Diazabicycloalkanes with nitrogen atoms in bridgehead positions. 16. Synthesis and properties of benzo[b]-1,4-diazabicyclo[2.2.2]octene and dibenzo[b,e]-1,4-diazabicyclo[2.2.2]octadiene,containing primary aromatic amino acids

4-Aminobenzo[1,2-b]-1,4-diazabicyclo[2.2.2]octene and 4-aminodibenzo[1,2-b, e]-1,4-diazabicyclo[2.2.2]octadiene have been prepared by cyclization reactions of N--chloroethyl derivatives of 1,2,4-triaminobenzene and aminophenazine, and subsequent catalytic hydrogenation of the corresponding 4-nitrobenzo[1,2-b]-1,4-diazabicyclo[2.2.2]octene and 4-benzylaminodibenzo[1,2-b,e]-1,4-diazabicyclo[2.2.2]-octadiene. Pyrimidines

It is shown that the reaction of benzalbisurea with 3-amino-5-hydroxy-1-R-pyrazoles gives 1-R-dipyrazolo[3,4-b:4,3-e]pyridine (R=CH₃) or a spiro(pyrazole-4,5-pyrimidine) derivative (R=C₆H₅). Similar reactions of benzalbisurea with 3-hydroxy-5-amino-1-methylpyrazole give 2-methyldipyrazole[3,4-b:4,3-e]pyridine or substituted tetrahydropyrazolo[3,4-d] pyrimidine. Reactions of 2-aryl (hetaryl) methylene-3-oxoquinuclidines with bifunctional nucleophilic reagents

The reactions of 2-aryl(hetaryl)methylene-3-oxoquinuclidines with hydrazine hydrate, thiourea, and phenylhydrazine gave, respectively, 7-phenyl-7,7a,dihydropyrazolo[3,4-b]quinuclidine, 7-(4-methoxyphenyl)-7,7a-dihydropyrazolo[3,4-b]quinuclidine, 7-(2-thienyl)-7,7a-dihydropyrazolo-[3,4-b]quinuclidine, 3,3-azinobis (2-benzylidenequinuclidine), 3,3-azinobis[2-(4-methoxybenzylidene)quinuclidine], 6-thio-8-phenyl-5,6,7,8-tetrahydropyrimido[5,4-b]quinuclidine, 2-benzylidene -3-oxoquinuclidine phenylhydrazone, and 2-(4-methoxybenzylidene)-3-oxoquinuclidine phenylhydrazone. Study of furan compounds

Research on synthesis of polycyclic spirans of the 1, 6-dioxaspiro-[4, 4]nonane group is continued. Electrolysis of methanol solutions of 2-furylcyclopentanol, 2-(5-methyl-furfuryl)cyclopentanol, and 2-furfuryl-1-indanol, gives, by intramolecular alkoxylation, spiro{perhydrocyclopenta[b]furan-2, 2-(5dihydrofuran)}, spiro{perhydrocyclopenta[b]furan-2, 2-(5-methoxy-5-methyl-2, 5-dihydrofuran)}, and spiro{2, 3, 3a, 8b-tetrahydro-4H-indeno[1, 2-b]furan-2, 2-(5-methoxy-2, 5-dihydrofuran)}, hitherto undescribed in the literature. Depending on the conditions, catalytic hydrogenation of these gives: spiro{perhydiocyclopenta[b]furan-2, 2-(5-methoxytetrahydrofuran)}, spiro{2, 3a, 8b-tetrahydro-4H-indeno[1, 2-b]furan-2, 2-(5-methoxytetrahydrofuran)}, spiro{perhydrocyclopenta[b]furan-2, 2-tetrahydrofuran}, and spiro{2, 3, 3a, 8b-tetrahydro-4H-indeno[1, 2-b]furan-2, 2-tetrahydrofuran}.For Part XXXI see [1].     

Formation of 5-oxoindeno[1,2-b]pyran derivatives in cyclization of 1,5-diketones of the indian-1,3-dione series

Indeno[1,2-b]pyrans, including 2-methylene and 2-acetoxy derivatives of 3-ethoxy-carbonyl-4-phenyl-5-oxo-3,4-dihydroindeno[1,2-b]pyran, are formed in the cyclization of 4-(indan-1,3-dion-2-yl)-4-phenyl-2-butanone and its 3-ethoxycarbonyl derivative in acetic anhydride in the presence of sulfuric acid.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 2, pp. 178–183, February, 1988.     

Azide-tetrazole tautomerism of diazidodiazines and their benzo analogs

It has been discovered with the aid of IR and NMR spectroscopy that 5-azidotetrazolo [1,5-a]quinazoline exists in solutions in a tautomeric equilibrium with ditetrazolo [1,5-a:1,5-c]quinazoline, and the thermodynamic and kinetic characteristics of the equilibrium have been determined. Synthesis and reactions of pyrrolo[2,3-c]azepine derivatives

The construction of the pyrrolo[2,3-c]azepine system by the reaction of 2-oxo-3-hydroxy-4-cyano-2H, 1,5,6,7-tetrahydroazepine with glycine ester and subsequent cyclization of the resulting N-substituted amino nitrile in an alcohol solution of sodium ethoxide was studied. The pyrrolo[2,3-c]azepine system was converted to the three-ring pyrimido[4,54,5]pyrrolo[2,3-c]azepine system, the alkylation of which gave N,N-dialkyl and N,N,N-trialkyl derivatives. Cyclization of 3-benzyl-4, 6-dioxo-5-(N,N-dimethyl)aminoethyl-6H,3,4,7,8,9,10-hexahydropyrimido[4,54,5]pyrrolo[2,3-c] azepine hydrochloride under the influence of phosphorus oxychloride gave the four-ring pyrazino[3,2,1-b,c]azepino[3,4-b]pyrrolo[3,2-d]-pyrimidine system. Podand-porphyrins with peripheral chelates

Alkylation of 5-(3-nitro-4-hydroxyphenyl)-10,15,20-triphenylporphyrin with chloropyridines[2,6-dichloromethyl-and 2-chloromethyl-6-(2-nitrophenoxymethylpyridine)] as well as condensation by a mixed-aldehyde method starting with formylpyridines [2-(2-nitro-4-formylphenoxymethyl)-6-(2-nitrophenoxy-methyl-) or 2,6-di(2-nitro-4-formylphenoxymethyl)pyridine], benzaldehyde, and pyrrole are used to synthesize previously unknown podand-porphyrins containing from one to four peripheral chelates.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 4, pp. 475–481, April, 1991.     

N-acetylindoxyl in the synthesis of new condensed heterocycles

Derivatives of a new heterocyclic system thieno[2,35,6]pyrimido[3,4-a]indole, were obtained by the reaction of substituted N,O-diacetylindoxyls with excess N-(2-methyl-3-ethoxycarbonyl-4-thienyl)hydrazine. Transformed steroids. 160. Reaction of 20,20-dimethoxy-16α,17α-epoxypregn-5-ene-3β, 21-diol-20-one with pyridine thiocyanate and carbethoxyhydrazine

Conclusion The reaction of 20,20-dimethoxy-16,17-epoxypregn-5-ene-3,21-diol-20-one with pyridine thiocyanate in the presence of carbethoxyhydrazine proceeds by two competing paths: cisopening of the oxide ring by a -SCN ion at the C¹⁷ atom, and substitution of one of the methoxyl groups by -NCS ion. As a result 2,20-dicarbethoxyhydrazones of pregn-5-ene-3,21-diol-20-one-[17,16-d]-1,3-oxathiolan-2-one and 20-methoxy-16,17-epoxypregn-5-ene-3-ol-[20,21-d]-1,3-oxazolidine-2-thione are formed.Translated from Izvestiya Akademii Nauk SSSR, Seriya Khimicheskaya, No. 4, pp. 911–914, April, 1987.     

Preparation and stereochemical characterization of some N-acyl-[1]benzopyrano[3,4-c]pyrazole derivatives from rotenoids

Summary Several new N-acyl derivatives of 1-(4-hydroxy-2,3-dihydrobenzofuran-5-yl)-7,8-di methoxy-1,9 b,2,3,3 a,4-hexahydro-[1]benzopyrano[3,4-c]pyrazole-1-ene have been prepared by appropriate chemical transformation of isohydrazones of rotenone and amorphigenin. A study of their¹H- and¹³C-NMR spectra confirmed the presence of the twocis 3a, 9b, 2 and 3a, 9b, 2 diastereomers in the parent isohydrazones and revealed the strong predominance of the conformers withendo orientation of the 3-NAc group. The conformations due to rotation about the 1,5-bond between rings C and D in the 4-OH and 4-O-substituted compounds were also determined by taking into account the anisotropic effect of aromatic rings A and D, and the hydrogen bond between 4-OH and the 2-N atom, as well as by inspecting the Dreiding models.

Darstellung und stereochemische Charakterisierung einiger N-Acyl-[1]benzopyrano[3,4-c]pyrazol-Derivate von Rotenoiden

Zusammenfassung Mittels geeigneter chemischer Transformationen von Isohydrazonen von Rotenon und Amorphigenin wurden einige neue N-Acyl-Derivate von 1-(4-Hydroxy-2,3-dihydrobenzofuran-5-yl)-7,8-dimethoxy-1,9 b,2,3,3 a,4-hexahydro-[1]benzopyrano[3,4-c]pyrazol-1-en hergestellt. Eine Untersuchung ihrer¹H und¹³C-NMR-Spektren zeigt die Gegenwart von zweicis 3a, 9b, 2- und 3a, 9b, 2-Diastereomeren in den Ausgangs-Isohydrazonen und eine starke Bevorzugung der Konformeren mitendo-Orientierung der 3-NAc-Gruppe. Die Konformationen bezüglich der Rotation um die 1,5-Bindung zwischen Ring C und D werden für die 4-OH und 4-O-substituierten Verbindungen unter Berücksichtigung von Anisotropie-Effekten der aromatischen A- und D-Ringe, der Wasserstoffbrücken zwischen 4-OH und dem 2-N Atom und auch der Betrachtung der entspechenden Dreiding-Modelle diskutiert.

     

Synthesis of 2,3-disubstituted 4-oxo-3,4,5,6-tetrahydrospiro-(benzo[h]quinazoline-5,1′-cyclopentanes)

3-Substituted 4-oxo-2-thioxo-1,2,3,4,5,6-hexahydrospiro(benzo[h]quinazoline-5,1-cyclopentanes) were obtained by the reaction of 4-amino-3-ethoxycarbonyl-1,2-dihydrospiro(naphthalene-2,1-cyclopentane) with methyl, phenyl, and benzyl isothiocyanates and cyclization of the obtained thioureas. Condensation of the products with various halides gave 2,3-substituted 4-oxo-3,4,5,6-tetrahydrospiro(benzo[h]quinazoline-5,1-cyclopentanes). The reaction of 4-oxo-2-thioxo-1,2,3,4,5,6-hexahydrospiro(benzo[h]quinazoline-5,1-cyclopentane) with 1,2-dibromoethane and 1,3-dibromopropane led to 6-oxo-7,8-dihydrospiro(benzo[h]thiazolidino[2,3-b]quinazoline-7,1-cyclopentane) and 7-oxo-8,9-dihydrospiro(benzo[h]-perhydrothiazino[2,3-b]quinazoline-8,1-cyclopentane) respectively.A. Intramolecular cyclization of N-phthalyl-β-aryl-β-alanine phenylhydrazide

The reaction of N-phthalyl--aryl--alanine N-methyl-N-phenylhydrazides with phosphorus oxychloride (at80 °C) is accompanied by further transformations of the initially formed 2-aminoindole derivatives and leads to isoindolo[1,22,3]-pyrimido[5,6-b]indole derivatives. Intermediate 2-aminoindoles were isolated at lower reaction temperatures. The hydrolysis of the isoindolo[1,22,3]pyrimido [5, 6-b]indole derivatives was studied. Also a few unknown compounds could be isolated and their structure elucidated; 9-methoxy-7-(3.4.5-trimethoxyphenyl)-8H-1.3-dioxolo[4.5-g] [1]-benzopyran-8-on (=5.3.4.5-tetramethoxy-6.7-methylenedioxyisoflavone (III A); 9-methoxy-7-(3.4-dimethoxyphenyl)-8H-1.3-dioxolo[4.5-g] [1]-benzopyran-8-on (=5.3.4-trimethoxy-6.7-methylenedioxyisoflavone(III B); 9-hydroxy-7-(p-hydroxyphenyl)-8H-1.3-dioxolo[4.5-g] [1]-benzopyran-8-on (=5.4-dihydroxy-6.7-methylenedioxyiosoflavone (IX); 5.7-dihydroxy-3-(3-hydroxy-4-methoxyphenyl)-6-methoxy-4H-1-benzopyran-4-on (=5.7.3-trihydroxy-6.4-dimethoxyisoflavone (XI B); 5.7-dihydroxy-3-(4-hydroxy-3-methoxyphenyl)-6-methoxy-4H-1-benzopyran-4-on (=5.7.4-trihydroxy-6.3-dimethoxyisoflavone (XI C).The structure determination was effected by combined spectroscopical and chemical methods.

Preparation of 8-(5-oxoindeno [3,2-b]-4-pyridyl)-1-naphthoic acids

The condensation of 1-(1,3-indanedion-2-ylidene)-2-acenaphthenone with imines of -dicarbonyl compounds gave 2,5-dioxospiro 1,4,acenaphthene-1,4-dihydroindeno-[3,2-b]pyridines. Oxidation of the pyridines with air oxygen at room temperature without catalysts brings about cleavage of the C-C bond to give 8-(5-oxoindeno[3,2-b]-4-pyridyl)-1-naphthoic acids.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 2, pp. 210–212, February, 1972.     

Thin-layer chromatographic separation of some closely related olefinic benzo- and naphtofurans

Summary A method is reported for the separation of 16 closely related 2-(1, 2-diphenylvinyl)-3-phenylbenzofuran and 2-(1, 2-diphenylvinyl)-1-phenylnaphtho[2,1-b] furan derivatives on thin-layers of silica gel-G.     

New monomers for the synthesis of heat-stable polyheterocycles

To obtain benzodioxazole analogs of the polypyromellitimides and polymers with alternating pyromellitimide and benzodioxazole links, we have synthesized 2,6-di(paminophyl)benzo[1,2-d5,4-d]dioxazole (I), 2,6-di[p-(p-aminophenoxy)phenyl]benzo-[1,2-d5,4-d]dioxazole (II), N,N-di(p-chloroformylphenyl)pyromellitdiimide (III) and N,N-di(p-chloroformylphenyl)-2,5-dimethoxycarbonyl terephthalamide (IV). 9-(2′,5′-Dimethyl-4′-pyridyl)fluorene in syntheses of fluorospirodihydrofuropyridine and pyridofluoranthene

1,2,5-Trimethyl-4-(9-fluorenylidene)piperidine, which is formed by condensation of fluorene with 1,2,5-trimethyl-4-piperidone, is converted catalytically to 9(2,5-dimethyl-4-pyridyl)fluorene, from which 2-methylpyrido[4,5-a]fluoranthene and its demethylated analog were obtained by catalytic dehydrocyclization. Oxidation of 9-(2,5-dimethy1-4-pyridyl)fluorene gave 9-(2,5-dimethyl-4-pyridyl)9-fluorenol and fluorene-9-spiro-4-(6-oxo-2-carboxypyrido[4,5-c]-4-6-dihydrofuran). 6-Methyl-2-phenyl-7-(9-fluorenyl)indolizine was synthesized by the Chichibabin method.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 8, pp. 1087–1090, August, 1978.     

Condensed heterocycles with a thiazole ring. 13. Azinothiazoloquinoxalinium salts

Synthesis and photochromic properties of spiroanthrooxazine

1,3,3-Trimethylspiro[indoline-2,3-3H-anthraceno[2,1-b][1,4]oxazine] was synthesized. The compound has photochromic properties in solutions and in polymeric matrices.
