Cellulose trisphenylcarbamate is regioselectively bonded to 3-aminopropyl silica gel and underivatized silica gel, respectively, at the 6-position of the primary hydroxyl group on the glucose unit of cellulose with 4,4'-diphenylmethane diisocyanate (DPDI) as a spacer. Enantioseparations are evaluated on these prepared chiral stationary phases (CSPs) with several organic acids as the modifiers in the mobile phase by high-performance liquid chromatography. The influence of the amount of DPDI used on chiral resolution is investigated. Also, the corresponding coated-type phase is also prepared for the aim of comparison. It is observed that the bonded-type phase shows a lower chiral recognition power but a better column efficiency than the coated-type phase under the liquid chromatographic mobile phase with hexane-alcohol. However, the bonded-type CSPs are compatible with a wider number of solvents such as tetrahydrofuran (THF) or chloroform, which generally result in the solubility or swelling of the cellulose derivatives on the coated-type CSPs. The results obtained from this study indicate that the bonded-type CSP may provide complementary enantioselectivity over the coated-type phase by adopting THF as a component in the mobile phase.  相似文献

Immobilized polysaccharide-based chiral stationary phases (CSPs) are gaining importance in the resolution of racemic compounds due to their stable nature on working with normal solvents and those prohibited for use with coated phases (tetrahydrofuran, chloroform, dichloromethane, acetone, 1,4-dioxane, ethyl acetate, and certain other ethers). This review discusses the use of immobilized polysaccharide CSPs in the chiral resolution of various racemates by liquid chromatography. The discussion includes immobilization methodologies, enantioselectivities, efficiencies, and a comparison of chiral recognition capabilities of coated vs. immobilized CSPs. Some applications of immobilized CSPs to the chiral resolution of racemic compounds are also presented.  相似文献

Immobilized polysaccharide‐based columns showed excellent enantioselectivity in normal phase separation mode. In this work, enantioseparation abilities of four immobilized polysaccharide‐derived chiral stationary phases (Chiralpak IA, Chiralpak IB, Chiralpak IC, and Chiralpak ID) toward 15 azole compounds were evaluated. Separation was carried out using n‐hexane as mobile phase with ethanol, 1‐propanol, 1‐butanol, and 2‐propanol as modifiers. And twelve compounds have achieved baseline separation with the resolutions ranging between 2.05 and 21.73. The enantioseparation on the four polysaccharide‐based chiral columns using different alcohol modifiers was compared. In general, the best separation performance was identified as Chiralpak IC, which was able to resolve 11 compounds to baseline and two partially under the screening conditions. Separation on Chiralpak IB was not satisfactory, because only four compounds were baseline separated.  相似文献

A sensitive and accurate LC method for the determination of AT13148 enantiomeric purity has been developed and validated. Baseline separation with a resolution higher than 1.8 was accomplished within 15 min using a Chiralpak AD-H column (250 × 4.6 mm; particle size 5 μm) and n-hexane: 2-propanol: diethylamine (85:15:0.1, v/v) as mobile phase at a flow rate of 1 mL min?1. Eluted analytes were monitored by UV absorption at 254 nm. The effects of mobile phase components, temperature and flow rate on enantiomeric selectivity and resolution of enantiomers were investigated. Calibration curves were plotted within the concentration range between 7 and 500 μg mL?1 (n = 11), and the recoveries between 98.24 and 100.99% were obtained, with relative standard deviation lower than 1.32%. LOD and LOQ for AT13148 were 2.46 and 7.38 μg mL?1 and for its enantiomer were 2.54 and 7.49 μg mL?1, respectively. It was demonstrated that the developed method was accurate, robust and sensitive for the determination of enantiomeric purity of AT13148, especially for the analysis of bulk samples.  相似文献

Chiral stationary phases (CSP) for LC had a major impact on pharmaceutical R&D when they first became commercially available in the 1980s. Even although the use of CSP in pharmaceutical R&D is now very much a mature area, there is still scope for using CSP more effectively to bring about efficiencies. One such instance is the possibility of combining the chiral LC test for the level of a trace enantiomeric impurity in a chiral drug substance and the LC test for related substances into one test. It was envisaged that this could be achieved by carrying out reversed-phase LC on an ODS silica/CSP coupled column system. In evaluating Chiralpak QD-AX, Cyclobond I 2000 DNP and Whelk-O1 CSP using a polar organic - aqueous mobile phase it was found that the Whelk-O1 CSP had good achiral selectivity, the required match of retentivity with the ODS silica material, ACE 5 C18 and also exhibited an encouraging degree of enantioselectivity in the reversed-phase mode. Following consideration of the selectivity of the ACE 5 C18 and Whelk-O1 phases it became apparent that it might be possible to achieve the desired goal of achieving both the enantiomeric impurity and related substances separations in one system by using the Whelk-O1 CSP on its own. This was subsequently demonstrated to be the case using S-naproxen, laevokalim and S-flurbiprofen as illustrative examples.  相似文献

Four chiral stationary phases, based on the phenylcarbamate derivatives of amylose or cellulose: Chiralcel OD-H, Chiralpak AD, Lux Cellulose-2 and Lux Amylose-2, were evaluated for the enantiomeric separation of an acetamide chiral intermediate, the (4S-trans)-4-(ethylamino)-4-(N-acetamide)-5,6-dihydro-(6S)-methyl-4H-thieno-[2,3-b]thiopyran-7,7-dioxide, using SFC. The effect of the different modifiers and temperatures, on the separation, was also studied. The chiral separation could not be achieved using the Chiralpak AD column, nevertheless the other columns provided excellent results with analysis times close to 6 min and resolutions higher than 2. The highest enantioresolutions and retentions were obtained with the Lux Cellulose-2 column and 2-propanol as organic modifier. The isoelution temperatures were estimated from the van't Hoff plots, and in all the cases they were above the temperature range studied which means that the enantiomeric separation was enthalpy driven.  相似文献

Fused silica capillary columns coated with several alkyl or acyl cyclodextrin derivatives, especially those of α- and β-cyclodextrins, are suitable for the enantiomer separation of a wide variety of volatile compounds of different molecular size and functionality. Positional isomers and more than 250 pairs of optical isomers have been resolved, including chiral hydrocarbons, acetals, ethers, epoxides, carbonates, lactones, esters, acids, ketones, aldehydes, alcohols, halocarbons, and also nitrogen-and sulfur-containing compounds. The physical properties of the cyclodextrin derivatives, even those obtained as viscous fluids, could be improved by dissolving them in polysiloxane liquid phases commonly used for GLC.  相似文献

Summary In preparative column liquid chromatography, it is necessary to work with conditions of mass overload to obtain high throughput per unit time. The influence of sample mass and of eluent composition on the separation of R,S-1-(1-naphthylethyl)propylamide on chiral 3,5-dinitrobenzoylphenylglycine-silica (Pirkle type stationary phase) was investigated. At a sample load of 200μg of racemate per gram of stationary phase, the column becomes overloaded. At higher sample loads the peaks become triangular; therefore the first peak is always pure up to loads of 10mg g−1, although resolution is low. The purity of the second peak depends on sample mass, but not on the strength (polarity) of the mobile phase. This is due to the effect that peak width, as well as resolution, decrease as the polarity of the eluent increases. In certain cases the polarimeter, used as a detector, can give more information on peak purity than the UV detector.  相似文献

In this work, the enantiomeric separation of ten anticholinergic drugs was first examined on two derivative polysaccharide chiral stationary phases (CSPs), i.e., Chiralpak ID and Chiralpak IA in the normal phase mode. Except for scopolamine hydrobromide, the remaining nine analytes could be completely separated with good resolutions using both columns under the optimized mobile phase conditions. And the enantiomeric discrimination ability of the studied CSPs towards nine analytes was in the order of Chiralpak ID > Chiralpak IA. The influences of organic modifier types, alcohol content, and base/acid additives on the enantiomeric separation were evaluated and optimized. According to the experimental results, the effect of the structures of analytes on enantiomeric separation was discussed. Additionally, the chiral recognition mechanisms were proposed based on the thermodynamic analysis of the experimental data.  相似文献

During the last decade, chiral monolithic stationary phases have been prepared and used for rapid enantioseparations in CEC and HPLC. Various chiral selectors are used to prepare these CSPs. The preparation, properties, and applications of these CSPs are discussed in this paper. Attempts have been made to describe optimization strategies and the chiral recognition mechanisms. A comparison of chiral separations in CEC and HPLC is described. Efforts have also been made to predict the future perspectives and challenges of chiral monolithic stationary phases. The most effective chiral selectors include polysaccharides, cyclodextrins, and macrocyclic glycopeptide antibiotics. These chiral phases produced acceptable analytical enantiomeric separation of a variety of racemates. However, the development of these CSPs for preparative‐scale separations is needed.  相似文献

The direct enantioseparation of a novel aminothiazolecarboxamide fungicide, ethaboxam, on polysaccharide-derived chiral stationary phases (CSPs) is described. Good resolution is achieved with several polysaccharide-derived CSPs. Chiralcel OD (OD-H) and Chiralpak AS are excellent for direct enantiomer separation of ethaboxam. The elution behavior and the effects of eluent composition on the resolution of ethaboxam are also investigated. The direct chiral separation by chiral stationary phases (CSPs) is one of the most important techniques to analyze enantiomeric purity as well as to get enantiomerically pure material quickly. Among various types of CSPs, polysaccharide type CSPs are well known by their versatility and durability. They are not only effective under normal-phase conditions, but also under reversed-phase conditions. In order to get a good separation under the reversed-phase conditions, it is the key to choose an appropriate mobile phase. For example, a simple mixture of water/acetonitrile or water/methanol are sufficient for a neutral analyte, while it is necessary to use an acidic solution instead of water for an acidic analyte and a solution of a chaotropic salt (or a basic solution) for a basic analyte, respectively. The paper also presents lists of more than 350 separation examples that include 22 validated methods for drug analyses from serum, plasma, or urine samples on polysaccharide type CSPs under reversed-phase conditions.  相似文献
