Here we report the detailed measurement of long-range heteronuclear spin-spin coupling constants, especially 2, 3JCH spin-spin couplings for eight different cyclopentane derivatives. These 2, 3JCH constants were shown to be a useful tool in the determination of the relative stereochemistry in these rings. The coupling constant measurements reported here are based on two different experiments: a 2D heteronuclear correlation experiment named G-BIRDR, X-CPMG-HSQMBC and the 2D-coupled gHSQC {1H-13C} experiment  相似文献

Individual transitions of magnetically equivalent spin systems such as methyl groups residing on isotropically tumbling molecules in solution usually cannot be observed as multiplet-split NMR lines. We propose a pair of NMR experiments, 2D [13C,1Halphaalpha]Methyl and [13C,1Hbetabeta]Methyl HSQC, to overcome this limitation and enable direct and selective observation of individual 1H transitions in 13C-labeled methyl spin systems. Immediate applications include quantitative measurements of 1H-1H residual dipolar couplings (RDC) and cross-correlated relaxation between 1H chemical shift anisotropy and 1H-1H dipole-dipole interactions. The use of the experiments for the measurement of RDCs is demonstrated with two proteins, one weakly aligned by means of Pf1 phages and the other using a naturally present paramagnetic heme group.  相似文献

Measurement of 1H-1H dipolar couplings in macromolecules, weakly oriented by a dilute liquid crystalline medium, is generally limited to the largest such interactions. By removing dipolar couplings to nearest neighbors, either by decoupling, deuteration, or both, more remote interactions become accessible. The approach is demonstrated for measurement of amide-amide interactions in the proteins calmodulin and ubiquitin and permits observation of direct dipolar couplings between protons up to 7 A apart. Quantitative evaluation of 1H-1H dipolar couplings measured in ubiquitin shows excellent agreement with its solution structure.  相似文献

By introducing dipolar recoupling methods to high-resolution magic-angle spinning (HRMAS) NMR spectroscopy, a class of experiments has been delevoped that allows the measurement of residual dipole-dipole couplings of approximately 1 Hz in weakly immobilized molecules. Using homonuclear 1H-1H recoupling, distances of up to approximately 8 A can be selectively determined, while heteronuclear 1H-13C recoupling provides access to dynamic order parameters of individual molecular segments on the order of approximately 10-3. The experiments are demonstrated on functionalized oligopeptides that are attached to polymer resins.  相似文献

Theoretical and experimental 2JHH coupling constants for six-membered rings containing oxygen or sulfur atoms were studied to investigate whether the 2JHH coupling constant can be used for stereoelectronic studies in heterocyclohexanes, instead of 1JCH, because it is well known that experimental measurements of 2JHH coupling constants at low temperature are much easier to determine than the corresponding 1JCH couplings. For all compounds studied here, the 2JHH coupling constants are affected by sigma*C-H antibonding occupancy together with bond angle effects. For cyclohexane and oxygen-containing compounds, the influence on the geminal coupling for Hax-C2-Heq and for X1-C2-X3 (X=O and C), bond angles are more pronounced than for the sulfur derivatives.  相似文献

We applied the POST-C7 DQ-dipolar recoupling pulse sequence to the measurement of (1)H-(1)H distances with high precision. The spectral resolution is enhanced by detecting the (1)H magnetization via (13)C signals. A least-squares fitting of the build-up curve of the transferred magnetization to the exact numerical simulations yielded a (1)H(alpha)-(1)H(beta) distance of 248 +/- 4 pm for fully (13)C-labeled L-valine. This distance agrees with the neutron diffraction study. The negative transferred magnetization clearly indicates that the direct DQ (1)H-(1)H dipolar couplings have the largest effect. The signal for the magnetization transfer builds up rapidly by the direct (1)H-(1)H dipolar coupling, and decreases to zero at longer mixing time when the relayed magnetization transfer becomes significant. This large intensity change of the signal leads to the high precision in the distance measurement. We inspected factors that limit the effective bandwidth of the POST-C7 recoupling for the (1)H and (13)C homonuclear spin systems. The spin interactions at times shorter than the cycle time of the C7 sequence were also evaluated to measure the distances. The carbon-detected 2D (1)H DQ mixing experiment was demonstrated for the measurement of multiple (1)H-(1)H distances.  相似文献

The Lewis X (LeX) determinant, a trisaccharide with the carbohydrate sequence Galbeta(1-->4)[Fucalpha(1-->3)]GlcNAcbeta, is believed to be responsible for Ca2+-mediated cell-cell interactions. In partly oriented phases composed of mixtures of penta(ethyleneglycol)monododecyl ether HO(CH2CH2O)5C12H25 and n-hexanol in the presence of Ca2+ ions, the variation of the residual dipolar couplings 1DCH of various CiHi vectors in LeX as a function of the concentration of the trisaccharide demonstrates the existence of very weak LeX-Ca2+-LeX complexes in solution. Synthetic 3-, 4-, and 6-deoxy-LeX variants were also shown to form complexes in the presence of calcium ions, despite the replacement of one of their hydroxyl groups by hydrogen atoms. This is the first direct observation in solution of a calcium-mediated interaction between LeX molecules.  相似文献

PASADENA hyperpolarization of succinic acid for MRI and NMR spectroscopy   总被引:1,自引:0,他引:1  
We use the PASADENA (parahydrogen and synthesis allow dramatically enhanced nuclear alignment) method to achieve 13C polarization of approximately 20% in seconds in 1-13C-succinic-d2 acid. The high-field 13C multiplets are observed as a function of pH, and the line broadening of C1 is pronounced in the region of the pK values. The 2JCH, 3JCH, and 3JHH couplings needed for spin order transfer vary with pH and are best resolved at low pH leading to our use of pH approximately 3 for both the molecular addition of parahydrogen to 1-13C-fumaric acid-d2 and the subsequent transfer of spin order from the nascent protons to C1 of the succinic acid product. The methods described here may generalize to hyperpolarization of other carboxylic acids. The C1 spin-lattice relaxation time at neutral pH and 4.7 T is measured as 27 s in H2O and 56 s in D2O. Together with known rates of succinate uptake in kidneys, this allows an estimate of the prospects for the molecular spectroscopy of metabolism.  相似文献

A large fraction of hydrogens in proteins and nucleic acids is of the methylene type. Their detailed study, however, in terms of structure and dynamics by NMR spectroscopy is hampered by their fast relaxation properties, which give rise to low sensitivity and resolution. It is demonstrated that six different relaxation interference processes, involving 1H-13C and 1H-1H dipolar interactions and 1H and 13C chemical shift anisotropy, can be used simultaneously to mitigate these problems effectively. The approach is applicable to the majority of NMR experiments commonly used to study side chain and backbone conformation. For proteins, its efficiency is evaluated quantitatively for two samples: the third IgG-binding domain from Streptococcal Protein G and the protein calmodulin complexed with a 26-residue target peptide. Gains in both resolution and sensitivity by up to factors of 3.2 and 2.0, respectively, are observed for Gly residues at high magnetic field strengths, but even at much lower fields gains remain substantial. The resolution enhancement obtained for methylene groups makes possible a detailed analysis of spectral regions commonly considered inaccessible due to spectral crowding. For DNA, the high resolution now obtainable for C5' sites permits an H5'/H5'-based sequential NOE assignment procedure, complementary to the conventional base-H1'/H2'/H2' pathway.  相似文献

The enflurane and ethoxyflurane 1H, 13C and 19F NMR spectra are examined—including sign determination of FF and FH couplings—and considered in the light of previously reported results for methoxyflurane. Conformational differences between methoxyflurane and the former two molecules are indicated by through space FH coupling constants and by the nonequivalence of geminal fluorine nuclei. A quantitative method to record (1)H-(13)C correlation NMR spectra (Q-HSQC) is presented. The suppression of (1)J(CH)-dependence is achieved by modulating the polarization transfer delays of HSQC. In addition, the effect of homonuclear couplings, as well as relaxation during the pulse sequence are discussed. We developed the Q-HSQC approach for the quantitative analysis of wood lignin, a complex polymer where it has been difficult to obtain reliable data on the relative amounts of different structural units. The current method is applicable to a variety of complex mixtures, where normal 1D (1)H- and (13)C-NMR methods fail.  相似文献

13C and 1H NMR spectroscopy have been used to study the orientational order which develops when a nematogenic compound, 4-pentyl-4'-cyanobiphenyl (5CB), approaches the transition from the isotropic to the nematic phase at T(NI). The experiments yield values of field induced dipolar couplings, (1), between all of the directly bonded carbon and hydrogen nuclei in the molecule, and 2D(HH)B, the geminal dipolar coupling between protons in the first methylene group in the alkyl chain. The temperature dependence of these couplings shows that in each case they follow a divergence behavior governed by (T - T*)(-1),where T* is a temperature determined from the experimental data and which is close to but less than T(NI). Experiments performed at spectrometer field strengths of 11.75 and 18.79 T confirm the prediction that the induced couplings should depend on the square of the applied field strength. It was found that, within experimental error, T* is the same for each field-induced coupling, and that T(NI) - T* is the same at 11.75 and 18.79 T. It is shown that the set of field-induced couplings 1D(CHi)B obtained at a temperature close to T(NI) can be used to derive a conformer distribution for 5CB in the isotropic phase.  相似文献

The application of selective population inversion (SPI) to the assignment of 13C resonances and to the determination of the relative sign of 13C? 1H coupling constants is demonstrated for 1-phenyl-1,2-dibromo-2-nitroethane. A negative sign for the geminal C-2? H-1 coupling constant has been determined from the 13C spectra. The relationship between SPI and TSI is discussed.  相似文献

A low radio frequency power polarization inversion spin exchange at the magic angle (PISEMA) pulse sequence is described for the measurement of heteronuclear dipolar couplings from solids. The method employs a time averaged nutation concept to significantly reduce the rf power required to spin-lock low gamma nuclear spins in PISEMA experiments. The efficacy of the 2D method is demonstrated on a single crystal of n-acetyl-L-(15)N-valyl-L-(15)N-leucine dipeptide to measure (1)H-(15)N dipolar couplings and a liquid crystal sample to measure (1)H-(13)C dipolar couplings.  相似文献

29Si-13C couplings were measured in para substituted silylated phenols, X--C6H4--O--SiR1R2R3 (X = NO2, CF3, Cl, F, H, CH3, CH3O). The SiR1R2R3 silyl groups included trimethylsilyl (Si(CH3)3, TMS), tert-butyldimethylsilyl (Si(CH3)2C(CH3)3, TBDMS), dimethylsilyl (SiH(CH3)2, DMS), and tert- butyldiphenylsilyl (Si(C6H5)2C(CH3)3, TBDPS). Previously developed (Si,C,Si)gHMQC methods and narrow 29Si lines allowed the determination of coupling constants over up to five bonds. Besides the number of intervening bonds between the silicon and carbon atoms, all the measurable couplings depend also on the nature of the substituents on the silicon. The two- and three-bond couplings are not affected by ring substitution in the para position. These properties render the 29Si-13C couplings suitable for line assignment in the spectra of silylated polyphenols. The experimental results are in reasonable agreement with theoretical calculations. The calculations show, in agreement with the data reported in the literature for couplings between other nuclei, that the two-bond and three-bond couplings, which are of similar magnitudes, are of opposite signs. If the signs of these geminal and vicinal couplings could be determined experimentally, they would greatly facilitate the line assignment. The four- and five-bond couplings are affected by the substituent X in a nontrivial manner.  相似文献

The acrylonitrile/methyl methacrylate copolymers of different monomer concentration were prepared by photo polymerization using uranyl ion as initiator. The carbon 13 and proton spectra of these copolymers are overlapping and complex. The complete spectral assignment of the 13C- and 1H-NMR spectra were done with the help of Distortionless Enhancement by Polarization Transfer (DEPT) and two dimensional 13C–1H Heteronuclear Single Quantum Correlation (HSQC) experiments. The methylene, methine and the methyl carbon resonances show both stereochemical (triad level) and compositional (dyad, triad, tetrad, pentad and hexad level) sensitivity. 2D Double Quantum Filtered Correlated Spectroscopy (DQFCOSY) experiment was used to ascertain the various geminal couplings between the methylene protons. © 1998 John Wiley & Sons, Inc. We have developed a solid-state NMR method for observing the signals due to 13C spins of a peptide in the close vicinity of 31P and 2H spins in deuterated phospholipid bilayers. The signal intensities in 13C high-resolution NMR spectra directly indicate the depolarization of 1H by 1H-31P and 1H-2H dipolar couplings under multiple-contact cross-polarization. This method was applied to a fully 13C-, 15N-labeled 14-residue peptide, mastoparan-X (MP-X), bound to phospholipid bilayers whose fatty acyl chains are deuterated. The 13C NMR spectra for the depolarization were simulated from the chemical shifts and structure of membrane-bound MP-X previously determined and the distribution of 2H and 31P spins in lipid bilayers. The minimization of RMSD between the simulated and the experimental spectra showed that the amphiphilic alpha-helix of MP-X was located in the interface between the water layer and the hydrophobic domain of the bilayer, with nonpolar residues facing the phosphorus atoms and alkyl chains of the lipids.  相似文献

Dipole-dipole couplings are selectively recoupled during the mixing period, while MAS ensures high resolution in the spectral dimensions. Incoherent dipolar exchange is replaced by amplified coherent processes, such that time scales for polarization transfer are shortened, and dipolar double-quantum techniques become applicable. A pulse sequence for the selective recoupling of heteronuclear dipolar interactions in mobile amorphous phase of powdered semicrystalline polymers is described. 1H-13C dipolar interactions are selectively measured by PISEMA-type sequence. Selection of 13C magnetization originating from amorphous phase is achieved by a train of saturation pulses followed by a short delay and a direct excitation pulse on 13C spins. The development of undesired net 13C magnetization during the recoupling sequence is prevented by the efficient "reverse" 13C --> 1H cross-polarization. The efficacy of the 2D method to measure 1H-13C dipolar couplings selectively for mobile components is demonstrated on powdered crystalline L-alanine, semicrystalline polyethylene, and nanocomposite polyamide-6/montmorillonite.  相似文献

We show that (13)C-(1)H dipolar couplings in fully protonated organic solids can be measured by applying a Symmetry-based Resonance-Echo DOuble-Resonance (S-REDOR) experiment at ultra-fast Magic-Angle Spinning (MAS). The (13)C-(1)H dipolar couplings are recovered by using the R12 recoupling scheme, while the interference of (1)H-(1)H dipolar couplings are suppressed by the symmetry properties of this sequence and the use of high MAS frequency (65 kHz). The R12 method is especially advantageous for large (13)C-(1)H dipolar interactions, since the dipolar recoupling time can be incremented by steps as short as one rotor period. This allows a fine sampling for the rising part of the dipolar dephasing curve. We demonstrate experimentally that one-bond (13)C-(1)H dipolar coupling in the order of 22 kHz can be accurately determined. Furthermore, the proposed method allows a rapid evaluation of the dipolar coupling by fitting the S-REDOR dipolar dephasing curve with an analytical expression.  相似文献
