In this study, an organic–inorganic hybrid monolithic capillary column was applied and optimized for the determination of cefdinir in plasma, and the electro‐osmotic flow that usually hinders migration in reverse polarity became a driving force. The Sample used for pharmacokinetic research was collected by microdialysis using phosphate buffer (pH 7.4) as perfusate, and a volume of 60 μL fluid was mixed with 140 μL of acetonitrile. By using a silica‐allyldimethyldodecylammonium monolithic column (100 μm inner diameter, 21 cm effective length and 31.2 cm total length), and a mobile phase consisting of phosphate and acetonitrile (pH 4.5, 50:50, v/v), at a voltage of 20 kV, the analytes were successfully separated with the background within 2.5 min. The detection wavelength was 214 nm. The calibration curve showed a good linearity (r2 = 0.9994) over the concentration range of 0.2–50 μg/mL. The proposed method showed good specificity, linearity, sensitivity, precision and recovery, and the introduction of field amplified sample stacking helped to improve the low recovery caused by microdialysis. This method was successfully applied to quantify cefdinir in rat plasma to support a pre‐clinical pharmacokinetic trial.  相似文献

A novel method combining molecular imprinting and SPE was developed in a capillary column for the determination of auramine O in shrimp. The capillary monolithic column was prepared by UV‐initiated in situ polymerization, using auramine O as template and methacrylic acid and ethylene dimethacrylate as functional monomer and cross‐linker, respectively. The properties of the prepared capillary monolithic column were investigated under the optimized conditions coupled with HPLC, and then the morphologies of the inner polymers were characterized by SEM. The calibration curve was expressed as A = 103C + 19.8 (r = 0.9992) with a linear range of 0.25–25.0 μg/mL, and the recoveries of auramine O at different concentrations in shrimp ranged from 90.5 to 92.4% with RSDs ranging from 2.1 to 4.4%. The capacities of the molecularly imprinted polymer and nonimprinted polymer columns were 0.722 and 0.147 μg/mg, respectively, and the LOD (S/N = 3) of auramine O in shrimp was 17.85 μg/kg. Under the selected conditions, the enrichment factors obtained were higher than 70‐fold. The results indicate that the prepared molecularly imprinted capillary monolithic column was reliable and applicable to the analysis of auramine O in shrimp.  相似文献

Water-compatible ciprofloxacin-imprinted monolithic columns were synthesized in water-containing systems for selective extraction of ciprofloxacin from human urine samples. Methanol-water (10:3, v/v) was used as a porogenic solvent and the obtained monolithic imprinted polymers reveal high selectivity to ciprofloxacin in an aqueous environment; the affinity can be easily controlled by adjusting the pH of the mobile phase. Owing to the unique porous structure and flow-through channels existing in the network skeleton of the monolithic MIP, urine samples could be directly injected into the column, proteins and other biological matrix were quickly washed out and ciprofloxacin was selectively retained and enriched. Good linearity was obtained from 0.08 to 400 mg/L (r=0.998) with the relative standard deviations less than 3.6%. The limit of detection of the method was 0.04 mg/L and the recoveries were more than 94.5% at three different concentrations. Moreover, by increasing the injection volume to 2.0 mL, the sensitivity of the method could be improved 100-fold according to the peak height of ciprofloxacin. This expedient greatly simplified the overall procedure, resulting in a rapid and efficient sample analysis while maintaining precision and accuracy.  相似文献

An automated system has been developed for the determination of trace tetracycline antibiotics (TCs) in egg samples, based on online molecularly imprinted solid-phase extraction (MISPE) coupling with high-performance liquid chromatography (HPLC). Oxytetracycline and chlortetracycline were chosen as mixed templates to synthesize highly selective molecularly imprinted polymers for online extraction. Under the optimal online MISPE-HPLC condition, 10 mL egg samples were injected into the MISPE column and then the matrix was washed out. By rotating the switching valve, TCs were transferred to the analytical column and then separated by HPLC. Because sample pretreatment and chromatographic separation were carried out simultaneously, the whole analytical time (18 min) was significantly shortened compared with conventional offline techniques. The detection limits ranged from 0.8 to 1.3 ng/g. The enhancement factors were in the range of 159-410. The spiked recoveries of TCs in real egg samples ranged from 91.6 to 107.6% and the relative standard deviations (RSDs) were not higher than 4.0%.  相似文献

A method was developed for the determination of ractopamine in pig urine using molecularly imprinted solid-phase extraction (MISPE) as the sample clean-up technique combined with high-performance liquid chromatography. The molecularly imprinted polymer (MIP) was synthesized in acetonitrile-triethylamine system using ractopamine (RAC) as the template and acrylamide as the monomer. The binding capacity of the polymer toward RAC was found to be about 2.57 mg of ractopamine/g of polymer. The optimal procedures for MISPE consisted of conditioning with 3 mL methanol, equilibrating with 3 mL of water, loading volume of <10 mL of aqueous sample (pH 7), washing with 3 mL water and 3 mL methanol, and eluting with 5 mL of 5% ammonia in methanol. In the four spiked samples with the levels of 0.01, 0.1, 1.0 and 5.0 μg/mL, the mean recoveries of analyte on the MIP were higher than 90% with relative standard deviation <10%, and significant differences between imprinted and non-imprinted materials were observed. The MIP selectivity was evaluated by checking 11 drugs with similar and different molecular structures to that of RAC. The characteristics of three-dimensional cavities and hydrogen bond interaction were regarded as the main factors that dominated the retention of RAC on the MISPE cartridge.  相似文献

This work describes an on-line molecularly imprinted solid-phase extraction (MISPE) method for spectrophotometric determination of nicotine in urine samples of smokers. This method is based on manganese (VII) to manganese (VI) reduction in an alkaline medium, promoted by nicotine. Two wash solutions (1:4 (v/v) acetonitrile:sodium hydroxide - pH 11.4, and nitric acid - pH 2.5) were employed to circumvent interferences. Aqueous solutions containing nicotine plus different possible concomitants (cotinine, anabasine, norcotinine and caffeine) were tested individually. The analytical calibration curve was prepared in urine samples collected from non-smokers and spiked with nicotine standard from 1.1 to 60 μmol L−1 (r2 > 0.998). The limit of quantification and the analytical frequency were 1.1 μmol L−1 and 11 h−1, respectively. The precision, evaluated using 3, 10 and 30 μmol L−1 nicotine in urine, was 10, 10 and 4% (intra-day precision) and 12, 13 and 5% (inter-day precision), respectively. Accuracy was checked through high performance liquid chromatography and the results did not present significant differences at the 95% confidence level according to the Student's t-test.  相似文献

A novel solid phase extraction (SPE) method for determination of tetracyclines (TCs) in milk and honey samples by molecularly imprinted monolithic column was developed. Using tetracycline (TC) as the template, methacrylic acid (MAA) as the functional monomer, ethylene glycol dimethacrylate (EGDMA) as the cross-linker, methanol as the solvent, cyclohexanol and dodecanol as the mixed porogenic solvents, a TC imprinted monolithic column was prepared by in situ molecular imprinting technique for the first time, and the optimal synthesis conditions and the selectivity of TC imprinted monolithic column were investigated. The interfering substances in food samples and TCs can be separated successfully on imprinted column. Molecularly imprinted solid phase extraction (MISPE) coupling with C18 column was used to determinate the TCs in milk and honey. The recoveries of this method for six tetracyclines antibiotics such as tetracycline (TC), oxytetracycline (OTC), minocycline (MINO), chlortetracycline (CTC), metacycline (MTC) and doxycycline (DTC) were investigated, and high recoveries of 73.3-90.6% from milk samples and 62.6-82.3% from honey samples were obtained. A method for determination of TCs at low concentration level in milk and honey samples was successfully developed by using the monolithic column as the precolumn for solid phase extraction of six TCs compounds.  相似文献

The highly selective, fast and effective sample pretreatment technique molecularly imprinted solid-phase extraction (MISPE) can overcome the low sensitivity of the highly efficient capillary electrophoresis-UV method (CE-UV). In this work, narrowly dispersible bisphenol A (BPA)-imprinted polymeric microspheres with a high capacity factor of k′ = 6.8 and an imprinted factor of I = 6.53 were investigated as selective solid-phase extraction (SPE) sorbents for use in extraction of BPA from different sample matrices (tap water, wastewater, Yangtze River water, soil from the Yangtze River, shrimp and human urine). Washing and eluting protocols of MISPE were optimized. Under optimal conditions, recoveries of MISPE were investigated. Recoveries were basically constant and the relative standard deviation (RSD) was lower than 5.8% when loading volumes changed from 1 to 50 mL. Recoveries ranged from 71.20% to 86.23% for different sample matrices. Compared with C18 SPE, MISPE had higher selectivity and recovery for BPA. BPA was determined with good accuracy and precision in different complex samples using CE-UV coupled with MISPE. Spiked recoveries ranged from 95.20% to 105.40%, and the RSD was less than 7.2%. Because a large loading volume was achieved, the enrichment efficiency of pretreatment and the sensitivity of this method were improved. The limits of detection of this MISPE-CE-UV method for BPA in tap water, wastewater, Yangtze River water, soil from the Yangtze River, shrimp and human urine were 3.0 μg L− 1, 5.4 μg L− 1, 6.9 μg L− 1, 2.1 μg L− 1, 1.8 μg L− 1 and 84 μg L− 1, respectively.  相似文献

A molecularly imprinted polymer has been synthesized to specifically extract adefovir, an antiviral drug, from serum and urine by dispersive solid‐phase extraction before high‐performance liquid chromatography with UV analysis. The imprinted polymers were prepared by bulk polymerization by a noncovalent imprinting method that involved the use of adefovir (template molecule) and functional monomer (methacrylic acid) complex prior to polymerization, ethylene glycol dimethacrylate as cross‐linker, and chloroform as porogen. Molecular recognition properties, binding capacity, and selectivity of the molecularly imprinted polymers were evaluated and the results show that the obtained polymers have high specific retention and enrichment for adefovir in aqueous medium. The new imprinted polymer was utilized as a molecular sorbent for the separation of adefovir from human serum and urine. The serum and urine extraction of adefovir by the molecularly imprinted polymer followed by high‐performance liquid chromatography showed a linear calibration curve in the range of 20–100 μg/L with excellent precisions (2.5 and 2.8% for 50 μg/L), respectively. The limit of detection and limit of quantization were determined in serum (7.62 and 15.1 μg/L), and urine (5.45 and 16 μg/L). The recoveries for serum and urine samples were found to be 88.2–93.5 and 84.3–90.2%, respectively.  相似文献

A rapid, simple and selective method based on molecularly imprinted, spin column extraction coupled with fluorescence detection was successfully established for the determination of 2,4-dinitrophenol in serum samples. The 2,4-dinitrophenol imprinted polymers exhibited highly selective recognition for the template molecule and the maximum adsorption capacity was 138.9 mg/g. The results indicated that when water is used as the loading solution, only 2,4-dinitrophenol could be adsorbed on the spin column without the remaining structural analogs(2-nitrophenol, 4-nitrophenol and phenol). After eluting with acetonitrile/acetic acid(9/1, v/v), 2,4-dinitrophenol in serum samples could be determined by using the fluorescence spectrometer, based on the fluorescence enhancement of fluorescein by the template molecule. Under the optimal conditions, the spiked recovery ranged from 95.8% to 103.4% and the detection limit was 1 nmol/L. The results confirmed the reliability and practicality of the protocol and revealed a good perspective of this method for biological sample analysis.  相似文献

A molecularly imprinted polymer has been synthesized for a selective on-line catechol extraction, followed by its spectrophotometric determination in guarana powder, mate tea and tap water samples. A clean-up column, containing a methacrylic polymer + C18 solid phase, was also used in order to enhance selectivity. The imprinted polymer was prepared by bulk polymerization using catechol as template and 4-vinylpyridine as the functional monomer. Permanganate solution was used as spectrophotometric reagent, where Mn(VII) was reduced to Mn(II) by catechol in an acid medium, causing color loss, which was monitored at 528 nm. Physical (extraction flow rate, elution flow rate, coil length) and chemical (nature and concentration of the eluent, potassium permanganate concentration) variables were optimized, and the selectivity was appraised using three molecules (4-chloro-2-methylphenol, 2-cresol, 2-methoxyphenol) similar to catechol. These molecules did not present interference in 1:8, 1:10 and 1:10 (catechol/concomitant) proportions, respectively. The analytical calibration curve ranged from 3.0 up to 100 μmol L− 1 (r > 0.999; seven concentrations levels, n = 3) and the limits of detection (LOD) and quantification (LOQ) were 0.8 and 2.7 μmol L− 1, respectively. Precision, expressed as RSD, was of 3.0% (20 μmol L− 1, n = 10), and the analytical frequency was 15 h− 1. Accuracy was checked by the HPLC technique and the results did not present significant difference at 95% confidence levels according to the t test.  相似文献

In this work, a novel molecularly imprinted polymer (MIP) monolithic column with integrated in‐column electroosmotic pump (EOP) was designed and successfully prepared to facilitate the capillary chromatography with MIP column. A silica‐based EOP was synthesized at the detection end of the MIP monolithic capillary column by so‐gel to provide the hydrodynamic driven force for the capillary chromatography. Because of large surface area and low fluidic resistance of the silica monolith,a strong and steady EOF was generated by silica‐based EOP, indicating that the EOP was quite compatible with MIP capillary column. With the sufficient EOF provided by EOP, the electro‐driven based capillary chromatographic separation of nitrophenol isomers was achieved in 4‐vinylpyridine‐based MIP monolithic capillary, which was originally proved infeasible because of the EOF shortage. No significant influence upon the specific recognition of the MIP was found due to the setting of EOP after the detection window of the column. The influence of experimental parameters on the EOF such as voltage and pH value of running buffer was investigated. The column was also evaluated by capillary liquid chromatographic mode to compare with EOP‐driven capillary chromatography. Higher column efficiency was obtained by EOP‐driven separation with improved peak shape. The results suggested that EOP‐supported technique would be a good way to solve the problem of weak EOF generation in electro‐driven capillary chromatography.  相似文献

The determination of acidic pharmaceuticals, such as non-steroidal anti-inflammatory drugs NSAIDs and clofibric acid (metabolite of clofibrate), at low ng L−1 levels in wastewater requires highly selective and sensitive analytical procedures. The removal of matrix components during sample preparation results in significant benefits towards reducing the matrix effects during LC-MS analysis. Therefore this work describes a simple method to enrich and clean up NSAIDs and clofibric acid from sewage water using molecularly imprinted solid-phase extraction (MISPE). Final analysis was performed by liquid chromatography-tandem mass spectrometry. The performance of this method has been evaluated in fortified tap and sewage water in terms of recovery, precision, linearity, and method quantification limit. Recovery for all compounds ranged in all matrices between 84 and 116% with intra-day R.S.D. values below 11.5%. Matrix effect evaluation demonstrated that even complex sample matrixes, such as pond or sewage water did not showed significant ion suppression/enhancement compared to tap water. The performance of the method was further emphasized by the study of pond water, which receives treated water from a sewage treatment plant in south Sweden. Raw sewage and treated water were also tested. In those samples, all acidic pharmaceuticals were detected in concentration above method quantification limits ranging from 5.1 to 5153.0 ng L−1.  相似文献

The selective extraction of baicalin is important to its quality control especially when the matrices are complicated. In this work, a novel molecularly imprinted polymer was prepared for the selective extraction of baicalin in herbs. The molecularly imprinted polymer was synthesized by the copolymerization of 4‐vinyl pyridine and ethylene glycol dimethacrylate in the presence of baicalin by a precipitation polymerization method. After the optimization of parameters for molecularly imprinted polymer preparation, including the functional monomer, porogen, sampling solvent, and washing solvent, good selectivity was obtained, with an imprinting factor of about 4, which is much better than that achieved by the bulk‐polymerization method. The performances of the prepared molecularly imprinted polymers were systematically investigated, including adsorption kinetics, isotherm experiment, and Scatchard analysis. On the basis of the good adsorptive capability of the prepared molecularly imprinted polymer, it was also applied for the pretreatment of baicalin in Scutellaria baicalensis Georgi. The result showed that most of the matrices were removed and baicalin was selectively enriched.  相似文献

This work reports the preparation of a molecularly imprinted polymer (MIP) for selective catalytic detection of serotonin (5-hydroxytryptamine, 5-HT). The process is based on the synthesis of polymers with hemin introduced as the catalytic center to mimic the active site of peroxidase. The copolymer MIP, containing artificial recognition sites for 5-HT, has been prepared by bulk polymerization using methacrylic acid (MAA) and hemin as the functional monomers, and ethylene glycol dimethacrylate (EGDMA) as the cross-linker. For the determination of 5-HT, a flow injection analysis system coupled to an amperometric detector was optimized using multivariate analysis. The effects of different parameters, such as pH, buffer flow rate, buffer nature, peroxide concentration and sample volume were evaluated. After optimizing the experimental conditions, a linear response range from 1.0 up to 1000.0 μmol L−1 was obtained with a sensitivity of 0.4 nA/μmol L−1. The detection limit was found to be 0.30 μmol L−1, while the precision values (n = 6) evaluated by relative standard deviation (R.S.D.) were, respectively, 1.3 and 1.7% for solutions of 50 and 750 μmol L−1 of 5-HT. No interference was observed by structurally similar compounds (including epinephrine, dopamine and norepinephrine), thus validating the good performance of the imprinted polymer. The method was applied for the determination of 5-HT in spiked blood serum samples.  相似文献

An efficient molecularly imprinted solid‐phase extraction protocol was developed for the separation of dopamine (DA) from human urine. After successful validation of the analytical method using high‐performance liquid chromatography coupled with fluorescence detection, a new strategy for the selective determination of DA in the presence of norepinephrine and epinephrine in human urine was presented. In the proposed protocol, the LODs and quantification for DA were 166 ± 36 and 500 ± 110 nmol/L, respectively, and the total recoveries of DA in the range of 1–15 μmol/L varied between 98.3 and 101.1%. DA was detected in the real urine samples at the level of 47–167 μg/L (0.250–0.895 μmol/L). The superiority of the novel analytical strategy was shown by comparison with the results obtained for a commercially available imprinted sorbent.  相似文献

Dexamethasone‐imprinted polymers were fabricated by reversible addition–fragmentation chain transfer polymerization on the surface of magnetic nanoparticles under mild polymerization conditions, which exhibited a narrow polydispersity and high selectivity for dexamethasone extraction. The dexamethasone‐imprinted polymers were characterized by scanning electron microscopy, transmission electron microscope, Fourier transform infrared spectroscopy, X‐ray diffraction, energy dispersive spectrometry, and vibrating sample magnetometry. The adsorption performance was evaluated by static adsorption, kinetic adsorption and selectivity tests. The results confirmed the successful construction of an imprinted polymer layer on the surface of the magnetic nanoparticles, which benefits the characteristics of high adsorption capacity, fast mass transfer, specific molecular recognition, and simple magnetic separation. Combined with high‐performance liquid chromatography, molecularly imprinted polymers as magnetic extraction sorbents were used for the rapid and selective extraction and determination of dexamethasone in skincare cosmetic samples, with the accuracies of the spiked samples ranging from 93.8 to 97.6%. The relative standard deviations were less than 2.7%. The limit of detection and limit of quantification were 0.05 and 0.20 μg/mL, respectively. The developed method was simple, fast and highly selective and could be a promising method for dexamethasone monitoring in cosmetic products.  相似文献

A method is proposed for the clean-up and preconcentration of natural and synthetic estrogens from aqueous samples employing molecularly imprinted polymer (MIP) as selective sorbent for solid-phase extraction (SPE). The selectivity of the MIP was checked toward several selected natural and synthetic estrogens such as estrone (E1), 17β-estradiol (β-E2), 17α-estradiol (α-E2), estriol (E3), 17α-ethinylestradiol (EE2), dienestrol (DIES) and diethylstilbestrol (DES). Ultrahigh pressure liquid chromatography (UHPLC) coupled to a TSQ triple quadrupole mass spectrometry (QqQ) was used for analysis of target analytes. The chromatographic separation of the selected compounds was performed in less than 2 min under isocratic conditions. The method was applied to the analysis of estrogens in spiked river and tap water samples. High recoveries (>82%) for estrone, 17β-estradiol, 17α-estradiol, estriol and 17α-ethinylestradiol were obtained. Lower but still satisfactory recoveries (>48%) were achieved for dienestrol and diethylstilbestrol. The method was validated and found to be linear in the range 50-500 ng L(-1) with correlation coefficients (R(2)) greater than 0.995 and repeatability relative standard deviation (RSD) below 8% in all cases. For analysis of 100-mL sample, the method detection limits (LOD) ranged from 4.5 to 9.8 ng L(-1) and the limit of quantitation (LOQ) from 14.9 to 32.6 ng L(-1). To demonstrate the potential of the MIP obtained, a comparison with commercially available C(18) SPE was performed. Molecularly imprinted SPE showed higher recoveries than commercially available C(18) SPE for most of the compounds. These results showed the suitability of the MIP-SPE method for the selective extraction of a class of structurally related compounds such as natural and synthetic estrogens.  相似文献

This work reports the preparation of molecularly imprinted polymer particles for the selective extraction and determination of four benzophenones from aqueous media. The polymer was prepared by using 4‐vinylpridine as functional monomer, ethylene glycol dimethacrylate as cross‐linker, acetonitrile as porogenic solvent and 2,2’,4,4’‐tetrehydroxybenzophenone as template. Good specific adsorption capacity (Qmax = 27.90 μmol/g) for 2,2’,4,4’‐tetrehydroxybenzophenone was obtained in the sorption experiment and good class selectivity for 2,2’,4,4’‐tetrehydroxybenzophenone, 2,4‐dihydroxybenzophenone, 2,2’‐dihydroxy‐4‐methoxybenzophenone, 2,2’‐dehydroxy‐4,4’‐dimethoxybenzophenone was demonstrated by the chromatographic evaluation experiment. Factors affecting the extraction efficiency of the molecularly imprinted solid‐phase extraction procedure were investigated systematically. An accurate and sensitive analytical method based on the molecularly imprinted solid‐phase extraction coupled with high‐performance liquid chromatography and diode array detection has been successfully developed for the simultaneous determination of four benzophenones from tap water and river water with method detection limits of 0.25–0.72 ng/mL. The recoveries of benzophenones for water samples at two spiking levels (500 and 5000 ng/mL for each benzophenone) were in the range of 86.9–103.3% with relative standard deviations (n = 3) below 9.2%.  相似文献

The aim of this work was to develop a method for the clean-up of a mycotoxin, i.e. Ochratoxin A (OTA), from cereal extracts employing a new molecularly imprinted polymer (MIP) as selective sorbent for solid-phase extraction (SPE) and to compare with an immunoaffinity column. A first series of experiments was carried out in pure solvents to estimate the potential of the imprinted sorbent in terms of selectivity studying the retention of OTA on the MIP and on a non-imprinted polymer using conventional crushed monolith. The selectivity of the MIP was also checked by its application to wheat extracts. Then, after this feasibility study, two different formats of MIP: crushed monolith and micro-beads were evaluated and compared. Therefore an optimization procedure was applied to the selective extraction from wheat using the MIP beads. The whole procedure was validated by applying it to wheat extract spiked by OTA at different concentration levels and then to a certified contaminated wheat sample. Recoveries close to 100% were obtained. The high selectivity brought by the MIP was compared to the selectivity by an immunoaffinity cartridge for the clean-up of the same wheat sample. The study of capacity of both showed a significant higher capacity of the MIP.  相似文献
