Traditionally, the raw materials for beer production are barley, hops, water, and yeast, but most brewers use also different adjuncts. During the alcoholic fermentation, the contribution of aroma compounds from other ingredients to the final beer flavor depends on the wort composition, on the yeast strain, and mainly on the process conditions. In this context, banana can also be a raw material favorable to alcoholic fermentation being rich in carbohydrates and minerals and providing low acidity. In this work, the objective was to evaluate the performance of wort adjusted with banana juice in different concentrations. For this, static fermentations were conducted at 15 °C at pilot scale (140 L of medium). The addition of banana that changed the concentration of all-malt wort from 10 °P to 12 and 15 °P were evaluated (°P is the weight of the extract or the sugar equivalent in 100 g solution, at 20 °C). The results showed an increase in ethanol production, with approximately 0.4 g/g ethanol yield and 0.6 g/L h volumetric productivity after 84 h of processing when concentrated wort was used. Thus, it was concluded that banana can be used as an adjunct in brewing methods, helping in the development of new products as well as in obtaining concentrated worts.  相似文献

Trifloxystrobin and tebuconazole are used for control of Sigatoka leaf spot disease of banana. This study was conducted to evaluate residue persistence of the fungicides in/on banana fruit, other edible parts and soil after spray application of the combination formulation, Nativo 75 WG, at the standard dose, 87.5 + 175 and double dose, 175 + 350 g a.i. ha?1. The fungicides were extracted from banana and soil with acetone, partitioned into dichloromethane and cleaned-up using activated charcoal for trifloxystrobin and primary/secondary amine (PSA) for tebuconazole samples. The limit of quantification of the method was 0.05 mg kg?1 for both fungicides. Initial residues of trifloxystrobin were 0.444 and 0.552 mg kg?1 in/on banana with peel (whole fruit), which reached <0.05 and 0.065 mg kg?1 after 30 days from treatment at the standard and double doses, respectively. Tebuconazole residues were 0.636 and 960 mg kg?1 initially and reduced to 0.066 and 0.101 mg kg?1 after 30 days. Trifloxystrobin and tebuconazole degraded with the half-life of about 11 days. Trifloxystrobin or its metabolite was not detected in the fruit pulp. Tebuconazole being systemic in nature moved to the fruit pulp which was highest on the 3rd day (0.103 and 0.147 mg kg?1) and remained for 15 days. Matured banana fruit, flower, pseudostem and field soil were free from fungicide residues. For consumption of raw banana 43 days pre-harvest interval (PHI) is required after treatment of the combination formulation. Therefore application of the fungicides towards maturity stage of the fruits may be avoided.  相似文献

In the present work, cassava leaves were treated with 0.5 kg ammonia/kg dry matter at 78 °C and 30% moisture content in a 2-kg reactor. Protein extraction was carried out with a calcium hydroxide solution (pH 10) for 30 min at several temperatures (30 °C, 45 °C, 60 °C, 75 °C, and 90 °C) and solid/liquid ratios (1:10 and 1:15) in a thermostatized bath. Soluble protein content of the extracts was determined by Lowry’s method. Dry substrate concentrations of 5%, 7.5%, and 10% and enzyme doses of 2 and 5 IU/g dry matter were used for the enzymatic hydrolysis in an orbital incubator at 50 °C and 100 rpm. Both cellulase and xylanase were used. Reducing sugars produced were determined with the dinitrosalicylic acid method. The highest protein extraction yield for the ammonia-treated leaves was 29.10%, which was 50% higher than with the untreated leaves (20%), and was obtained at 90 °C with a 1:10 solid/liquid ratio. The concentrate had a protein content of 36.35% and the amino acid profile was suitable for swine and poultry. The highest sugar yield was 54.72% with respect to theoretical and was obtained with 5% solids and an enzyme dose of 5 IU/g dry matter. This yield was 3.4 times higher than the yield of the untreated leaves (16.13%). These results indicate that cassava leaves have a great potential for animal feeding and ethanol production. Both protein extraction and sugar yields may be enhanced by optimizing the ammonia treatment.  相似文献

An extraction method based on matrix solid-phase dispersion was developed to determine carbofuran, pyrimethanil and tetraconazole in banana using gas chromatography–mass spectrometry. The best results were obtained using 2.0 g of banana, 1.0 g of silica as dispersant sorbent and n-hexane:ethyl acetate (1:4, v/v) as eluting solvent. The method was validated using banana samples fortified with pesticides at different concentration levels (0.05–2.0 mg kg?1). Average recoveries (four replicates) ranged from 68 to 111%, with relative standard deviations between 6.6 and 20.5%. Detection and quantification limits for banana ranged from 0.02 to 0.05 and 0.05 to 0.10 mg kg?1, respectively.  相似文献

Chinese purple yam is famous for therapeutic and nutritional values in lowering blood glucose, blood pressure and even preventing and treating cardiovascular diseases. However, traditional extraction techniques for the functional polyphenolic compounds mostly utilise unfriendly organic solvent and easily cause degradation of polyphenols. In this study, a novel ionic liquid-based ultrasound-assisted extraction (IL-UAE) technique was utilised to obtain polyphenols. The optimal extraction condition included: solid–liquid ratio (0.05 g/mL), ionic liquid concentration (1.05 M), extraction temperature (67°C) and ultrasonic time (37 min). The maximum free radical scavenging activity (86.21%) and total antioxidant activity (2.21 mM) were achieved and matched well with the predicted values, superior to that of the extract from regular ethanol-based UAE. IL-UAE could be a rapid and green technique for efficient extraction of polyphenols from purple yam with low solvent consumption and few structural deterioration, exhibiting application potential in food and pharmaceutical industries.  相似文献

Forest biomass is a promising resource for future biofuels and bioproducts. Pre-pulping extraction of hemicellulose by alkaline (Green Liquor) pretreatment produces a neutral-pH extract containing hemicellulose-derived oligomers. A near-term option for use of this extract is to hydrolyze the oligomers to fermentable monomer sugars. Chips of mixed northern hardwoods were cooked in a rocking digester at 160 °C for 110 min in Green Liquor at a concentration of 3% Na2O equivalent salts on dry wood. The mass of wood extracted into the Green Liquor extract was approximately 11.4% of the debarked wood mass, which resulted in a dilute solution of oligomeric hemicelluloses sugars. The concentration of the extract was increased through partial evaporation prior to hydrolysis. Dilute sulfuric acid hydrolysis was applied at conditions ranging from 100 to 160 °C, 2% to 6% (w/v) H2SO4, and 2- to 258-min residence time. The maximum fermentable sugar concentration achieved from evaporated extract was 10.7 g/L, representing 90.7% of the maximum possible yield. Application of the biomass pretreatment severity function to the hydrolysis results proved to offer a relatively poor prediction of temperature and reaction time interaction. The combined severity function, which incorporates reaction time, temperature, and acid concentration, did prove to provide a useful means of trading off the combined effects of these three variables on total sugar yields.  相似文献

Aqueous extracts of Neolamarckia cadamba fruits prepared at different maturity stages were used for the analysis of various phytochemicals, and their antioxidant and antibacterial activities were determined. Ripe fruit extract had highest phenolics (3.14 mM GAE/ g fruit extract) with caffeic acid, tannic acid, syringic acid and quercetin as major phenolic compounds. The ripe fruit extract showed lowest IC50 values in DPPH radical scavenging assay (231.33 μg fruit extract/ mL), and highest ABTS radical scavenging activity (111.18 μM TEAC/g). Immature fruit extract showed lowest minimum inhibitory concentration against tested bacteria, and the antibacterial activity was probably due to membrane permeation, as was evident by leakage of genetic material and reduction in propidium iodide uptake by bacterium; and by inhibition of sugar and amino acid uptake. The appreciable amount of phenolic compounds and biological activities in the aqueous extracts of N. cadamba fruits suggests it's potential application as natural preservative.  相似文献

Generally the metallurgical industries generate huge amounts of harmful environmental wastes, which may be solids, liquids, or gases in their nature. The present study aims to recovery sulfuric acid (227 g/l) from wastes generated during hydrometallurgical digestion of titanium ores with sulfuric acid for TiO2 production. For this purpose Alamine 336/kerosene solvent was used. Various extraction parameters as Alamine 336 concentration, shaking time, type of diluent, and O/A phase ratio were studied and optimized. Under the optimum conditions, Mc-Cabe Thiele diagram results indicated that, after four extraction stages the acid concentration in aqueous phase was reduced from the initial value of 227 g/l to about 17 g/l. Stripping of the loaded sulfuric acid from the organic phase was done with warm water (60°C). Stripping parameters as water temperature, stripping time, and A/O phase ratio were studied. Under the stripping the loaded acid concentration in organic phase was reduced from 210 to 6 g/l which matched theoretically by McCabe Thiele diagram.  相似文献

Optimization of cultural conditions for enhanced cellulase production by Aspergillus niger NS-2 were studied under solid-state fermentation. Significant increase in yields (CMCase 463.9?±?20.1 U/g, FPase 101.1?±?3.5 U/g and β-glucosidase 99?±?4.0 U/g) were obtained under optimized conditions. Effect of different nutritional parameters was studied to induce the maximum production of cellulase complex. Scale-up studies for enzyme production process were carried out. Characterization studies showed that enzymes produced by A. niger NS-2 were highly temperature- and pH stable. At 50 °C, the half life for CMCase, FPase, β-glucosidase were approximately 240 h. Cellulases from A. niger NS-2 were stable at 35 °C for 24 h over a broader pH range of 3.0–9.0. We examined the feasibility of using steam pretreatment to increase the saccharification yields from various lignocellulosic residues for sugar release which can potentially be used in bioethanol production. Saccharification of pretreated dry potato peels, carrot peels, composite waste mixture, orange peels, onion peels, banana peels, pineapple peels by crude enzyme extract from A. niger NS-2, resulted in very high cellulose conversion efficiencies of 92–98 %.  相似文献

Gamma irradiation in combination with hot water dipping treatment was tested for maintaining storage quality and extending the shelf life of peach fruit. The matured peaches fruits (local variety no, Tex-A-6-69) were first dipped in hot water having temperature of 0, 40 and 60 °C for 60 seconds and they were exposed to 0.5 and 1.0 kGy doses of gamma radiation and stored in paper cartons under ambient (temperature 25 ± 2 °C, RH 70 %) conditions for their physico-chemical evaluations on weekly basis. The combine effect of hot water dipping treatment and irradiation for peach was study for the first time in Pakistan. The parameters studied included % weight loss, % ash content, % moisture content, total soluble solids (TSS), titratable acidity and ascorbic acid. The sensory indices studied were size, shape, color and overall acceptability. The sensory evaluation values at 0 days for the controlled peach sample were recorded for comparison. Statistical analysis was performed to find determinates of irradiation alone and in combination with hot water dipping techniques on peaches. Studies revealed that irradiation treatment in combination with hot water treatments significantly (LSD at 5 %) maintained the storage quality of peach fruit under ambient conditions. Data obtained for weight loss, moisture, shape and overall acceptability showed significant results while non-significant values were recorded for ash, TSS and size. Overall, consumers rated the acceptability of treated peaches higher than untreated peaches. This particular type of research also helps in improving one’s country export quality of fruits.  相似文献

Very high gravity (VHG) wheat mashes containing more than 300 g of dissolved solids per liter were prepared and fermented with active dry yeast at 20, 25, 30, and 35°C with and without yeast extract as nutrient supplement. At 20°C, mashes with 38% (w/v) dissolved solids end-fermented without any nutrient supplementation and maximum ethanol yields of 23.8% (v/v) were obtained. With increasing temperatures, the sugar consumption decreased. Addition of yeast extract stimulated the rate of fermentation at all temperatures, but did not increase the total amount of sugar consumed. The stimulatory effect of yeast extract on cell multiplication decreased with increasing sugar concentration, and virtually no difference in cell number was observed between yeast extract-supplemented and unsupplemented mashes at sugar concentrations above 33% (w/v). The fermentative capacity of the yeast (expressed as maximum specific rate of sugar consumption) remained the same at all sugar concentrations in unsupplemented mashes, but decreased in yeast extract-supplemented mashes at sugar concentrations below 33% (w/v). When the sugar concentration was above 33% sugar (w/v), the fermentative capacity in yeast extract-supplemented mashes was greater than that observed in unsupplemented samples.  相似文献

Orange fruit is one of the most popular types of fruit in the world, and its juice is the main product of its processing. This study aimed to evaluate a simple, fast, and eco-friendly methodology, atmospheric solids analysis probe mass spectrometry (ASAP-MS), to assess the chemical profile of four oranges varieties (Valencia, Folha Murcha, Pera, and Iapar). The oranges' varieties were evaluated for the physicochemical composition (extraction yield, pH, total titratable acidity, total soluble solids [TSS], °Brix), ratio (TSS/TA), and bioactive compounds (ASAP-MS analysis). The characterization of oranges resulted in great values of oranges yield according to the varieties (44.00–48.10% [w/w], adequate and characteristic acidity [0.73–1.35%], soluble solids content (10.24–13.80°Brix), pH (3.30–3.96), and ratio (7.59–19.90) level for this fruit. This powerful method showed that all analysis procedures were simple, fast, and easy because there is no need to prepare the sample and the analysis time lasted 2 min. Besides, results obtained exhibited a vast array of chemical groups. Principal component analysis (PCA) defined and distinguished the varieties of the orange. Therefore, ASAP-MS and PCA showed that they are very attractive candidates for routine analysis to monitor the varieties of the orange with its pronounced advantages, besides being contributing to the environment because it does not use any quantities of organic solvents. Coffee is one of the most important agricultural products in Brazil. More than 50 % of the coffee fruit is not used for the production of commercial green coffee and is therefore discarded, usually ending up in the environment. The goal of this work was to select an efficient process for obtaining coffee pulp extract and to evaluate the use of this extract in bioethanol production. The effects of heat treatment and trituration on the yield and composition of the extract were investigated by measuring the amounts of reducing sugars, starch, pectin, and phenolic compounds. The extraction process was most efficient at room temperature using grinding followed by pressing. Five different fermentation media were tested: sugarcane juice or molasses diluted with water or with coffee pulp extract and a medium with only coffee pulp extract. Batch fermentations were carried out at 30 °C for 24 h, and samples were taken to obtain measurements of the total reducing sugars, cell count, and ethanol concentration. The addition of coffee pulp extract did not influence the fermentation or yeast viability, and it can thus be mixed with sugarcane juice or molasses for the production of bioethanol, with a yield of approximately 70 g/L.  相似文献

In this study, biosurfactant-producing bacteria was evaluated for biosurfactant production by using banana peel as a sole carbon source. From the 71 strains screened, Halobacteriaceae archaeon AS65 produced the highest biosurfactant activity. The highest biosurfactant production (5.30 g/l) was obtained when the cells were grown on a minimal salt medium containing 35 % (w/v) banana peel and 1 g/l commercial monosodium glutamate at 30 °C and 200 rpm after 54 h of cultivation. The biosurfactant obtained by extraction with ethyl acetate showed high surface tension reduction (25.5 mN/m), a small critical micelle concentration value (10 mg/l), thermal and pH stability with respect to surface tension reduction and emulsification activity, and a high level of salt tolerance. The biosurfactant obtained was confirmed as a lipopeptide by using a biochemical test FT-IR, NMR, and mass spectrometry. The crude biosurfactant showed a broad spectrum of antimicrobial activity and had the ability to emulsify oil, enhance PAHs solubility, and oil bioremediation.  相似文献

Hydrogen cyanamide is a plant-growth regulator used on fruit crops, and has a high toxicity. In this study, a simple and sensitive method for detecting hydrogen cyanamide in fruit was developed. In the proposed method, cyanamide was extracted with water from homogenized fruit samples (grape, kiwi, and peach). The extract was purified through twice liquid–liquid extraction. Then, the purified extract was derivatized with 6-aminoquinolyl-N-hydroxysuccinimidyl carbamate. Finally, the cyanamide derivative was analyzed using HPLC with fluorescence detection. This method was validated at 2, 0.2, and 0.01 mg kg−1 cyanamide. The recovery rates were between 87.1 and 96.6 %, and the intraday and interday reproducibilities were within 1.8 and 2.2 %, respectively. The limits of detection and quantification were 2.4 and 8 μg kg−1 cyanamide, respectively. This method was applied to explore the residue degradation of cyanamide in the field. The results show that the half-life of cyanamide residues on grapes was <2.0 days. Compared with the previous methods, this simple method is sensitive and can be applied for detecting cyanamide in a wide range of fruits.

Cachaça is the typical and genuine denomination for the sugar cane beverage produced in Brazil. It has an alcoholic content between 38 and 48 % by volume, at 20 °C, and is obtained from the distillation of fermented sugar cane juice with the possible addition of up to 6 g/L of sugar, expressed by sucrose. This paper aims to outline the approach taken for the certification of the mass fraction of individual alcohols and their associated uncertainties in a cachaça certified reference material (CRM) produced by the Organic Analysis Laboratory of the Chemical Metrology Division of Inmetro. The requirements of ISO Guide 31, ISO Guide 34 and ISO Guide 35 were followed for the certification. This included the assessment of the degree of homogeneity of the material, the short-term stability study to determine the transport conditions, the long-term stability study to establish the shelf life and storage conditions of the CRM and the characterization process. The certified values were determined by two analytical techniques: gas chromatography coupled to mass spectrometry and gas chromatography with flame ionization detection. In order to support the routine testing of cachaça, in helping analytical laboratories validate and standardize their methods, the certified reference material (CRM 02.1/10.003b) was developed and certified for the mass fractions of methanol (9.18 mg/100 g ± 0.82 mg/100 g), 2-butanol (4.15 mg/100 g ± 0.40 mg/100 g), 1-butanol (1.30 mg/100 g ± 0.18 mg/100 g), isobutanol (24.0 mg/100 g ± 1.0 mg/100 g) and 1-propanol (29.4 mg/100 g ± 1.3 mg/100 g) in a spiked cachaça.  相似文献

In this study the antioxidant and cytotoxicity activity of the Adonidia merrillii fruits were investigated using different solvent polarities (methanol, ethyl acetate and water). The results showed that the total phenolic and flavonoid contents of the methanolic extract was higher compare with other extract with respective values of 17.80 ± 0.45 mg gallic acid equivalents/g dry weight (DW) and 5.43 ± 0.33 mg rutin equivalents/g DW. Beside that The RP-HPLC analyses indicated the presence of gallic acid, pyrogallol, caffeic acid, vanillic acid, syringic acid, naringin and rutin. In the DPPH, NO2 and ABTS scavenging assays, the methanolic extract exhibited higher antioxidant activity as compared to the ethyl acetate and water extracts. The extracts exhibited moderate to weak cytotoxic activity in the assays using human hepatocytes (Chang liver cells) and NIH/3T3 (fibroblasts cell) cell lines. The findings showed the Adonidia merrillii fruit extracts to possess considerable antioxidant and cytotoxicity properties. The fruit, therefore, is a potential candidate for further work to discover antioxidant and cytotoxic drugs from natural sources.  相似文献

In this study, a simple, rapid, low cost, sensitive and environmentally friendly technique, supramolecular solvent microextraction (SM-SME) followed by high performance liquid chromatography-ultraviolet has been proposed to extract carbaryl from water samples. Parameters, affecting the SM-SME performance such as the weight of decanoic acid (DeA), volume of tetrahydrofuran (THF), pH and salt concentration, were studied and optimised. The effect of the pH on the extraction efficiency was evaluated by one–factor-at-a-time methodology, but the other variables were optimised by a face-centred cube central composite design methodology. Optimum extraction conditions were obtained: DeA: 70 mg; THF: 650 µL; salt concentration: 10% (w/v) NaCl and pH = 2–4), and the performance of the proposed method was evaluated. Under the optimum conditions, good linearity (1.0–500 µg L?1, r2 = 0.9994) was obtained. Limit of detection and limit of quantification were 0.3–1.0 µg L?1, respectively. Also, the recoveries of the carbaryl were obtained in the ranged from 96% to 105%. Finally, proposed method was successfully applied for the determination of the carbaryl in the water samples of farms run-off and rivers and satisfactory results were obtained.  相似文献

The objective of this study was to evaluate the ethanol production from the sugars contained in the sugarcane bagasse hemicellulosic hydrolysate with the yeast Pichia stipitis DSM 3651. The fermentations were carried out in 250-mL Erlenmeyers with 100 mL of medium incubated at 200 rpm and 30 °C for 120 h. The medium was composed by raw (non-detoxified) hydrolysate or by hydrolysates detoxified by pH alteration followed by active charcoal adsorption or by adsorption into ion-exchange resins, all of them supplemented with yeast extract (3 g/L), malt extract (3 g/L), and peptone (5 g/L). The initial concentration of cells was 3 g/L. According to the results, the detoxification procedures removed inhibitory compounds from the hemicellulosic hydrolysate and, thus, improved the bioconversion of the sugars into ethanol. The fermentation using the non-detoxified hydrolysate led to 4.9 g/L ethanol in 120 h, with a yield of 0.20 g/g and a productivity of 0.04 g L?1 h?1. The detoxification by pH alteration and active charcoal adsorption led to 6.1 g/L ethanol in 48 h, with a yield of 0.30 g/g and a productivity of 0.13 g L?1 h?1. The detoxification by adsorption into ion-exchange resins, in turn, provided 7.5 g/L ethanol in 48 h, with a yield of 0.30 g/g and a productivity of 0.16 g L?1 h?1.  相似文献

This article discusses the extraction and characterization of new natural fiber extracted from red coconut empty fruit bunch. The physicochemical, mechanical, and thermal properties of alkali-treated red coconut empty fruit bunch fibers (ARCEFBFs) were reported and compared with other natural fibers for the first time. Cellulose content (65.02 wt%), wax (0.32 wt%), density (1.421 g/cc), and tensile strength (1299.49 MPa) were identified in ARCEFBFs. Fourier transform infrared spectroscopy and X-ray diffraction analysis confirmed that ARCEFBFs are rich in cellulose content with crystallinity index of 53.6%. Thermogravimetric analysis revealed that these fibers are thermally stable until 270.48°C.  相似文献
