Thioredoxin (Trx) plays an important role in achieving redox balances in cells and protecting the cells from oxidative damage. However, little is known about how arsenic affects Trx chemically. It is conceivable that trivalent arsenicals may bind to Trx, which has a highly conserved -CysGlyProCys- sequence. The objective of this study is to characterize the binding of seven arsenic species with Trx from E. coli and humans, using two mass spectrometry techniques. The arsenic-Trx complexes and the free arsenicals were well separated by size-exclusion liquid chromatography (LC) and detected with inductively coupled plasma mass spectrometry (ICPMS). The LC/ICPMS analyses showed that the trivalent arsenic species were able to form complexes with both human and E. coli Trx. Determination of binding constants indicated that affinity to Trx was higher for monomethylarsonous acid (MMA(III)) and phenylarsine oxide (PhAs(III)) than inorganic arsenite (iAs(III)) and dimethylarsinous acid (DMA(III)), probably because MMA(III) and PhAs(III) were able to form stable complexes by binding to two vicinal cysteines in the -CysGlyProCys- region of the Trx. The complexes of arsenicals with both human and E. coli Trx were further characterized by nano-electrospray tandem mass spectrometry. Binding stoichiometries for different arsenic species were consistent with the available cysteine residues in the Trx. Mass spectral evidence also suggests that the pentavalent arsenicals could be reduced by Trx. This study provides the first detailed chemical characterization of the interactions between Trx and arsenic species.  相似文献

Ion-pair reverse-phase HPLC-inductively coupled plasma (ICP) MS was employed to determine arsenite [As(III)], dimethyl arsenic acid (DMA), monomethyl arsenic (MMA) and arsenate [As(V)] in Chinese brake fern (Pteris vittata L.). The separation was performed on a reverse-phase C18 column (Haisil 100) by using a mobile phase containing 10 mM hexadecyltrimethyl ammonium bromide (CTAB) as ion-pairing reagent, 20 mM ammonium phosphate buffer and 2% methanol at pH 6.0. The detection limits of arsenic species with HPLC-ICP-MS were 0.5, 0.4, 0.3 and 1.8 ppb of arsenic for As(III), DMA, MMA, and As(V), respectively. MMA has been shown for the first time to experimentally convert to DMA in the Chinese brake fern, indicating that Chinese brake fern can convert MMA to DMA by methylation.  相似文献

Rabbit metallothionein (MT) 2A complexes with Cd(II), Zn(II), Ag(I), Cu(I), Hg(II), arsenite, monomethylarsonous acid (MMA), and dimethylarsinous acid (DMA) have been examined using ion-mobility measurements and mass spectrometry in a triple-quadrupole mass spectrometer equipped with a segmented second quadrupole that doubled as an ion-mobility cell [Guo, Y.; Wang, J.; Javahery, G.; Thomson, B. A.; Siu, K. W. M. An Ion-Mobility Spectrometer with Radial Collisional Focusing. Anal. Chem.2005, 77, 266-275]. The metal ions confer conformational rigidity on the MT complexes, which counteracts Coulombic repulsion among protons added as a result of electrospray. Triply and quadruply protonated Cd(7)MT2A have smaller cross-sections than the Cd(7)MT2A structure deduced from published NMR data. For the 6+ ions, the As(6)MT2A complex has a cross-section of 790 A(2); the MMA(10)MT2A complex, 920 A(2); and the DMA(20)MT2A complex, 1220 A(2). This increase in cross-section of the As(III) species, from As(3+) to MMA to DMA, is interpreted as a consequence of decreasing multiple coordination and increasing number of methyl groups.  相似文献

建立了一种利用高效液相色谱-双通道原子荧光检测联用同时进行砷和硒形态分析的方法。以10 mmol/L NH4H2PO4溶液(pH 5.6)(添加2.5%(体积分数)的甲醇)为流动相,在12 min内同时分离了三价砷(As(III))、一甲基砷(MMA)、二甲基砷(DMA)、五价砷(As(V))、硒代胱氨酸(SeCys)、硒代蛋氨酸(SeMet)和四价硒[Se(IV)]等化合物。As(III)、DMA、MMA、As(V)、SeCys、SeMet和Se(IV)的检出限分别为1,3,2,3,4,18和3 μg/L (进样量为200 μL),5次测定的相对标准偏差为1.9%~6.1%(As 100 μg/L, Se 300 μg/L)。应用该方法对人体尿样及硒酵母片中砷和硒的形态进行了分析,目标物在尿样中的加标回收率为83%~108%,在硒酵母片中的加标回收率为88%~105%。实验结果表明,该方法可用于尿样及药品中砷和硒形态的日常分析。该方法减少了样品的分析时间和试剂用量,降低了工作强度,提高了工作效率。  相似文献

This study aimed to establish complementary high performance liquid chromatography (HPLC) methods including three modes of separation: ion pairing, cation exchange, and anion exchange chromatography, with detection by inductively coupled plasma mass spectrometry (ICPMS). The ion pairing mode enabled the separation of inorganic arsenate (As(V)), monomethylarsonic acid (MMA(V)), and dimethylarsinic acid (DMA(V)). However, the ion pair mode was unable to differentiate inorganic arsenite (As(III)) from arsenobetaine (AsB); instead, cation exchange chromatography was used to isolate and quantify AsB. Anion exchange chromatography was able to speciate all of the aforementioned arsenic species. Potential inaccurate quantification problem with urine sample containing elevated concentration of AsB, which eluted immediately after As(III) in anion exchange or ion pairing mode, was overcame by introducing a post-column hydride generation (HG) derivatization step. Incorporating HG between HPLC and ICPMS improved sensitivity and specificity by differentiating AsB from hydride-forming arsenic species. This paper emphasizes the usefulness of complementary chromatographic separations in combination with HG-ICPMS to quantitatively determine concentrations of As(III), DMA(V), MMA(V), As(V), and AsB in the sub-microgram per liter range in human urine.  相似文献

The effects on the absorbance signals obtained using HG-AAS of variations in concentrations of the reaction medium (hydrochloric acid), the reducing agent [sodium tetrahydroborate(III); NaBH(4)], the pre-reducing agent (l-cysteine), and the contact time (between l-cysteine and arsenic-containing solutions) for the arsines generated from solutions of arsenite, arsenate, monomethylarsonic acid (MMA), and dimethylarsenic acid (DMA), have been investigated to find a method for analysis of the four arsenic species in environmental samples. Signals were found to be greatly enhanced in low acid concentration in both the absence (0.03-0.60 M HCl) and the presence of l-cysteine (0.001-0.03 M HCl), however with l-cysteine present, higher signals were obtained. Total arsenic content and speciation of DMA, As(III), MMA, and As(V) in mixtures containing the four arsenic species, as well as some environmental samples have been obtained using the following conditions: (i) total arsenic: 0.01 M acid, 2% NaBH(4), 5% l-cysteine, and contact time<10 min; (ii) DMA: 1.0 M acid, 0.3-0.6% NaBH(4), 4.0% l-cysteine, and contact time <5 min; (iii) As(III): 4-6 M acid and 0.05% NaBH(4) in the absence of l-cysteine; (iv) MMA: 4.0 M acid, 0.03% NaBH(4), 0.4% l-cysteine, and contact time of 30 min; (v) As(V): by difference. Detection limits (ppb) for analysis of total arsenic, DMA, As(III), and MMA were found to be 1.1 (n=7), 0.5 (n=5), 0.6 (n=7), and 1.8 (n=4), respectively. Good percentage recoveries (102-114%) of added spikes were obtained for all analyses.  相似文献

The stability of arsenic species (arsenate [As(V)], monomethylarsonate [MMA], dimethylarsinate [DMA] and arsenite [As(III)]) in two types of urban wastewater samples (raw and treated) was evaluated. Water samples containing a mixture of the different arsenic species were stored in the absence of light at three different temperatures: +4 degrees C, +20 degrees C and +40 degrees C. At regular time intervals, arsenic species were determined by high performance liquid chromatography (HPLC)-hydride generation (HG)-atomic fluorescence spectrometry (AFS). The experimental conditions for the separation of arsenic species by HPLC and their determination by AFS were directly optimised from wastewater samples. As(III), As(V), MMA and DMA were separated on an anion exchange column using phosphate buffer (pH 6.0) as the mobile phase. Under these conditions the four arsenic species were separated in less than 10 min. The detection limits were 0.6, 0.9, 0.9 and 1.8 micro g L(-1) for As(III), DMA, MMA and As(V), respectively. As(V), MMA and DMA were found stable in the two types of urban wastewater samples over the 4-month period at the three different temperatures tested, while the concentration of As(III) in raw wastewater sample decreased after 2 weeks of storage. A greater stability of As(III) was found in the treated urban wastewater sample. As(III) remained unaltered in this matrix at pH 7.27 over the period studied, while at lower pH (1.6) losses of As(III) were detected after 1 month of storage. The results show that the decrease in As(III) concentration with time was accompanied by an increase in As(V) concentration.  相似文献

Determination of arsenic species by large-volume field amplified stacking injection-capillary zone electrophoresis (LV-FASI-CZE) is reported in this paper. Whole column injection was employed. The optimum buffer pH for the separation of weak acids was discussed. It was found that the optimum buffer to analyze the stacked arsenate (As(V)), monomethylarsonate (MMA), and dimethylarsinate (DMA) was 25 mM phosphate at pH 6.5. However, the optimum buffer to analyze the concentrated arsenite (As(III)) was 20 mM phosphate - 10 mM borate at pH 9.28. The limits of detection of the method developed were 0.026 mg/L for As(III), 0.023 mg/L for As(V), 0.043 mg/L for MMA, and 0.018 mg/L for DMA. An enrichment factor of 34-100 for several arsenic species was obtained. In the end, this method was applied to determine the arsenic concentration in the environmental reference materials to show the usefulness of the method developed.  相似文献

Arsenic retention on natural red earth (hereafter NRE) was examined as a function of pH, ionic strength, and initial arsenic loading using both macroscopic and spectroscopic methods. Proton binding sites on NRE were characterized by potentiometric titrations yielding an average pH(zpc) around 8.5. Both As(III)- and As(V)-NRE surface configurations were postulated by vibration spectroscopy. Arsenic speciation in paddy rice is of considerable interest due to its impact on the food safety and human health. In this study, a simple methodology was developed to simultaneously extract and analyze As species in rice from China. Arsenic species, including arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA), and monomethylarsonic acid (MMA), were extracted by methanol-water (50:50, v/v) containing 0.02 mol L?1 nitric acid with a microwave-assisted procedure, and then determined by high performance liquid chromatography–hydride generation-atomic fluorescence spectrometry (HPLC–HG-AFS). The results showed that the method has good efficiency (>90%) for rice, indicating that there were no significant losses or transformations of arsenic during sample treatment and analysis. The limits of quantification (LOQ) of the method were 8.0, 20, 12, and 12 ng g?1 for As(III), As(V), DMA, and MMA, respectively. When this method was applied to the analysis of rice, As(III) had the highest concentration, followed by DMA, As(V), and MMA. The estimated weekly intake of inorganic As from rice by Chinese people accounted for 11.83% of the provisional tolerable weekly intake. The As speciation results in this study suggest that the risk associated with As in rice to human health may be negligible.  相似文献

Arsenic (As) is a toxic trace element that occurs naturally in groundwater and soils. Understanding the reactions of arsenite (As(III)) and arsenate (As(V)) with soil and mineral surfaces is critical for predicting the fate and transport of As in the environment and developing better ways to remediate As-contaminated areas. This investigation uses X-ray absorption near edge spectroscopy (XANES) to evaluate the solid phase oxidation state and mineral surface binding sites in three agricultural soil samples from California, USA by fitting linear combinations of XANES spectra derived from several synthetic and well characterized As(III)- and As(V)-treated model compounds (Fe and Al metal hydroxides and aluminosilicate illite clay mineral). The results suggest that As(III) is either partially or completely oxidized to As(V) when reacted with soil in an aqueous, batch reaction. The As(III)-treated Aiken soil was composed of 60% As(III) attached to surfaces similar to lepidocrocite (γ-FeOOH)) and 40% As(V) attached to aluminosilicate (illite). The Fallbrook soil completely oxidized As(III) and the product was As(V) adsorbed on Al hydroxide (gibbsite, γ-Al(OH)3) (62%), illite (16%), and lepidocrocite (γ-FeOOH) (22%). The reaction of As(III) with Wyo soil resulted in 42% As(III) adsorbed on surface similar to goethite and 58% As(V) adsorbed on lepidocrocite. Arsenic(V) adsorption on soil resulted in stable As(V) surface complexes that were well described by XANES spectra from As(V) adsorption complexes on gibbsite, illite, and lepidocrocite.  相似文献

Wool or hair fibre is a metabolically dead material after it has left the epidermis. During growth the fibre in the root is a metabolically very active organ, which is highly influenced by the health status of the living being. Arsenic is one of the elements that is easily taken up by the cells of the root and stored in the fibre afterwards. Here we show that arsenic can quantitatively be extracted by boiling the wool fibre or hair in water. The high intake of arsenic species by the sheep of North Ronaldsay (the seaweed-eating sheep) leads to a high arsenic concentration in wool (mean 5.2+/-2.3 mug g(-1)). The wool of lambs of these sheep, which are not exposed to seaweed, contains about 10 times less arsenic, which is still elevated compared to uncontaminated wool. The arsenic species identified in wool extract are arsenite (As(III)), arsenate (As(V)), monomethylarsonic acid (MMA(V)) and monomethylarsonious acid (MMA(III)) as minor species. The major species is dimethylated arsenic DMA in its tri- and pentavalent form (dimethylarsinous acid (DMA(III)) and dimethylarsinic acid (DMA(V))) accounting for 85% of the specified arsenic in the wool which reflects the amount of dimethylated species (i.e. the arsenoribofuranosides) taken up by seaweed being the main food source of the sheep. However, there are unknown arsenic species in the extract, which are not eluting from a strong anion exchange column. In vitro incubation experiments with this kind of wool showed that it has reducing properties but no demethylation was recorded. The absorption ability of the wool for methylated arsenic species is negligible, while inorganic arsenic is easier to be absorbed in the fibre (11-17%). This means that the species integrity is only guaranteed in terms of the degree of methylation but not in terms of their redox status.  相似文献

The weakest step in the analytical procedure for speciation analysis is extraction from a biological material into an aqueous solution which undergoes HPLC separation and then simultaneous online detection by elemental and molecular mass spectrometry (ICP-MS/ES-MS). This paper describes a study to determine the speciation of arsenic and, in particular, the arsenite phytochelatin complexes in the root from an ornamental garden plant Thunbergia alata exposed to 1 mg As L(-1) as arsenate. The approach of formic acid extraction followed by HPLC-ES-MS/ICP-MS identified different As(III)-PC complexes in the extract of this plant and made their quantification via sulfur (m/z 32) and arsenic (m/z 75) possible. Although sulfur sensitivity could be significantly increased when xenon was used as collision gas in ICP-qMS, or when HR-ICP-MS was used in medium resolution, the As:S ratio gave misleading results in the identification of As(III)-PC complexes due to the relatively low resolution of the chromatography system in relation to the variety of As-peptides in plants. Hence only the parallel use of ES-MS/ICP-MS was able to prove the occurrence of such arsenite phytochelatin complexes. Between 55 and 64% of the arsenic was bound to the sulfur of peptides mainly as As(III)(PC(2))(2), As(III)(PC(3)) and As(III)(PC(4)). XANES (X-ray absorption near-edge spectroscopy) measurement, using the freshly exposed plant root directly, confirmed that most of the arsenic is trivalent and binds to S of peptides (53% As-S) while 38% occurred as arsenite and only 9% unchanged as arsenate. EXAFS data confirmed that As-S and As-O bonds occur in the plants. This study confirms, for the first time, that As-peptides can be extracted by formic acid and chromatographically separated on a reversed-phase column without significant decomposition or de-novo synthesis during the extraction step.  相似文献

We developed two separation methods using CZE with UV detection for the determination of the most common inorganic and methylated arsenic species and some phenylarsenic compounds. Based on the separation method for anions using hydrodynamic sample injection the detection limits were 0.52, 0.25, 0.27, 0.12, 0.37, 0.6, 0.6, 1.2 and 1.0 mg As/L for phenylarsine oxide (PAO), p-aminophenylarsonic acid (p-APAA), o-aminophenylarsonic (o-APAA), phenylarsonic acid (PAA), 4-hydroxy-3-nitrobenzenearsonic acid (roxarsone), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenite or arsenious acid (As(III)) and arsenate (As(V)), respectively. These detection limits were improved by large-volume sample stacking with polarity switching to 32, 28, 14, 42, 22, 27, 26 and 27 microg As/L for p-APAA, o-APAA, PAA, roxarsone, MMA, DMA, As(III) and As(V), respectively. We have applied both methods to the analysis of the arsenic species distribution in aqueous soil extracts. The identification of the arsenic species was validated by means of both standard addition and comparison with standard UV spectra. The comparison of the arsenic species concentrations in the extracts determined by CZE with the total arsenic concentrations measured by inductively coupled plasma-atomic emission spectroscopy (ICP-AES) indicated that CZE is suited for the speciation of arsenic in environmental samples with a high arsenic content. The extraction yield of phenylarsenic compounds from soil was derived from the arsenic concentrations of the aqueous soil extracts and the total arsenic content of the soil determined by ICP-AES after microwave digestion. We found that 6-32% of the total amount of arsenic in the soil was extractable by a one-step extraction with water in dependence on the type of arsenic species.  相似文献

采用微波辅助提取-液相色谱-氢化物发生-原子荧光光谱法(LC-HG-AFS)联用技术分析了太湖沉积物中砷的形态[亚砷酸(As(III))、二甲基砷酸钠(DMA)、一甲基砷酸二钠(MMA)和砷酸As(V)]。测得沉积物中以无机砷为主,且以As(V)居多。选定以1mol/L的磷酸和0.1mol/L抗坏血酸为提取液,在微波辅助萃取(功率为60W,时间12min)下,萃取率达79.84%~91.57%,回收率在94.78%~107.6%之间。4种砷的形态在0~160μg/L之间时线性良好,检测限为0.6~2.3μg/L,相对标准偏差RSD为1.62%~2.20%。方法具有简便、快速、灵敏的特点。  相似文献

A novel procedure was developed for the determination of arsenite (As(III)), arsenate (As(V)), monomethylarsonic (MMA) and dimethylarsinic acid (DMA) with ion chromatography-hydride generation-atomic fluorescence spectrometry (IC-HG-AFS) by employing a new gas-liquid separator (GLS). The effective separation of the four arsenic species was achieved in about 12 min. With a sample loading volume of 20 μl, the measurable minimum for As(III), DMA, MMA and As(V) were 0.02, 0.045, 0.043 and 0.166 ng, respectively, along with relative standard deviations of 1.1, 1.1, 1.7 and 2.2% at the 100 μg l−1 level (n = 6) for As(III), DMA, MMA and As(V), respectively. The present procedure was applied for the speciation of arsenic in underground water and in urine samples, and the sum of the four arsenic species by IC-HG-AFS was in good agreement with the total value by HG-AFS.  相似文献

The simultaneous separation and determination of arsenite As(III), arsenate As(V), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB), and arsenocholine (AsC) in rice samples have been carried out in one single anion‐exchange column run by high‐performance liquid chromatography with inductively coupled plasma mass spectrometry. To estimate the effect of variables on arsenic (As) speciation, the chromatographic conditions including type of competing anion, ionic strength, pH of elution buffer, and flow rate of mobile phase have been investigated by a univariate approach. Under the optimum chromatographic conditions, baseline separation of six As species has been achieved within 10 min by gradient elution program using 4 mM NH4HCO3 at pH 8.6 as mobile phase A and 4 mM NH4HCO3, 40 mM NH4NO3 at pH 8.6 as mobile phase B. The method detection limits for As(III), As(V), MMA, DMA, AsB, and AsC were 0.4, 0.9, 0.2, 0.4, 0.5, and 0.3 μg/kg, respectively. The proposed method has been applied to separation and quantification of As species in real rice samples collected from Hunan Province, China. The main As species detected in all samples were As(III), As(V) and DMA, with inorganic As accounting for over 80% of total As in these samples.  相似文献

An analytical method based on microwave decomposition and flow injection analysis (FIA) coupled to hydride generation atomic absorption spectrometry (HGAAS) is described. This is used to differentiate arsenite [As(III)], arsenate [As(V)], monomethylarsonic acid (MMA) and dimethylarsinic acid (DMA) from organoarsenic compounds usually present in seafood. Without microwave digestion, direct analysis of urine by HGAAS gives the total concentration of As(III), As(V), MMA and DMA because organoarsenic compounds such as arsenobetaine, usually found in most seafood, are not reducible upon treatment with borohydride and therefore cannot be determined by using the hydride generation technique. The microwave oven digestion procedure with potassium persulfate and sodium hydroxide as decomposition reagents completely decomposes all arsenicals to arsenate and this can be measured by HGASS. Microwave decomposition parameters were studied to achieve efficient decomposition and quantitative recovery of arsenobetaine spiked into urine samples. The method is applied to the determination of urinary arsenic and is useful for the assessment of occupational exposure to arsenic without intereference from excess organoarsenicals due to the consumption of seafood. Analysis of urine samples collected from an individual who ingested some seafood revealed that organoarsenicals were rapidly excreted in urine. After the ingestion of a 500-g crab, a 10-fold increase of total urinary arsenic was observed, due to the excretion of organoarsenicals. The maximum arsenic concentration was found in the urine samples collected approximately between 4 to 17 hr after eating seafood. However, the ingestion of organoarsenic-containing seafoods such as crab, shrimp and salmon showed no effect on the urinary excretion of inorganic arsenic, MMA and DMA.  相似文献

Arsenic speciation in rice has received attention due to its impact on food safety and human health. In this study, a sensitive method was developed for the determination of inorganic arsenic in rice using online anion suppression with ion chromatography and inductively coupled plasma mass spectrometry. HCl of 0.01?mol/L was the optimal extracting agent, and 38?mmol/L sodium carbonate and 15?mmol/L sodium acetate were used as the mobile phase to separate dimethylarsinic acid (DMA), arsenite, monomethylarsonic acid (MMA), and arsenate. The results showed that there were no significant losses or transformations with the anion suppressor and an improvement in sensitivity. The limits of quantification were 0.1?µg/L for DMA, As(III) and MMA, and 0.2?µg/L for As(V). The procedure was used to determine inorganic arsenic in rice; As(III) and DMA were the primary forms present. The reproducibility from seven measurements showed that the relative standard deviation was less than 1.68%. The recoveries were from 99.76 to 110.42%. The present work offers a new approach for the determination of inorganic arsenic in rice.  相似文献

A pre-oxidation procedure which converts arsenite [As(III)] into arsenate [As(v)] was investigated in urinary arsenic speciation prior to on-line photo-oxidation hydride generation with ICP-MS detection. This sample pre-oxidation method eliminates As(III) and As(v) preservation concerns and simplifies the chromatographic separation. Four oxidants, Cl2, MnO2, H2O2 and I3-, were investigated. Chlorine (ClO-aq) and MnO2 selectively converted As(III) into As(v) in pure water samples, but the conversion was inefficient in the complex urine matrix. Oxidation of As(III) by H2O2 was least affected by the urine matrix, but the removal of excess H2O2 at pH 10 proved difficult. The most appropriate oxidant for the selective conversion of As(III) into As(v) with minimal interference from the urine matrix is I3- at pH 7. Unlike H2O2, excess oxidant can be easily removed by the addition of S2O3(2-). The I3-(-)S2O3(2-) treatment on a fortified sample of reconstituted NIST SRM 2670 freeze dried urine indicated that arsenobetaine (AsB), dimethlyarsinic acid (DMA), monomethylarsonic acid (MMA) and As(v) were not chemically degraded with recoveries ranging from 95 to 102% for all arsenicals. Sample clean-up involved pH adjustment prior to C18 filtration in order to achieve efficient As(III) conversion and quantitative recoveries of AsB and DMA. The concentrations determined in NIST SRM 2670 freeze dried urine were AsB 17.2 +/- 0.5, DMA 56 +/- 4 and MMA 10.3 +/- 0.3 with a combined total of 83 +/- 5 micrograms L-1 (+/- 2 sigma).  相似文献
