The interactions of scopoletin to bovine serum albumin (BSA) and human serum albumin (HSA) have been investigated by spectroscopic methods. The fluorescence tests indicated that the formation mechanism of scopoletin–BSA/HSA complexes belonged to the static quenching. The displacement experiments suggested that scopoletin primarily bound to tryptophan residues of BSA/HSA within site I (subdomain IIA). The binding distance of scopoletin to BSA/HSA was 2.38/2.34 nm. The thermodynamic parameters (ΔG, ΔH and ΔS) calculated on the basis of different temperatures revealed that the binding of BSA–scopoletin was mainly depended on van der Waals interaction and hydrogen bond, and yet the binding of HSA–scopoletin was strongly relied on the hydrophobic interaction and electrostatic interaction. The results of synchronous fluorescence, 3D fluorescence, UV–vis absorption, and FT-IR spectra showed that the conformations of BSA and HSA altered with the addition of scopoletin. In addition, the effects of some common ions on the binding constants of scopoletin to proteins were also investigated.  相似文献

The structural changes in the protein macromolecules caused by polycyclic aromatic hydrocarbon (PAH) ecotoxicants were studied using the data on intrinsic fluorescence of proteins and fluorescence of PAH molecules introduced into proteins. A luminescence method for PAH determination in proteins was developed and used to study the interaction of two PAHs (pyrene and anthracene) with proteins of two types (bovine serum albumin and human serum albumin). The results were interpreted using the Stern–Volmer fluorescence quenching model. The association constants and the number of binding sites in the protein–ligand complexes were calculated. The binding of PAHs with proteins was described based on the static version of quenching with formation of nonfluorescent complexes of protein fluorophores with PAHs.  相似文献

The interaction of ilaprazole (IPZ), ilaprazole sulfone (IPZO) and ilaprazole sulfide (IPZI) with bovine serum albumin (BSA), and the effect of IPZO and IPZI on the interaction of IPZ with BSA have been investigated by fluorescence, synchronous fluorescence, ultraviolet–visible (UV–vis), Fourier transform infrared spectroscopy (FT-IR) and circular dichroism (CD). The results indicated that IPZ, IPZO and IPZI had a strong ability to quench the intrinsic fluorescence of BSA, and the binding affinities were significantly affected by structures in the order IPZ>IPZO>IPZI, while the van der Waals force and hydrogen bond played major roles in their binding with BSA. The analysis of synchronous fluorescence, FT-IR and CD spectra showed the change in secondary structure of BSA upon interaction with IPZ, IPZO or IPZI. Site marker competitive experiments indicated that their binding to BSA primarily took place in subdomain IIA. The presence of IPZO and IPZI decreased the quenching constants of IPZ with BSA by about 68.4% and 95.1%, respectively, which possibly resulted from the existence of competitive binding between IPZ and its metabolites with BSA. However, IPZO and IPZI did not change the quenching mechanism of IPZ with BSA, while all the fluorescence quenching was initiated by static quenching procedure combined with non-radiative energy transfer. Our results may have relevant insight into IPZ's bioavailability and efficacy affected by its metabolites.  相似文献

We have studied quenching of the fluorescence of the diprotonated form of 3,7,13,17-tetramethyl-2,8,12,18-tetrabutylporphyrin by iodide ions in solutions. We have established that there is both static quenching of fluorescence when nonfluorescent complexes are formed with the iodide ions, as a result of an internal heavy atom effect, and also dynamic diffusion-controlled quenching. 对甘草酸单铵盐(MAG)-牛血清白蛋白(BSA)体系的荧光光谱进行了研究,采用同步荧光技术考察了甘草酸单铵盐对牛血清白蛋白构象的影响,认为甘草酸单铵盐(MAG)对牛血清白蛋白(BSA)体系荧光猝灭是由于生成了超分子复合物的静态猝灭,求得MAG-BSA的形成常数KA及热力学函数ΔGH和ΔS,根据热力学函数确定了超分子间的作用力类型为静电作用力,依据Forster非辐射能量转移机制,确定了给体-受体间的结合距离和能量转移效率。  相似文献

Naringenin, a flavanone compound highly enriched in grapefruits, has been identified as a possible inhibitor of cell proliferation; and thus has the potential to act as an antitumorigenic agent. In this study, the binding of naringenin to bovine serum albumin (BSA) was studied at the physiological conditions (pH=7.40) by fluorescence and UV-vis spectroscopy. Naringenin strongly quenches the intrinsic fluorescence of BSA, and a decrease in the fluorescence quenching constant was observed together with an increase in temperature, which indicates that the fluorescence quenching of BSA by naringenin is a result of the formation of naringenin-BSA complex. Binding parameters calculating from Stern-Volmer method and Scatchard method showed that naringenin bind to BSA with the binding affinities of the order 104 L mol−1. Thermodynamic parameters such as ΔG, ΔH and ΔS, were calculated at different temperatures, showing that electrostatic interactions were mostly responsible for the binding of naringenin to BSA. Site marker competitive displacement experiments demonstrating that naringenin bind with high affinity to site I (subdomain IIA) of BSA. Furthermore, the effect of metal ions to naringenin-BSA system was studied, and the specific binding distance r (3.30 nm) between donor (Trp-212) and acceptor (naringenin) was obtained according to fluorescence resonance energy transfer (FRET).  相似文献

The mechanism of interaction of an antidepressant, fluoxetine hydrochloride (FLX) with bovine serum albumin (BSA) has been studied by different spectroscopic techniques under physiological conditions. FLX was found to quench the intrinsic fluorescence of protein by static quenching mechanism. The binding constant ‘K’ was found to be 7.06×103 M−1 at 296 K. The value of ‘n’ close to unity revealed that the BSA has a single class of binding site for FLX. Based on thermodynamic parameters, hydrogen bonding and van der Waals forces were proposed to operate between BSA and FLX. The change in conformation of protein was noticed upon its interaction with the drug. From displacement studies it was concluded that the FLX bound to protein at site I. The effects of various common metals ions on the binding were also investigated.  相似文献

应用荧光光谱法研究了牛血清蛋白与荧光增白剂CBS-X、BBU、VBL的相互作用.通过Stern-Volmer方程、Lineweaver-BurK方程和双对数曲线进行计算,研究了FWA对BSA内源荧光的猝灭机制.FWA对BSA内源荧光的猝灭主要为静态猝灭和荧光共振能量转移猝灭.测定了荧光增白剂CBS-X、BBU、VBL对BSA的猝灭常量和扩散常量(283 K),确定了荧光增白剂与BSA结合位点数均为1.根据Frster非辐射能量转移理论,计算了BSA与荧光增白剂分子间的结合距离和能量转移效率.通过测定283 K和298 K时供体与受体分子间结合常量,计算了BSA与荧光增白剂作用的热力学参量.BSA与FWA作用的ΔH<0,ΔS>0,并以此确定了BSA 与荧光增白剂分子主要通过静电力进行作用.  相似文献

The fluorescence and ultraviolet spectroscopies were explored to study the interaction between edaravone (EDA) and bovine serum albumin (BSA) under imitated physiological condition. The experimental results show that the fluorescence quenching mechanism between EDA and BSA is a combined quenching (dynamic and static quenching). The binding constants, binding sites, and the corresponding thermodynamic parameters (ΔG, ΔH, and ΔS) of the interaction system were calculated at different temperatures. According to Förster non-radiation energy transfer theory, the binding distance between EDA and BSA was calculated to be 3.10 nm. The effect of EDA on the conformation of BSA was analyzed using synchronous fluorescence spectroscopy. In addition, the effects of some common metal ions Mg2+, Ca2+, Cu2+, and Ni2+ on the binding constant between EDA and BSA were examined.  相似文献

应用荧光光谱法研究了meso-四(3,5-二溴-4-羟基苯基)卟啉[T(DBHP)P]与牛血清白蛋白(BSA)之间的结合反应,基于T(DBHP)P对BSA内源荧光的猝灭机理,测定了两者之间在不同温度下的结合常数,温度为27 ℃时,荧光猝灭法测得反应的结合常数为K=1.30×106 L·mol-1,温度为48 ℃时,K=6.32×105 L·mol-1,结合常数随温度升高而减小,由此判定该猝灭类型为静态猝灭。根据Frster非辐射能量转移理论,确定了T(DBHP)P与BSA之间的能量转移效率E=0.91,能量给体(BSA)与受体[T(DBHP)P]之间的结合距离r=2.39 nm<7 nm,符合非辐射能量转移条件。依据热力学参数ΔG<0,ΔH<0,ΔS>0确定了T(DBHP)P与BSA之间的作用力主要是静电引力。同时,利用同步荧光光谱,考察了T(DBHP)P对BSA构象的影响,结果发现,T(DBHP)P的加入使BSA构象发生变化,BSA内部残基所处环境的疏水性降低。  相似文献

The interaction between silver nanoparticles (SNPs) and Bovine Serum Albumin (BSA) was investigated at physiological pH in an aqueous solution using fluorescence spectroscopy. The analysis of fluorescence spectrum and fluorescence intensity indicates that SNPs have a strong ability to quench the intrinsic fluorescence of BSA by both static and dynamic quenching mechanisms. Resonance light scattering (RLS) spectra indicated the formation of a complex between BSA and SNP. The number of binding sites ‘n’ and binding constants ‘K’ were determined at different temperatures based on fluorescence quenching. The thermodynamic parameters namely ∆H°, ∆G°, ∆S° were calculated at different temperatures and the results indicate that hydrophobic forces are predominant in the SNP-BSA complex. Negative ∆G° values imply that the binding process is spontaneous. Synchronous fluorescence spectra showed a blue shift which is indicative of increasing hydrophobicity.  相似文献

Using as an example the concentration dependence of the fluorescence quenching constants for quenching by Cd2+ ions for different weight fractions of humic acid, we have demonstrated the application of the fluorescence quenching method for studying the effect of association on the structure of humic acids. We have established that the quenching constants for the light fraction (5–30 kD) decrease as the humic acid concentration increases and the quenching constants increase for the heavy fraction (60–100 kD). The observed dependences are interpreted in terms of the idea of oppositely directed effects of association on the structure of the environment of fluorophores located within the core and peripheral regions of the humic acid macromolecule.  相似文献

The interaction between ilaprazole and bovine serum albumin (BSA) has been investigated in the absence and presence of Cu2+ or Fe3+ by means of fluorescence spectroscopy. The fluorescence intensity of BSA decreased remarkably with no obvious BSA maximum emission wavelength shift by adding ilaprazole. Similar fluorescence shape with larger quenching extent of BSA was observed with increasing concentrations of ilaprazole in the presence of Cu2+ or Fe3+. The quenching constants and affinities of ilaprazole with BSA in the presence of Cu2+ and Fe3+ decreased. The decreased affinity and unchangeable binding distance in the presence of metal ions may result from a competitive binding between ilaprazole and metal ions. The results indicated that the presence of Cu2+ or Fe3+ could improve ilaprazole′s maximum effects, which may have relevant consequence in rationalizing dosage for patients with gastric and duodenal ulcers.  相似文献

Chloramphenicol (CHL) and quinolone drugs like ofloxacin (OFLX), lomefloxacin (LMX) and ciprofloxacin (CPFX) can all quench the fluorescence of bovine serum albumin (BSA) in the aqueous solution of pH=7.40. This quenching effect becomes more significant when CHL and quinolone drugs coexist. Based on this, further studies on the interactions between CHL and quinolone drugs using fluorescence spectrum are established. The results showed that the interaction between the drugs would increase the binding constant and binding stability of the drug and protein, thus reducing the amount of drugs transported to their targets. Therefore, free drug concentration at targets would decrease, reducing the efficacy of the drugs. It indicated that there exists antagonistic action between drugs. The results also showed that the quenching mechanism of BSA by the drugs is a static procedure. The number of binding sites is 1 in various systems. Due to the existence of the antagonistic action between drugs, the binding distance r is reduced. Studies utilizing synchronous spectra showed that the antagonistic action between the drugs would affect the conformation of BSA, making protein molecules extend and hydrophobic decrease. The order of antagonistic action between CHL and quinolone drugs is: CPFX>OFLX>LMX with presence of BSA.  相似文献

The interactions of human (HSA) and bovine (BSA) serum albumins with 1-cyanoethyl-5-chlorouracil (CECU) were investigated by fluorescence spectroscopy, UV absorption spectroscopy, and molecular modeling methods under the simulated physiological conditions. The results of fluorescence measurements indicate that CECU has a strong ability to quench the intrinsic fluorescence of both HSA and BSA through a static quenching procedure. The binding constants (K) at different temperatures and thermodynamic parameters, enthalpy change (ΔH), and entropy change (ΔS) were calculated according to fluorescence data. The results show that hydrophobic interaction is a predominant intermolecular force for stabilizing the complex, which is in agreement with the results of molecular modeling study. In this paper, interaction of Schiff base and its metal complexes carrying naphthalene ring in the structure with bovine serum albumin (BSA) were investigated using UV-vis absorption, fluorescence spectroscopies and molecular docking methods. The effect on the binding mechanism and properties of these compounds containing metal-free, iron and copper ions were also investigated. The fluorescence spectroscopy results showed that fluorescence intensity of BSA in the presence of different concentration of ligands was decreased through a static quenching mechanism. Binding constants (KSV, Kbin and Ka) and thermodynamic parameters (ΔG, ΔH and ΔS) for the ligand-protein interactions were also determined. ΔG values of ligand-protein interaction were calculated in the range ? 6.3 to ?5.5 kcal/mol. These negative values showed that binding process is spontaneous and, hydrogen bonding and van der Waals force were main interaction of the protein and ligands. ΔH and ΔS value were also calculated in the range of 1.10 to 1.26 kJ/mol and 0.133 to 0.135 kJ/mol. K, respectively. These positive values indicated that the binding process between ligands and BSA are endothermic and electrostatic interaction, respectively.  相似文献

应用荧光光谱法、紫外分光光度法研究了在不同酸度、温度条件下,西那沙星(Sinafloxacin)与牛血清白蛋白(bovine serum albumins,BSA)的相互作用,研究表明:西那沙星对BSA内源荧光的猝灭机制属于形成复合物所引起的静态猝灭,利用荧光猝灭双倒数图计算了西那沙星与牛血清白蛋白之间的结合常数KD,根据Fōrster非辐射能量转移理论计算出西那沙星与牛血清白蛋白结合时授体-受体间的结合距离r=3.64 nm、能量转移效率E=0.163,表明两者之间有较强的作用,并根据热力学参数确定了西那沙星与牛血清白蛋白之间的主要作用力类型为静电作用力,同时采用同步荧光、三维荧光技术考察了西那沙星对BSA构象的影响。此外,讨论了共存Cu2+,Fe3+,Zn2+,Mg2+对西那沙星与BSA结合作用的影响。为探讨西那沙星在生物体内与蛋白质的作用机理和生物学效应提供了理论依据。  相似文献

Three novel transition metal complexes (Hapy)(2)[M(DCA)(2)]·6H(2)O (M?=?Mn(II) (1), Ni(II) (2), Cu(II) (3); DCA?=?demethylcantharate, 7-oxabicyclo[2.2.1]heptane-2,3-dicarboxylate, C(8)H(8)O(5); Hapy?=?2-aminopyridine acid, C(5)H(7)N(2)) were synthesized and characterized by elemental analysis, infrared spectra, thermogravimetric analysis and X-ray diffraction. DNA binding properties of the complexes were investigated by electronic absorption spectra, fluorescence spectra and viscosity measurements. Results indicated the complexes could bind to DNA through partial intercalation mode with binding constants K ( b )/(L·mol(-1)) of 1.91?×?10(4) (1), 5.13?×?10(4) (2) and 1.12?×?10(5) (3) at 298?K. Meanwhile, the interactions of the complexes with BSA were also studied by fluorescence spectra. The results suggested that the complexes could quench the fluorescence of BSA through static quenching with the binding constants K ( A )/(L·mol(-1)) of 1.44?×?10(6) (1), 1.14?×?10(7) (2) and 2.98?×?10(4) (3). And the main contribution was tryptophan residues of BSA. The antiproliferative activity test revealed that complexes showed more intense inhibition ratios against human hepatoma cells lines and human gastric cancer cells lines in vitro. Copper(II) complex (3) possesses the strongest inhibition ratio against human hepatoma cells.  相似文献
