Superparamagnetic dextran nanoparticles were prepared by coprecipitation. Effects of concentration of dextran, amount of ironic salts, stirring speed, concentration of ammonium hydroxide and mole ratio of Fe3+/Fe2+ on the effective diameter of dextran magnetic nanopartides (DMNP) were studied. Dextran immunological magnetic nanoparticles (DIMNP) were formed by the reaction of the monoclonal anti-human mammary cancer antibody with DMNP oxidized by sodium periodate, and the properties of magnetic susceptibility, shape and retention of antibody activity were investigated. The in vitro cancer antigen binding ability of DIMNP was demonstrated by radioimmunoassay, and the in vivo magnetic localization and antibody targeting ability of radiolabeled DIMNP were studied.  相似文献

Biodegradable PLA composites were prepared using microcrystalline cellulose (MCC) and silver (Ag) nanoparticles. The main objective of the present study is to develop new biopolymer composites with good mechanical properties, thermal stability, maintaining the optical transparency and also providing antimicrobial properties through silver nanoparticle introduction. Composites were prepared with 1%wt of Ag nanoparticles and 5%wt of MCC using a twin-screw microextruder; film parameters were optimized in order to obtain a thickness range between 20 and 60 μm.PLA composites maintained optical transparency properties of the matrix, while MCC was able to reduce polymer permeability. Thermal analysis revealed that MCC increased PLA crystallinity and the mechanical properties of the composites demonstrated that tensile modulus was improved by microcrystalline cellulose.  相似文献

A novel nano in situ gel forming system of 5-Fluorouracil (5-FU) was investigated for its potential use for conjunctival/corneal squamous cell carcinoma (CCSC). The study was conducted in two steps, in the first step PLA nanoparticles were prepared and characterized; in the second step the drug loaded PLA nanoparticles were dispersed in sodium alginate solution yielding the modified nano in situ system, which were evaluated in rabbit eye. Size and morphology of prepared PLA particles were verified by using dynamic light scattering (DLS), atomic force microscope (AFM) and scanning electron microscope (SEM). In vitro and in vivo study of free 5-FU, PLA nanoparticles and modified nano in situ system were conducted in simulated tear fluid and in rabbit eye respectively. PLA nanoparticles were in size range of 128-194 nm with spherical shape and smooth surface with narrow size distribution. No polymer drug interaction was found as confirmed by FTIR, NMR and DSC. XRD of PLA nanoparticles confirmed that 5-FU was present in the crystalline state. In vitro experiments indicated a diffusion controlled release of 5-FU from both PLA nanoparticles and modified nano in situ system with high burst effect. Modified nano in situ gel system (MNS) significantly increased the Cmax and AUC0-8 in aqueous humor as compared to 5-FU solution and PLA nanoparticles. Higher 5-FU level in aqueous humor was possibly because of increased retention time of gel matrix-embedded drug loaded nanoparticles. Overall results showed the potential of MNS for ophthalmic delivery in the therapy of CCSC.  相似文献

The assembly of poly(lactic acid) (PLA) nanoparticles on a 12-aminodecanoic acid (ADA) self-assembled monolayer (SAM) is described. Assembly is accomplished through electrostatic interactions between the positively charged SAM and the negatively charged PLA nanoparticles. The strategy used involves two steps in which a preliminary electrochemical coating of the ADA SAM is followed by a second step that involves immersing the SAM in a solution containing gold or PLA nanoparticles. The SAM was characterized by using cyclic voltammetry (CV), X-ray photoelectron spectroscopy (XPS), FTIR spectroscopy, and contact angle measurements, whereas scanning electron microscopy (SEM) was used to image the nanoparticles after electrostatic attachment was achieved. We found that the surface coverage of the nanoparticles could be controlled by modulating the electrostatic interactions between the negatively charged particles and the positively charged SAM surface by varying the pH of the nanoparticle solution, the immersion time, and the number of cyclic voltammetry scans under which the SAM was formed.  相似文献

Pyrene-loaded biodegradable polymer nanoparticles were prepared by incorporating pyrene into the polymer nanoparticles formulated from amphiphilic diblock copolymer, methoxy poly(ethylene glycol)–poly(lactic acid) (MePEG–PLA). Their morphological structure and physical properties were characterized by nuclear magnetic resonance (NMR), dynamic light scattering, fluorescence spectroscopy, transmission electronic microscopy and zeta potential measurements. Further, MePEG–PLA nanoparticles containing pyrene as fluorescent marker were administered intranasally to rats, and the distribution of nanoparticles in the nasal mucosa and the olfactory bulb were visualized by fluorescence microscopy. NMR results confirmed that MePEG–PLA copolymer can form nanoparticles in water, and hydrophilic PEG chains were located on the surface of the nanoparticles. The particle size, zeta potential and pyrene loading efficiency of MePEG–PLA nanoparticles were dependent on the PLA block content in the copolymer. Following nasal administration, the absorption of nanoparticles across the epithelium was rapid, with fluorescence observed in the olfactory bulb at 5 min, and a higher level of fluorescence persisted in the olfactory mucosa than that in the respiratory mucosa. These results show that pyrene could serve as a useful fluorescence probe for incorporation into polymer nanoparticles to study tissue distribution and MePEG–PLA nanoparticles might have a great potential as carriers of hydrophobic drugs.  相似文献

The aim of the present work was to evaluate if the presence of a polyethylenglycol (PEG) coating around PLA nanoparticles would affect their interaction with biological surfaces, following oral administration to rats. For this purpose, a model antigen, 125I-radiolabeled tetanus toxoid, was encapsulated in PLA and PLA-PEG nanoparticles by a modified water-in-oil-in-water solvent evaporation technique. Firstly, the stability of the nanoparticles in simulated gastrointestinal fluids was evaluated. Results showed an interaction between the nanoparticles and the enzymes of the digestive fluids, this interaction being considerably reduced by the PEG coating around the particles. On the other hand, the PLA forming the nanoparticles was found to be only slightly degraded (9% converted to lactate for PLA nanoparticles and 3% for PLA-PEG nanoparticles) and that the encapsulated tetanus toxoid remained mostly associated to the nanoparticles upon incubation in the digestive fluids for up to 4 h. Finally, the in vivo experiments showed that, after oral administration to rats, the levels of encapsulated radioactive antigen in the blood stream and lymphatics were higher for PLA-PEG nanoparticles than for PLA nanoparticles. In conclusion, the PLA-PEG nanoparticles have a promising future as protein delivery systems for oral administration.  相似文献

In this study, a highly toughened PLA was prepared through physical melt-blending with EVA at the presence of hydrophilic nanosilica and SEBS-g-MA block copolymer compatibilizer. The effect of nanosilica and compatibilizer on the morphology, mechanical properties, and linear rheology of the PLA/EVA blends was also investigated. According to TEM images, nanosilica was selectively located in the PLA matrix while some were placed on the interface between the two polymers as was also predicted by thermodynamic and kinetic analysis. Upon the addition of nanoparticles, the interfacial adhesion between the phases was enhanced and the average droplet size decreased. Interestingly, incorporation of SEBS-g-MA induced morphological changes as the spherical EVA droplets turned into a cylindrical shape. DSC results indicated that blending with EVA copolymer resulted in the reduction of crystallization of PLA matrix; however, the crystallinity increased at the presence of nanoparticles up to 5 wt%. The addition of compatibilizer considerably hindered the crystallization of the PLA phase. PLA/EVA blend containing optimum levels of nanosilica exhibited considerably enhanced tensile toughness, elongation at break, and impact strength. On the other hand, the simultaneous addition of nanoparticles and SEBS-g-MA led to synergistic toughening effects and the compatibilized blend containing nanosilica exhibited excellent impact toughness. For instance, the elongation at break of the compatibilized PLA/EVA blend containing the optimal content of nanosilica was increased from 7% to 121% (compared to neat sample). The notched Izod impact strength was also increased from 5.1 to 65 kJ/m2. Finally, the microstructure of the blends was assessed by rheological measurements.  相似文献

PLA nanocomposite films with multifunctional characteristics such as mechanical, anti-UV, antibacterial, electrical, gas barrier properties are potentially of high interest as packaging biomaterials. Occasionally, desired and beneficial effects obtained by addition nanofillers come along with some drawbacks, leading to the sharp drop in the molecular weights of the polyester chains, and consequently an important loss of mechanical and thermal properties. Novel PLA-ZnO nanocomposite films were produced by melt-compounding PLA with 0.5–3% ZnO rod-like nanoparticles. The surface treatment of nanofiller by silanization (with triethoxy caprylylsilane) was necessary to obtain a better dispersion and to limit the decrease of molecular mass of PLA. The morphology, molecular, thermo-mechanical and transport properties to water vapor of PLA-ZnO films were analyzed with respect to the neat PLA. According to DSC and to XRD, the produced films were essentially amorphous. The changes in PLA permeation properties were strongly dependent on temperature and nanofiller loading. The well dispersed ZnO nanoparticles within the polyester matrix were effective in increasing the tortuosity of the diffusive path of the penetrant molecules. The activation energy remained similar for PLA and PLA-1% ZnO, but was found greater at higher loading of ZnO (3%), confirming the increased difficulty of travelling molecules to diffuse through PLA. In comparison to the neat PLA (presenting no antimicrobial efficacy), the nanocomposites were active against both Gram-positive and Gram-negative bacteria, stronger antibacterial activity being evidenced after 7 days elapsed time. By considering the multifunctional properties of PLA-ZnO nanocomposites, the films produced by extrusion can be considered a promising alternative as environmental-friendly packaging materials.  相似文献

A new phosphorus‐based organic additive (PDA) was designed and successfully synthesized using a three‐component reaction for improvement of the thermal and combustion resistance of polylactic acid (PLA). For compensate for mechanical properties of PLA, hydroxyapatite nanoparticles was modified via in situ surface modification with PDA and was used for preparation of PLA nanocomposites. The structure and morphology as well as thermal, combustion, and mechanical properties of the all PLA systems were investigated. The X‐ray diffraction (XRD) and field‐emission scanning electron microscopy (FE‐SEM) results indicated that the presence of PDA as surface modifier has been necessary for a desirable dispersion of hydroxyapatite (HA) nanoparticles in the PLA matrix. The thermal, combustion, and mechanical properties of the PLA system films were investigated using thermogravimetric analysis (TGA), microscale combustion calorimeter (MCC), and tensile test, respectively. The initial decomposition temperature and char residue of PLA containing 6 mass% of PDA along with 2 mass% HA nanoparticles were increased 20°C and 12% respectively, compared with that of the neat PLA. The peak of heat release rate was decreased from 566 W/g for the neat PLA to 412 W/g for PLA containing 2 mass% of PDA along with 6 mass% HA nanoparticles. By incorporation of only 2 mass% HA nanoparticles and 6 mass% of PDA, the tensile strength was obtained 51 MPa higher than that of the neat PLA.  相似文献

Polylactic acid (PLA) was modified by poly (butylene adipate-co-terephthalate) (PBAT) and nano-attapulgite (AT) using the melt blending technique. Ethylene-butyl acrylate-glycidyl methacrylate (E-BA-GMA) was used as a compatibilizer which can bond the AT nanoparticles with PLA/PBAT matrix by interaction between the epoxy and hydroxyl groups. The effects of the AT content on the mechanical properties, thermal properties, crystallinity and morphology of PLA/PBAT/ATT nanocomposites were investigated. The results showed that the tensile strength, elongation at break and impact strength of PLA/PBAT could be simultaneously increased by incorporating AT nanoparticles. PLA/PBAT/AT nanocomposites possessed higher thermal stability than pure PLA/PBAT. In the ternary composite system of PLA/PBAT/AT, AT acted as a heterogeneous nucleating agent and was able to increase the crystallization temperature. When the AT content was low (≤2.5 wt%), AT nanoparticles could uniformly disperse in the PLA/PBAT matrix. In general, AT was an effective filler to reinforce and toughen PLA/PBAT blend simultaneously, and the PLA/PBAT/AT nanocomposite with 2.5 wt% AT exhibited a good combination of strength and toughness.  相似文献

Low molecular weight (M(w)) poly(L-lactic acid) (PLA) nanoparticles were coated with polyelectrolytes (PEs) by layer-by-layer (LbL) technique using a filtration approach. Poly(allylamine hydrochloride) and poly(sodium 4-styrenesulfonate) were applied as PEs in coating. LbL coating is aimed to use in producing (nano)particulate drug delivery systems with improved biocompatibility and sustained or targeted release of drug substances. Nanoparticles of rapidly biodegradable polymers, like the low M(w) PLA, open up a possibility to control the release of the encapsulated substance by the coating, but set challenges to the coating process due to increased aggregation tendency and degradation rate of the polymer. When the core PLA nanoparticles were prepared by nanoprecipitation, surface properties of the nanoparticles were affected by solvent selection. Successful LbL coating of the PLA nanoparticles was obtained only with chloroform, but not with dichloromethane as the solvent during nanoprecipitation. Reason for this was found to be the more charged surface of the nanoparticles prepared with chloroform compared to the nanoparticles prepared with dichloromethane.  相似文献

Maleic anhydride (MA) grafted poly(lactic acid) (PLA) (PLA-g-MA) was synthesized from PLA. Proton nuclear magnetic resonance confirmed the grafting of the MA. PLA-g-MA and PLA were used to prepare polymeric nanoparticles. Particle size distributions were measured by dynamic light scattering, and colloidal stability was determined by (zeta) ζ-potential. The ζ-potential becomes more negative for PLA-g-MA than PLA nanoparticle dispersions, due to the presence of deprotonated carboxylic acid groups on the backbone of the PLA and confirms the MA grafting results. Maleic anhydride grafted on PLA backbone improves the covalent conjugation with ovalbumin (OVA) compared to OVA physically adsorbed on the particles. The chemical conjugation was carried out via amide linkages between the carboxylic groups of the nanoparticles, activated with 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide, and the amino groups of the protein. The amount of protein conjugated was measured by using the bicinchoninic acid method and is threefold higher compared to the adsorbed OVA. Moreover, the PLA-g-MA nanoparticles increased the amount of conjugated OVA by 36 wt% compared to PLA nanoparticles. OVA adsorption and OVA conjugation provided colloidal dispersions with excellent stability.  相似文献

In the presence of surfactant, water-insoluble poly(D,L-lactide) (PLA) was dispersed into narrowly distributed nanoparticles stable in water via microphase inversion. The structure and degradation of such formed nanoparticles were investigated by a combination of static and dynamic laser light scattering. Our results revealed that the degradation rate increased with the temperature and pH so that the degradation could be regulated from minutes to days. Using anionic sodium dodecyl sulfate (SDS) as stabilizer resulted in a slower degradation than using cationic hexadecyltrimethylammonium bromide (HTAB). The phthalocyanine chromophores (PC) could be encapsulated inside these PLA nanoparticles. The degradation of individual PLA nanoparticles led to a controllable releasing of PC. The absorption and fluorescence studies revealed a correlation between the degradation and the releasing of PC. Our results showed that a higher PC/PLA ratio could lead to a faster degradation.  相似文献

The purpose of this investigation was to design novel pentablock copolymers (polylactide–polycaprolactone–polyethylene glycol–polycaprolactone–polylactide) (PLA–PCL–PEG–PCL–PLA) to prepare nanoparticle formulations which provide continuous delivery of steroids over a longer duration with minimal burst effect. Another purpose was to evaluate the effect of poly(l-lactide) (PLLA) and poly(d,l-lactide) (PDLLA) incorporation on crystallinity of pentablock copolymers and in vitro release profile of triamcinolone acetonide (selected as model drug) from nanoparticles. PLA–PCL–PEG–PCL–PLA copolymers with different block ratio of PCL/PLA segment were synthesized. Release of triamcinolone acetonide from nanoparticles was significantly affected by crystallinity of the copolymers. Burst release of triamcinolone acetonide from nanoparticles was significantly minimized with incorporation of proper ratio of PDLLA in the existing triblock (PCL–PEG–PCL) copolymer. Moreover, pentablock copolymer-based nanoparticles exhibited continuous release of triamcinolone acetonide. Pentablock copolymer-based nanoparticles can be utilized to achieve continuous near–zero-order delivery of corticosteroids from nanoparticles without any burst effect.  相似文献

Capillary electrophoretic (CE) methods were used for the quantitative determination of model drugs [salbutamol sulphate (SS), sodium cromoglycate (SCG) and beclomethasone dipropionate (BDP)] in poly(D,L-lactic acid) (PLA) nanoparticles, which were prepared by the nanoprecipitation method. Zeta potential and size distribution of the nanoparticles were determined by electrophoretic mobility determinations and photon correlation spectroscopy, respectively. Interactions between the drugs, the PLA nanoparticles and the fused-silica capillary were investigated by electrokinetic capillary chromatography (EKC). A quantitative CE method was developed for salbutamol sulphate and sodium cromoglycate, and the linearity and repeatability of migration times, peak areas and peak heights were determined. Microemulsion electrokinetic chromatography was used for the quantitative determination of beclomethasone dipropionate. According to this study, the applied electromigration techniques were suitable for the interaction, drug entrapment and dissolution studies of pharmaceutical nanoparticles. The results suggest that even quantitation of the drug located inside the nanoparticles was possible. Encapsulation of the more hydrophilic model drugs (SS, SCG) in the PLA nanoparticles was less efficient than in the case of BDP.  相似文献

Composite fibers composed of poly(l-lactide)-grafted hydroxyapatite (PLA-g-HAP) nanoparticles and polylactide (PLA) matrix were prepared by electro-spinning. Environmental scanning electron microscope (ESEM) and transmission electron microscopy (TEM) were employed to investigate the morphology of the composite fibers and the distribution of PLA-g-HAP nanoparticles in the fibers, respectively. At a low content (∼4 wt%) of PLA-g-HAP, the nanoparticles dispersed uniformly in the fibers and the composite fibrous mats exhibited higher strength properties, compared with the pristine PLA fiber mats and the simple hydroxyapatite/PLA blend fiber mats. But when the content of PLA-g-HAP further increased, the nanoparticles began to aggregate, which resulted in the deterioration of the mechanical properties of the composite fiber mats. The degradation behaviors of the composite fiber mats were closely related to the content of PLA-g-HAP. At a low PLA-g-HAP content, degradation may be delayed due to the reduction of autocatalytic degradation of PLA. When PLA-g-HAP content was high, degradation rate increased because of the enhanced wettability of the composite fibers and the escape of the nanoparticles from fiber surfaces during incubation.  相似文献

The morphology and thermal stability of melt-mixed poly(lactic acid) (PLA)/poly(ε-caprolactone) (PCL) blend nanocomposites with small amounts of TiO2 nanoparticles were investigated. The nanoparticles were mostly located in the PLA phase, with good dispersion of individual particles, although significant aggregation was also visible. The thermal stability and degradation behaviour of the different samples were studied using thermogravimetric analysis (TGA) and TGA-Fourier-transform infrared (FTIR) spectroscopy. Neat PCL showed better thermal stability than PLA, but the degradation kinetics revealed that PLA had a higher activation energy of degradation than PCL, indicating its degradation rate more strongly depends on temperature, probably because of a more complex degradation mechanism based on chain scission and re-formation. Blending of PLA and PCL reduced the thermal stabilities of both polymers, but the presence of TiO2 nanoparticles improved their thermal stability. The nanoparticles also influenced the volatilization of the degradation products from the blend, acted as degradation catalyst and/or retarded the escape of volatile degradation products.  相似文献

Polylactic acid (PLA) films containing 1 wt % and 3 wt % of lignin nanoparticles (pristine (LNP), chemically modified with citric acid (caLNP) and acetylated (aLNP)) were prepared by extrusion and characterized in terms of their overall performance as food packaging materials. Morphological, mechanical, thermal, UV–Vis barrier, antioxidant and antibacterial properties were assayed; appropriate migration values in food simulants and disintegration in simulated composting conditions were also verified. The results obtained indicated that all lignin nanoparticles succeeded in conferring UV-blocking, antioxidant and antibacterial properties to the PLA films, especially at the higher filler loadings assayed. Chemical modification of the fillers partially reduced the UV protection and the antioxidant properties of the resulting composites, but it induced better nanoparticles dispersion, reduced aggregates size, enhanced ductility and improved aesthetic quality of the films through reduction of the characteristic dark color of lignin. Migration tests and disintegration assays of the nanocomposites in simulated composting conditions indicated that, irrespectively of their formulation, the multifunctional nanocomposite films prepared behaved similarly to neat PLA.  相似文献
