Acetals and ethers,XVII. One- or two-step syntheses of 2-(2-alkoxyethyl)-1,3-dioxacyclanes

2-(2-Alkoxyethyl)-1,3-dioxanes (1) were prepared by ap-toluenesulfonic acid-catalyzed, one-step reaction of propenal with a mixture of aliphatic alcohol and trimethylene glycol in good yields. The transacetalization reaction of 1,1,3-trialkoxypropanes (3) with ethylene glycol or propylene-(1,2)glycol afforded good yields of pure 2-(2-alkoxyethyl)-1,3-dioxolanes (5 or6), respectively. This reaction proceeds through an intermediate 1,3-dialkoxy-1-(2-hydroxyalkoxy)-propane.

Ein- oder Zweistufensynthese von 2-(2-Alkoxyethyl)-1,3-dioxacyclanen

Zusammenfassung In der durchp-Toluolsulfonsäure — katalysierten, direkten Reaktion von Propenal mit einem Gemisch von aliphatischem Alkohol und Trimethylenglykol wurden die entsprechenden 2-(2-Alkoxyethyl)-1,3-dioxane (1) in guten Ausbeuten erhalten. Die Umacetalisierung von 1,1,3-Trialkoxypropanen (3) mit Ethylenglykol oder 1,2-Propylenglykol lieferte 2-(2-Alkoxyethyl)-1,3-dioxolane (5 oder6) in guten Ausbeuten. Die Umacetalisierungsreaktion von 1,1,3-Trialkoxypropanen verläuft über 1,3-Dialkoxy-1-(2-hydroxyalkoxy)-propane als Zwischenprodukte.

     

Interaction of benzimidazole-2-thione with propargyl bromide and 1,3-dibromopropyne

Interaction of benzimidazole-2-thione with propargyl bromide under found conditions permitted the direct synthesis of 2-(2-propynylsulfanyl)-3H-1,3-benzimidazolium bromide, the base 2-(2-propynylsulfanyl)-1H-1,3-benzimidazole, stable crystalline 2-(1,2-propadienylsulfanyl)-1H-1,3-benzimidazole, and 3-methyl[1,3]thiazolo[3,2-a][1,3]benzimidazole. The reaction of benzimidazole-2-thione with 1,3-dibromopropyne in chloroform or absolute methanol gave 2-(3-bromo-2-propynylsulfanyl)-3H-1,3-benzimidazolium bromide, and in absolute methanole in the presence of a twofold excess of sodium methoxide the reaction proceeded stereo-and regioselectively to give 3-[(Z)-bromomethylidene][1,3]thiazolo[3,2-a][1,3]benzimidazole. 1-(2-Quinoxalyl)-, 1-[3,5-Di(trifluoromethyl)phenyl]-, 1-(2-Carboxyphenyl)-, and 1-Ethoxycarbonyl-4-oxo-4,5,6,7-tetrahydroindazoles

The corresponding 1-(2-quinoxalyl)-, 1-[3,5-di(trifluoromethyl)phenyl]-, and 1-ethoxycarbonyl-3-methyl-4-oxo-4,5,6,7-tetrahydroindazoles have been obtained from reactions of 2-acetyl-1,3-cyclohexanedione, its 5,5-dimethyl and 5-(2-furyl) derivatives, with 2-hydrazinoquinoxaline, 3,5-di(trifluoromethyl)phenylhydrazine, and ethoxycarbonylhydrazine. On interaction with ethoxycarbonylhydrazine the intermediate 2-[1-(-ethoxycarbonyl)hydrazino]ethylidene-1,3-cyclohexanediones were also isolated. From the potassium salt of 2-formyldimedone and 2-carboxyphenylhydrazine hydrochloride, 2-(2-carboxyphenyl)hydrazinomethylene-5,5-dimethyl-1,3-cyclohexanedione was obtained, the cyclization of which in ethanol in the presence of HCl led to 1-(2-carboxyphenyl)- and 1-(2-ethoxycarbonylphenyl)-6,6-dimethyl-4-oxo-4,5,6,7-tetrahydroindazole.     

Synthesis of 3-[2-(1,3-butadienyl)]-1H-indoles en route to murrapanine analogue

A three-step procedure has been developed for the synthesis of 3-[2-(1,3-butadienyl)]-1H-indoles. TBAF was proved to be an effective reagent for dehydrobromination and carbomethoxy deprotection in one step to give 3-[2-(1,3-butadienyl)]-1H-indoles from the corresponding bromo-derivatives. Suitably substituted indolyl-1,3-butadiene has been successfully applied to prepare murrapanine analogue via Diels-Alder reaction.     

Synthesis of 2-oxazolidinones from (1S,2S)-2-amino-1-(4-nitrophenyl)-1,3-propanediol

Synthesis and X-ray diffraction study of 5-(8-methoxy-2-methyl-6,7-methylenedioxy-1,2,3,4-tetrahydroisoquinolin-1-yl)-1,3-dimethylbarbituric acid

The reaction of cotarnine with 1,3-dimethylbarbituric acid afforded 5-(8-methoxy-2-methyl-6,7-methylenedioxy-1,2,3,4-tetrahydroisoquinolin-1-yl)-1,3-dimethylbarbituric acid. Its crystal structure was established by X-ray diffraction analysis and the structure in solution was studied by ¹H NMR spectroscopy. This compound has a zwitterionic structure. In the crystal, the molecules are linked in dimers by intermolecular hydrogen bonds, and in solutions, the dimers occur in equilibrium with the monomers.     

α-Ureidoalkylation of 1,3-bis(hydroxymethyl)-imidazolidin-2-one

The α-ureidoalkylation of imidazolidine-2,4-dione, urea, carboxylic acid amides, and sulfonamides has been studied using 1,3-bis(hydroxymethyl)-imidazolidin-2-one as ureidoalkylating agent. Reactions of cyclic 1,3-dicarbonyl compounds with 1,2(1,4)-dihydro-1-methyl-2(4)-methylene-N-heterocycles. A new access to 6,12-methano-dibenz[d,g]-[1,3]oxazocinones

Summary The enamine-type methylene-N-heterocycles1–5 react with cyclic 2-ethoxymethylene-1,3-dicarbonyl compounds6 to give 2-[2-(hetarylidene)ethylidene]-1,3-dicarbonyl compounds7–14. The result of the reactions between 1,2-dihydro-1-methyl-2-methylene-quinoline (1a) and cyclic 1,3-dicarbonyl compounds depends on the nature of the dihydro intermediatesA/B. Dehydrogenation of keton intermediatesA results in 2-(1,2-dimethyl-4(1H)-quinolylidene)-1,3-dicarbonyl compounds17–21. Reaktionen cyclischer 1,3-Dicarbonylverbindungen mit 1,2(1,4)-Dihydro-1-methyl-2(4)-methylen-N-heterocyclen. Ein neuer Zugang zu 6,12-Methano- dibenz[d,g][1,3]oxazocinonen

Zusammenfassung Aufgrund ihres Enamincharakters reagieren die Methylen-N-heterocyclen1–5 mit cyclischen 2-Ethoxymethylen-1,3-dicarbonylverbindungen6 zu den 2-[2-(Hetaryliden)ethyliden]-1,3-dicarbonylverbindungen7–14. Das Ergebnis der Reaktionen zwischen 1,2-Dihydro-1-methyl-2-methylen-chinolin (1a) und cyclischen 1,3-Dicarbonylverbindungen hängt von der Natur der zwischenzeitlich entstehenden DihydroverbindungenA/B ab. Die Intermediat-KetoneA gehen durch Dehydrierung während der Reaktion in die 2-(1,2-Dimethyl-4(1H)chinolyliden)-1,3-dicarbonylverbindungen17–21 über. Die Intermediat-EnoleB mit sechsgliedrigem Dicarbonylring bilden in intramolekularer Reaktion die 6,12-Methano-dibenz[d,g][1,3]oxazocinone22–25, deren Struktur am Beispiel der Verbindung23 durch¹H-NMR sowie durch Röntgenkristallstrukturanalyse bewiesen wird.

     

Laser-enhanced ionization and laser-induced atomic fluorescence as element-specific detection methods for gas chromatography. Application to organotin analysis

The applicability of (1R,2R)-1,3-diacetoxy-1-(4-nitrophenyl)-2-propyl isothiocyanate [(R,R)-DANI] as a recently developed chiral derivatizing agent for the enantioseparation of a series of β-blockers is described. The thiourea diastereomers formed were analyzed by reversed-phase high-performance liquid chromatography, mixtures of water and methanol or acetonitrile being used for elution. Conditions of derivatizations (temperature, reagent excess and reaction time) were optimized, and the effects of organic modifiers on the retention and separation were investigated; the diastereomers could readily be baseline separated with methanol-containing mobile phases with resolutions between 1.58 and 2.72.     

Reactions of 1,3-bis(ω-bromoalkyl)-6-methyluracils with 2-(dialkylamino)ethylphosphonates and 2-(dialkylamino)ethyl phosphates

1,3-Bis(4-bromobutyl)-6-methyluracil reacts with diethyl 2-(dimethylamino)ethylphosphonate to form a bisquaternary ammonium salt, whereas the reaction of 1,3-bis-(6-bromohexyl)-6-methyluracil with diethyl 2-(diethylamino)ethyl phosphate gives 1,3-bis-[(6-diethoxyphosphoryloxy)hexyl]-6-methyluracil and 1,1,4,4-tetraethylpiperazinium dibromide. Structure elucidation of acetylation products of 2-acyl-1,3-indanediones by correlation of infrared spectral data

Summary 2-(1-Acetoxyalkylidene)- and 2-(1-acetoxybenzylidene)-1,3-indanediones (1a–1e) were proven to be the products of acetylation of 2-acyl-1,3-indanediones (2a–2e) by ketene using a detailed investigation and correlation analysis of infrared spectral data as well as¹H-NMR and¹³C-NMR spectra. Study by means of CNDO/2 and MMPI methods also demonstrates that the structure1 is more stable as the alternative one of 2-acyl-3-acetoxy-2-indene-1-ones (5). It was shown that the recently proposed general correlations v(C=O)_s vs. v(C=O)_as and v(C=O) vs. X⁺(R) as well as the mechanical anharmonicities of asymmetric C=O stretching vibration can be successfully used as a tool of structural diagnostics of cyclic 1,3-dicarbonyl compounds.

Strukturaufklärung von Acetylierungsprodukten von 2-Acyl-1,3-indandionen mittels Korrelation von Infrarot-Daten

Zusammenfassung 2-(1-Acetoxyalkyliden)- und 2-(1-Acetoxybenzyliden)-1,3-indandione (1a–1e) wurden mittels einer detaillierten infrarot-spektroskopischen Untersuchung (IR-Korrelation) und¹H-NMR und¹³C-NMR Spektroskopie als Acetylierungsprodukte von 2-Acyl-1,3-indandionen (2a–2e) mit Keten nachgewiesen. CNDO/2- und MMPI-Rechnungen zeigten auch, daß Struktur1 stabiler ist, als die der alternativen 2-Acyl-3-acetoxy-2-inden-1-one5. Es wird gezeigt, daß die kürzlich vorgeschlagenen allgemeinen Korrelationen v(C=O)_s gengen v(C=O)_as und v(C=O) gegen X ⁺(R) und auch die mechanischen Anharmonizitäten der asymmetrischen C=O Streckschwingung erfolgreich als Werkzeug zur Strukturaufklärung cyclischer 1,3-Dicarbonyl-Verbindungen eingesetzt werden können.

     

Synthesis of novel 3-(1,3-thiazol-2-yl)-7,8-dihydroquinoline-2,5(1H,6H)-diones

An efficient method for the synthesis of novel 3-(1,3-thiazol-2-yl)-7,8-dihydroquinoline-2,5(1H,6H)-diones from various 2-dimethylaminomethylidenecyclohexane-1,3-diones, (1,3-thiazol-2-yl)acetonitriles, and dimethylformamide dimethyl acetal was developed. These transformations proceeded through intermediate 2-[2-(4-aryl-1,3-thiazol-2-yl)-2-cyanoethenyl]-3-oxocyclohex-1-en-1-olates. They were isolated as piperidinium salts and used in further heterocyclization reactions with aromatic amines, giving novel 1-aryl-3-(1,3-thiazol-2-yl)-7,8-dihydroquinoline-2,5(1H,6H)-diones. These compounds were also obtained by preparative three-step “one pot” synthesis under controlled microwave irradiation. Published in Russian in Izvestiya Akademii Nauk. 选取模型化合物,用AMl MO方法从理论上研究了手性的5-(L-盖氧基)-2(5—H)-呋喃酮与偶极硝酮体系环加成反应的机理。优化了反应过程各驻点的几何构型,计算了反应的活化焓、活化熵和反应速率常数,结果表明,该反应为同面的协同过程,非对映专一性产物的形成估计是由于盖氧基及硝酮分子上苯取代基的空间效应所致。     

Synthesis and structure of 1,3-dialkyl-4-(sulfonylimino)imidazolidin-2-ones

The reactions of 1,3-dialkyl-4,5-dihydroxyimidazolidin-2-ones with alkyl sulfamides have been studied and previously undescribed 1,3-dialkyl-4-(alkylaminosulfonylimino)imidazolidin-2-ones have been obtained. CeCl3·7H2O catalyzed C(sp2)−CN bond construction on water: Synthesis of (Z)-2-(2-Oxoindolin-3-ylidene)-2-arylacetonitriles

An environmentally friendly method for the construction of C(sp²)–CN bond on water has been described, and (Z)-2-(2-oxoindolin-3-ylidene)-2-arylacetonitriles were synthesized by using CeCl₃· 7H₂O as a catalyst. The reaction offers access to alkenyl nitriles in good-to-excellent yield. The investigations will be beneficial to reduce the use of toxic organic solvents and explore the efficient catalytic processes on water.     

Synthesis,characterization, and DNA-binding of a four-coordinate cobalt(II) complex with 1,3-bis(1-ethylbenzimidazol-2-yl)-2-oxopropane

A new four-coordinate cobalt(II) complex with 1,3-bis(1-ethylbenzimidazol-2-yl)-2-oxopropane (Etobb), Co(Etobb)Cl₂, has been synthesized and characterized by elemental analysis, electrical conductivities, infrared, and UV-Vis spectral measurements. The crystal structure has been determined by single-crystal X-ray diffraction. Cobalt(II) is a distorted tetrahedral geometry, surrounded by two nitrogens from Etobb and two chlorides. DNA-binding properties of Etobb and its Co(II) complex have been investigated by electronic absorption, fluorescence, and viscosity measurements. The experimental results suggest that the ligand and its Co(II) complex bind to DNA via intercalation, and the binding affinity of the Co(II) complex to DNA is greater than Etobb.     

Synthesis of 3-[4-(1-Benzofuran-2-yl)-1,3-thiazol-2-yl]-2-(4-aryl)-1,3-thiazolidin-4-one Derivatives as Biological Agents

A protocol for the synthesis of 3-[4-(1-benzofuran-2-yl)-1,3-thiazol-2-yl]-2-(4-aryl)-1,3-thiazolidin-4-one derivatives (5a–e) has been developed from 1-(1-benzofuran-2-yl)-2-bromoethanone (2),which served as a key intermediate for the synthesis of the title compounds. The reaction of compound 2 with thiourea furnished 4-(1-benzofuran-2-yl)-1,3-thiazol-2-amine 3, which upon further reaction with various aromatic aldehydes, gave Schiff bases 4a–e. These Schiff bases, when treated with thioacetic acid in the presence of catalytic amount of anhydrous ZnCl₂, yielded thiazolidinone derivatives 5a–e. All the newly synthesized compounds have been characterized by analytical and spectral data and screened for their antimicrobial and analgesic activity.

新型2-(2'-取代亚肼基)-1,3-二硫杂环戊烷衍生物的合成和菌活性研究

通过结构改造并结合活性基团的拼接合成了11个未见文献报道的新型2-(2'-取代亚肼基)-1,3-二硫杂环戊烷类化合物, 所有的目标物的结构都经¹H NMR, MS, IR和元素分析证实, 选择了其中5个化合物对7种细菌进行了抑菌活性测试, 并与市售农药20%三环唑、70%威尔达甲托的抑菌活性进行对比, 发现它们比市售农药有更为广泛的杀菌谱, 对照农药只对金黄色葡萄球菌有良好的抑菌作用而这些化合物则对所选择的七个菌种均有良好的抑菌作用. 其中化合物2-(对氟苯氧乙酰亚肼基)-1,3-二硫杂环戊烷(4c)的杀菌谱很广, 它对金色葡萄球菌、大肠杆菌、白色念珠菌的抑杀活性都很好.
