This work contains the results of studies on the influence of newly synthesized lysosomotropic substances (lysosomotropes) on human erythrocytes. Six homologous series of the compounds differing in the alkyl chain length and counterions were studied. They were found to hemolyse erythrocytes and to change their osmotic resistance. The observed hemolytic effects were dependent both on the compounds structure (polar head dimension and alkyl chain length of compound) and its form (the kind of the counterion). In parallel, the influence of lysosomotropes on fluidity of the erythrocyte membrane was studied. Three different fluorescent probes were used; 1,6-diphenyl-1,3,5-hexatriene (DPH), 1-(4-trimethylammoniumphenyl)-6-phenyl-1,3,5-hexatriene, p-toluenesulfonate (TMA-DPH) and 6-dodecanoyl-2-dimethylaminonaphthalene (laurdan). Their anisotropy (DPH and TMA-DPH) or general polarization (laurdan) values after incorporation into ghost erythrocyte membranes were measured. The results obtained show that fluidity changes accompanied the effects observed in hemolytic experiments both quantitatively and qualitatively.  相似文献

In this work, erythrocytes from carp were used as a nucleated cell model to test the hypothesis that the phthalocyanines (zinc - ZnPc and chloroaluminium -AlClPc) enhance ultrasonically induced damage in vitro. In order to confirm and complete our earlier investigation, the influence of ultrasound (US) and phthalocyanines (Pcs) on unresearched cellular components, was studied. Red blood cells were exposed to 1 MHz continuous ultrasound wave (0.61 and/or 2.44 W/cm2) in the presence or absence of phthalocyanines (3 μM). To identify target cell damage, we studied hemolysis, membrane fluidity and morphology of erythrocytes. To demonstrate the changes in the fluidity of plasma membrane we used the spectrofluorimetric methods using two fluorescence probes: 1-[4-(trimethylamino)phenyl]-6-phenyl-1,3,5,-hexatriene (TMA-DPH) and 1,6-diphenyl-1,3,5-hexatriene (DPH). The effect of US and Pcs on nucleated erythrocytes morphology was estimated on the basis of microscopic observation.The enhancement of ultrasonically induced membrane damage by both phthalocyanines was observed in case of hemolysis, and membrane surface fluidity, in comparison to ultrasound. The authors also observed changes in the morphology of erythrocytes. The obtained results support the hypothesis that the Pcs enhance ultrasonically induced cell damage in vitro.Furthermore, the influence of ultrasound on phthalocyanines (Pcs) in medium and in cells was tested. The authors observed changes in the phthalocyanines absorption spectra in the medium and the increase in the intensity of phthalocyanines fluorescence in the cells. These data can suggest changes in the structure of phthalocyanines after ultrasound action.  相似文献

荧光偏振法研究脉冲电场对酿酒酵母细胞膜流动性影响   总被引:1,自引:1,他引:1  
以DPH(1,6-二苯基-1,3,5-己三烯)为荧光探剂,采用荧光偏振法探讨了脉冲电场(0~25 kV·cm-1,0~266 ms)对酿酒酵母细胞膜流动性影响。经5 kV·cm-1电场处理后,酿酒酵母细胞膜的流动性显著减小,并且随电场强度和处理时间的增加而减小;通过平板计数法和紫外分光光度计法分别检测了脉冲电场对酿酒酵母细胞存活对数及膜通透性影响。结果显示,5 kV·cm-1虽然只能使少量的酵母致死,却能使酵母细胞膜的通透性显著增加,膜流动性显著降低。并且细胞的存活率随电场强度增大而减小,细胞膜的通透性随电场强度增大而增大。这表明细胞膜的流动性降低与细胞膜的通透性升高成正相关,与细胞的存活率成负相关。由此推测脉冲电场在对酿酒酵母灭菌过程中,细胞膜是其作用的一个关键位点,膜流动性减小,细胞膜通透性增强,是细胞死亡的主要原因。  相似文献

荧光偏振光谱法探测光动力过程中癌细胞膜的流动性   总被引:2,自引:0,他引:2  
以DPH作荧光探剂,用荧光偏振光谱法研究了金属酞菁配合物苯硫基钛菁锌(C56H32N8S4Zn)在光动力过程中,不同光照时间对乳腺癌细胞膜流动性的影响。实验结果表明,经光照激发光敏剂金属酞菁后,荧光标记团的偏振度增大,癌细胞膜流动性降低,微黏度升高。说明这是光动力治疗的疗效所在。用酶联免疫检测(MTT法)不同光照时间对细胞存活的影响,结果显示癌细胞最低存活率与膜流动性降低成正相关。这表明光动力过程在一个相近的程度影响癌细胞膜的流动性和增殖,提示光动力过程可能通过影响细胞膜流动性引起细胞增殖变化,即细胞膜是光动力作用的一个位点。  相似文献

Lipid bilayers have been largely used as model systems for biological membranes. Hence, their structures, and alterations caused on them by biological active molecules, have been the subject of many studies. Accordingly, fluorescent probes incorporated into lipid bilayers have been extensively used for characterizing lipid bilayer fluidity and/or polarity. However, for the proper analysis of the alterations undergone by a membrane, a comprehensive knowledge of the fluorescent properties of the probe is fundamental. Therefore, the present work compares fluorescent properties of a relative new fluorescent membrane probe, 2-amino-N-hexadecyl-benzamide (Ahba), with the largely used probe 6-dodecanoyl-N,N-dimethyl-2-naphthylamine (Laurdan), using both static and time resolved fluorescence. Both Ahba and Laurdan have the fluorescent moiety close to the bilayer surface; Ahba has a rather small fluorescent moiety, which was shown to be very sensitive to the bilayer surface pH. The main goal was to point out the fluorescent properties of each probe that are most sensitive to structural alterations on a lipid bilayer. The two probes were incorporated into bilayers of the well-studied zwitterionic lipid dimyristoyl phosphatidylcholine (DMPC), which exhibits a gel-fluid transition around 23 °C. The system was monitored between 5 and 50 °C, hence allowing the study of the two different lipid structures, the gel and fluid bilayer phases, and the transition between them. As it is known, the fluorescent emission spectrum of Laurdan is highly sensitive to the bilayer gel-fluid transition, whereas the Ahba fluorescence spectrum was found to be insensitive to changes in bilayer structure and polarity, which are known to happen at the gel-fluid transition. However, both probes monitor the bilayer gel-fluid transition through fluorescence anisotropy measurements. With time-resolved fluorescence, it was possible to show that bilayer structural variations can be monitored by Laurdan excited state lifetimes changes, whereas Ahba lifetimes were found to be insensitive to bilayer structural modifications. Through anisotropy time decay measurements, both probes could monitor structural bilayer changes, but the limiting anisotropy was found to be a better parameter than the rotational correlation time. It is interesting to have in mind that the relatively small fluorophore of Ahba (o-Abz) could possibly be bound to a phospholipid hydrocarbon chain, not disturbing much the bilayer packing and being a sensitive probe for the bilayer core.  相似文献

利用氮氧自由基自旋标记物 CAT-16 和 5-DSA 较为详尽地探讨了两类含羟基化合物 - 羟胺衍生物与苯酚取代物对红细胞膜流动性变化的影响. 实验结果表明:当非双亲性的小分子苯酚取代物插入红细胞膜内,膜的整体有序性降低和流动性增加;羟胺衍生物可作为促氧化剂诱导产生过氧化自由基,促使膜内磷脂间发生化学偶联,从而降低生物膜的内部流动性. 在红细胞膜的表面,标记物分子运动基本不受偶联影响,综合效应来自小分子插入导致的膜流动性增加.  相似文献

应用1,6-二苯基-1,3,5-已三烯(DPH)为荧光探针,超氧阴离子自由基和羟基自由基致鼠红细胞膜氧化损伤为模型,研究了茶叶花粉黄酮对鼠红细胞膜氧化损伤的影响.结果表明,超氧阴离子自由基和羟基自由基均能引起鼠红细胞膜脂质过氧化反应,脂质过氧化产物丙二醛含量显著升高,膜脂流动性下降.将鼠红细胞膜预先用茶叶花粉黄酮处理后,膜脂的MDA含量明显下降,呈现剂量与效应关系,膜脂流动性显著提高,表明茶叶花粉黄酮对超氧阴离子自由基和羟基自由基引起的鼠红细胞膜的氧化损伤有保护作用.  相似文献

Membrane fluidity measurements based on excimer formation of pyrene and pyrene derivatives as a measure of lateral diffusion yield a decreased fluidity in the presence of proteins [1,2]. It was the aim of our study to investigate whether the reduced excimer formation is due to a rigidifying effect of proteins on the whole membrane or if the fluorophore mobility is hindered mainly in the immediate protein environment. Resonance energy transfer in microsomal membranes between intrinsic tryptophan residues and pyrene was used to study the excimer formation rate in the vicinity of proteins. The excimer-to-monomer fluorescence ratio after excitation via resonance energy transfer decreased compared to that observed for the direct excitation. The results suggest that, because of the reduced fluidity in the neighborhood of proteins, pyrene and pyrenedodecanoic acid do not diffuse homogeneously in the membrane plane.  相似文献

争光霉素A6是争光霉素的组分之一。它能够选择性影响小鼠腹水肝癌细胞膜的流动性,而对正常小鼠的肝和骨髓细胞膜没有影响。A6可使癌变细胞膜类脂由液晶态到液态的相变点降低2℃。当A6结合持续过热时,可产生协同作用,使癌变细胞膜流动性的速率发生明显的改变。  相似文献

A method by which it is possible to characterize the membranes of biological samples on the basis of the EPR spectral lineshape simulation of membrane-dissolved nitroxide spin probes is described. The presented simulation procedure allows the determination of the heterogeneous structure of biological membranes and fluidity characteristics of individual membrane domains. The method can deal with isotropic and anisotropic orientations of nitroxides introduced into the biological samples described by restricted fast motion with a correlation time between 0.01 and 10 ns. The linewidths of the Lorentzian lineshapes are calculated in a restricted fast-motion approximation. In the special case of samples with high concentrations of nitroxides or in the presence of paramagnetic ions, the lineshapes are calculated directly from the exchange-coupled Bloch equations. The parameters describing ordering, relaxation, polarity, and the portions of the individual spectral components are extracted by optimizing the simulated spectra to the experimental spectrum with either a Simplex or a Monte Carlo algorithm. To improve the algorithm's efficiency, a new way of characterizing the goodness of fits is introduced. The new criterion is based on the standard least-squares function, but with special weighting of the partial sums. Its benefits are confirmed with membrane spectral simulation. Two classes of examples-simulation and optimizations of synthetic spectra to evaluate the accuracy of the optimization algorithms and simulation and optimization of EPR spectra of nitroxides in liposome suspensions in the presence of a broadening agent and in human leukocytes are shown.  相似文献

The hemolytic toxicity of tributyllead (TBL) and triphenyllead (TPhL) chlorides and its prevention by dithiotreitol (DTT), diethylenetriaminepentamethylenephosphonic acid pentasodium (PMP) and sodium disulfide (Na2S) was studied. It was found that both TBL and TPhL efficiently hemolyzed pig erythrocytes when used in micromolar concentrations; tributyllead chloride being about twice more efficient than triphenyllead chloride. The hemolytic efficiency of these compounds was blocked by PMP, DTT and Na2S in a concentration-dependent manner. However, significant differences in anti-hemolytic efficiency of these compounds were found. Namely, DTT and Na2S were very efficiently protecting erythrocytes against the action of organoleads, while the PMP protection was weak. Also, differences between DTT and Na2S protective efficiency were found. They more efficiently prevented erythrocyte hemolysis by TPhL than by TBL. Moreover, erythrocytes were better protected against the action of TBL by Na2S than by DTT. Such differentiation may be connected with possible differences in localization of the organolead compounds and protective agents in the erythrocyte membrane. To check these possibilities a series of experiments was performed using the fluorescence technique and various fluorimetric probes. These measurements enabled to determine fluidity changes induced in erythrocyte membranes by the organoleads and the protective compounds and to formulate some remarks concerning the differences in the mechanism of interaction of the organoleads with these membranes.  相似文献

As an alternative to conventional sterilization methods the plasma ions created from high voltage produce radicals which have a direct impact on microorganisms in water. Cell membrane fluidity is an important factor in survival as the fluidity allows for essential cell functions in which pH and fatty acids play a major role. This study investigated the effect of plasma application upon pH and fatty acid profiles of Escherichia coli and Staphylococcus aureus. Results showed that plasma sterilization is not predominantly a result of changes to pH levels or fatty acid contents in the bacterial cell membrane.  相似文献

The dynamic behaviour of three hydrophilic probes (two dyes and one fluorescently-labelled protein) inserted in the water layers of lyotropic lamellar phases has been studied by confocal fluorescence recovery experiments. Two different, ionic (AOT/NaCl/ H(2)O and non-ionic ( C(12)E(5) /hexanol/ H(2)O host systems were studied. The confinement effect has been carefully monitored using the swelling properties of the lamellar phases. In all cases, we measure the evolution of the probe diffusion coefficient in the layer plane D ( perpendicular) versus the separation between the membranes d(w). Depending on the composition of the lamellar phase, this distance can be continuously adjusted from 500A to about 20A. For all systems, we observe a first regime, called dilute regime, where the diffusion coefficient decreases almost linearly with 1/d (w) . In this regime, the Faxén theory for the friction coefficient of a spherical particle symmetrically dragged between two rigid walls can largely explain our results. More unexpectedly, when the membranes are non-ionic, and also quite flexible ( C(12)E(5) /hexanol in water), we observe the existence of a second, concentrated (or confined) regime, where the diffusion coefficient is nearly constant and different from zero for membrane separations smaller than the particle size. This new regime can be heuristically explained by simple arguments taking into account the membrane fluidity.  相似文献

The paper reports the results of our study on the dynamics of model phospholipid membranes studied by 31P NMR spectroscopy. The 31P NMR spectra of multilamellar vesicles in temperatures below the main phase transition of PC are reported. The 31P NMR spectra revealed changes caused by an increase of the membrane fluidity when polysialic acid (polySia) was applied as a modifying agent. The presence of polySia in the external environment of the phospholipid vesicles changes the motional freedom in the region of phosphate group of lipids. Increase of polysialic acid concentration changes structural properties of a membrane by increasing its fluidity.  相似文献

本文使用TEMPO和脂肪酸氮氧自由基自旋标记脂质体和正常的与癌变的鼠肝细胞膜,研究了Vc对膜流动性的影响,探讨有关Vc抗肿瘤作用的机理。实验表明:Vc可以影响膜类脂和膜蛋白的运动,提高癌变细胞膜的流动性,并随Vc浓度的增加其作用速度加快,但对正常肝细胞膜流动性则无明显影响。此外,在一定浓度范围内,Vc对正常的与癌变的肝细胞膜的掺入能力有显著差异。  相似文献

本文用圆二色光谱(CD)研究了交变应力对烟草细胞膜蛋白结构的影响 采用本实验室研制的强声波发生装置来产生交变应力场,研究了不同强度和频率的交变应力场作用后烟草细胞膜蛋白结构的变化 研究结果表明:细胞膜蛋白结构变化与应力的频率和强度密切相关,一定频率和强度范围内的交变应力能使得烟草细胞膜蛋白的二级结构发生明显的变化,表现为α螺旋的增加和 β转角的减少 这反映了细胞膜蛋白和磷脂相互作用的增强和细胞膜流动性的增加,进而为细胞的生长和分裂提供有利的条件 从细胞和分子水平研究交变应力对植物生长、发育的影响及其作用机制是一个很有意义的尝试.  相似文献

Polyethyleneimine (PEI) is one of the very efficient nonviral vectors being developed and tested for artificial gene transfer into target cells. One of its serious limitations is the significant cytotoxicity of the large amounts of free PEI in the mixtures of DNA and PEI used for transfection. To further investigate the cellular effects of free PEI, we have analyzed the PEI-induced alterations of various cell parameters such as membrane heterogeneity and fluidity, cytoplasmic pH, and plasma membrane potential in a variety of cells such as Swiss 3T3 fibroblast, Chinese hamster ovary, insect cells SF9, plant cell line BY2, and Saccharomyces cerevisae. Fluorescence probes such as Nile red, SNARF-1, and cyanine dye DiSC2(3), coupled with the technique of picosecond time-resolved fluorescence microscopy, were used in estimating the above-mentioned cell parameters. It was found that the cell membranes were largely unperturbed by PEI. However, the cytoplasmic pH showed an increase of 0.1–0.4 units when the cells were treated with PEI. The plasma membrane potential was found to be depolarized in S. cerevisae and Swiss 3T3 cells. These results suggest that the cytotoxic effects of PEI may partly originate from inhibition of regulation of cytoplasmic pH and plasma membrane potential. Further, it is proposed that the resultant cell alterations favors the transfection process.  相似文献
