A liquid chromatographic method for the determination of closantel residues in milk and tissues is developed and validated. An acetonitrile-acetone solution (80:20, v/v) is used for the extraction of closantel residues from milk and animal tissues, and the extract is purified by solid-phase extraction with Oasis MAX cartridges and a mixture of formic acid-acetonitrile (5:95, v/v) as the elution solution. A C(18) bonded silica column is used for chromatographic separation. The mobile phase consists of acetonitrile-water (85:15, v/v) containing 0.05% triethylamine at pH 2.5, adjusted with phosphoric acid with the flow-rate set at 1.0 mL/min. Using the fluorescence emission of closantel at lambda(ex) = 335 nm and lambda(ex) = 510 nm, the calibration curve is linear, with a correlation coefficient of 0.9999 over the concentration range of 10-5000 microg/kg for the tissue sample and 10-5000 microg/L for the milk sample. The detection limit (s/n = 3) is 3 microg/kg for tissue sample and 3 microg/L for milk sample. The intra- and inter-day repeatabilities are between 3.35-7.66% and 4.04-8.67%, respectively. The proposed method enables the quantitative determination of closantel residues at levels as low as 10 microg/kg in animal tissue samples and 10 microg/L in milk samples.  相似文献

A high-performance liquid chromatographic multiresidue method was developed for the determination of 8 penicillin compounds (benzylpenicillin, phenoxymethylpenicillin, ampicillin, amoxicillin, nafcillin, oxacillin, cloxacillin, and dicloxacillin) at trace levels in muscle tissue. This method involves extraction of the penicillins with phosphate buffer pH 9 followed by cleanup and concentration on a C18 solid-phase extraction column and reaction with benzoic anhydride at 50 degrees C for 5 min and with 1,2,4-triazole and mercury(II) chloride solution pH 9 at 65 degrees C for 10 min. The derivatized compounds are eluted on a C8 column with a mobile phase containing acetonitrile and phosphate buffer (pH 6; 0.1 mol/L) loaded with sodium thiosulfate and ion-pairing tetrabutylammonium hydrogenosulphate. The method detection limit is approximately 3-11 micrograms/kg and the limit of determination was evaluated down to 25 micrograms/kg in line with the criteria of the EU decision No. 93/256/EEC.  相似文献

A liquid chromatographic method with fluorescence detection was developed for simultaneous determination of norfloxacin, ofloxacin, ciprofloxacin, pefloxacin, lomefloxacin, danofloxacin, enrofloxacin, sarafloxacin, difloxacin, oxolinic acid, and flumequine in milk The samples were extracted with 10% trichloroacetic acid/acetonitrile (9 + 1, v/v) and cleaned by Strata-X reversed-phase solid-phase extraction cartridges. The 11 quinolones were separated on a reversed-phase C18 column (Hypersil BDS-C18) with mobile phase gradient elution and detected with fluorescence by means of a wavelength program. The recoveries for milk fortified with the 11 quinolones at 3 levels were 69-88% with acceptable relative standard deviations of <9% (intraday) and <14% (interday). The limits of detection were 23 microg/L for enrofloxacin, and 1-9 microg/L for the other 10 quinolones.  相似文献

High-performance liquid chromatography with diode-array detection (HPLC-DAD) was optimised and validated for the determination of tetracyclines in bovine milk and tissues. Milk and tissue samples were extracted and purified using a solid-phase extraction HLB Oasis cartridge and analysed using HPLC-DAD set at 365 nm. The analyses were carried out using the mobile phase of 0.01 M oxalic acid-acetonitrile-methanol (60:25:15, v/v/v) on a C8 column (250 x 4.6 mm I.D., 5 microm). Recoveries of tetracyclines from spiked samples at the three concentrations (0.5, 1 and 1.5) of the maximum residues limits (corresponding to 100 microg/kg for milk and the muscle) were higher than 81.1% in milk and 83.2% in muscle. The method was successfully validated for bovine milk and muscle in compliance with requirements set by draft SANCO/ 1805/ 2000 European Decision. The decision limit (CCalpha) was in the range 113.2-127.2 microg/kg and 107.7-129.9 micro/kg for all compounds in milk and muscle, respectively. The detection capability (CCbeta) was in the range 117.2-131.3 microg/kg and 114.9-133.1 microg/kg for all compounds in milk and muscle, respectively.  相似文献

建立了同时测定牛奶中残留的9种磺胺类药物的固相萃取-高效液相色谱分析方法。牛奶样品经磷酸盐缓冲液稀释后高速离心去除脂肪,过C18小柱,用水淋洗,甲醇洗脱,洗脱液经氮气吹干后用乙酸乙酯溶解,并过氨基固相萃取小柱净化,用正己烷及水淋洗,以甲醇-乙腈-水(含1%乙酸)(体积比为1∶1∶8)洗脱,洗脱液用于高效液相色谱分析。采用Inertsil ODS-3 C18柱分离,以水-乙酸和甲醇-乙腈为流动相进行梯度洗脱,二极管阵列检测器检测,外标法定量。9种磺胺类药物标准曲线的线性回归系数均在 0.9999 以上,线性范围为25~5000 μg/L,检出限为1.7~2.8 μg/L,定量限为5.7~9.2 μg/L。在10,20,40 μg/L 添加水平下的添加回收率为72.1%~88.3%,相对标准偏差为2.3%~5.0%。该方法具有快速、灵敏的特点,符合现行兽药残留分析的要求。  相似文献

A method for the determination of trace levels of triazines and organophosphorus pesticides in water is presented. The extraction method is based on a solid-phase extraction on C-18 bound silica SPE cartridges. A precolumn filled with Merck C-18 bound silica and home-made C-18 bound silica have been tested at a flow-rate of 3 ml/min with comparable preconcentration yields. A SIM-MS method using a (15)N labelled internal standard has been developed for the organophosphorus pesticides. Detection limits lower than 1 microg/L have been obtained. Separations have been carried out on a conventional GC column OV 17 (1 m) and a capillary column OV 17 (25 m) with a temperature program from 150 degrees C (2 min) to 300 degrees C (rate of 6 degrees C/min).  相似文献

A high-performance liquid chromatographic (HPLC) method based on solid-phase extraction was developed for the determination of cefazolin, cefoperazone, cefquinome and ceftiofur in raw bovine milk. The milk fat was removed by centrifugation and the cephalosporins were extracted in acetonitrile. The extract was cleaned up by solid-phase extraction on an octadecyl sorbent. The compounds were separated by ion-paired gradient HPLC on a phenyl column with ultraviolet detection at 270 nm. The limits of detection estimated by a conservative model were 11 microg/kg for cefazolin and cefoperazone and 7 microg/kg for cequinome and ceftiofur. The mean recoveries were 86-88% for cefazolin, 91-93% for cefoperazone, 69-72% for cefquinome and 84-88% for ceftiofur in the concentration range 20-200 microg/kg.  相似文献

A comprehensive analytical method has been developed and validated for the simultaneous determination of seventeen glucocorticoid residues in eggs and milk. The mass spectrometer parameters, the composition of the mobile phase and the sample preparation method were firstly optimized to obtain maximum sensitivity. The samples were deconjugated with beta-glucuronidase/arylsulfatase enzyme and concentrated using an Oasis HLB solid-phase extraction cartridge, followed by cleanup with a dual Sep-pak silica and aminopropyl cartridge. The analytes were quantified by ultra-performance liquid chromatography (using a C18 column)/electrospray ionization tandem mass spectrometry (UPLC/ESI-MS/MS) operating in the negative ion mode. The assay for the 17 glucocorticoids was linear over the range of 1-200 microg/L for milk and egg samples with a high correlation coefficient (>0.99). The limits of quantification (LOQs) for the target analytes were 0.04-1.27 microg/kg for the egg samples and 0.03-0.73 microg/kg for the milk samples. The average extraction recoveries of the glucocorticoids from eggs and milk at two concentration levels (spiked at 0.40 and 2.00 microg/kg) were 65.6-118.7% and 61.5-119.6%, respectively, with relative standard deviations between 1.8-17.0% and 2.4-18.4%, respectively. Because of its high sensitivity, good precision and specificity, the method was found to be suitable for trace analysis of synthetic and natural glucocorticoids in complex biosamples such as eggs and milk.  相似文献

A collaborative study involving 14 laboratories was conducted to determine residues of ampicillin in porcine muscle tissue by using a liquid chromatographic method developed for multipenicillin analysis that can quantitate 8 penicillin compounds (benzylpenicillin, phenoxymethylpenicillin, ampicillin, amoxicillin, nafcillin, oxacillin, cloxacillin, and dicloxacillin) at trace levels in muscle tissue. This method involves extraction of the penicillins with phosphate buffer, pH 9, followed by cleanup and concentration on a C18 solid-phase extraction column and reaction with benzoic anhydride at 50 degrees C and with 1,2,4-triazole and mercury(II) chloride solution, pH 9.0, at 65 degrees C. The derivatized compounds are eluted isocratically on a C8 column with a mobile phase of acetonitrile and phosphate buffer (pH 6; 0.1 M) containing sodium thiosulfate and the ion-pair reagent tetrabutylammonium hydrogen sulfate. The penicillins are detected by UV absorption at 325 nm. The limit of detection and the limit of determination (quantitation) of the method were calculated to be approximately 3-5 and 25 microg/kg, respectively, in accordance with the criteria of European Union (EU) Decision No. 93/256/EEC. In this first interlaboratory study, collaborators were instructed to monitor 4 different penicillin compounds (benzylpenicillin, phenoxymethylpenicillin, ampicillin, and amoxicillin) by analyzing 8 blind samples of muscle tissue in triplicate. These samples were prepared from 2 materials containing different concentrations of incurred ampicillin (63.5 microg/kg for material No. 1 and 358.1 microg/kg for material No. 2) and 1 blank material. The repeatability relative standard deviation and the reproducibility relative standard deviation were 10.2 and 17.4%, respectively, for material No. 1 and 7.0 and 16.0%, respectively, for material No. 2. These results demonstrate that the method is suitable for the determination of ampicillin residues in muscle tissue at the EU maximum residue limit (50 microg/kg) and above. However, the identification of positives by this procedure may need additional confirmation by techniques with greater specificity, such as liquid chromatography combined with mass spectrometry, or tandem mass spectrometry. Investigations regarding the basis of interlaboratory testing studies will further demonstrate the suitability of multiresidue methodology for detecting and quantitating other compounds in the family of penicillin antibiotics.  相似文献

建立了高效液相色谱-电喷雾电离质谱法同时测定性保健品中违禁药物脱水吗啡、西地那非和前列地尔的分析方法。样品用甲醇超声提取,用Zorbax Eclipse XDB-C18柱分离,以乙腈-0.5%甲酸水溶液作为流动相,以保留时间和质荷比对分离出的组分予以定性确证,用峰面积进行定量。结果表明,脱水吗啡、西地那非和前列地尔的线性范围分别为50.0~5000.0 μg/L,10.0~1000.0 μg/L和40.0~4000.0 μg/L,检出限(以信噪比为3计)分别为20.0,4.0和10.0 μg/L。样品的加标平均回收率为89%~95%,相对标准偏差为9.5%~11%。该方法简便、快速、灵敏,适用于含中药成分的性保健品中这3种药物的同时分析。  相似文献

A method based on solid-phase microextraction (SPME) and gas chromatography with flame ionization detection (GC-FID) has been optimized for the determination of benzene, toluene, ethylbenzene and xylenes (BTEX) in water released from a waste treatment plant. The extraction step was optimized using fractional factorial and central composite designs including the following experimental factors: saline concentration; extraction time; desorption time; agitation velocity; headspace volume. A multiple function was used to describe the experimental conditions for simultaneous extraction of the compounds. The procedure, based on direct SPME at 50 degrees C, using a polydimethylsiloxane fiber, showed good linearity (r>0.997 over a concentration range 2-200 microg L(-1)) and repeatability (relative standard deviation (RSD)<4.23%) for all compounds, with limits of detection ranging from 0.05 to 0.28 microg L(-1), and limits of quantification ranging from 0.14 to 0.84 microg L(-1). Concentrations of the target compounds in these samples were between 145.8 and 1891 microg L(-1).  相似文献

A liquid chromatography-mass spectrometry (LC/MS) method was developed for the separation and determination of linear alkylbenzenesulfonates (C10-C14 LAS) in environmental water samples using a hydrophilic polymer column (Shodex Mspak GF-310 4D). This method involves a solid-phase extraction of the LAS samples with a Sep-Pak PS-2 cartridge. The LAS components were separated on the column with a mobile phase of 29% (w/v) acetonitrile-water containing 0.8 mM di-n-butylammonium acetate and 0.2 M acetic acid, and were detected by mass spectrometry with electrospray ionization. Detection limits of the developed method based on selected ion monitoring (SIM) technique for the C10-C14 LAS standards were 13-47 ng L(-1). The concentrations of the C10-C14 LAS in the environmental water samples ranged between 5-317 microg L(-1) for a river water sample and 0.4-6.4 microg L(-1) for a seawater sample. Linear relationships between the logarithms of retention factors and the alkyl chain lengths for each phenyl positional isomer of LAS could successfully be used for the identification of the isomer peaks.  相似文献

A new chromogenic reagent, 2-(2-quinolylazo)-5-diethylaminophenol (QADEAP), was synthesized. A sensitive, selective and rapid method has been developed for the determination of microg/L level silver ion based on the rapid reaction of silver(l) with QADEAP and the solid phase extraction of the colored chelate with C18 cartridge. The QADEAP reacts with Ag(l) to form a violet chelate of a molar ratio 1:2 (silver to QADEAP) in pH 3.5-8.0. This chelate was prconcentrated by solid phase extraction with C18 cartridge. An enrichment factor of 100 was achieved. The molar absorptivity of the chelate is 1.30 x 10(5) L mol(-1) cm(-1) at 590 nm in measured solution. Beer's law is obeyed in the range of 0.01-0.6 microg/ml. The relative standard deviation for eleven replicate samples of 0.01 microg/ml is 1.15%. The detection limit is 0.02 microg/L in the original samples. This method was applied to the determination of microg/L level silver ion in water with good results.  相似文献

A high-performance liquid chromatographic method was optimized and validated for the determination of atenolol and chlorthalidone (CT) in human breast milk. The milk samples were extracted and purified using ACN and phosphoric acid for precipitation of proteins followed by removal of ACN and milk fats by extraction with methylene chloride. The samples were applied, after an extraction procedure, to a cyanide column using a mobile phase consisting of ACN/water (35:65 v/v) and buffered at pH 4.0 with flow rate of 1.0 mL/min. Quantitation was achieved with UV detection at 225 nm using guaifenesin as the internal standard. The effectiveness of protein precipitation and clean up procedure were investigated. The method was validated over the range of 0.3-20 microg/mL for atenolol and 0.25-5 microg/mL for CT.  相似文献

The use of a different optimization procedure that involves Experimental Design (ED) and Artificial Neural Networks (ANN) for the off-line coupling solid-phase microextraction-micellar electokinetic chomatography (SPME-MEKC) is presented. This combination of ED and ANN, mathematical tools not previously used in SPME-MEKC optimization, allowed us to obtain good extraction efficiencies in the SPME procedure for the determination of a group of eleven triazine herbicides in groundwater samples. Both extraction and desorption steps were carried out by solution stirring at 900 rpm. Optimal conditions for the off-line SPME procedure were: extraction with a poly(dimethylsiloxane)/divinylbenzene SPME fiber for 120 min, 10% (w/v) NaCl, desorption time 40 min, and 70% (v/v) of methanol/buffer as desorption mixture. Detection limits lay between 0.80 microg L(-1) and 4.89 microg L(-1). Finally, the optimized method was applied to the determination of these compounds in spiked and non-spiked groundwater samples using a previously optimized MEKC separation.  相似文献

An integrated solid-phase spectrophotometry/ FIA method is proposed for the determination of the synthetic colorant matter Quinoline Yellow (QYWS) in the presence of its unsulfonated derivative QYSS. The procedure is based on the retention and preconcentration of the low level QYSS on a C-18 silica gel minicolumn, followed by sequential measurement of its absorbance at lambda = 410 nm after its elution with methanol. The applicable concentration range, the detection limit and the relative standard deviation were the following: for QYWS, from 0.10 to 30.0 mg L(-1); 0.013 mg L(-1); and 0.6%; and for QYSS, between 10 and 1.000 microg L(-1); 2 microg L(-1); and 1.3%, respectively. The method was applied to the determination of small amounts of QYSS present in QYWS in Colognes. Percentages of recovery between 98% and 99% were obtained in all instances. The method was also satisfactorily applied to the determination of these compounds in samples of commercial Colognes comparing the results for QYWS with those offered by an HPLC reference method and also validating the results chemometrically.  相似文献

Dihydrostreptomycin (DHS) is an aminoglycoside antibiotic used in veterinary medicine in combination with benzylpenicillin for the treatment of bacterial infections in cattle, pigs and sheep. A method to determine its residues in edible tissues of cattle, as well as in milk, was developed and validated. Extraction of DHS from the tissues was performed using a liquid extraction with a 10 mM phosphate buffer containing 2% (w/v) trichloroacetic acid, while milk samples were treated with a 50% (w/v) trichloroacetic acid solution, followed by a solid-phase clean-up procedure on a carboxypropyl (CBA) weak cation exchange column. Ion-pair chromatography, using a mixture of 20 mM pentafluoropropionic acid in water and acetonitrile as the mobile phase, was used to retain DHS and the internal standard streptomycin (STR) on a Nucleosil (5 microm) reversed-phase C18 column. The components were detected and quantified by electrospray ionization (ESI) tandem mass spectrometry. The method could be validated according to EC (European Community) requirements with respect to linearity, trueness and precision, the latter evaluated at the maximum residue limit (MRL) - 1000 ng g(-1) for kidney, 500 ng g(-1) for muscle, liver and fat, and 200 ng g(-1) for milk -, at one-half of the MRL and at one and a half times the MRL. A limit of quantification of 10 ng g(-1) and 1 ng ml(-1) was obtained for all tissues and for milk, respectively, which is far below one-half of the MRL as requested, while the limit of detection was in the low ppb range, varying between 1.9 and 4.2 ng g(-1) for the different tissues tested, and being 0.6 ng ml(-1) for milk. The method was used for the monitoring of DHS residues in incurred tissue and milk samples coming from cattle medicated with DHS in combination with benzylpenicillin by intramuscular injection, in order to evaluate withdrawal times.  相似文献

A liquid chromatographic (LC) method was developed for fast and simple measurement of retinyl palmitate (vitamin A) in fortified milk. Retinyl acetate internal standard was added to a test portion of milk followed by extraction into hexane. The hexane extract was analyzed by LC using a normal-phase silica gel column equilibrated with mobile phase (conditioned hexane-isopropanol, 99.85 + 0.15, v/v) about 1 h before injections. The retinyl palmitate concentration was calculated by using a relative response factor determined with calibration standards. In the collaborative study, 11 laboratories analyzed 13 pairs of fluid milk materials in blind duplicate. Twelve of the materials were composed of skim milk (< 0.5% fat), 1% fat milk, 2% fat milk, and 1% fat chocolate milk. Each material was fortified at 3 concentrations of retinyl palmitate of approximately 581 microg/L (1000 IU/qt), 1163 microg/L (2000 IU/qt), and 2236 microg/L (4000 IU/qt). The 13th material, unfortified skim milk, served as a matrix blank. Repeatability standard deviations (RSDr) without outliers ranged from 1.5 to 5.7% and reproducibility standard deviations (RSDR) without outliers ranged from 5.0 to 22.7%. cis-Isomers co-eluted with the predominant trans-retinyl palmitate isomer and were included in the results reported by all the collaborative laboratories. Endogenous long-chain esters from milk fat were also measured with the retinyl palmitate additive. The Study Director recommends that this method for determination of retinyl palmitate in fluid milk by LC be adopted First Action.  相似文献

Linear alkylbenzene sulfonates (LAS) have been determined in samples of the influent and the effluent, and in the sludge, from sewage-treatment plants (STP). LAS and sulfophenyl carboxylate compounds (SPC) were isolated by solid-phase extraction (SPE) with the polymeric phase Isolute ENV, then determined by liquid chromatography-electrospray mass spectrometry (LC-ESI-MS). The method enabled unequivocal identification of C10-C13 LAS by monitoring the ion at m/z 183 and the base peak corresponding to the [M-H]- ion. Average recoveries varied from 77-93% and the linear range of the method varied from 0.2 to 10 microg L(-1), with a limit of detection ranging from 10 ng L(-1) to 1.5 microg L(-1) when 200 mL waste water were preconcentrated. For sewage sludge, recoveries varied from 58 to 90% and the linear range was between 0.2 and 100 microg L(-1), with a detection limit ranging from 0.4 to 120 microg kg(-1) when 2.5 g sewage sludge was extracted. Unequivocal identification and determination of some metabolites of the LAS, the sulfophenyl carboxylate compounds (SPC), was achieved by monitoring [M-H]- ions.  相似文献
