The indole nucleus of Z-Trp-OBzl is modified by acylation of the indole nitrogen using Boc-N-methyl butyric acid followed by catalytic hydrogenation and introduction of the Fmoc group. The resulting derivative, Fmoc-Trp(Boc-Nmbu)-OH, is incorporated into peptide chains via solid-phase peptide synthesis (SPPS). After assembly of the peptide chain, the Boc group is cleaved by treatment with TFA. The peptide is isolated with the tryptophan residue modified with a cationic 4-(N-methylamino) butanoyl group, which improves the solubility of the peptide during HPLC purification. On treatment of the purified peptide at pH 9.5, the Nmbu group undergoes an intramolecular cyclization reaction; this results in the fully deprotected peptide and N-methylpyrrolidone.  相似文献

A new cyclic octreotide-like octapeptide was prepared by incorporation of an unnatural tetrazolyl amino acid, an analog of Fmoc-l-lysine, into the peptide chain. The new tetrazolyl amino acid, (S)-2-{[2-(9H-fluoren-9-yl)acetoxy]amino}-6-(1H-tetrazol-1-yl)hexanoic acid, was obtained by azidation of Fmoc-l-lysine trifluoroacetate with sodium azide in the presence of triethyl orthoformate in glacial acetic acid. The linear peptide sequence was prepared using efficient Fmoc/t-Bu solid-phase peptide synthesis (SPPS). Cyclization of the octapeptide was carried out via oxidation with iodine. The structure and purity of the cyclic octapeptide were confirmed by LC–MS, MALDI-TOF MS/MS analysis as well as 1D/2D 1H and 13C NMR spectroscopy.  相似文献

Ribosomally produced peptides that contain D-amino acids have been isolated from a number of vertebrate and invertebrate sources. In each case, the D-amino acids are introduced by a posttranslational modification of a parent peptide containing only amino acids of the L-configuration. The only known enzyme to catalyze such a reaction is the peptide epimerase (also known as peptide isomerase) from the venom of the funnel web spider, Agelenopsis aperta. This enzyme interconverts two 48-amino-acid-long peptide toxins that differ only by the stereochemistry at a single serine residue. In this paper we report the synthesis and testing of two pentapeptide analogues that contain modified amino acids at the site normally occupied by the substrate serine residue. When the L-chloroalanine-containing peptide 3 was incubated with the epimerase it was converted into the dehydroalanine-containing peptide 4 via an elimination of HCl. The dehydroalanine peptide 4 was independently synthesized and found to act as a potent inhibitor of the epimerase (IC50 = 0.5 microM). These results support a direct deprotonation/reprotonation mechanism in which a carbanionic intermediate is formed. The observed inhibition by 4 can be attributed to the sp(2)-hybridization of the alpha-carbon in the dehydroalanine unit that mimics the planar geometry of the anionic intermediate.  相似文献

In the present investigation, we studied the enzymatic synthesis of monoacylglycerols (MAG) and diacylglycerols (DAG) via the esterification of saturated fatty acids (stearic, palmitic and an industrial residue containing 87% palmitic acid) and glycerol in a solvent‐free system. Three immobilized lipases (Lipozyme RM IM, Lipozyme TL IM and Novozym 435) and different reaction conditions were evaluated. Under the optimal reaction conditions, esterifications catalyzed by Lipozyme RM IM resulted in a mixture of MAG and DAG at high conversion rates for all of the substrates. In addition, except for the reaction of industrial residue at atmospheric pressure, all of these products met the World Health Organization and European Union directives for acylglycerol mixtures for use in food applications. The products were quantified by 13C NMR, with the aid of an external reference signal which was generated from a sealed coaxial tube filled with acetonitrile‐d3. After calibrating the area of this signal using the classical external reference method, the same coaxial tube was used repeatedly to quantify the reaction products. In the protein chemical synthesis via native chemical ligation (NCL) method with three peptide segments, the N-terminal cysteine residue of middle segment is generally protected by thiazolidine ring. In this paper, we show the novel method for thiazolidine ring opening using 2,2′-dipyridyl disulfide (DPDS). The N-terminal thiazolidine was converted into S-pyridylsulfenylated cysteine residue with DPDS under acidic conditions, and this N-terminally Cys peptide protected with disulfide was applicable to NCL reaction without purification and deprotection steps. DPDS treatment did not remove other Cys protecting groups generally used for regioselective disulfide bond formation reactions. These results indicate that this thiazolidine ring opening reaction is quite useful for the protein chemical synthesis with three-segment NCL strategy.  相似文献

The synthesis of lipid core peptide (LCP) vaccines using stepwise solid-phase peptide synthesis commonly results in products which are difficult to purify to homogeneity. A new technique for synthesizing highly-pure, mono-epitopic, multi-valent LCP-systems using native chemical ligation is presented. Various conditions were assessed for ligating four copies of a thioester-modified 88/30 serotype group A streptococcal peptide antigen onto an LCP-system featuring four cysteine residues. Overall, the vaccine was synthesized in high purity (>99%), and high yield (90%) when the ligation reaction was performed in the presence of 1% sodium dodecyl sulfate and at elevated temperatures (37 °C).  相似文献

Trimethyl alpha-amino derivatives of peptides (penta to deca) with a permanent positive charge on their alpha-amino groups were prepared by in vacuo reaction with iodomethane and subjected to matrix-assisted laser desorption/ionization mass spectrometry (MALDI-MS). Compared to the unmodified peptide, the signal intensity of the trimethyl alpha-amino derivative in MALDI-MS is increased by at least an order of magnitude. Similarly, an octapeptide with a trimethylated epsilon-amino group derived from the solitary lysine residue of the B-chain of insulin also shows the same relative increase in signal intensity. Another advantage of the in vacuo methylation procedure is that trimethylation of a peptide amino group can be carried out readily with a combination of isotopes (13)CH(3)I and (12)CH(3)I or CD(3)I and CH(3)I, yielding a doublet signal either 3 or 9 units apart, respectively. The presence or absence of such a doublet signal can be used as a criterion to discriminate between peptide and non-peptide signals in the mass spectrum.  相似文献

Formation of peptide thioesters, based on an N to S acyl shift mediated by an auxiliary, N-4,5-dimethoxy-2-mercaptobenzyl (Dmmb) group, under acidic conditions, is described. The protected peptide was assembled on a hydroxymethylphenylacetamidomethyl resin via an N-Dmmb-amino acid residue according to standard Fmoc solid-phase peptide synthesis following treatment with trifluoroacetic acid. The peptide α-thioester was released from the resin by reaction with 2-mercaptoethanesulfonic acid in the presence of N,N-diisopropylethylamine.  相似文献

Rheumatoid arthritis (RA) is an inflammatory autoimmune disease affecting about 0.24 % of the world population. Protein arginine deiminase type 4 (PAD4) is believed to be responsible for the occurrence of RA by catalyzing citrullination of proteins. The citrullinated proteins act as autoantigens by stimulating an immune response. Citrullinated α-enolase has been identified as one of the autoantigens for RA. Hence, α-enolase serves as a suitable template for design of potential peptide inhibitors against PAD4. The binding affinity of α-enolase-derived peptides and PAD4 was virtually determined using PatchDock and HADDOCK docking programs. Synthesis of the designed peptides was performed using a solid phase peptide synthesis method. The inhibitory potential of each peptide was determined experimentally by PAD4 inhibition assay and IC50 measurement. PAD4 assay data show that the N-P2 peptide is the most favourable substrate among all peptides. Further modification of N-P2 by changing the Arg residue to canavanine [P2 (Cav)] rendered it an inhibitor against PAD4 by reducing the PAD4 activity to 35 % with IC50 1.39 mM. We conclude that P2 (Cav) is a potential inhibitor against PAD4 and can serve as a starting point for the development of even more potent inhibitors.  相似文献

The reaction of activated ruthenocene carboxylic acid with the resin-bound peptide octreotate yields, after cleavage and purification by preparative HPLC, the first ruthenocenoyl peptide bioconjugate 1. Octreotate is a chemically stabilized analogue of somatostatine. It is a cyclic octapeptide with a disulfide bond and has been previously used for molecular diagnostics due to the fact that somatostatine receptors are over-expressed by a variety of cancer cells. Conjugate 1 was obtained in good yield and purified by preparative HPLC to >95% purity as judged by analytical HPLC. It has been identified by HPLC, IR and mass spectrometry (ESI and MALDI-TOF). The peptide’s NMR signals are assigned by standard 2D methods. In addition, the 1H NMR spectrum of 1 shows characteristic signals for the metallocene between 5.1 and 4.3 ppm. Compound 1 thus is a new example of tumor-targeted organometallic ruthenium bioconjugates.  相似文献

A method for the solid phase synthesis of substituted arginine containing peptides starting from an isothiocitrulline precursor is described. In this procedure, a peptide containing one or more protected ornithine residue(s) was assembled on a solid support. Following selective deprotection, ornithine residue(s) was (were) converted into S-methyl-isothiocitrulline in three steps. Subsequent reaction with primary or secondary amines afforded mono and disubstituted arginine-containing derivatives, respectively. Using lysine instead of ornithine afforded substituted homoarginine-containing derivatives.  相似文献

Dimedone is the most widely used chemical probe for detection of cysteine sulfenic acid in peptides and proteins. The reaction of dimedone with cysteine sulfenic acid results in the formation of unique cysteine dimedone motif containing thioether bridge. Based on the structure of cysteine dimedone residue in polypeptide, a new building block of Fmoc-Cys(Dmd)-OH was developed for solid phase synthesis of peptide cysteine dimedone. Mass spectrometric sequencing of synthetic peptides have confirmed successful incorporation of cysteine dimedone in peptide chain using HBTU/HOBt as a coupling agent. The new method permits synthesis of peptides containing both cysteine thiol and cysteine dimedone in the same sequence which was difficult to achieve by conventional methods. The synthetic peptide of glutathione cysteine dimedone was used as a standard in probing the air-mediated oxidation of thiol to disulfide form of glutathione. The co-elution of standard peptide and reaction mixture of oxidation of glutathione in presence of dimedone using RP-HPLC have confirmed the formation of glutathione cysteine sulfenic as an intermediate in the air-mediated oxidation of glutathione. The synthetic peptides of cysteine dimedone may find application in the field of redox proteomics and generation of antibodies against modified cysteine residue.  相似文献

Peptide and protein lipoxidation is a deleterious process which has been related to several degenerative conditions. In the present study, the interaction of lipid secondary oxidation products with peptides was investigated by evaluating the modifications occurring to angiotensin II (Ang-II) in the presence of an oxidizing polyunsaturated glycerophospholipid (1-palmitoyl-2-arachidonoyl-glycerophosphatidylcholine, PAPC). PAPC oxidation was promoted by Fenton chemistry and the oxidation products were incubated with Ang-II. The reaction products were finally analysed by off-line nanospray high-performance liquid chromatography/matrix-assisted laser desorption/ionization tandem mass spectrometry (nano-HPLC/MALDI-TOF-MS/MS). Ang-II was found to form adducts with 26 different aldehydes, leading to 37 distinct reaction products. Modification of Ang-II occurred through reaction with reactive carbonyl species (RCS) originating from fatty acyl chain cleavage, while interactions with the oxidized phospholipid could not be detected. Adduction was observed to occur both by Michael and Schiff base mechanisms, most prevalently taking place at the peptide N-terminus or the arginine residue. Histidine modification could only be demonstrated to occur via Michael addition with two aldehydes: 4-hydroxy-2-nonenal (HNE) and 2-octenal. The highly reactive 4-oxo-2-nonenal (ONE) was shown to react preferentially with the arginine side chain, while malondialdehyde addition could only be confirmed at the N-terminus. Aspartic acid oxidative decarboxylation, amino acid side chain oxidation, multiple adduction or peptide cross-links could not be perceived. The inability to detect these reaction products is indicative of their low abundance or non-existence in competitive reaction conditions. The multiplicity of peptide modifications described emphasizes the complexity of lipoxidation, the effects of which are not possible to fully understand by the evaluation of independent reaction products.  相似文献

HOBt,HOSu,HOOBt是多肽合成中常用的试剂,它们常被用作羧基活化试剂和消旋抑制剂。首次发现HOBt,HOSu,HOOBt在碱的存在下可与氯代烷烃溶剂发生反应,并系统地研究了不同的反应条件对产率的影响和对多肽合成的影响。发现反应的产率与碱的用量,反应温度,反应物等因素有关,碱性越强,反应速度越快产率越高,HOBt较易反应,HOSu较为稳定,二氯甲烷易生成双取代产物,1,2-二氯乙烷易生成单代产物,所有产物均经硅胶柱层析分离纯化,共合成8个经合物并通过光谱鉴定。虽然在模型二肽BocAlaPheOCH3合成反应中检测到少量副产物的产生,但二肽的产率>82%。  相似文献

The solid-phase synthesis of "unnatural" amino aldehydes, amino ketones, peptide aldehydes, and peptide ketones was accomplished from commercially available resin in a series of room temperature reactions. The initial step involved addition of an "unnatural" side chain to the N-terminus of a benzophenone imine-activated Weinreb resin-bound amino acid or peptide derivative. The alkylated imine was hydrolyzed, and the amine was converted to the Boc-, Cbz-, or naphthoyl derivative. The resin-bound substrate was then cleaved with DIBAL-H or a Grignard reagent to give the amino aldehyde, amino ketone, peptide aldehyde, or peptide ketone products. Twenty-four reactions were carried out simultaneously using a "Billboard" reaction apparatus to give products in 27-87% (59% average) isolated yield.  相似文献

Self-decomposition of the nickel(III) doubly deprotonated peptide complex of Gly2HisGly occurs by base-assisted oxidation of the peptide. At < or =p[H+] 7.0, the major pathway is a four-electron oxidation (via 4 Ni(III) complexes) at the alpha carbon of the N-terminal glycyl residue. The product of this oxidation is oxamylglycylhistidylglycine, which hydrolyzes to yield ammonia and oxalylglycylhistidylglycine. Both of these peptide products decompose to give isocyanatoacetylhistidylglycine. A small amount (2%) of oxidative decarboxylation also is observed. In another major pathway above p[H+] 7.0, two Ni(III)-peptide complexes coordinate via an oxo bridge in the axial positions to form a reactive dimer species. This dimer generates two Ni(II)-peptide radical intermediates that cross-link at the alpha carbons of the N-terminal glycyl residues. In 0.13 mM Ni(III)-peptide at p[H+] 10.3, this pathway accounts for 60% of the reaction. The cross-linked peptide is subject to oxidation via atmospheric O2, where the 2,3-diaminobutanedioic acid is converted to a 2,3-diaminobutenedioic acid. The products observed at 相似文献

Sequence determination of peptides blocked by amino-terminal pyrrolidone carboxylic acid (PCA) has in the past been hampered by the lack of a reliable and efficient method for removing this residue. We report here a rapid, efficient enzymatic method for removal of PCA. Reversed-phase high-performance liquid chromatography is used to monitor the reaction and to separate unblocked peptide from the reaction product. The method allows direct sequence analysis of newly purified PCA blocked peptides, eliminating the need for complicated enzymatic digestions and chromatography to determine the amino-terminal residue. Only a few micrograms of peptide are required instead of the several milligrams needed in the past.  相似文献

The Boc-N-methyl-N-[2-(methylamino)ethyl]carbamoyl group (Boc-Nmec) is reported as a new side chain-protective group for tyrosine in Fmoc solid-phase peptide synthesis. Tyrosine is incorporated into the peptide as Fmoc-Tyr(Boc-Nmec)-OH by standard coupling methods. During the cleavage of the peptide from the resin with TFA the Boc group is simultaneously cleaved while the cationic N-methyl-N-[2-(methylamino)ethyl]carbamoyl group remains attached to the tyrosine residue, thereby increasing the solubility of the peptide. After purification of the peptide, the Nmec protective group can be cleaved under neutral or mild alkaline conditions via an intramolecular cyclization reaction.  相似文献

The enzymatic synthesis of the cholecystokinin octapeptide (CCK‐8) is reported. The target octapeptide CCK‐8 is the minimum active sequence with the same biological activity as naturally occurring cholecystokinin and is a potential therapeutic agent in the control of gastrointestinal function as well as a drug candidate for the treatment of epilepsy and obesity. The protected CCK‐8 was obtained by incubation of Bz‐Arg‐Asp(OEt)‐Tyr‐Met‐OAl and Gly‐Trp‐Met‐Asp(OMe)‐Phe‐NH2 with immobilized α‐chymotrypsin. The Bz‐Arg group was used as an N‐terminal protecting group in the synthesis of the tripeptide fragment. The protected CCK‐8 was treated with trypsin to remove the Bz‐Arg group successfully. Free or immobilized enzymes were used as catalysts. The effect of the acyl donor ester structure, the C(α) protecting group of the nucleophile, reaction media, enzyme, and the carrier of the enzymes on the outcome of the coupling reaction was studied.  相似文献
