change in the temperature (25 and 45 °C), was initially studied. For comparison, the effect of temperature on the copolymer conformation/hydrodynamic size was determined with the aid of asymmetrical flow‐field flow fractionation and light scattering. To get more information about the hydrophilic/hydrophobic properties of the synthesized block copolymers, they were used as a pseudostationary phase in electrokinetic chromatography for the separation of some model compounds, that is, benzoates and steroids. Of particular interest was to find out whether a change in the length or concentration of the poly(N‐isoprolylacrylamide) block would affect the separation of the model compounds. Overall, our results show that capillary electrophoresis and asymmetrical flow‐field flow fractionation are suitable methods for characterizing conformational changes of such diblock copolymers.  相似文献

A phase with both hydrophobic and hydrophilic functionalities has been synthesized by modification of ground silica monolith particles with C18 and 1‐[3‐(trimethoxysilyl)propyl] urea ligands. A series of phases was prepared by changing the ratio of the two ligands to determine the optimal ratio in view of separation efficiency. The resultant optimized stationary phase was packed in narrow‐bore glass‐lined stainless‐steel columns (1 × 300 mm and 2.1 × 100 mm) and used for the separation of synthetic peptides and proteins. The average numbers of theoretical plates (N) of 52 100/column (174 000/m, 5.75 µm plate height) and 35 500/column (118 000/m, 8.47 µm plate height) were achieved with the 300 mm column at a flow rate of 25 µL/min (0.86 mm/s) in 60:40 v/v acetonitrile/30 mM aqueous ammonium formate for the mixture of peptides (Thr‐Tyr‐Ser, Val‐Ala‐Pro‐Gly, angiotensin I, isotocin, and bradykinin) and for the mixture of proteins (myoglobin, human serum albumin, and insulin), respectively. Fast analysis of the peptides and proteins was also carried out at a flow rate of 0.9 mL/min (6.88 mm/s) with the 100 mm column and all the analytes were eluted within 2 min with good separation efficiency.  相似文献
