Cholesterol oxidase biosensor has been constructed by using bovine serum albumin and glutaraldehyde as cross linker to immobilize cholesterol oxidase and cholesterol esterase on a glassy carbon electrode modified with Nafion and methyl viologen. The biosensor has been used to determine total cholesterol in blood. The linear range of the determination is 2.5×10~7 to 1.0×10-4 mol/L. The detection limit is about 5.0×10~8 mol/L. The response time is 12 s. This biosensor has the advantage of high selectivity, sensitivity and short response time.  相似文献

Myoglobin (Myb) of horse heart is incorporated on multi‐walled carbon nanotubes (MWNTs) and immobilized at a glassy carbon (GC) electrode surface. Its electrochemical behavior and enzyme activity are characterized by employing electrochemical methods. The results indicate that MWNTs can obviously promote the direct electron transfer between Myb and electrode, and that the Myb on MWNTs behaves as an enzyme‐like activity towards the electrochemical reduction of nitric oxide (NO). Accordingly, an unmediated NO biosensor is constructed. Experimental results reveal that the peak current related to NO is linearly proportional to its concentration in the range of 2.0×10?7–4.0×10?5 mol/L. The detection limit is estimated to be 8.0×10?8 mol/L. Considering a relative standard deviation of 2.1% in seven independent determinations of 1.0×10?5 mol/L NO, this biosensor shows a good reproducibility. The biosensor based on Myb/MWNTs modified electrode can be used for the rapid determination of trace NO in aqueous solution with a good stability, nice selectivity and easy construction.  相似文献

A bienzyme biosensor in which the enzymes β‐galactosidase (β‐Gal), fructose dehydrogenase (FDH), and the mediator tetrathiafulvalene (TTF) were coimmobilized by cross‐linking with glutaraldehyde atop a 3‐mercaptopropionic acid (MPA) self‐assembled monolayer on a gold disk electrode, is reported. The working conditions selected were Eapp=+0.10 V and (25±1) °C. The useful lifetime of one single TTF‐β‐Gal‐FDH‐MPA‐AuE was surprisingly long, 81 days. A linear calibration plot was obtained for lactulose over the 3.0×10?5–1.0×10?3 mol L?1 concentration range, with a limit of detection of 9.6×10?6 mol L?1. The effect of potential interferents (lactose, glucose, galactose, sucrose, and ascorbic acid) on the biosensor response was evaluated. The behavior of the SAM‐based biosensor in flow‐injection systems in connection with amperometric detection was tested. The analytical usefulness of the biosensor was evaluated by determining lactulose in a pharmaceutical preparation containing a high lactulose concentration, and in different types of milk. Finally, the analytical characteristics of the TTF‐β‐Gal‐FDH‐MPA‐AuE are critically compared with those reported for other recent enzymatic determinations of lactulose.  相似文献

The carbon nanotubes decorated nanoplatinum (CNT‐Pt) were prepared using a chemical reduction method and a novel base electrode was constructed by intercalating CNT‐Pt on the surface of a waxed graphite electrode. The results showed that the nano‐particles of platinum at a waxed graphite electrode exhibits high catalytic activity for the reduction of hydrogen peroxide. The cholesterol oxidase (ChOx), chosen as a model enzyme, was immobilized with sol‐gel on the CNT‐Pt base electrode to construct a biosensor. The current response of the biosensor for cholesterol was very rapid (<20 s). The linear range for cholesterol measurement was 4.0×10?6 mol/L ?1.0×10?4 mol/L with a detection limit of 1.4×10?6 mol/L. The experiments also showed that the ChOx/sol‐gel/CNT‐Pt biosensor was sensitive and stable in detecting cholesterol in serum samples.  相似文献

Hemoglobin was entrapped in composite electrodeposited chitosan‐multiwall carbon nanotubes (MCNTs) film by assembling gold nanoparticles and hemoglobin step by step. In phosphate buffer solution (pH 7), a pair of well‐defined and quasireversible redox peaks appeared with formal potential at ?0.289 V and peak separation of 100 mV. The redox peaks respected for the direct electrochemistry of hemoglobin at the surface of chitosan‐MCNTs‐gold nanoparticles modified electrode. The parameters of experiments have also been optimized. The composite electrode showed excellent electrocatalysis to peroxide hydrogen and oxygen, the peak current was linearly proportional to H2O2 concentration in the range from 1×10?6 mol/L to 4.7×10?4 mol/L with a detection limit of 5.0×10?7 mol/L, and this biosensor exhibited high stability, good reproducibility and better selectivity. The biosensor showed a Michaelis–Menten kinetic response as H2O2 concentration is larger than 5.0×10?4 mol/L, the apparent Michaelis–Menten constant for hydrogen peroxide was calculated to be 1.61 μmol/L.  相似文献

In this paper, a pre‐anodized inlaying ultrathin carbon paste electrode (PAIUCPE) with 316L as a matrix was constructed by a simple and fast electrochemical pretreatment. Using xanthine (Xa) and hypoxanthine (HXa) as the target compounds, the pH effects compositions of buffer solution, the accumulation times, hydrogen bond catalysis, degree of auxiliary electrode reaction on the size of peak currents (Ip) of Xa and HXa was discussed in detail. Also, it was proposed that Xa and HXa were respectively absorbed at the surface of PAIUCPE through hydrogen bonding. The influencing mechanisms of the PAIUCEP on electrochemical oxidation of Xa and HXa were explained in detail. Moreover, the linear relationships for the Xa and HXa were obtained in the range of 6×10?8–3×10?5 mol/L and 2×10?7–7×10?5 mol/L, respectively. The detection limits for the Xa and HXa were 1.2×10?8 mol/L and 5.7×10?8 mol/L, respectively. Moreover, this proposed method could be applied to determine the Xa and HXa in human urine simultaneously with satisfactory results.  相似文献

In this paper, nichrome was adopted as a substrate, to fabricate the pre‐anodized inlaying ultrathin carbon paste electrode (PAIUCPE). The electrochemical behaviors and diffusion mechanisms of three dihydroxybenzene isomers at the electrode were carefully investigated. The effect of pH on oxidation peak current was also detailedly explained. The results were shown that oxidation peak current not only related to the reaction of electroactive materials at the working electrode, but also depended on the reaction variable of reduction at the auxiliary electrode. The oxidation peaks of hydroquinone (HQ), catechol (CC) and resorcinol (RC) located at 0.181 V, 0.288 V and 0.736 V. For CC, RC and HQ, the oxidation peak currents were linear to the concentrations at the range of 5.0 × 10?6~5.0 × 10?4 mol/L, 3.0 × 10?6~5.0 × 10?4 mol/L and 4.0 × 10?6~4.0 × 10?4 mol/L with the detection limits of 2.0 × 10?7 mol/L, 1.2 × 10?7 mol/L and 1.2 × 10?7 mol/L, respectively. The proposed method was successfully applied in the simultaneous determination of dihydroxybenzene isomers in artificial sewage samples with satisfactory results.  相似文献

A uric acid biosensor was fabricated by the Langmuir–Blodgett (LB) technique to immobilize the uricase on chitosan/Prussian blue (CS/PB) prefunctionalized indium-tin oxide (ITO) electrode. The effects of ionic strengths, acidity of subphase, and uricase amount on the film were studied. The electrochemical properties of the uricase/n-nonadecanoic acid (UOx/NA) LB film proved that CS/PB was a good electro-catalyst for the reduction of hydrogen peroxide produced by enzymatic reaction of UOx, and protein molecules retained their natural electro-catalytic activity. The linear range of uric acid detection was from 5 × 10?6 mol/L to 1.15 × 10?3 mol/L with a detection limit of 1.8 × 10?7 mol/L.  相似文献

A cholesterol biosensors fabricated by immobilization of cholesterol oxidase (ChOx) in a layer of silicic sol‐gel matrix on the top of a Prussian Blue‐modified glassy carbon electrode was prepared. It is based on the detection of hydrogen peroxide produced by ChOx at ?0.05 V. The half‐lifetime of the biosensor is about 35 days. Cholesterol can be determined in the concentration range of 1×10?6?8×10?5 mol/L with a detection limit of 1.2×10?7 mol/L. Normal interfering compounds, such as ascorbic acid and uric acid do not affect the determination. The high sensitivity and outstanding selectivity are attributed to the Prussian Blue film modified on the sensor.  相似文献

A mediator-free glucose biosensor, termed a “third-generation biosensor,” was fabricated by immobilizing glucose oxidase (GOD) directly onto an oxidized boron-doped diamond (BDD) electrode. The surface of the oxidized BDD electrode possesses carboxyl groups (as shown by Raman spectra) which covalently cross-link with GOD through glutaraldehyde. Glucose was determined in the absence of a mediator used to transfer electrons between the electrode and enzyme. O2 has no effect on the electron transfer. The effects of experimental variables (applied potential, pH and cross-link time) were investigated in order to optimize the analytical performance of the amperometric detection method. The resulting biosensor exhibited fast amperometric response (less than 5 s) to glucose. The biosensor provided a linear response to glucose over the range 6.67×10−5 to 2×10−3 mol/L, with a detection limit of 2.31×10−5 mol/L. The lifetime, reproducibility and measurement repeatability were evaluated and satisfactory results were obtained.  相似文献

Four kinds of xanthine oxidase (XOD) based amperometric biosensors were fabricated and their analytical performances were compared. Polypyrrole (PPY)/XOD biosensor was constructed by electrochemical oxidation of pyrrole in the solution containing xanthine oxidase and pyrrole in this paper. Colloidal Au was then immobilized on the biosensor. On the other hand, electron mediator, Prussian Blue (PB), was deposited on the electrode before the immobilization of PPY/XOD to enhance electron‐transfer rate and current response. The results showed that PPY/XOD, PPY/XOD/Au‐colloid, PB/PPY/XOD and PB/PPY/XOD/Au‐colloid biosensors exhibit good response to xanthine in 1×10?6 M and 2×10?5 M and Michaelis‐Menten constants (Km) of these biosensors were 242.2, 113.4, 144.5, 43.2 μmol?L?1, respectively. The dependence of current responses with applied voltages was discussed, and different mechanisms of these biosensors were discussed. It has been found that colloidal Au can enhance the current response at the same concentration of xanthine solution and decrease the energy‐barrier of electron‐transfer reaction on the electrode.  相似文献

An amperometric cholesterol biosensor based on immobilization of cholesterol oxidase in a Prussian blue (PB)/polypyrrole (PPy) composite film on the surface of a glassy carbon electrode was fabricated. Hydrogen peroxide produced by the enzymatic reaction was catalytically reduced on the PB film electrode at 0 V with a sensitivity of 39 μA (mol/L)?1. Cholesterol in the concentration range of 10?5 ? 10?4 mol/L was determined with a detection limit of 6 × 10?7 mol/L by amperometric method. Normal coexisting compounds in the bio‐samples such as ascorbic acid and uric acid do not interfere with the determination. The excellent properties of the sensor in sensitivity and selectivity are attributed to the PB/PPy layer modified on the sensor.  相似文献

Gold modified nanoporous silica based magnetic microparticles have been prepared as support for the immobilization of the enzyme horseradish peroxidase (HRP). The enzyme modified gold microparticles were retained onto the surface of a solid carbon paste electrode with the help of a permanent magnet. The analytical performances of the resulting biosensor were characterized by studying hydroquinone (HQ) and hydrogen peroxide. The former was monitored by the direct electroreduction of the biocatalytically generated quinone. Several experimental parameters influencing the biosensor response were investigated. A linear response to HQ was obtained in the concentration range comprised between 5×10?7 and 4.5×10?6 M with a detection limit of 4×10?7 M. The enzyme electrode provided a linear response to hydrogen peroxide over a concentration range comprised between 5×10?7?1.3×10?4 M with a detection limit of 4×10?7 M. The inhibition of the biosensor response in the presence of thiols e.g. cysteine, captopril, glutathione and Nacystelyn (NAL) has been pointed out.  相似文献

A novel biopolymer/room‐temperature ionic liquid composite film based on carrageenan, room temperature ionic liquid (IL) [1‐butyl‐3‐methylimidazolium tetra?uoroborate ([BMIM]BF4)] was explored for immobilization of hemoglobin (Hb) and construction of biosensor. Direct electrochemistry and electrocatalytic behaviors of Hb entrapped in the IL‐carrageenan composite ?lm on the surface of glassy carbon electrode (GCE) were investigated. UV‐vis spectroscopy demonstrated that Hb in the IL‐carrageenan composite ?lm could retain its native secondary structure. A pair of well‐de?ned redox peaks of Hb was obtained at the Hb‐IL‐carrageenan composite ?lm modi?ed electrode through direct electron transfer between the protein and the underlying electrode. The heterogeneous electron transfer rate constant (ks) was 2.02 s?1, indicating great facilitation of the electron transfer between Hb and IL‐carrageenan composite film modi?ed electrode. The modi?ed electrode showed excellent electrocatalytic activity toward reduction of hydrogen peroxide with a linear range of 5.0×10?6 to 1.5×10?4 mol/L and the detection limit was 2.12×10?7 mol/L (S/N=3). The apparent Michaelis‐Menten constant KMapp for hydrogen peroxide was estimated to be 0.02 mmol/L, indicating that the biosensor possessed high af?nity to hydrogen peroxide. Selected from a series of structurally related heteroaromatic thiols, a newly synthesized reagent 2‐amino‐5‐mercapto‐[1,3,4] triazole (MATZ) was used to fabricate self‐assembled monolayers (SAMs) on gold electrode for the first time. The MATZ/Au SAMs was characterized by electrochemical methods and scanning electronic microscopy (SEM). In 0.04 mol/L Britton–Robinson buffer solution (pH 5), the electrochemical behavior of dopamine showed a quasireversible process at the MATZ/Au SAMs with an electrode kinetic constant 0.1049 cm/s. However, the electrochemical reaction of uric acid at the SAMs electrode showed an irreversible oxidation process, the charge‐transfer kinetics of uric acid was promoted by the SAMs. By Osteryoung square‐wave voltammetry (OSWV), the simultaneous determination of dopamine and uric acid can be accomplished with an oxidation peak separation of 0.24 V, the peak current of dopamine and uric acid were linearly to its concentration in the range of 2.5×10?6–5.0×10?4 mol/L for dopamine and 1×10?6–1×10?4 mol/L for uric acid with a detection limit of 8.0×10?7 mol/L for dopamine and 7.0×10?7 mol/L for uric acid. The MATZ/Au SAMs electrode was used to detect the content of uric acid in real urine and serum sample with satisfactory results.  相似文献

A laccase-based biosensor was developed by specific enzyme adsorption on screen-printed working electrodes of DROPSENS cells, and stabilized with Nafion 0.1% membrane. The electrode was characterized with respect to response time, sensitivity, linear range, detection limit, pH dependence, interferences, and long-term stability. The tested substrates were catechol, rosmarinic acid, caffeic acid, chlorogenic acid, and gallic acid. The optimized biosensor proved the following characteristic performances: the apparent Michaelis Menten calculated considering rosmarinic acid substrate 8.3 × 10?6 mol L?1 (r = 0.995, n = 6); the dynamic range of biosensor response for rosmarinic acid 7 × 10?7 ? 1.5 × 10?6 mol L?1; the detection limit for rosmarinic acid 1.19 × 10?7 mol L?1 (RSD = 1.08%, n = 3). It was noticed that the biosensor reaches systematically 90% to 94.3% from the response obtained by LC-DAD-ESI-MS for real samples.  相似文献

The fabrication and application of a new electrochemical detector for use in HPLC is presented. The detector consists of an electrode modified with a composite film composed of acetylene black and dihexadecyl hydrogen phosphate. The electrochemistry of xanthine and hypoxanthine at this chemically modified electrode (CME) was investigated by cyclic voltammetry. It is found that the CME exhibits efficient electrocatalytic activity towards xanthine and hypoxanthine, with good sensitivity, stability, and lifetime. The linear ranges cover three orders of magnitude, and the detection limits are 6.0 × 10?8 mol L?1 for xanthine and 2.5 × 10?7 mol L?1 for hypoxanthine (at an S/N ratio of 3). The method was coupled to in-vivo microdialysis sampling and successfully applied to quantify xanthine and hypoxanthine in rat striatal microdialysates of freely moving rats.  相似文献

The present work describes the development of a nanocomposite system and its application in construction of a new amperometric biosensor applied in the determination of total polyphenolic content from propolis extracts. The nanocomposite system was based on covalent immobilization of laccase on functionalized indium tin oxide nanoparticles and it was morphologically and structural characterized. The casting of the developed nanocomposite system on the surface of a screen-printed electrode was used for biosensor fabrication. The analytical performance characteristics of the settled biosensor were determined for rosmarinic acid, caffeic acid and catechol (as laccase specific substrate). The linearity was obtained in the range of 1.06×10?6 ? 1.50×10?5 mol L?1 for rosmarinic acid, 1.90×10?7 ? 2.80×10?6 mol L?1 for caffeic acid and 1.66×10?6 ? 7.00×10?6 mol L?1 for catechol. A good sensitivity of amperometric biosensor 141.15 nA µmol?1 L?1 and fair detection limit 7.08×10?8 mol L?1 were obtained for caffeic acid. The results obtained for polyphenolic content of propolis extracts were compared with the chromatographic data obtained by liquid-chromatography with diode array detection.   相似文献

We report on a novel electrochemical method to detect trace pentachlorophenol (PCP) by using a chitosan (CS) modified carbon paste electrode (CS/CPE). Compared with that at a bare carbon paste electrode (CPE), the current response was greatly improved at the CS/CPE due to the enhancement effect of CS. Under optimal working conditions, the oxidation peak current of PCP was proportional to its concentration in the range of 1.0 × 10?7 to 5.0 × 10?6 and 5.0 × 10?6 to 1.0 × 10?4 mol/L, with an extremely low detection limit of 4.0 × 10?8 mol/L. Our method was successfully used to detect the PCP concentration in vegetable samples.  相似文献
