The synthesis and structure-activity-relationship (SAR) for a series of N-substituted piperazinyl carbamoyl 7-15 and piperazinyl acetyl 18-26 derivatives of tetrahydropapaverine have been carried out. The general synthetic methods of carbamoyl tetrahydropapaverine analogues involve N-substituted piperazines and carbamoyl imidazole tetrahydropapaverine as starting materials. Another route for synthesizing these compounds, involving the formation of carbamoyl imidazole piperazine has also been explored. Acylation of tetrahydropapaverine followed by substitution with various piperazinyl moities afforded the acetyl tetrahydropapaverine derivatives. Variously substituted piperazines have been used to monitor the effect of electron releasing and electron withdrawing substituents upon the antispasmodic activity of the molecules. Effect of varying electron densities on the antispasmodic activity, by altering the position of these groups on the benzene ring has also been monitored. Pharmacological methods involve the in vitro antispasmodic activity studies on a freshly removed guinea pig ileum using a force displacement transducer amplifier connected to a physiograph. Among the analogues synthesized in the present study, a promising compound 7, a potent muscle relaxant as compared to papaverine has been obtained.  相似文献

Carbamoyl phosphonate derivatives of methacrylic acid and α-methyl styrene were synthesized in good yield by reacting dimethyl and diethylphosphite with isocyanato ethylmethacrylate (IEM) or with meta-isopropenyl-α,α-dimethylbenzylisocyanate (m-TMI). The IEM derivatives yield soluble homopolymers with common radical initiators. The m-TMI carbamoyl phosphonates were copolymerized with styrene and with acrylates yielding soluble materials with better than 50 mol % incorporation of the phosphonate monomer. The side chain carbamoyl phosphonate group drastically lowers the Tg relative to those of the parent polymers. Thermogravimetric studies suggest the NHC(O)? P(O)(OR)2 bond to be stable to about 225°C. The radiopacifying salt UO2(NO3)26 H2O yields transparent films with the phosphonate-containing polymers. The structures of 2‐[N‐(2‐chlorophenyl)carbamoyl]benzenesulfonamide and 2‐[N‐(4‐chlorophenyl)carbamoyl]benzenesulfonamide, both C13H11ClN2O3S, are stabilized by extensive intra‐ and intermolecular hydrogen bonds. In both structures, sulfonamide groups are hydrogen bonded via the N and O atoms and form chains of molecules. The carbamoyl groups are also hydrogen bonded, involving the O and N atoms, further strengthening the polymeric chains running along the c and a axes in the 2‐ and 4‐chloro derivatives, respectively. Carbamoylsulfonamide derivatives are novel compounds with a great potential for medicinal applications.  相似文献

A method for producing tri- and tetrasubstituted ureas from carbamoyl imidazolium salts is presented. Carbamoyl imidazolium salts are prepared from the reaction of N,N carbonyldiimidazole (CDI) with secondary amines, followed by alkylation with iodomethane. These stable salts can be stored for extended periods and are effective electrophilic carbamoylation reagents. Primary and secondary amines add to carbamoyl imidazolium salts at room temperature to give tri- and tetrasubstituted ureas in excellent yields. This reaction was used to synthesize ureas using both liquid-liquid extraction and solid-phase extraction (cation exchange) purification techniques. Liquid-liquid extraction affords the product ureas more cleanly than cationic exchange. A series of urea compounds were synthesized using parallel synthesis techniques in high yields and with suitable purity for routine in vitro biological tests. These studies validate the utility of carbamoyl imidazolium salts as useful building blocks for combinatorial library synthesis.  相似文献

The Heterolytic Fragmentation of Benzoin-O-(carbamoyl)oximes While the known heterolytic fragmentation reactions give only three, thermal decomposition of benzoin-O-(carbamoyl)oximes results in at least four fragments: nitrile or isocyanide, carbonyl compound, CO2 and amine. This exception is due to the transformation of the nucleofugal group 3 into the unstable carbamic acid and its decomposition (s. Scheme 1). Since only the configuration of benzoin (E)-O-(carbamoyl)oximes is satisfactory for concerted reactions, we conclude that the nitrile producing fragmentation of these (E)-compounds is concerted, whereas in the isocyanide producing fragmentation of the corresponding (Z)-compounds several steps are involved. – In contrast to the benzoin-O-(carbamoyl)oximes the pyrolysis of benzil-(E)-O-(methylcarbamoyl)oxime starts with the elimination of methyl isocyanate and the following fragmentation is that of the oxime.  相似文献

The structures of 2‐[(2,3‐dimethylphenyl)carbamoyl]benzenesulfonamide, 2‐[(3,4‐dimethylphenyl)carbamoyl]benzenesulfonamide and 2‐[(2,6‐dimethylphenyl)carbamoyl]benzenesulfonamide, all C15H16N2O3S, are stabilized by extensive intra‐ and intermolecular hydrogen bonds. In all three structures, the sulfonamide and carbamoyl groups are involved in hydrogen bonding. In the 2,3‐dimethyl and 2,6‐dimethyl derivatives, dimeric units and chains of molecules are formed parallel to the c axis. In the 3,4‐dimethyl derivative, the hydrogen bonding creates tetrameric units, resulting in macrocyclic R44(22) rings that form sheets in the ab plane. The three analogues are closely related to the fenamate class of nonsteroidal anti‐inflammatory drugs.  相似文献

Re(CO)2(NO)(PPh3)2 reacts with aroyl azides RCON3 (R = C6H5, p-CH3C6H4) in benzene to form isocyanate complexes of formula Re(CO)(NO)-(PPh3)2(RCONCO) (I). When the reaction is carried out in protic solvents such as ethanol, carbamoyl derivatives of formula Re(NCO)(NO)(PPh3)2-(CONHCOR) (II) are obtained, which give Re(NCO)(NO)(PPh3)2(CO)(NHCOR) when dissolved in chloroform, a terminal carbonyl ligand being formed from the carbamoyl group.I can be transformed into II by reaction with gaseous HCl, via [Re(CO)-(NO)(PPh3)2 {C(OH)=NCOR}]+Cl- followed by anion exchange with NaN3. II reacts with mineral acids HX (X = Cl, BF4) to give amide derivatives of formula [Re(NCO)(NO)(PPh3)2(CO)(NH2COR)]+ X- which when X = Cl can be easily transformed into Re(NCO)(NO)(PPh3)2(CO)Cl, the amide ligand being removed. Both the protonation reactions of I and II are reversible. IR and 1H NMR data of the new compounds and the mechanisms of formation of I and II are reported and discussed.  相似文献

A simple, efficient, and general method has been developed for the synthesis of carbamoyl and thiocarbamoyl phosphonic esters using CaCl2 as an efficient Lewis base catalyst. Carbamoyl and thiocarbamoyl phosphonic esters were obtained in good yield (37%–65%) and purity under mild conditions by the reaction of diethyl phosphite with isocyanates and isothiocyanates in the presence of CaCl2. This method is easy, rapid, and good‐yielding reaction for the synthesis of carbamoyl and thiocarbamoyl phosphonic esters. © 2009 Wiley Periodicals, Inc. A green and simple method has been developed for efficient preparation of diverse annulated 2-amino-3-cyano-4H-pyran derivatives in the presence of a low loading of tetraethylammonium 2-(carbamoyl)benzoate (TEACB), as a bifunctional organocatalyst, under solvent-free conditions using the ball milling technique. This procedure is a clean, transition-metal-free, and environmentally friendly approach that offers many advantages including short reaction times, high to quantitative yields, low cost, and straightforward work-up.  相似文献

Molecular structure and vibrational frequencies of carbamoyl azide NH2CO-NNN have been investigated with ab initio and density functional theory (DFT) methods. The molecular geometries for all the possible conformers of the molecule were optimized using DFT-B3LYP, DFT-BLYP and MP2 applying the standard 6-311++G** basis set. From the calculations, the molecule was predicted to exist predominantly in cis conformation with the cis-trans rotational barrier of about 7.91-9.10 kcal/mol depending on the level of theory applied. The vibrational frequencies and the corresponding vibrational assignments of carbamoyl azide in Cs symmetry were examined theoretically and the calculated Infrared and Raman spectra of the molecule in the cis conformation were plotted. Observed frequencies for normal modes were compare with those calculated from normal mode coordinate analysis carried out on the basis of ab initio and DFT force fields using the standard 6-311++G** basis set of the theoretical optimized geometry. Theoretical IR intensities and Raman activities are reported.  相似文献

We present an efficient,fast and simple strategy of generating the intermediate carbamoyl chlorides from secondary amines using stoichiometric amounts of bis(trichloromethyl)carbonate(BTC) in solution and solvent-free conditions with excellent yields.The results obtained showed the yield increasing on whether a base was used.Finally,an efficient and rapid synthesis of variety carbamate derivatives was developed by the reaction with a high variety of different alcohols,phenols,diols and this intermediate at room temperature with grinding and in solvent-free conditions under microwave irradiation.The presence of various safe bases is shown to be effective in reducing the reaction times,increasing the yields and easing purification.The present method does not involve any hazardous phosgene.  相似文献

Reduction of tri-O-acetyl-beta-L-fucopyranosyl cyanide with Pd-hydrogen, in the presence of Ac(2)O and Boc(2)O, gave N-protected-mono- and -di-(2,3,4-tri-O-acetyl-beta-L-fucopyranosylmethyl)-amines, which allow for the syntheses of small cluster oligosaccharide mimetics of fucopyranosylomethyl-substituted ureas. From di-(2,3,4-tri-O-acetyl-beta-L-fucopyranosylmethyl) amine was also prepared a carbamoyl chloride as a potentially useful synthon for preparation of more complex C-glycosidic conjugates.  相似文献

An efficient and convenient access to 2-tert-butylisoindolin-1-ones via an oxidative radical cyclization process from stable carbamoylxanthates, derived from secondary tert-butylamines, is described. The proposed mechanism for this transformation involves, the generation of a carbamoyl radical, its cyclization to the aromatic system, and the dilauroyl peroxide (DLP) mediated rearomatization to generate the isoindolinone ring system. Additionally, the syntheses of cichorine and 4-hydroxyisoindolin-1-one natural products were carried out to underscore the synthetic potential of this xanthate-based carbamoyl radical-oxidative cyclization.  相似文献

Efficient and rapid preparations of carbamoyl chlorides and unsymmetrical ureas from tertiary amines and phosgene or its safe equivalent triphosgene [bis(trichloromethyl)carbonate, BTC] are described. First, the reaction of stoichiometric amounts of phosgene with secondary amines was revisited, and it was shown that the formation of carbamoyl chlorides in high yields required careful adjustments of experimental conditions and the use of pyridine as an HCl scavenger. A phosgene-mediated dealkylation of triethylamine was observed when this base was used instead of pyridine. Taking advantage of this observation, a strategy of synthesis of carbamoyl chlorides from tertiary amines and phosgene has been developed. N-Alkyl-N-benzyl(substituted)tetrahydroisoquinolines, -piperazines, -piperidines, or -anilines were treated with stoichiometric amounts of phosgene (or BTC) in CH(2)Cl(2). Tertiary amines bearing electron-enriched benzyl group(s) afforded carbamoyl chlorides in excellent yields and without any contamination by symmetrical ureas. Subsequent additions of primary or secondary amines to these carbamoyl chlorides produced unsymmetrical ureas in single-pot and high-yielding operations. This methodology was applied in (11)C-chemistry. From [(11)C]phosgene, a common precursor used in the preparation of radiotracers for positron emission tomography, a rapid and efficient synthesis of (11)C-carbamoyl chlorides and (11)C-unsymmetrical ureas derived from tetrahydroisoquinoline and piperazine is described. The first example of (11)C-amide formation from the reaction of a (11)C-carbamoyl chloride and an organometallic (cyanocuprate or a Grignard reagent in the presence of a nickel catalyst) is also presented.  相似文献
