Summary Direct chiral-phase HPLC methods have been developed for the determination of flurbiprofen and its major metabolites, namely 4′-hydroxyflurbiprofen and 3′-hydroxy-4′-methoxyflurbiprofen, in biological fluids using a derivatized amylose chiral stationary phase (CSP; Chiral-pak AD). Quantification of all three analytes, both free and conjugated, in urine was carried out following liquid-liquid extraction using tandem ultraviolet (UV) and fluorescence detection. Determination of flurbiprofen and the 4′-hydroxy-metabolite in plasma utilized the same CSP but required modification in the mobile phase composition and sole use of fluorescence detection. The urine assay was linear (r>0.998) between 0.05–10 μg mL−1, 0.1–20 μg mL−1 and 0.01–2 μg mL−1 for the enantiomers of flurbiprofen, 4′-hydroxyflurbiprofen and 3′-hydroxy-4′-methoxyflurbiprofen respectively. The plasma assay was linear (r>0.997) between 0.1–6 μg mL−1 and 0.01–0.6 μg mL−1 for the enantiomers of flurbiprofen and 4′-hydroxyflurbiprofen respectively. Both assays, typically yielded within- and between-day imprecision and accuracy values less than 10% for the enantiomers of the different analytes. Initial volunteer studies suggest that the disposition of flurbiprofen displays modest enantioselectivity in humans.  相似文献

Summary The chromatographic resolution of the enantiomers of sulindac has been achieved using a Chiralpak AD CSP (10 μm, 250×4.6 mm) with a mobile phase of hexane: ethanol (85∶15 v/v) containing trifluoroacetic acid (0.05% v/v) at a flow rate of 1.0 mL min−1. Under these conditions the enantiomers eluted with separation and resolution factors of 1.43 and 2.46 respectively. Semipreparative isolation of the enantiomers and their characterization by circular dichroism spectroscopy and NMR, in the presence of a chiral shift reagent, indicated that the elution order was (−)-(S)- before (+)-(R)-sulindac. The enantiomeric composition of sulindac in urine following administration of the racemic drug to man was determined by sequential achiral-chiral chromatography. Achiral analysis was carried out using a Spherisorb S5 ODS2 stationary phase (5 μm, 250×4.6 mm) and a mobile phase of aqueous acetic acid (2% v/v; pH 3.5): acetonitrile: THF (50∶48∶2 by volume) at a flow rate of 1.0 mL min−1. The HPLC eluate containing sulindac (retention time 4.9 min) was collected and following workup, the enantiomeric composition of the drug was determined using the CSP. Over the 24 h collection period sulindac was excreted predominantly as theR-enantiomer, but the enantiomeric composition was found to vary markedly with time which is presumably associated with the complex metabolism of the drug.  相似文献

The influence of the mobile phase composition and the column temperature on the retention and the enantioselectivity are investigated. Summary Direct separations of the enantiomers of cetirizine and related compounds have been achieved by reversed-phase HPLC on the Chiralcel OD-R, a polysaccharide-derived chiral stationary phase; the mobile phase was usually perchlorate solution supplemented with acetonitrile. Resolution of the enantiomers of cetirizine and related compounds was good. The effect of the acetonitrile content of the mobile phase was investigated, and the effect of the structure of the chiral compounds on their behavior on the Chiralcel OD-R column is discussed.  相似文献

Summary In preparative column liquid chromatography, it is necessary to work with conditions of mass overload to obtain high throughput per unit time. The influence of sample mass and of eluent composition on the separation of R,S-1-(1-naphthylethyl)propylamide on chiral 3,5-dinitrobenzoylphenylglycine-silica (Pirkle type stationary phase) was investigated. At a sample load of 200μg of racemate per gram of stationary phase, the column becomes overloaded. At higher sample loads the peaks become triangular; therefore the first peak is always pure up to loads of 10mg g−1, although resolution is low. The purity of the second peak depends on sample mass, but not on the strength (polarity) of the mobile phase. This is due to the effect that peak width, as well as resolution, decrease as the polarity of the eluent increases. In certain cases the polarimeter, used as a detector, can give more information on peak purity than the UV detector.  相似文献

The direct enantioseparation of duloxetine and its R-enantiomer was achieved by HPLC using hydroxypropyl-β-cyclodextrin (HP-β-CD) as a chiral selector and a vancomycin chiral stationary phase (Chirobiotic V). Operational parameters, such as the concentration of HP-β-CD, buffer pH, organic modifiers, temperature and flow rate, were varied in order to achieve the desired retention time and resolution. These two enantioseparation methods developed gave a baseline resolution of the enantiomers. Finally, the HPLC-CSP method was selected to determine the enantiomeric purity of duloxetine drug substance due to its much shorter analysis time and better resolution. The limit of detection of this method was 0.06 μg mL−1.  相似文献

Summary Optical isomers of substituted binaphthyl, compounds such as 2,2-dihydroxy-1,1-dinaphthyl and 1,1-binaphthyl-2,2-diyl hydrogenphosphate are, separated by HPLC using micellar bile-salt mobile phases. Operating conditions which affect the optical resolution of these enantiomers are examined. The largest separation factor achieved for these enantiomers is 2.74. The elution order of the latter enantiomers changes with mobile phase composition.  相似文献

Summary The preparation and properties of chiral bonded phases with amino acid groups are described. These phases were used in non-polar eluents and in aqueous systems in the presence of Cu2+ ions in the ligand-exchange mode. The optimization of chiral resolution is demonstrated for both cases. With non-polar eluents similarity between bonded groups and solutes is required. The elution could be accelerated by non-protonic moderators. Ligand-exchange separation is influenced by the copper content of the eluent and the stationary phase, the organic moderator concentration, the pH and ionic strength of the buffer and by the temperature. Structural requirements of both the bonded groups and of the solutes for chiral separation are discussed.  相似文献

Reasonable recognition is achieved in the separation of organosilicon compounds on cellulose carbamate. Summary Chromatographic separation of racemic amines, carboxylic acids and alcohols can be achieved with excellent resolution as measured in terms of the chromatographic separation factor by derivatization with a homochiral oxazolidin-2-one easily prepared in three steps fromendo-borneol. The resolved materials can be isolated in excellent yields by cleavage of the resultant diastereomers using conventional methods, which also allow recovery of the chiral resolving agent for re-use.  相似文献

A stereoselective high-performance liquid chromatographic method has been established for chiral separation of melatoninergic derivatives with one or two chiral centers, new agonist and antagonist ligands for melatonin receptors. Reversed-phase separations were performed on cellulose-based chiral stationary phases—tris-3,5-dimethylphenylcarbamate (Chiralcel OD-RH) or tris-methylbenzoate (Chiralcel OJ-R). Water–modifier (methanol or acetonitrile) mixtures in different proportions were used as mobile phases. The effects of organic mobile-phase modifier concentration, temperature, and compound structure were examined. Baseline separation (RS > 1.5) was readily obtained for many of the compounds.  相似文献

Summary An analytical approach has been developed for the one step determination of enantiomeric amphetamine composition in plasma, using on-line, pre-column solid phase derivatization with reversed phase HPLC separation. The high molecular weight protein components were excluded by the small pore structure of the polymer and washed out of the reaction column before derivatization. Spiked amphetamine in human plasma was extracted and derivatized by the polystyrene based FMOC-L-prolyl solid phase reagent. The derivatized diastereomers were separated on a conventional ODS column with an ACN/H2O mobile phase. No kinetic resolution or racemization was observed in this solid phase derivatization. Calibration plots and reproducibility experiments were performed to demonstrate the validity of the new approach. Automation of the procedure provided a simple and reproducible method for direct chiral recognition in plasma samples.  相似文献

Summary High-performance liquid chromatographic and gas chromatographic methods were developed for the separation of unusual secondary aromatic amino acids. Amino acids containing 1,2,3,4-tetrahydroisoquinoline, 1,2,3,4-tetrahydronorharmane-1-carboxylic acid and 1,2,3,4-tetrahydro-3-carboxy-2-carboline moieties were synthetized in racemic or chiral forms. The high-performance liquid chromatography was carried out either on a teicoplanin-containing chiral stationary phase or on an achiral C18 column. In the latter case the diastereomers of the amino acids formed by precolumn derivatization with the chiral reagents 2,3,4,6-tetra-O-acetyl-β-D-glucopyranosyl isothiocyanate or 1-fluoro-2,4-dinitrophenyl-5-L-alanine amide were separated. The gas chromatographic analyses were based on separation on a Chirasil-L-Val column. Two crystalline degradation products (CDP-1-M and CDP-1-m) are formed from vancomycin by hydrolytic loss of ammonia. The CDP are structurally similar to vancomycin, with two carboxyl groups, but are biologically inactive. In this work the CDP of vancomycin have been covalently linked to silica gel and their ability to resolve racemic mixtures, their stability, and their loading has been evaluated by HPLC. Elemental analysis and scanning electron microscopy were used to monitor the surface of this new stationary phase. Enantiomeric separation was achieved for some compounds that have not been previously been separated on other chiral stationary phases.  相似文献

Summary Enantiomers of several sulfoxides and some selenoxides can be easily separated by using a new chiral stationary phase (CSP-DACH-DNB) containing the 3,5-dinitrobenzoyl derivative of R,R-(-)1,2-diamino-cyclohexane as selector, covalently bonded to the siliceous matrix. The easy operative conditions and the high enantioselectivity values (α) allow a direct transfer of the analytical separations to a semi-preparative and preparative scale.  相似文献
