A fast and simple gas chromatography-mass spectrometry (GC-MS) method for determination of forty-one pesticide residues in maize is introduced. The sample preparation involves liquid-liquid partitioning with acetonitrile in presence of anhydrous MgSO(4) and NaCl (QuEChERS) followed by dispersive liquid-liquid microextraction (DLLME) using carbon tetrachloride as extractive solvent and the extract obtained by QuEChERS as dispersive solvent. The main factors influencing DLLME efficiency including extractive solvent type and volume as well as the volume of dispersive solvent were evaluated in this study. The DLLME procedure effectively provides an enrichment of the extract and a cleanup of certain polar matrix components, which can maximize the sensitivity when a single quadrupole MS is used. For validation purposes, recoveries studies were carried out at two concentration levels, yielding recovery rates in the range 70-120% for 82% of the analytes. A good linearity and precision, with relative standard deviations generally below 20% were obtained for all forty-one pesticides. The limits of detection obtained were lower than 19 μg kg(-1) for more than 63% of the analytes. In two of a total of ten samples of maize, residues of lindane, tefluthrin, pirimicarb, folpet and bifenthrin were found, although at levels below the maximum limit established for this kind of samples.  相似文献

3-Chloropropane-1,2-diol (3-MCPD) and 1,3-dichloro-2-propanol (1,3-DCP) were determined for the first time in bakery foods using pressurized liquid extraction (PLE) combined with in situ derivatization and GC-MS analysis. This one-step protocol uses N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA) as silylation reagent. Initially, screening experimental design was applied to evaluate the effects of the variables potentially affecting the extraction process, namely extraction time (min) and temperature (°C), number of cycles, dispersant reagent (diatomaceous earth in powder form and as particulate matter with high pore volume Extrelut NT) and percent of flush ethyl acetate volume (%). To reduce the time of analysis and improve the sensitivity, derivatization of the compounds was performed in the cell extraction. Conditions, such as the volume of BSTFA, temperature and time for the in situ derivatization of analytes using PLE, were optimized by a screening design followed to a Doehlert response surface design. The effect of the in-cell dispersants/adsorbents with diatomaceous earth, Florisil and sodium sulfate anhydrous was investigated using a Box-Behnken design. Using the final best conditions, 1 g of sample dispersed with 0.1 g of sodium sulfate anhydrous and 2.5 g diatomaceous earth was extracted with ethyl acetate. 1 g of Florisil, as clean-up adsorbent, and 70 μL of BSTFA were used for 3 min at 70°C. Under the optimum conditions, the calibration curves showed good linearity (R(2)>0.9994) and precision (relative standard deviation, RSD≤2.4%) within the tested ranges. The limits of quantification for 1,3-DCP and 3-MCDP, 1.6 and 1.7 μg kg(-1), respectively, are far below the established limits in the European and American legislations. The accuracy, precision, linearity, and limits of quantification provided make this analytical method suitable for routine control. The method was applied to the analysis of several toasted bread, snacks, cookies and cereal samples, none of which contained chloropropanols at concentrations above the legislation levels.  相似文献

The phenomenon known as "matrix-induced enhancement effect" is not only observed in the analysis of pesticides in food, but also in Chinese herbs. Several approaches have been proposed to overcome the matrix-induced effect, but each method has serious limitations. Compared with standard calibration methods, the procedure with adding analyte protectants offers a more convenient and effective route to solve the problem. In the current study, we have analyzed 195 types of pesticides in Chinese herbs by gas chromatography-mass spectrometry (GC-MS), and the compounds that are susceptible to matrix effect were picked up and confirmed. In addition, several analyte protectants were evaluated and the most effective combination was determined. D-ribonic acid-γ-lactone (2 mg/ml) and D-sorbitol (1 mg/ml) were shown to be the best analyte protectants for the analysis of most pesticides.  相似文献

Tetrodotoxin (TTX), a toxic compound found in some puffers can cause death to humans through consumption. We have developed a simplified method for the screening of TTX in puffers using GC-MS. A puffer tissue of 0.5g was treated with 5mL of 0.1% acetic acid, followed by alkaline hydrolysis, LLE or liquid-liquid extraction and N-methyl-N-TMS-trifluoroacetamide derivatization. The developed method used only a small sample and solvent, simplified LLE and derivatization procedures and short chromatographic analysis (8.2min). All of these contribute to cost-saving, enhanced sample throughput and high sensitivity of the screening assay. The developed method was validated and proved to be within the acceptable range.  相似文献

A method was developed to employ National Institute of Standards and Technology (NIST) 2008 retention index database information for molecular retention matching via constructing a set of empirical distribution functions (DFs) of the absolute retention index deviation to its mean value. The effects of different experimental parameters on the molecules' retention indices were first assessed. The column class, the column type, and the data type have significant effects on the retention index values acquired on capillary columns. However, the normal alkane retention index (I(norm)) with the ramp condition is similar to the linear retention index (I(T)), while the I(norm) with the isothermal condition is similar to the Kováts retention index (I). As for the I(norm) with the complex condition, these data should be treated as an additional group, because the mean I(norm) value of the polar column is significantly different from the I(T). Based on this analysis, nine DFs were generated from the grouped retention index data. The DF information was further implemented into a software program called iMatch. The performance of iMatch was evaluated using experimental data of a mixture of standards and metabolite extract of rat plasma with spiked-in standards. About 19% of the molecules identified by ChromaTOF were filtered out by iMatch from the identification list of electron ionization (EI) mass spectral matching, while all of the spiked-in standards were preserved. The analysis results demonstrate that using the retention index values, via constructing a set of DFs, can improve the spectral matching-based identifications by reducing a significant portion of false-positives.  相似文献

A solid‐phase extraction (SPE) method was developed to extract 14 pesticides simultaneously from environment samples using cigarette filter as the sorbent before gas chromatography‐mass spectrometry (GC‐MS) analysis. Parameters influencing the extraction efficiency, such as the sample loading flow rate, eluent and elution volume, were optimized. The optimum sample loading rate was 3 mL/min, and the retained compounds were eluted with 6 mL of eluent at 1 mL/min under vacuum. Good linearity was obtained for all the 14 pesticides (r2>0.99) from 0.1 to 20 μg/L for water and from 2 to 400 μg/kg for soil samples. The detection limits (signal‐to‐noise=3) of the proposed method ranged from 0.01 to 0.20 μg/L for water samples and from 0.42 to 6.95 μg/kg for soil samples. The developed method was successfully applied for determination of the analytes in real environmental samples, and the mean recoveries ranged from 76.4 to 103.7% for water samples and from 79.9 to 105.3% for soil samples with the precisions (relative standard deviation) between 2.0 and 13.6%.  相似文献

Simple and rapid quantitative determination of fatty-acid-based biofuels is greatly important for the study of genetic engineering progress for biofuels production by microalgae. Ideal biofuels produced from biological systems should be chemically similar to petroleum, like fatty-acid-based molecules including free fatty acids, fatty acid methyl esters, fatty acid ethyl esters, fatty alcohols and fatty alkanes. This study founded a gas chromatography-mass spectrometry (GC-MS) method for simultaneous quantification of seven free fatty acids, nine fatty acid methyl esters, five fatty acid ethyl esters, five fatty alcohols and three fatty alkanes produced by wild-type Synechocystis PCC 6803 and its genetically engineered strain. Data obtained from GC-MS analyses were quantified using internal standard peak area comparisons. The linearity, limit of detection (LOD) and precision (RSD) of the method were evaluated. The results demonstrated that fatty-acid-based biofuels can be directly determined by GC-MS without derivation. Therefore, rapid and reliable quantitative analysis of fatty-acid-based biofuels produced by wild-type and genetically engineered cyanobacteria can be achieved using the GC-MS method founded in this work.  相似文献

The fatty acid composition of milk is of considerable interest due to their nutritional and functional properties. Although rapid milk fat separation and transesterification procedures have been developed, the overall procedure remains time consuming, specially, for the analysis of a large number of samples. In this work, a fast and simple method for direct profiling of fatty acids from milk using thermochemolysis has been developed. This method has the capability of directly analyse fatty acids from one drop of milk without fat extraction or cleanup. Our approach for thermochemolysis is based on thermal desorption integrated with a cold trap inlet. The optimized method does not present isomerisation/degradation of polyunsaturated fatty acid and shows milk fatty acid profiles comparable to the conventional method based on fat extraction and alkaline transesterification. Overall, this method has demonstrated significant potential for high throughput analysis of fatty acids in milk.  相似文献

An exhaustive GC-MS acquisition study was performed, for the simultaneous analysis of natural and synthetic steroids and cholic acids (in order to insert them into the last tierce of our multiresidue analysis system), such as androsterone, β-estradiol, transdehydroandro-sterone, transdehyroandrosterone, mestranol, dihydrotestosterone, ethinylestradiol, testosterone, norethisterone, estriol, 4-androstene-3,17-dione, gestodene, levonorgestrel, etonogestrel, coprostanol, progesterone, cholesterol, medroxyprogesterone-acetate, lithocholic acid, stigmasterol, cholic acid, chenodeoxycholic acid, β-sitosterol, ursodeoxycholic acid, 3-hydroxy-7-ketocholic acid and dehydrocholic acid, in total 26 compounds. As novelties to the field, for the trimethylsilyl (TMS) oxime ether/ester derivatives of steroids and cholic acids, at first, a tandem mass spectrometric (MS/MS), multiple reaction monitoring (MRM) type acquisition method has been developed in a single run; also for the first time, the three acquisition techniques, the full scan (FS), the selective ion monitoring (SIM), in our case the multiple ion monitoring (MIM) and the currently optimized MRM methods, have been compared; all three, in parallel, under strictly the same derivatization/instrumental conditions, both in matrix free solutions and municipal wastewater from two Hungarian wastewater treatment plants (WWTPs). Critical evaluation of the three acquisition protocols was collated on their analytical performances and validated under the same conditions. The data of six point calibration curves for FS, MIM and MRM methods, showed that both R2 (0.9995, 0.9858, 0.9975) and RSD (5.3, 5.8, 5.0), for two parallel derivatizations, each injected three times, proved to be independent of the acquisition processes. Whereas, for the method limit of quantification (LOQ) and the instrument limit of quantification (ILQ) values showed considerable differences. LOQ data, were decreasing in the FS, MIM, MRM line (expressed in ng/L), for all steroids and cholic acids. The same trend was determined in terms of the ILQ values. The practical utility of the optimized acquisition techniques was confirmed by the quantitation of the steroids and cholic acids contents of wastewater samples. Results confirmed the importance of the MRM acquisition method, even in comparison to the MIM one: with particular interest in selected cases: avoiding the extreme overestimation of the β-estradiol (156-1325%) and that of the ethinylestradiol (582-831%) concentrations in the wastewater samples.  相似文献

A method for analysing carbamates and other relatively polar pesticides by LC–MS–MS with electrospray ionisation has been developed. The method is based on extraction by ultrasonication using a methanolic ammonium acetate–acetic acid buffer. After centrifugation the samples are filtered in Miniprep filter HPLC vials and detected by LC–MS–MS. To compensate for variations in the MS response [13C6]-carbaryl was used as internal standard and matrix-matched pesticide solutions were used as external standards for the quantification. The method has been validated for the matrices apple, avocado, carrot, lettuce, orange, potato and wheat at the spiking levels—0.02; 0.04 and 0.20 mg kg−1. Recoveries were generally in the range 70–120%. Results from participation in three intercomparisons proved the accuracy of the method. As the analytical procedure does not include any concentration or cleanup steps, it is easy and fast to perform, making it applicable for routine analysis in large pesticide monitoring programmes.  相似文献

A sensitive and selective gas chromatography with mass spectrometry method was developed for the simultaneous determination of three organophosphorus pesticides, namely, chlorpyrifos, malathion, and diazinon in three different food commodities (milk, apples, and drinking water) employing solid‐phase extraction for sample pretreatment. Pesticide extraction from different sample matrices was carried out on Chromabond C18 cartridges using 3.0 mL of methanol and 3.0 mL of a mixture of dichloromethane/acetonitrile (1:1 v/v) as the eluting solvent. Analysis was carried out by gas chromatography coupled with mass spectrometry using selected‐ion monitoring mode. Good linear relationships were obtained in the range of 0.1–50 μg/L for chlorpyrifos, and 0.05–50 μg/L for both malathion and diazinon pesticides. Good repeatability and recoveries were obtained in the range of 78.54–86.73% for three pesticides under the optimized experimental conditions. The limit of detection ranged from 0.02 to 0.03 μg/L, and the limit of quantification ranged from 0.05 to 0.1 μg/L for all three pesticides. Finally, the developed method was successfully applied for the determination of three targeted pesticides in milk, apples, and drinking water samples each in triplicate. No pesticide was found in apple and milk samples, but chlorpyrifos was found in one drinking water sample below the quantification level.  相似文献

The rotating disk sorptive extraction (RDSE) technique was applied to the determination of pesticides in aqueous samples. Pesticides of different polarities were considered in this study: chlorpyrifos, diazinon, fenvalarate, cyhalothrin, cypermethrin, lindane and malathion. The sorptive/desorptive behavior of the pesticides was studied using a rotating disk containing a polydimethylsiloxane (PDMS) phase on one of its surfaces. The analyte polarity was a significant factor in the extraction time; shorter extraction times were required for the more apolar pesticides. The optimum variables for the extraction of all analytes were: extraction time of 3 h, sample volume of 25 mL, rotational velocity of the disk 1250 rpm, desorption time of 30 min using methanol. For pesticides with values of Log Kow > 4, the extraction time can be reduced to 30 min for a quantitative extraction. Under these conditions, recoveries between 76% and 101% were obtained for the target pesticides, and the repeatability of the methodology, expressed as relative standard deviation, was determined to be between 10% and 20%. Additionally, the limits of detection of the analytes were lower than 3.1 μg L−1. The extraction method developed using the RDSE was compared to a stir bar sorptive extraction (SBSE) under the same conditions. It can be observed that the extraction using the rotating disk offers higher recoveries because of its higher PDMS volume and its higher surface area to volume ratio that allows for improved mass transfer.  相似文献

An effective low cost sample preparation methodology for the determination of regulated fragrance allergens in leave-on and rinse-off cosmetics has been developed applying, for the first time, matrix solid-phase dispersion (MSPD) to this kind of analytes and samples. The selection of the most suitable extraction conditions was made using statistical tools such as ANOVA, as well as a factorial multifactor experimental design. These studies were carried out using real cosmetic samples. In the final conditions, 0.5 of sample, previously mixed with 1g of anhydrous Na(2)SO(4), were blended with 2g of dispersive sorbent (Florisil), and the MSPD column was eluted with 5 mL of hexane/acetone (1:1). The extract was then analyzed by GC-MS without any further clean-up or concentration step. Accuracy, precision, linearity and detection limits (LODs) were evaluated to assess the performance of the proposed method. Quantitative recoveries (>75%) were obtained and RSD values were lower than 10% in all cases. The quantification limits were well below those set by the international cosmetic regulations, making this multi-component analytical method suitable for routine control. In addition, the MSPD method can be implemented in any laboratory at low cost since it does not require special equipment. Finally, a wide variety of cosmetic products were analyzed. All the samples contained several of the target cosmetic ingredients, with and average number of seven. The total fragrance allergen content was in general quite high, even in baby care products, with values close to or up to 1%, for several samples, although the actual European Cosmetic Regulation was fulfilled.  相似文献

A rapid method, based on sonication-assisted extraction in small columns (SAESC) and subsequent quantification and identification by gas chromatography-mass spectrometry (GC-MS), was developed for the determination of cyclic and linear siloxanes in soil. In the experiments with spiked samples (10-50 ng g(-1)), the recovery of cyclic and linear siloxanes ranged from 87.7 to 108.0% and from 84.9 to 107.6%, respectively. The validated method was used to determine the levels of these compounds in various types of soil samples collected from different locations in Spain. The cyclic siloxanes, decamethylcyclopentasiloxane (D5) and dodecamethylcyclohexasiloxane (D6) were detected in all the soil samples analyzed at concentrations from 9.2 to 56.9 ng g(-1) for D5 and from 5.8 to 27.1 ng g(-1) for D6 in agricultural soils and from 22 to 184 ng g(-1) for D5 and from 28 to 483 ng g(-1) for D6 in industrial soils. The total linear siloxanes concentrations (L5-L14) (sum of the 10 congeners) ranged from 191 to 292 ng g(-1) in agricultural soils and from 1411 to 8532 ng g(-1) in industrial soils.  相似文献

Gas chromatography-mass spectrometry (GC-MS) with electron impact (EI), positive-ion chemical ionization (PCI) and negative-ion chemical ionization (NCI) were applied as confirmatory techniques for residue analysis of chlorotriazine pesticides, their degradation products and organophosphorus pesticides in soil samples. Clean-up was effected using a Florisil column with subsequent analysis by GC with a nitrogen-phosphorus detector. GC-MS with the EI mode of operation is the common mode of confirmation for all the pesticides. Further confirmation by either GC-MS with PCI and NCI for chlorotriazines and organophosphorus pesticides, respectively, is recommended. The method was applied to the determination of residue levels of atrazine, deethylatrazine, deisopropylatrazine, simazine, fenitrothion and tetrachlorvinphos in several soil samples at levels from 5 ng g?1 to 9 μg g?1.  相似文献

A multiresidue method was developed for the quantification and confirmation of 70 pesticides in paddy field water. After its filtration, water was injected directly in a liquid chromatograph coupled to a hybrid triple quadrupole-linear ion trap-mass spectrometer (QqLIT). The list of target analytes included organophosphates, phenylureas, sulfonylureas, carbamates, conazoles, imidazolinones and others compounds widely used in different countries where rice is cropped. Detection and quantification limits achieved were in the range from 0.4 to 80 ng L(-1) and from 2 to 150 ng L(-1), respectively. Correlation coefficients for the calibration curves in the range 0.1-50 μg L(-1) were higher than 0.99 except for diazinon (0.1-25 μg L(-1)). Only 9 pesticides presented more than 20% of signal suppression/enhancement, no matrix effect was observed in the studied conditions for the rest of the target pesticides. The method developed was used to investigate the occurrence of pesticides in 59 water samples collected in paddy fields located in Spain and Uruguay. The study shows the presence of bensulfuron methyl, tricyclazole, carbendazim, imidacloprid, tebuconazole and quinclorac in a concentration range from 0.08 to 7.20 μg L(-1).  相似文献

Beer stability is a major concern for the brewing industry, as beer characteristics may be subject to significant changes during storage. This paper describes a novel non-targeted methodology for monitoring the chemical changes occurring in a lager beer exposed to accelerated aging (induced by thermal treatment: 18 days at 45 °C), using gas chromatography-mass spectrometry in tandem with multivariate analysis (GC-MS/MVA). Optimization of the chromatographic run was performed, achieving a threefold reduction of the chromatographic time. Although losing optimum resolution, rapid GC runs showed similar chromatographic profiles and semi-quantitative ability to characterize volatile compounds. To evaluate the variations on the global volatile signature (chromatographic profile and m/z pattern of fragmentation in each scan) of beer during thermal deterioration, a non-supervised multivariate analysis method, Principal Component Analysis (PCA), was applied to the GC-MS data. This methodology allowed not only the rapid identification of the degree of deterioration affecting beer, but also the identification of specific compounds of relevance to the thermal deterioration process of beer, both well established markers such as 5-hydroxymethylfufural (5-HMF), furfural and diethyl succinate, as well as other compounds, to our knowledge, newly correlated to beer aging.  相似文献

A method for separation and identification of peaks in essential oil samples based on rapid repetitive heart-cutting using multidimensional gas chromatography (MDGC)-mass spectrometry (MS) coupled with a cryotrapping interface is described. Lavender essential oil is analyzed by employing repetitive heart-cut intervals of 1.00 and 1.50 min, achieved in a parallel MDGC-MS/GC-FID experiment. The number of peaks that were detected in 1D GC operation above a given response threshold more than tripled when MDGC-MS employing the cryotrapping module method was used. In addition, MDGC-MS enabled detection of peaks that were not individually evident in 1D GC-MS, owing to effective deconvolution in time of previously overlapped peaks in 1D GC. Thus separation using the cryomodulation approach, without recourse to using deconvolution software, was possible. Peaks widths decreased by about 5-7-fold with the described method, peak capacity increased from about 9 per min to 60 per min, and greater sensitivity results. Repeatability of retention times for replicate analyses in the multidimensional mode was better than 0.02% RSD. The present study suggests that the described heart-cutting technique using MDGC-MS can be used for general improvement in separation and identification of volatile compounds.  相似文献

An analytical method for the determination of UV filters in soil and sediment has been developed and validated considering benzophenones (BP) and salicylates as target analytes. Soil and sediment samples were extracted with ethyl acetate-methanol (90:10, v/v) assisted with sonication, performing a simultaneous clean-up step. Quantification of these compounds was carried out by gas chromatography-mass spectrometry (GC-MS) after derivatization of the extracts with N,O-bis(trimethylsilyl)trifluoroacetamide (BSTFA). Recoveries from spiked soil samples ranged from 89.8% to 104.4% and they were between 88.4% and 105.3% for spiked sediment samples. The effect of the residence time and soil moisture content on the recovery of these compounds was also studied. The precision, expressed as relative standard deviation, was in all cases below 6.1% and the limits of detection (S/N=3) varied from 0.07 to 0.10 ng g(-1) and from 0.11 to 0.28 ng g(-1) for soils and sediments, respectively. The validated method was applied to the analysis of five benzophenone and two salicylate UV filters in soil and sediment samples collected in different areas of Spain.  相似文献
