The synthesis and self‐assembly properties in aqueous solutions of novel amphiphilic block copolymers composed of one hydrophilic, pH and temperature responsive poly(dimethyl amino ethyl methacrylate) (PDMAEMA) block and one weakly hydrophobic, water insoluble, potentially thermoresponsive poly(hydroxy propyl methacrylate) (PHPMA) block, are reported. The block copolymers were prepared by RAFT polymerization and were molecularly characterized by size exclusion chromatography, NMR, and FTIR spectroscopies. The PDMAEMA‐b‐PHPMA amphiphilic block copolymers self‐assemble in different nanostructured aggregates when inserted in aqueous media. The effects of different solubilization protocols, as well as the effects of solution temperature and pH on the structure of the aggregates, are studied by light scattering and fluorescence spectroscopy measurements. Experimental results indicate that there is a number of solution preparation and physicochemical parameters that allow the control and manipulation of the structure and thermoresponsive properties of PDMAEMA‐b‐PHPMA aggregates in aqueous media. © 2018 Wiley Periodicals, Inc. The azobenzene‐based amphiphilic copolymers have drawn significant attention as a kind of multi‐responsive smart materials. The demand on deeper investigation of how the azobenzene group influences the micelles formation and light‐responsive behavior on molecular level is very urgent. In this article, multi‐responsive block copolymers, poly (acrylic acid)‐block‐poly[4'‐[[(2‐Methacryloyloxy)ethyl]ethylainino]azobenzene‐co‐poly (ethylene glycol) methyl ether methacrylate] (PAA‐b‐P (AzoMA‐co‐PEGMA)), with pH‐, light‐ and reduction‐responsiveness were synthesized by the monomers of AzoMA, PEGMA and acrylic acid via reversible addition‐fragmentation chain transfer polymerization (RAFT). The amphiphilic block copolymer presented aggregation‐induced emission effect, and it was pH, light, and reduction responsive. The results showed that the micelle size decreased with the decreasing of pH within a certain range. However, the particle size of micelles increased significantly when the pH was 4. Once adding reduction agent, the micelles were disassembly. Fluorescent molecule of Nile red was selected as a hydrophobic guest molecule to study the properties of encapsulating and releasing abilities of block copolymer micelles for guest molecules. The results showed that the loading capacity of three kinds of copolymer micelles was closely related to the aggregates formed by the hydrophobic block, mainly azobenzene block. Besides, the block copolymer micelles could release a certain amount of Nile red under the irradiation of UV light, the reduction with Na2S2O4 as reductant, and the exposure to alkaline environment. The mechanism of how the different status of azobenzene group influenced the self‐assembly and multi‐responsive behavior was explored on molecular level.  相似文献

Amphiphilic diblock copolymers that contained hydrophilic poly[bis(potassium carboxylatophenoxy)phosphazene] segments and hydrophobic polystyrene sections were synthesized via the controlled cationic polymerization of Cl3P?NSiMe3 with a polystyrenyl–phosphoranimine as a macromolecular terminator. These block copolymers self‐associated in aqueous media to form micellar structures which were investigated by fluorescence spectroscopy, dynamic light scattering, and transmission electron microscopy. The size and shape of the micelles were not affected by the introduction of different monovalent cations (Li+, K+, Na+, and Cs+) into the stable micellar solutions. However, exposure to divalent cations induced intermicellar crosslinking through carboxylate groups, which caused precipitation of the ionically crosslinked aggregates from solution. This micelle‐coupling behavior was reversible: the subsequent addition of monovalent cations caused the redispersion of the polystyrene‐block‐poly[bis(potassium carboxylatophenoxy)phosphazene] (PS–KPCPP) block copolymers into a stable micellar solution. Aqueous micellar solutions of PS–KPCPP copolymers also showed pH‐dependent behavior. These attributes make PS–KPCPP block copolymers suitable for studies of guest retention and release in response to ion charge and pH. © 2005 Wiley Periodicals, Inc. A series of well‐defined, fluorinated diblock copolymers, poly[2‐(dimethylamino)ethyl methacrylate]‐b‐poly(2,2,2‐trifluoroethyl methacrylate) (PDMA‐b‐PTFMA), poly[2‐(dimethylamino)ethyl methacrylate]‐b‐poly(2,2,3,4,4,4‐hexafluorobutyl methacrylate) (PDMA‐b‐PHFMA), and poly[2‐(dimethylamino)ethyl methacrylate]‐b‐poly(2,2,3,3,4,4,5,5‐octafluoropentyl methacrylate) (PDMA‐b‐POFMA), have been synthesized successfully via oxyanion‐initiated polymerization. Potassium benzyl alcoholate (BzO?K+) was used to initiate DMA monomer to yield the first block PDMA. If not quenched, the first living chain could be subsequently used to initiate a feed F‐monomer (such as TFMA, HFMA, or OFMA) to produce diblock copolymers containing different poly(fluoroalkyl methacrylate) moieties. The composition and chemical structure of these fluorinated copolymers were confirmed by 1H NMR, 19F NMR spectroscopy, and gel permeation chromatography (GPC) techniques. The solution behaviors of these copolymers containing (tri‐, hexa‐, or octa‐ F‐atom)FMA were investigated by the measurements of surface tension, dynamic light scattering (DLS), and UV spectrophotometer. The results indicate that these fluorinated copolymers possess relatively high surface activity, especially at neutral media. Moreover, the DLS and UV measurements showed that these fluorinated diblock copolymers possess distinct pH/temperature‐responsive properties, depending not only on the PDMA segment but also on the fluoroalkyl structure of the FMA units. © 2009 Wiley Periodicals, Inc.  Two pH-responsive amphiphilic diblock copolymers, namely polyisobutylene-block-poly[2-(N,N-dimethylamino)ethyl methacrylate] (PIB-b-PDMAEMA) and polyisobutylene-block-poly(metharylic acid) (PIB-b-PMAA), were synthesized via oxyanion-initiated polymerization, and their multiple self-assembly behaviors have been studied. An exo-olefin-terminated highly reactive polyisobutylene (HRPIB) was first changed to hydroxyl-terminated PIB (PIB-OH) via hydroboration-oxidation of C=C double bond in the chain end, and then reacted with KH to yield a potassium alcoholate of PIB (PIB-O-K+). PIB-O-K+ was immediately used as a macroinitiator to polymerize DMAEMA monomer, resulting in a cationic diblock copolymer PIB-b-PDMAEMA. With the similar synthesis procedure, the anionic diblock copolymer PIB-b-PMAA could be prepared via a combination of oxyanion-initiated polymerization of tert-butyl methacrylate (tBMA) and subsequent hydrolysis of tert-butyl ester groups in PtBMA block. The functional PIB and block copolymers have been fully characterized by 1H-NMR, FT-IR spectroscopy, and gel permeation chromatography (GPC). These samples allowed us to systematically investigate the effects of block composition on the pH responsivity and various self-assembled morphologies of the copolymers in THF/water mixed solvent. Transmission electron microscopy (TEM) images revealed that these diblock copolymers containing small amount of original PIB without exo-olefin-terminated group are able to self-assemble into micelles, vesicles with different particle sizes and cylindrical aggregates, depending on various factors including block copolymer composition, solvent polarity and pH value.  相似文献

FT-IR, DSC, and NMR techniques allowed the structural characterization of four copolymers formed by styrene and methacrylic units (methacrylic acid (MAA), dimethylamine ethyl methacrylate (DMAEMA), sodium methacrylate (MANa), and 1-hydroxyethyl methacrylate (HEMA). The copolymer composition was studied by Fourier transform-infrared spectroscopy (FT-IR) and nuclear magnetic resonance (NMR) spectroscopy. The thermal behavior of the block copolymers was analyzed by differential scanning calorimetry (DSC). Three of the four copolymers showed two transitions caused by changes in the polymer heat capacity (ΔCp) of each block. Diffusion-ordered spectroscopy (DOSY) experiments were used to distinguish copolymer from homopolymer mixtures. Finally, the triad-level stereosequences of styrene-methacrylic copolymers were obtained using 13C NMR. The results indicate that by increasing the alkyl-substituent length in the methacrylic block, the probability of syndiotactic polymerization increases.  相似文献

Diblock copolymers of poly[2‐(dimethylamino)ethyl methacrylate]‐block‐poly[di(ethylene glycol) methyl ether methacrylate], PDMAEMA‐b‐PDEGMA, were synthesized by reversible addition–fragmentation chain transfer polymerization. The block ratio was varied to study the influence on the lower critical solution temperature and the corresponding phase transition in water. Therefore, turbidimetry, differential scanning calorimetry (DSC), dynamic light scattering (DLS), and laser Doppler velocimetry were applied. Additionally, asymmetric flow field‐flow fractionation (AF4) coupled to DLS and multiangle laser light scattering (MALLS) was established as an alternative route to characterize these systems in terms of molar mass of the polymer chain and size of the colloids after the phase transition. It was found that AF4–MALLS allowed accurate determination of molar masses in the studied range. Nevertheless, some limitations were observed, which were critically discussed. The cloud point and phase transition of all materials, as revealed by turbidimetry, could be confirmed by DSC. For block copolymers with block ratios in the range of 50:50, a thermo‐induced self‐assembly into micellar and vesicular structures with hydrodynamic radii (Rh) of around 25 nm was observed upon heating. At higher temperatures, a reordering of the self‐assembled structures could be detected. The thermo‐responsive behavior was further investigated in dependence of pH value and ionic strength. Variation of the pH value mainly influences the solubility of the PDMAEMA segment, where a decrease of the pH value increases the transition temperature. A novel kind of pH‐sensitive brush copolymer [poly(2‐hydroxyethyl methacrylate)‐graft‐poly(ethylethylene phosphate)]‐block‐poly[2‐(dimethylamino)ethyl methacrylate] [(PHEMA‐g‐PEEP)‐b‐PDMAEMA] with biodegradable polyphosphoester as the side chains, and its self‐assembled aggregates were developed for nonviral gene delivery. The brush copolymers were synthesized via a combination of single‐electron transfer living radical polymerization and ring‐opening polymerization. The chemical structures of these brush copolymers were characterized by FTIR, 1H NMR, and 31P NMR measurements. The critical aggregation concentration values of (PHEMA‐g‐PEEP)‐b‐PDMAEMA in pH 7.4 buffer solution were determined by the fluorescence probe technique. The interaction of (PHEMA‐g‐PEEP)‐b‐PDMAEMA and DNA was studied by agarose gel retardation assay, and the formed complexes were further investigated by means of zeta potential, dynamic light scattering, and transmission electron microscopy measurements. In addition, the in vitro cytotoxicity and transfection tests indicated that these brush copolymers showed low toxicity and favorable transfection efficiency to HeLa cells. All these results demonstrated that these biocompatible brush copolymers may be a promising candidate as nonviral polymeric gene vector. © 2013 Wiley Periodicals, Inc. Well‐defined amphiphilic block copolymers were prepared by ring opening metathesis polymerization and their stimuli responsive behavior of formed micelles in aqueous solution was investigated. The hydrophobic core of the micelles consists of either a poly[5,6‐bis(ethoxymethyl)bicyclo[2.2.1]hept‐2‐ene]‐block with a glass transition Tg at room temperature or a poly[endo,exo[2.2.1]bicyclohept‐5‐ene‐2,3‐diylbis (phenylmethanone)] with a Tg of 143 °C. For the polyelectrolyte shell, the precursor block poly[endo,exo[2.2.1]bicyclohept‐5‐ene‐2,3‐dicarboxyclic tert‐butylester] was transformed into the free acidic block by cleavage of the tert‐butyl groups using trifluoroacetic acid. Micellar solutions were prepared by dialysis, dissolving the copolymers in dimethyl sulfoxide which was subsequently replaced by water. All polymers form micelles with radii between 10 and 20 nm at a pH‐value below 5, where the carboxylic acid groups are in the protonated state. The block copolymer micelles show a strong increase of the hydrodynamic radius with increasing pH‐value, due to the repulsion among the formed carboxylate anions resulting in a stretching of the polymer chains. In this state, the micelles exhibit responsive behavior to ionic strength where a contraction of the micelles is observed as the carboxylate charges are balanced by sodium ions, whereas no changes of the hydrodynamic radius on addition of salt are observed at low pH. © 2009 Wiley Periodicals, Inc. We report on the synthesis of novel amphiphilic hyperbranched copolymers, namely Hyperbranched-[poly (ethylene glycol) methyl ether methacrylate-co-lauryl methacrylate] (H-[P(OEGMA-co-LMA)]), obtained by reversible addition-fragmentation chain transfer (RAFT) polymerization utilizing the divinyl monomer, ethylene glycol dimethacrylate (EGDMA) as the branching agent. Molecular characterization by size exclusion chromatography (SEC) and proton nuclear magnetic resonance (1H-NMR) spectroscopy indicated the success of the polymerization. The self-assembly behavior in aqueous media was investigated by light scattering techniques and fluorescence spectroscopy. The hyperbranched copolymers form multimolecular aggregates of nanoscale dimensions with a low critical aggregation concentration. In addition, the model hydrophobic drug, curcumin (CUR), known also for its intrinsic fluorescence properties, was used in order to investigate the H-[P(OEGMA-co-LMA)] copolymers drug encapsulation ability. Curcumin is successfully loaded into the polymeric nanoparticles, as confirmed by dynamic light scattering and UV–Vis spectroscopy. Interestingly, curcumin hydrophobic interactions with the hyperbranched copolymers result in more well-defined co-assembled nanostructures, in terms of size and size distribution. The mixed copolymer-CUR nano-assemblies consist of small size nanoparticles (<100 nm) which exhibit relatively high size uniformity, colloidal stability and fluorescent properties. Overall, results signify that the biocompatible H-[P(OEGMA-co-LMA)] nanostructures could potentially serve as nanocarrier systems for drug delivery and bio-imaging applications.  相似文献

The aim of this work was the development of a versatile route for the preparation of temperature‐ and pH‐responsive hydrogels with small dimensions. The copolymerization of N,N‐dimethylaminoethyl methacrylate with various amounts (5 and 10 mol %) of dimethylmaleimidoethyl methacrylate in solution with 2,2′‐azobisisobutyronitrile as an initiator is described. The structural and molecular characterization of the copolymers was performed with proton nuclear magnetic resonance, Fourier transform infrared, and ultraviolet spectroscopy, as well as size exclusion chromatography. Differential scanning calorimetry and thermogravimetry were used for the thermal characterization of the copolymers. Micro‐ and nanohydrogels of the copolymers were prepared by photocrosslinking. The gels obtained by photocrosslinking were characterized with a combination of surface plasmon resonance and optical waveguide spectroscopy, dynamic light scattering, and scanning electron microscopy. A novel amphiphilic copolymer poly(ethylene glycol)-block-poly(N,N-dimethylamino-2-ethylmethacrylate)-blockpoly[6-(4-methoxy-azobenzene-4’-oxy) hexyl methacrylate](PEG-b-PDMAEMA-b-PMMAzo) was prepared by ATRP polymerization.The self-assembly and responsive behaviors were investigated by SEM,TEM,LLS and UV-Vis spectra.The results indicated that the copolymers can self-assemble into spherical structures in aqueous media.The aggregate size can be tuned by pH and temperature.The trans-cis isomerization behavior of the formed aggregates was also examined.Upon irradiation with a linear polarized light,the elongation degree of the aggregates was increased with the irradiation time.  相似文献

A series of well-defined poly[(ethylene oxide)-b-2-(dimethylamino)ethyl methacrylate] (PEO-b-PDMAEMA) diblock copolymers were synthesized by atom transfer radical polymerization (ATRP) techniques. Post-polymerization reactions were performed to transform a portion of the tertiary amine groups of the PDMAEpsilonMA into phosphorozwitterions. The aggregation behavior of the prepared zwitterionic block copolymers was investigated by static and dynamic light scattering techniques at 25 and 37 degrees C, in weakly basic and acidic aqueous solutions. Antiparasitic drugs used for the treatment of Leishmania were incorporated into the copolymer aggregates. The effect of the solution pH, the zwitterion content, temperature, and the quantity of the incorporated drug on the aggregation behavior of the copolymers was tested.  相似文献

Two new poly(ethylene oxide)-poly(styrene oxide) triblock copolymers (PEO-PSO-PEO) with optimized block lengths selected on the basis of previous studies were synthesized with the aim of achieving a maximal solubilization ability and a suitable sustained release, while keeping very low material expense and excellent aqueous copolymer solubility. The self-assembling and gelling properties of these copolymers were characterized by means of light scattering, fluorescence spectroscopy, transmission electron microscopy, and rheometry. Both copolymers formed spherical micelles (12-14 nm) at very low concentrations. At larger concentration (>25 wt%), copolymer solutions showed a rich phase behavior, with the appearance of two types of rheologically active (more viscous) fluids and of physical gels depending on solution temperature and concentration. The copolymer behaved notably different despite their relatively similar block lengths. The ability of the polymeric micellar solutions to solubilize the antifungal drug griseofulvin was evaluated and compared to that reported for other structurally-related block copolymers. Drug solubilization values up to 55 mg g−1 were achieved, which are greater than those obtained by previously analyzed poly(ethylene oxide)-poly(styrene oxide), poly(ethylene oxide)-poly(butylene oxide), and poly(ethylene oxide)-poly(propylene oxide) block copolymers. The results indicate that the selected SO/EO ratio and copolymer block lengths were optimal for simultaneously achieving low critical micelle concentrations (cmc) values and large drug encapsulation ability. The amount of drug released from the polymeric micelles was larger at pH 7.4 than at acidic conditions, although still sustained over 1 day.  相似文献

Here we report the synthesis and solution characterization of a novel series of AB diblock copolymers with neutral, water‐soluble A blocks consisting of N,N‐dimethylacrylamide and pH‐responsive B blocks of N,N‐dimethylvinylbenzylamine. To our knowledge, this represents the first example of an acrylamido–styrenic block copolymer prepared directly in a homogeneous aqueous solution. The best blocking order [with poly(N,N‐dimethylacrylamide) as a macro‐chain‐transfer agent] yielded well‐defined block copolymers with minimal homopolymer impurities. The reversible aggregation of these block copolymers in aqueous media was studied with 1H NMR spectroscopy and dynamic light scattering. Finally, an example of core‐crosslinked micelles was demonstrated by the addition of a difunctional crosslinking agent to a micellar solution of the parent block copolymer. © 2004 Wiley Periodicals, Inc. The synthesis of new stimulus-responsive block copolymer gelators using atom transfer radical polymerisation (ATRP) in either methanol or 2-propanol/water mixtures at 20 °C is described. Bifunctional and trifunctional initiators were used to prepare ABA triblock and I(BA)3 three-arm star diblock copolymers, respectively, using a ‘one-pot’ ATRP protocol, in which the central block comprised poly(glycerol monomethacrylate) and the outer blocks comprised pH-responsive poly[2-(diethylamino)ethyl methacrylate] or poly[2-(diisopropylamino)ethyl methacrylate]. These copolymers dissolve molecularly in acidic solution but formed free-standing gels at around neutral pH on addition of base. Gel strength was judged by both tube inversion experiments and shear rheometry measurements and a comparison between the linear and star architectures was made.  相似文献

Polydimethylsiloxane (PDMS) block copolymers were synthesized by using PDMS macroinitiators with copper-mediated living radical polymerization. Diamino PDMS led to initiators that gave ABA block copolymers, but there was low initiator efficiency and molecular weights are somewhat uncontrolled. The use of mono- and difunctional carbinol–hydroxyl functional initiators led to AB and ABA block copolymers with narrow polydispersity indices (PDIs) and controlled number-average molecular weights (Mn's). Polymerization with methyl methacrylate (MMA) and 2-dimethylaminoethyl methacrylate (DMAEMA) was discovered with a range of molecular weights produced. Polymerizations proceeded with excellent first-order kinetics indicative of living polymerization. ABA block copolymers with MMA were prepared with between 28 and 84 wt % poly(methyl methacrylate) with Mn's between 7.6 and 35 K (PDI <1.30), which show thermal transitions characteristic of block copolymers. ABA block copolymers with DMAEMA led to amphiphilic block copolymers with Mn's between 9.5 and 45.7 K (PDIs of 1.25–1.70), which formed aggregates in solution with a critical micelle concentration of 0.1 g dm−3 as determined by pyrene fluorimetry experiments. A series of novel temperature‐ and pH‐responsive graft copolymers, poly(L ‐glutamic acid)‐g‐poly(N‐isopropylacrylamide), were synthesized by coupling amino‐semitelechelic poly(N‐isopropylacrylamide) with N‐hydroxysuccinimide‐activated poly(L ‐glutamic acid). The graft copolymers and their precursors were characterized, by ESI‐FTICR Mass Spectrum, intrinsic viscosity measurements and proton nuclear magnetic resonance (1H NMR). The phase‐transition and aggregation behaviors of the graft copolymers in aqueous solutions were investigated by the turbidity measurements and dynamic laser scattering. The solution behavior of the copolymers showed dependence on both temperature and pH. The cloud point (CP) of the copolymer solution at pH 5.0–7.4 was slightly higher than that of the solution of the PNIPAM homopolymer because of the hydrophilic nature of the poly(glutamic acid) (PGA) backbone. The CP markedly decreased when the pH was lowered from 5 to 4.2, caused by the decrease in hydrophilicity of the PGA backbone. At a temperature above the lower critical solution temperature of the PNIPAM chain, the copolymers formed amphiphilic core‐shell aggregates at pH 4.5–7.4 and the particle size was reduced with decreasing pH. A series of fluoroalkyl end-capped diblock copolymers of poly[2-(N,N-dimethylamino)ethyl methacrylate] (PDMAEMA or PDMA) and poly[2-(N,N-diethylamino)ethyl methacrylate] (PDEAEMA or PDEA) have been synthesized via oxyanion-initiated polymerization, in which a potassium alcoholate of 4,4,5,5,6,6,7,7,7-nonafluoro-1-heptanol (NFHOK) was used as an initiator. The chemical structures of the NFHO-PDMA-b-PDEA and NFHO-PDEA-b-PDMA depended on the addition sequence of the two monomers and the feeding molar ratios of [DMA] to [DEA] during the polymerization process. These copolymers have been characterized by (1)H NMR and (19)F NMR spectroscopy and gel permeation chromatography (GPC). The aggregation behavior of these copolymers in aqueous solutions at different pH media was studied using a combination of surface tension, fluorescence probe, and transmission electron microscopy (TEM). Both diblock copolymers exhibited distinct pH/temperature-responsive properties. The critical aggregation concentrations (cacs) of these copolymers have been investigated, and the results showed that these copolymers possess excellent surface activity. Besides, these fluoroalkyl end-capped diblock copolymers showed pH-induced lower critical solution temperatures (LCSTs) in water. TEM analysis indicated that the NFHO-PDMA(30)-b-PDEA(10) diblock copolymers can self-assemble into the multicompartment micelles in aqueous solutions under basic conditions, in which the pH value is higher than the pKa values of both PDMA and PDEA homopolymers, while the NFHO-PDEA(10)-b-PDMA(30) diblock copolymers can form flowerlike micelles in basic aqueous solution.  相似文献

The desorption and subsequent pH-responsive behavior of selectively quaternized poly(2-(dimethylamino)ethyl methacrylate)-block-poly(2-(diethylamino)ethyl methacrylate) (PDMA-PDEA) films at the silica/aqueous solution interface has been characterized. The copolymer films were prepared at pH 9, where micelle-like surface aggregates are spontaneously formed on silica. The subsequent rinse with a copolymer-free electrolyte solution adjusted to pH 9 causes partial desorption of the weakly or non-quaternized copolymers, but negligible desorption for the highly quaternized copolymers. Further rinsing with a pH 4 electrolyte solution results in additional desorption and extension (swelling) of the remaining adsorbed copolymer film normal to the interface. This pH-responsive behavior is reversible for two pH cycles (9-4-9-4) as monitored by both quartz crystal microbalance with dissipation monitoring (QCM-D) and also zeta potential measurements. The magnitude of the pH-responsive behavior depends on the mean degree of quaternization of the PDMA block. Moreover, a combination of contact angle data, zeta potential measurements and in situ atomic force microscopy (AFM) studies indicates that the pH-responsive behavior is influenced not only by the number of cationic binding sites on the adsorbed copolymer chains but also by the adsorbed layer structure.  相似文献
