A regioselective synthesis of 2-aminotriazinones 6a-d is reported, by reaction of 3-methylthio-1,2,4-triazinones 5a-d with O-(2,4-dinitrophenyl)hydroxylamine (2) as an amino-transfer agent. A spectroscopic study and an unequivocal synthesis of 2-amino-4-methyl-6-phenyl-1,2,4-triazinedione ( 11a ) has shown the site of amination to be N2 of the 1,2,4-triazinone ring. Subsequent reaction of 2-amino-1,2,4-triazinones 6a-b with amines, followed by ring closure with aliphatic acids provided [1,2,4]triazolo[2,3-b][1,2,4]triazine-7(1H)ones 13a-e. Conversion of [1,2,4]triazolo[2,3-b][1,2,4]triazinone 13c to unsubstituted [1,2,4]triazolo[2,3-6][1,2,4]triazine (15) was attained.  相似文献

In the current study, a convenient and simple way is presented to synthesize a novel type of supported heterogeneous organocatalyst in 21-81% yield by the copolymerization of 9-amino-9-deoxy-epi-cinchonine organocatalyst with acrylonitrile using AIBN as radical initiator. The chemical compositions (x/y) and weight-average molecular weights of copolymers 1a-d were determined by (1)H NMR and GPC analysis respectively. Their porous and layered structure, and surface morphology were characterized by nitrogen adsorption-desorption, XRD and TEM. In the asymmetric aldol addition of p-nitrobenzaldehyde to cyclohexanone and 1-hydroxy-2-propanone in water, all the supported organocatalysts 1a-d afforded excellent isolated yields (90.2-94.7%) and stereoselectivities (96.8-97.8%ee anti, anti/syn = 91/9). The highest catalytic property (96% yield, anti/syn = 90/10 and 99%ee anti) in water as the sole solvent was achieved under the optimized conditions. Compared with cyclohexanone, cyclopentanone and acetone showed the less desired enantioselectivities in the same aldol reactions. At the end of the aldol reaction, the copolymer-supported organocatalyst 1a was readily recovered in 95-98% yield from reaction mixture by simple filtration using an organic membrane. Even in the fifth run, there was no significant loss in catalytic activity and stereocontrol (94.3% yield, 97.2%ee anti, anti/syn = 90/10). After continuous reuse five times, there was some drop in catalytic activity and stereoselectivity.  相似文献

Chemoselective SN2' condensation of primary enolates of alkyl methyl ketones 2a-e with dimethyl bromomethylfumarate (1) followed by highly diastereoselective NaBH4 reduction of the ketone function in the formed ketodiesters 3a-e and the regioselective in situ lactonization of the unisolable intermediates 4a-e exclusively furnished the cis-3,5-disubstituted gamma-butyrolactones (+/-)-5a-e in very good yields. Similarly, the face-selective coupling reaction of cyclohexanone enolate with 1 to form a mixture of diastereomers in an 8:2 ratio followed by a highly selective reductive cyclization of 9 plus 10 exclusively provided the cis-octahydrobenzofuran (+/-)-12 in 70% overall yield.  相似文献

l-Phenyl-lH-tetrazol-5-yIhydrazine (2) was reacted with nitrous acid to yield 1-phenyl-lH-tetrazolo[4,5-d]tetrazole (3). l-Arylidene-2-(l-phenyl-lH-tetrazol-5-yl)hydrazines (4) were generally reactive towards electrophilic reagents. When treated with bromine in acetic acid, 4 yielded mixtures of 1-arylidene-2-[1-(4-bromophenyl)-lH-tetrazol-5-yl]hydrazines (5a-d) and 2-[1-(4-bromophenyl)-lH-tetrazol-5-yl]hydrazidic bromides (6a-d). Solvolysis of 6a-d in aqueous acetone yielded 5-aryl-1-(4-bromophenyl)-1,2,4-triazolo[4,3-d]tetrazoles (7a-d). The structures of the synthesized compounds were confirmed on the basis of elemental analysis, IR and 1H NMR data.  相似文献

The preparation of 1,2,4-triazolo[1,5-c]quinazolines 4a-d , 5 , 8a-d by cyclocondensation of 1a-c with carboxylic acids and carboxylic anhydrides, respectively, is described. By different pathways, the 5-thioxo-5,6-dihydro-1,2,4-triazolo[1,5-c]quinazolines 4a-d react with hydrazine hydrate or amines with the formation of 5-substituted 1,2,4-triazolo[1,5-c]quinazolines 9 and 10a-d . Cyclocondensation of 9 with carboxylic acids, carboxylic anhydrides, and nitrous acid, respectively, leads to the new anellated heterocycles bis-1,2,4-triazolo[4,3-a:1,5-c]quinazoline 13 and tetrazolo [1,5-a]-1,2,4-triazolo[1,5-c]quinazoline ( 14 ).  相似文献

A new route to C-6-selenenyl analogs of compound 1a from 5-alkyl-6-chlorouracils 6a-b has been described. A mild and highly efficient synthesis of 1-(alkoxymethyl)-5-alkyl-6-(arylselenenyl)uracils 8a-e has been accomplished from 6a-b in good yields using a two step procedure. Silylation of 5-alkyl-6-chlorouracils 6a-b using N,O-bis(trimethylsilyl)acetamide followed by regioselective alkylation of the silylated intermediate with ethyl or benzyl chloromethyl ether in dichloromethane afforded the desired 1-(alkoxymethyl)-5-alkyl-6-chlorouracils 7a-d in 88–94% yields. Compounds 7a-d readily underwent addition-elimination reaction with an appropriate arylselenol in the presence of ethanolic sodium hyroxide to produce the corresponding 1-(alkoxymethyl)-5-alkyl-6-(arylselenenyl)uracils 8a-e in excellent yields (94–99%).  相似文献

L-proline amides derived from various chiral beta-amino alcohols that bear substituents with various electron natures at their stereogenic centers are prepared and evaluated for catalyzing the direct Aldol reaction of 4-nitrobenzaldehyde with acetone. Catalysts with strong electron-withdrawing groups are found to exhibit higher catalytic activity and enantioselectivity than their analogues with electron-donating groups. The presence of 2 mol % catalyst 4g significantly catalyzes the direct Aldol reactions of a wide range of aldehydes with acetone and butanone, to give the beta-hydroxy ketones with very high enantioselectivities ranging from 96% to >99% ee. High diastereoselectivity of 95/5 was observed for the anti Aldol product from the reaction of cyclohexanone, and excellent enantioselectivity of 93% ee was provided for anti Aldol product from the reaction of cyclopentanone.  相似文献

6-Substituted benzyl-4-phenyl-3-thioxo-2,3,4,5-tetrahydro-1,2,4-triazin-5-ones 3a-d were prepared and converted into their corresponding 3-methylthio derivatives 4a-d . Reaction of compounds 4a-d with hydrazine hydrate gave the corresponding 4-amino-3-anilino-4,5-dihydro-1,2,4-triazin-5-ones 5a-d . 6-Substituted benzyl-4-phenyl-2,3,4,5-tetrahydro-1,2,4-triazin-3,5-diones 9a-c were synthesized and allowed to react with hydrazine hydrate to give the corresponding 6-substituted benzyl-4-amino-2,3,4,5-tetrahydro-1,2,4-triazin-3,5-diones 10a-c . The biological evaluation of some of these triazines is described. All compounds were screened for antiviral, antibacterial, antimycobacterial, antifungal and antiyeast activity. No important biological activity was found.  相似文献

A reaction of 2-aminobenzenesulfonamide ( 1 ) with 2-chloroethyl or 3-chloropropyl isothiocyanate in isopropanol afforded 2-(2′-chloroethylthioureido)- and 2-(3′-chloropropylthioureido)benzenesulfonamides ( 2a,b ) in 67% and 55% yield respectively. Treatment of 2a,b with triethylamine in methanol at room temperature furnished 3-(2′-aminoethylthio)- and 3-(3′-aminopropylthio)-2H-1,2,4-benzothiadiazine 1,1-dioxides ( 9a,b ) in quantitative yield. Heating 2b to reflux in methanol under neutral conditions gave 9b but in the form of the hydrochloride 8b which could be converted into the free base 9b by treating with ammonia water. When compounds 2a,b were treated with triethylamine in methanol at elevated temperature, 3-(2′-mercaptoethylamino)- and 3-(3′-mercaptopropylamino)-2H-1,2,4-benzothiadiazine 1,1-dioxides ( 10a,b ) were obtained in good yield. Alternatively, 10a,b could also be prepared from 9a,b in 95% and 77% yield respectively.  相似文献

A robust, regioselective synthesis of 3-amino-1,2,4-triazoles is described. This reaction employs a key intermediate 2, which is coupled to carboxylic acids in good yields to afford intermediates 3a-d. These entities, in turn, react with a variety of hydrazines or hydrazine hydrochlorides to provide proposed intermediates 4a-j, which under microwave conditions cyclize to the desired 3-amino-1,2,4-triazoles (compounds 5a-j). This approach permits the rapid synthesis of regioselective N1-substituted 3-amino-1,2,4-triazoles, and is shown to afford a variety of such compounds in 34-70% isolated yields.  相似文献

The chemical behavior of 4-methyl and 4-phenyl-5-oxo-1H-4,5-dihydro-1,2,4-benzotriazepines towards methylating and acidic agents has been investigated. The 4-methyl derivative, when treated with methyl fluorosulfonate furnished, after crystallization from water, a quaternary salt (2,4-dimethyl-5-oxo-1H-4,5-dihydro-1,2,4-benzotriazepinium fluorosulfonate), whereas from the 4-phenyl derivative, a complex mixture was obtained, which, after boiling with water, afforded 2-methylindazolone, aniline fluorosulfonate and formic acid. In acidic medium the 4-methyl derivative underwent an isomerization reaction yielding 1-imino-3-methylquinazolin-4-one, but the 4-phenyl derivative exclusively yielded products resulting from ring opening. In alkaline medium, both compounds gave hydrolytic cleavage products.  相似文献

In this study appropriate hydrazide compounds, furan-2-carboxylic acid hydrazide (1) and phenylacetic acid hydrazide (2) were converted into 1,4-substituted thiosemicarbazides 4a-e and 5a-e and 4-amino-5-(furan-2-yl or benzyl)-4H-1,2,4-triazole-3-thiols 7 and 10. The 1,4-substituted thiosemicarbazides were then converted into 5-(furan-2-yl or benzyl)-4-(aryl)-4H-1,2,4-triazole-3-thiols 8a-e and 9a-e. In addition, the azomethines 11a-d and 12a-d were prepared from the corresponding arylaldehydes and the 4-amino-5-(furan-2-yl or benzyl)-4H-1,2,4-triazole-3-thiols 7 and 10. The structures of all the synthesized compounds were confirmed by elemental analyses, IR, (1)H-NMR and(13) C-NMR spectra.  相似文献

It was shown that pyrimido[4,5-e]-1,2,4-triazine-6,8-diones enter the reversed azadiene synthesis reaction with ketones and vinyl ethyl ether in the presence of diethylamine or boron trifluoride etherate, and also with enamines. As a result of the reaction, pyrido[2,3-d]pyrimidine-2,4-diones are formed in good yield. Pyrimido[5,4-e]-1,2,4-triazine-5,7-diones do not undergo such reactions with acetone. Reactions of 4,4'-bipyridine (1) with excess of polyfluoroalkyl iodides (2a-d) at 100-110 degrees C, under neat conditions, led to the formation of monoquaternary salts (3a-d) in > 90% yields. Salts 3a-d were metathesized with LiN(SO2CF3)2 either in water or water/acetone mixtures to form ionic liquids (4a-d), respectively, in > 88% yields. When 1 was reacted with 2.5 equivalent of 2a-c in DMF at 110 degrees C, the diquaternary salts 5a-c were formed in > 85% yields. Alternatively, 5a-c were also synthesized by heating a mixture of 3a-c and 2a-c (1.25 equivalent) in DMF. The metathesis reaction of 5a-c with LiN(SO2CF3)2 produced dicationic ionic liquids (6a-c) in > 86% yield.  相似文献

The reaction of 3-ethoxycarbonyl-5-perfluoroalkyl-1,2,4-oxadiazoles with hydroxylamines has been investigated, evidencing the possibility of competitive reaction paths. Nucleophilic addition of hydroxylamine to the electrophilic C(5) of the 1,2,4-oxadiazole ring, followed by ring-opening and ring-closure with enlargement, leads to high yield and in very mild experimental conditions to the formation of 5-hydroxyamino-3-perfluoroalkyl-6H-1,2,4-oxadiazin-6-ones, one of these presenting water gelation ability. In turn, reactions with N-methylhydroxylamine lead the exclusive formation of 4-perfluoroacylamino-2-methyl-2H-1,2,5-oxadiazol-3-ones through the well known Boulton-Katritzky rearrangement.  相似文献

Three-component reaction of 1-[(4-chlorophenyl)sulfanyl]acetone, malononitrile, and substituted aromatic aldehydes in the presence of sodium ethoxide under simple mixing at ambient temperature for 5–8 min afforded highly functionalized 4H-pyrans in good to excellent yields. 1,3-Dipolar cycloaddition of nitrile oxides over the nitrile functionality of the 4H-pyrans furnished 1,2,4-oxadiazoles in moderate yields.
