The polyzwitterionic brushes comprised of poly(2‐methacryloyloxyethyl phosphorylcholine) (pMPC) segments, which are used for surface modification of polymers and biocompatible coatings, were investigated. In this work, reverse surface‐initiated atom transfer radical polymerization (RATRP) of zwitterionic 2‐methacryloyloxyethyl phosphorylcholine (MPC) is employed to tailor the functionality of graphene oxide (GeneO) in a well‐controlled manner and produce a series of well‐defined hemocompatible hybrids (termed as GeneO‐g‐pMPC). The complexes were characterized by FT‐IR, XRD, and Raman. Results show that MPC has been coordinated on the graphene oxide sheet. Thermal stability of the nanocomposites in comparison with the neat copolymer is revealed by thermogravimetric analysis and differential thermal analysis. Scanning electron microscopy and transmission electron microscope images of the nanoconposite displays pMPC chains were capable of existing on GeneO sheet by RATRP. The biocompatibility properties were measured by plasma recalcification profile tests, hemolysis test, and MTT assays, respectively. The results confirm that the pMPC grafting can substantially enhance the hemocompatibility of the GeneO particles, and the GeneO‐g‐pMPC hybrids can be used as biomaterials without causing any hemolysis. With the versatility of RATRP and the excellent hemocompatibility of zwitterionic polymer chains, the GeneO‐g‐pMPC nanoparticles with desirable blood properties can be readily tailored to cater to various biomedical applications. The crystal unit‐cell structures and the isothermal crystallization kinetics of poly(L ‐lactide) in biodegradable poly(L ‐lactide)‐block‐methoxy poly(ethylene glycol) (PLLA‐b‐MePEG) diblock copolymers have been analyzed by wide‐angle X‐ray diffraction and differential scanning calorimetry. In particular, the effects due to the presence of MePEG that is chemically connected to PLLA as well as the PLLA crystallization temperature TC are examined. Though we observe no variation of both the PLLA and MePEG crystal unit‐cell structures with the block ratio between PLLA and MePEG and TC, the isothermal crystallization kinetics of PLLA is greatly influenced by the presence of MePEG that is connected to it. In particular, the equilibrium melting temperature of PLLA, T, significantly decreases in the diblock copolymers. When the TC is high so that the crystallization is controlled by nucleation, because of the decreasing T and thereafter the nucleation density with decreasing PLLA molecular weight, the crystallinity of PLLA also decreases with a decrease in the PLLA molecular weight. While, for the lower crystallization temperature regime controlled by the growth mechanism, the crystallizability of PLLA in copolymers is greater than that of pure PLLA. This suggests that the activation energy for the PLLA segment diffusing to the crystallization site decreases in the diblocks. © 2006 Wiley Periodicals, Inc. Microwave (MW)‐assisted ring‐opening polymerization (ROP) provides a rapid and straightforward method for engineering a wide array of well‐defined poly(3‐hydroxyalkanoate)‐b‐poly(D,L ‐lactide) (PHA‐b‐PLA) diblock copolymers. On MW irradiation, the bulk ROP of D,L ‐lactide (LA) could be efficiently triggered by a series of monohydroxylated PHA‐based macroinitiators previously produced via acid‐catalyzed methanolysis of corresponding native PHAs, thus affording diblock copolyesters with tunable compositions. The dependence of LA polymerization on temperature, macroinitiator structure, irradiation time, and [LA]0/[PHA]0 molar ratio was carefully investigated. It turned out that initiator efficiency values close to 1 associated with conversions ranging from 50 to 85% were obtained only after 5 min at 115 °C. A kinetic investigation of the MW‐assisted ROP of LA gave evidence of its “living”/controlled character under the experimental conditions selected. 2‐Methacryloyloxyethyl phosphorylcholine (MPC), a biomimetic monomer, was grafted from silicon wafer surfaces at room temperature by combining self‐assembly of initiator and surface‐initiated atom transfer radical polymerization. Two methods were used to control the grafting process. One was to add free initiator to the reaction system; the other was to add excess deactivator. The grafting densities up to 0.3 chains/nm2 were obtained. The surface thickness increased linearly with MPC conversion. The thickness depended on catalyst and monomer concentrations, as well as activator/deactivator ratio. Poly(MPC) layers of >100 nm thick were obtained by optimizing the polymerization conditions. A second block of either poly(MPC) or poly[2‐(dimethylamino)ethyl methacrylate] was also grown from the grafted poly(MPC), demonstrating the system livingness. X‐ray photoelectron spectroscopy was used to examine the surface chemical compositions showed good agreement with the theoretical values. © 2004 Wiley Periodicals, Inc. ABA block copolymers of methyl methacrylate and methylphenylsilane were synthesized with a methodology based on atom transfer radical polymerization (ATRP). The reaction of samples of α,ω‐dihalopoly(methylphenylsilane) with 2‐hydroxyethyl‐2‐methyl‐2‐bromoproprionate gave suitable macroinitiators for the ATRP of methyl methacrylate. The latter procedure was carried out at 95 °C in a xylene solution with CuBr and 2,2‐bipyridine as the initiating system. The rate of the polymerization was first‐order with respect to monomer conversion. The block copolymers were characterized with 1H NMR and 13C NMR spectroscopy and size exclusion chromatography, and differential scanning calorimetry was used to obtain preliminary evidence of phase separation in the copolymer products. © 2002 Wiley Periodicals, Inc. Linear and four‐armed poly(l ‐lactide)‐block‐poly(d ‐lactide) (PLLA‐b‐PDLA) block copolymers are synthesized by ring‐opening polymerization of d ‐lactide on the end hydroxyl of linear and four‐armed PLLA prepolymers. DSC results indicate that the melting temperature and melting enthalpies of poly (lactide) stereocomplex in the copolymers are obviously lower than corresponding linear and four‐armed PLLA/PDLA blends. Compared with the four‐armed PLLA‐b‐PDLA copolymer, the similar linear PLLA‐b‐PDLA shows higher melting temperature (212.3 °C) and larger melting enthalpy (70.6 J g?1). After these copolymers blend with additional neat PLAs, DSC, and WAXD results show that the stereocomplex formation between free PLA molecular chain and enantiomeric PLA block is the major stereocomplex formation. In the linear copolymer/linear PLA blends, the stereocomplex crystallites (sc) as well as homochiral crystallites (hc) form in the copolymer/PLA cast films. However, in the four‐armed copolymer/linear PLA blends, both sc and hc develop in the four‐armed PLLA‐b‐PDLA/PDLA specimen, which means that the stereocomplexation mainly forms between free PDLA molecule and the inside PLLA block, and the outside PDLA block could form some microcrystallites. Although the melting enthalpies of stereocomplexes in the blends are smaller than that of neat copolymers, only two‐thirds of the molecular chains participate in the stereocomplex formation, and the crystallization efficiency strengthens. © 2014 Wiley Periodicals, Inc. Novel thermo‐responsive poly(N‐isopropylacrylamide)‐block‐poly(l ‐lactide)‐block‐poly(N‐isopropylacylamide) (PNIPAAm‐b‐PLLA‐b‐PNIPAAm) triblock copolymers were successfully prepared by atom transfer radical polymerization of NIPAAm with Br‐PLLA‐Br macroinitiator, using a CuCl/tris(2‐dimethylaminoethyl) amine (Me6TREN) complex as catalyst at 25 °C in a N,N‐dimethylformamide/water mixture. The molecular weight of the copolymers ranges from 18,000 to 38,000 g mol?1, and the dispersity from 1.10 to 1.28. Micelles are formed by self‐assembly of copolymers in aqueous medium at room temperature, as evidenced by 1H NMR, dynamic light scattering (DLS) and transmission electron microscopy (TEM). The critical micelle concentration determined by fluorescence spectroscopy ranges from 0.0077 to 0.016 mg mL?1. 1H NMR analysis in selective solvents confirmed the core‐shell structure of micelles. The copolymers exhibit a lower critical solution temperature (LCST) between 32.1 and 32.8 °C. The micelles are spherical in shape with a mean diameter between 31.4 and 83.3 nm, as determined by TEM and DLS. When the temperature is raised above the LCST, micelle size increases at high copolymer concentrations due to aggregation. Poly(D ,L ‐lactide) and poly(D ,L ‐lactide‐co‐glycolide) with various composition and with one methacrylate and one carboxylate end group were synthesized and grafted onto poly(vinyl alcohol) (PVA) via the carboxylate group. The graft copolymers were crosslinked via the methacrylate groups using a free radical initiator. The polymer networks were characterized by means of NMR and studied qualitatively by means of IR spectroscopy. The influence of the glycolide content in the polyester grafts and of the number of ester units in the grafts on thermal properties and swellability were studied as well. The high swellability in water is characteristic of all hydrogels. Differential scanning calorimetry (DSC) showed a single glass transition temperature that occurs in the range between 51 and 69 °C. Thermogravimetric analysis (TGA) of the networks showed the main loss in weight in the temperature range between 290 and 370 °C. © 2007 Wiley Periodicals, Inc. This paper aims to report the fabrication of biodegradable thin films with micro‐domains of cylindrical nanochannels through the solvent‐induced microphase separation of poly(L ‐lactide)‐block‐poly(ethylene glycol)‐block‐poly(L ‐lactide) (PLA‐b‐PEG‐b‐PLA) triblock copolymers with different block ratios. In our experimental scope, an increase in each of the block lengths of the PLA and PEG blocks led to both a variation in the average number density (146 to 32 per 100 µm2) and the size of the micro‐domains (140 to 427 nm). Analyses by atomic force microscopy (AFM) and fluorescence microscopy indicated that the hydrophilic PEG nanochannels were dispersed in the PLA matrix of the PLA‐b‐PEG‐b‐PLA films. We demonstrated that the micro‐domain morphology could be controlled not only by the block length of PEG, but also by the solvent evaporation conditions.

In the present work, sequential interpenetrating polymer networks (IPNs) based on silicone and poly(2‐methacryloyloxyethyl phosphorylcholine) (PMPC) were developed with improved protein resistance. The structure and morphology of the IPNs were characterized by Fourier transform infrared spectroscopy (FTIR) and transmission electron microscopy (TEM). The results showed that the IPNs exhibited heterogeneous morphology. The bulk properties such as water content, ion permeability, and mechanical strength of the IPNs were determined by gravimetric method, ionoflux measurement technique, and tensile tester, respectively. The surface characteristics of the IPNs were investigated by X‐ray photoelectron spectroscopy (XPS) and contact angle measurements. XPS analysis suggested that PMPC was present on the surface as well as in the bulk material. The IPNs possessed more hydrophilic surface than pristine silicone revealed by contact angle measurements. Bovine serum albumin (BSA) was used as a model protein to evaluate protein resistance by a bicinchoninic acid assay method. The result revealed that the protein adsorption on the IPNs was significantly reduced compared to pristine silicone. These results suggest that the IPNs based on silicone and PMPC may be developed as novel ophthalmic biomaterials. Copyright © 2010 John Wiley & Sons, Ltd.  相似文献

The block glycopolymer, poly(2‐(α‐d ‐mannopyranosyloxy)ethyl methacrylate)‐b‐poly(l ‐lactide) (PManEMA‐b‐PLLA), was synthesized via a coupling approach. PLLA having an ethynyl group was successfully synthesized via ring‐opening polymerization using 2‐propyn‐1‐ol as an initiator. The ethynyl functionality of the resulting polymer was confirmed by MALDI‐TOF mass spectroscopy. In contrast, PManEMA having an azide group was prepared via AGET ATRP using 2‐azidopropyl 2‐bromo‐2‐methylpropanoate as an initiator. The azide functionality of the resulting polymer was confirmed by IR spectroscopy. The Cu(I)‐catalyzed 1,3‐dipolar cycloaddition between PLLA and PManEMA was performed to afford PManEMA‐b‐PLLA. The block structure was confirmed by 1H NMR spectroscopy and size exclusion chromatography. The aggregating properties of the block glycopolymer, PManEMA16kb‐PLLA6.4k (M n,PManEMA = 16,000, M n,PLLA = 6400) was examined by 1H NMR spectroscopy, fluorometry using pyrene, and dynamic light scattering. The block glycopolymer formed complicated aggregates at concentrations above 21 mg·L?1 in water. A monomode microwave reactor was used for the synthesis of designed star‐shaped polymers, which were based on dipentaerythritol with six crystallizable arms of poly(ε‐caprolactone)‐b‐poly(L ‐lactide) (PCL‐b‐PLLA) copolymer via a two‐step ring‐opening polymerization (ROP). The effects of irradiation conditions on the molecular weight were studied. Microwave heating accelerated the ROP of CL and LLA, compared with the conventional heating method. The resultant hexa‐armed polymers were fully characterized by means of FTIR, 1H NMR spectrum, and GPC. The investigation of thermal properties and crystalline behaviors indicated that the crystalline behaviors of polymers were largely depended on the macromolecular architecture and the length of the block chains. © 2010 Wiley Periodicals, Inc. The synthesis and characterization of a photocleavable block copolymer containing an ortho‐nitrobenzyl (ONB) linker between poly(methyl methacrylate) and poly(d ‐lactide) blocks is presented here. The block copolymers were synthesized via atom transfer radical polymerization (ATRP) of MMA followed by ring‐opening polymerization (ROP) of d ‐Lactide and ROP of d ‐lactide followed by ATRP of MMA from a difunctional photoresponsive ONB initiator, respectively. The challenges and limitations during synthesis of the photocleavable block copolymers using the difunctional photoresponsive ONB initiator are discussed. The photocleavage of the copolymers occurs under mild conditions by simple irradiation with 302 nm wavelength UV light (Relative intensity at 7.6 cm: 1500 μW/cm2) for several hours. © 2013 Wiley Periodicals, Inc. Biodegradable star‐shaped poly(ethylene glycol)‐block‐poly(lactide) copolymers were synthesized by ring‐opening polymerization of lactide, using star poly(ethylene glycol) as an initiator and potassium hexamethyldisilazide as a catalyst. Polymerizations were carried out in toluene at room temperature. Two series of three‐ and four‐armed PEG‐PLA copolymers were synthesized and characterized by gel permeation chromatography (GPC) as well as 1H and 13C NMR spectroscopy. The polymerization under the used conditions is very fast, yielding copolymers of controlled molecular weight and tailored molecular architecture. The chemical structure of the copolymers investigated by 1H and 13C NMR indicates the formation of block copolymers. The monomodal profile of molecular weight distribution by GPC provided further evidence of controlled and defined star‐shaped copolymers as well as the absence of cyclic oligomeric species. The effects of copolymer composition and lactide stereochemistry on the physical properties were investigated by GPC and differential scanning calorimetry. For the same PLA chain length, the materials obtained in the case of linear copolymers are more viscous, whereas in the case of star copolymer, solid materials are obtained with reduction in their Tg and Tm temperatures. © 2007 Wiley Periodicals, Inc. Using 2‐chloropropionamide derivative of poly(propyleneimine) dendrimer DAB‐dendr‐(NH2)32 (DAB‐32‐Cl) as the macroinitiator, atom transfer radical polymerization of styrene was successfully carried out in DMF medium. The monodisperse poly(propyleneimine)–polystyrene (dendrimer–PSt) particles with diameters smaller than 100 nm could be prepared. The morphology, size, and size distribution of the dendrimer–PSt particles were characterized by transmission electron microscopy (TEM) and photon correlation spectroscopy (PCS). The effects of reaction temperature, the ratio of St/macroinitiator, and reaction time on the size, and size distribution of the dendrimer–PSt nanoparticles were investigated. In a selective solvent (DMF/H2O), polymers can self‐assemble into different aggregate configurations such as regular microsphere and wire‐like thread. © 2008 Wiley Periodicals, Inc. Phosphorylcholine‐containing poly(L‐lactide) (PLLA‐PC) was synthesized by ring‐opening polymerization of L‐lactide in the presence of glycerophosphorylcholine originated from egg lecithin. Self‐assembling micelles were then obtained by film hydration, ultrasonication and stirring. Transmission electron microscopy and confocal laser scanning microscopy confirmed the micellar structure with hydrophobic core and hydrophilic shell. The critical micellar concentration (CMC) value of PLLA‐PC was only 1/50 that of naturally occurring PC, in agreement with a better surfactant property of the former. Dynamic light scattering showed that the size and size distribution of micelles varied with dilution, but the CMC was independent of the concentration of NaCl solution within 0.9 wt%, indicating that the micelles could be stable upon intravenous injection. In addition, the micelle solution could be stored at 4 °C over 30 days without any noticeable changes, whereas at 37 °C, the size, size distribution and the number of micelles decreased over time due to degradation. The solubility of clofazimine, a highly hydrophobic drug, was found to be 11.9 µg/ml in the PLLA‐PC micellar solution, which was 40 times that in pure water. This preliminary study suggests that PLLA‐PC micelles present a great potential as delivery system for hydrophobic drugs. Copyright © 2011 John Wiley & Sons, Ltd.  相似文献

A novel class of biomimetic glycopolymer–polypeptide triblock copolymers [poly(L ‐glutamate)–poly(2‐acryloyloxyethyllactoside)–poly(L ‐glutamate)] was synthesized by the sequential atom transfer radical polymerization of a protected lactose‐based glycomonomer and the ring‐opening polymerization of β‐benzyl‐L ‐glutamate N‐carboxyanhydride. Gel permeation chromatography and nuclear magnetic resonance analyses demonstrated that triblock copolymers with defined architectures, controlled molecular weights, and low polydispersities were successfully obtained. Fourier transform infrared spectroscopy of the triblock copolymers revealed that the α‐helix/β‐sheet ratio increased with the poly(benzyl‐L ‐glutamate) block length. Furthermore, the water‐soluble triblock copolymers self‐assembled into lactose‐installed polymeric aggregates; this was investigated with the hydrophobic dye solubilization method and ultraviolet–visible analysis. Notably, this kind of aggregate may be useful as an artificial polyvalent ligand in the investigation of carbohydrate–protein recognition and for the design of site‐specific drug‐delivery systems. © 2004 Wiley Periodicals, Inc. To develop new hemopurification systems based on cellulose membrane, we synthesized a graft copolymer of cellulose with poly(2‐methacryloyloxyethyl phosphorylcholine) (MPC) by a metal‐catalyzed atom transfer radical polymerization process in homogeneous media. First, cellulose was dissolved in a DMAc/LiCl solution system, and it reacted with 2‐bromoisobutyloyl bromide to produce macroinitiator (cell‐BiB). Then, MPC was polymerized to the cellulose backbone in a homogeneous DMSO/methanol mixture solution in the presence of cell‐BiB. Characterization with FT‐IR, NMR, and GPC measurements showed that there obtained a graft copolymer of cellulose backbone and poly(MPC) side chains (cell‐PMPC) with well‐defined structure, indicating a controlled/“living” radical polymerization. The proteins adsorption studies showed that cellulose membranes modified by the as‐prepared cell‐PMPC owns good protein adsorption resistance. © 2008 Wiley Periodicals, Inc. Polymerization of 2‐methacryloyloxyethyl phosphorylcholine (MPC) was kinetically investigated in ethanol using dimethyl 2,2′‐azobisisobutyrate (MAIB) as initiator. The overall activation energy of the homogeneous polymerization was calculated to be 71 kJ/mol. The polymerization rate (Rp) was expressed by Rp = k[MAIB]0.54±0.05 [MPC]1.8±0.1. The higher dependence of Rp on the monomer concentration comes from acceleration of propagation due to monomer aggregation and also from retardation of termination due to viscosity effect of the MPC monomer. Rate constants of propagation (kp) and termination (kt) of MPC were estimated by means of ESR to be kp = 180 L/mol · s and kt = 2.8 × 104 L/mol · s at 60 °C, respectively. Because of much slower termination, Rp of MPC in ethanol was found at 60 °C to be 8 times that of methyl methacrylate (MMA) in benzene, though the different solvents were used for MPC and MMA. Polymerization of MPC with MAIB in ethanol was accelerated by the presence of water and retarded by the presence of benzene or acetonitrile. Poly(MPC) showed a peculiar solubility behavior; although poly(MPC) was highly soluble in ethanol and in water, it was insoluble in aqueous ethanol of water content of 7.4–39.8 vol %. The radical copolymerization of MPC (M1) and styrene (St) (M2) in ethanol at 50 °C gave the following copolymerization parameters similar to those of the copolymerization of MMA and St; r1 = 0.39, r2 = 0.46, Q1 = 0.76, and e1 = +0.51. © 2000 John Wiley & Sons, Inc.
