The reaction of 3-methyl-1-phenyl-5-pyrazolene with arylideneamino nitriles in alcohol leads to the formation of 3-methyl-4(1-aryl-2,2-dicyanoethyl)-5-pyrazolones, which are readily cyclized in the presence of bases to the corresponding 6-amino-5-cyano-3-methyl-4-aryl- 1H,4H-pyrazolo[3,4-b]pyrans. The title compounds 6 have been prepared in a one-step procedure from the corresponding 4-aryl substituted 5-alkoxycarbonyl-6-methyl-3,4-dihydropyridones 4 in good yields. Quantum chemical calculations reveal a non-planar molecule with a distorted dihydropyridone ring and two favoured conformations. The 13C nmr data and theoretical calculations support a strong push-pull effect on the olefinic moiety.  相似文献

Substituted 2-aminobenzo[b]pyrans were synthesized by three-component condensation of aromatic aldehydes, cyanoacetic acid derivatives, and cyclic 1,3-diketones. The molecular and crystal structure of 2-amino-3-(2-methoxyethoxycarbonyl)-4-(2-nitrophenyl)-5-oxo-5,6,7,8-tetrahydro-4H-benzo[b]pyran was established by X-ray diffraction analysis.  相似文献

10-Methyl-2-chloro- and 2-chloro-3-methyl-3,4-diazaphenoxazines as well as 2-oxo-3,10-dimethyl-2,3-dihydro-3, 4-diazaphenoxazine were obtained by methylation of 2-chloro-3,4-diazaphenoxazines. Depending on the conditions, alkylation of 2-chloro-3,4-diazaphenoxazine with -diethylaminoethyl chloride gave 1-ethylimidazolino[1,21,2]pyridazo[3,4-b]-1,4-benzoxazine or 2-ethoxy-10-(-diethylaminoethyl)-3,4-diazaphenoxazine. The first asymmetric synthesis of 3-alkoxycarbonyl-2-amino-4-aryl-4H-naphtho[1,2-b]pyrans, by Michael addition of 1-naphthol to chirally modified arylidenecyanoacetates 6 and 7 , is described. Good yields and low diastereomeric excesses have been obtained in the 1,4-conjugate additions. The absolute stereochemistry at C-4 in major isomers of pyrans 8 and 9 has been assigned as 5 by X-ray analysis of major pyran 8 .  相似文献

In this paper , 10 novel 6-aryl-3-(5'-phenyl-2H-tetrazole-2'-methylene)-S-triazolo[3, 4-b]-1,3,4-thiadiazoles have been synthesized by the condensation of 3-(5'-phenyl-2H-tetrazole-2'-methylene)-4-amino-5-mercapto-1,2,4-triazole with various aromatic carboxylic acids in the presence of phosphorus oxychloride. The structure of these compounds was determined by elemental analysis , UV, IR, 1H NMR and MS. Their spectral properties were also discussed.  相似文献

A one-step procedure was developed for the synthesis of 5-acetyl-2-amino-4-aryl-3-cyano-4H-pyrano[3,2-b]indoles involving the three-component reaction of 1-acetylindol-3(2H)-one with aromatic aldehydes and malononitrile in ethanol in the presence of triethylamine as the catalyst. The structure of 5-acetyl-2-amino-4-(4"-chloro-3"-nitrophenyl)-3-cyano-4H-pyrano[3,2-b]indole was established by X-ray diffraction analysis.  相似文献

This study presents an efficient synthesis of 3-[6-(substituted-phenyl)-[1,2,4]triazolo[3,4-b][1,3,4] thiadiazol-3-yl]-1H-indazole via dehydrative condensation with cyclization of 4-amino-5-(1H-indazol-3-yl)-4H-[1,2,4]triazole-3-thiol and fluorinated or nonfluorinated carboxylic acids in presence of phosphorous oxychloride. The multistep reaction pathway proceeds through different compounds. Present synthesis has the advantages of easily accessible starting materials, convenient synthesis, simple reaction condition, wider substrate scope, and higher yield (75% to 90% isolated).  相似文献

The fused heterocycle 4-(phenylsulfonyl)-4H-furo[3,4-b]indole, which is an indole-2,3-quinodimethane synthetic analogue, is prepared in five steps from indole in 46% yield. A similar sequence is used to synthesize C-3 derivatives (3-methyl, 3-phenyl, and 3-heptyl). Thus, indole-3-carbaldehyde (1) is protected as the N-phenylsulfonyl derivative 2 and converted to the ethylene acetal 6. Lithiation at C-2 followed by treatment with an aldehyde affords the expected hydroxy acetals 7 and 8. Exposure to acid effects cyclization to the furoindoles 5 and 9. Furthermore, C-1 lithiation of furo[3,4-b]indole 9c followed by treatment with methyl iodide affords disubstituted furo[3,4-b]indole 10.  相似文献

Olefinic diols, prepared from (R)-(+)-2,2-dimethyl-1,3-dioxolane-4-carboxaldehyde via olefination and hydrolysis, were converted into enantiomerically pure hydroxy substituted tetrahydrofuran derivatives by cyclization using N-phenylselenophthalimide and BF3. The PhSe group in the C-4 position of these tetrahydrofurans was then substituted by an allyl group using allyltributylstannane in the presence of AIBN. The selenium promoted cyclizations of the allyl tetrahydrofurans in which the OH and the allyl groups are trans to each other formed the enantiopure perhydrofuro[3,4-b]pyrans, while the cyclization of the allyl tetrahydrofurans in which the OH and the allyl groups are cis gave rise to the perhydrofuro[3,4-b]furans. These bicyclic products were finally deselenenylated with triphenyltin hydride and AIBN.  相似文献

The reactions of 6-methoxybenzo[b]furan-3(2H)-one with 2-aryl-1,1-dicyanoethylenes and malononitrile or with aromatic aldehyde and two moles of malononitrile afford 2-amino-4-aryl-1,3-dicyano-7-methoxydibenzo[b,d]furans. 以2-溴甲基-6,7-亚甲二氧基-3-喹啉酸乙酯(1) 为原料,经三步合成了2-叔丁基-4-氟-苯并氧杂卓并[3,4-b]喹啉并[9,11]二氧戊环-14(6H)-酮(4).其中,6-(2-氟-4-叔丁基-苯氧甲基)-[1,3]二氧戊环并[4,5-g]喹啉-7-羧酸是经一锅法合成的.它的分子内闭环反应是在Eaton's reagent(P2O5-CH3SO3H)的作用下,在较温和的条件下进行的.所得新化合物3和4的结构经IR、1H NMR和元素分析得以证实.  相似文献

A survey of literature1 revealed that s-triazolo[3,4-b]-1,3,4-thiadiazole, an interesting fused system of s-triazole and 1,3,4-thiadiazole rings, has received much attention during recent years on account of its prominent utilizations as antifungal, antiinflammatory, antiviral, analgesic and anthelmintic agents probably resulting from its planner and compact structure. Our earlier work on the synthesis of this class of heterocycles showed antibacterial, herbicidal and plant growth regulative properties2-3 for the compounds. 1,2,3-Triazole derivatives have found their wide use in medicine, agriculture and industry4-5. Incorporation of 1,2,3-triazole moiety into the 6-position of this ring system may lead to achieving compounds of better biological activities. In view of the above findings coupled with scanty reports on these condensed compounds carrying 6-heterocyclic groups, we wish to report here the condensation of 4-amino-5-mercapto-3-(4-pyridyl)-l,2,4-triazole(2) with 1-aryl-4-carboxy-5-methyl-1,2,3-triazoles(1a-j) as a part of our continuing interest in this area.  相似文献
