Synthesis of Pyrazolo[3,4-b]- and Pyrido[2,3-b]-1,5-benzodiazepines

Abstract

3-Dimethylaminomethyleno-4-phenyl-1H-1,5-benzodiazepin-2-one (1) was synthesized and reacted with hydrazines, active nitriles, and amino-heterocyclic compounds to give fused heterocyclic compounds 2–14.

Synthesis of [1,2-3H2]-polystyrene

The synthesis of [1,2-³H₂]-polystyrene consisted of a two step reaction. First catalytic tritium gas addition to phenylacetylene was used to prepare [1,2-³H₂]-styrene and then it was polymerized to [1,2-³H₂]-polystyrene in the present of an initiator.     

Synthesis of functionalized 2,3-dihydrothieno[2,3-d]pyrimidin-4(1H)-ones and their recyclization to 2,3-dihydrothieno[3,4-d]pyrimidin-4(1H)-ones

By the reaction of 6-aryl(alkyl)amino-5-cyano-2,3-dihydro-1,3-thiazin-4(10)-ones with α-halogenoketones in the presence of triethylamine, 2,3-dihydrothieno[2,3-d]pyrimidin-4(1H)-ones have been synthesized and their acid-catalyzed recyclization to 2,3-dihydrothieno[3,4-d]pyrimidin-4(1H)-ones has been found and studied. © 2006 Wiley Periodicals, Inc. New organic nitrates. II. Synthesis of 2H-pyrido[2,3-e]-1,3-oxazine-2,4(3H)-diones, 2H-pyrido[4,3-e]-1,3-oxazine-2,4(3H)-diones, 2H-pyrido[4,3-e]-1,3-oxazin-4(3H)-ones, 2H-thieno[2,3-e]-1,3-oxazin-4(3H)-ones and 2H-thieno[3,4-e]-1,3-oxazin-2,4(3H)-diones

The preparation and the physico-chemical characterization of 2H-pyrido[2,3-e]-1,3-oxazine-2,4(3H)-diones, 2H-pyrido[4,3-e]-1,3-oxazine-2,4(3H)-diones, 2H-pyrido[4,3-e]-1,3-oxazin-4(3H)-ones, 2H-thieno[2,3-e]-1,3-oxazin-4(3H)-ones and 2H-thieno[3,4-e]-1,3-oxazine-2,4(3H)-diones are reported.     

Synthesis of 2-Trihalomethyl-3,4-dihydrothieno[2,3-d]pyrimidin-4-ones

At room temperature, N-(1-chloro-2,2,2-trihaloethylidene)-O-methyl urethanes react with 2-aminothiophenes to form N-(2-thienyl)-N'-(methoxycarbonyl)trihaloacetamidines, which when heated in boiling toluene undergo ring closure to form 2-trihalomethyl-3,4-dihydrothieno[2,3-d]pyrimidin-4-ones.     

Synthesis of (2S,3S)-[3-(2)H1]-4-methyleneglutamic acid and (2S,3R)-[2,3-(2)H2]-4-methyleneglutamic acid

(2S,3S)-[3-(2)H1]-4-Methyleneglutamic acid 1a and (2S,3R)-[2,3-(2)H2]-4-methyleneglutamic acid 1b have been synthesised for use in biosynthetic and metabolic studies.     

Synthesis of 3-formylbenzo[b]furan and 1-methyl-3,4-dihydrobenzo[b]-furo[2,3-c]pyridine

Starting from the readily available 3-methylcoumarilate, 3-formylbenzo[b]furan was synthesized in high yield. The latter could be converted to 1-methyl-3,4-dihydrobenzo[b]furo-[2,3-c]pyridine in moderate yield.     

Synthesis of derivatives of pyrido[3,4-b]- and azepino[3,4-b]pyrrolo[2,3-d]pyrimidines from enamines of α-oxo lactams

The reaction of enamines of some -oxo lactams with 1,3-dimethyl- and 1,3-dibenzyl-4-hydrazinouracils gave the uracilyl-4-hydrazones of these lactams, the catalytic and thermal indolization of which yielded derivatives of pyrido[3,4-b]- and azepino[3,4-b]pyrrolo[2,3-d]pyrimidines. 螺(口恶)嗪类化合物是在螺吡喃基础上发展起来的一类新的光致变色化合物，与螺吡喃相比，具有化学性质稳定，光响应快，抗疲劳性好等优点，在光信息记录、非银感成像、非线性光电器件、光过滤器、防伪以及装饰等领域具有很大的应用潜力［1］。     

Synthesis of 2-R-fluorantheno[2,3-d]- and 2-R-fluorantheno[3,2-d]oxazoles

2-R-Fluorantheno[2,3-d]- and 2-R-fluorantheno[3,2-d]oxazoles were synthesized, and their UV and IR spectra were studied. Ten new compounds are described.Translated from Khimiya Geterotsiklicheskikh Soedinenii, No. 7, pp. 905–907, July, 1974.     

Hydroxyiminoisoquinolin-3(2H)-ones. 9. Synthesis of some oxazolo[5,4-c]-, thiazolo[5,4-c]- and 2,3-dihydro-1H-[1,4]oxazino[2,3-c]isoquinolines

4-Acetylamino-1-phenyl-1,4-dihydro-3(2H)-isoquinolinones and 4-amino-1-phenylisoquinolin-3-ol, prepared from the corresponding 4-hydroxyimino compounds, were converted into new isoquinolines containing fused oxazole, thiazole and 1,4-oxazine rings.     

Synthesis of 4-(2-chloroethyl)-2,3-dihydro[1,4]oxazino[2,3-b]quinoline and 4-(2-chloroethyl)2,3-dihydropyrido[2,3-b][1,4]oxazine

The title compounds were synthesized from 3-[bis(2-hydroxyethyl)amino]quinolin-2(1H)-one 11a and 3-[bis(2-hydroxyethyl)amino]pyridin-2(1H)-one 18 respectively. The preparation involved a tandem chlorination/cyclization reaction.     

Convenient Synthesis of 2,2-Dimethyl-3,4-dihydro-2H-pyrano[2,3-b]quinolines

A convenient general synthesis of 2,2-dimethyl-3,4-dihydro-2H-pyrano[2,3-b]quinolines using the Wittig reaction is described. The o-nitrobenzaldehydes (1a–d) on reaction with phosphorane 2 provided ( E )-ethyl-α-(2,2-dimethylprop-2-ene)-2-nitrocinnamates (3a–d) in excellent yields, which on cyclization with polyphosphoric acid followed by reductive cyclization using Fe/HCl afforded dihydropyranoquinolines (5a–d). Alternatively, the pyranoquinolines 5a–d were also synthesised from esters 3a–d by employing domino reductive cyclization in a single step.
