Poly(lactide-co-glycolide) (PLGA) nanoparticles (NPs) with surface modification of heparin were fabricated by microemulsion–diffusion method. These novel colloidal particles were stabilized by lecithin and Tween 80. The effects of lecithin on the loading of heparin onto PLGA NPs and on the surface conductance were analyzed. The electronic micrographs revealed that spherical colloids were prepared and the incorporation of heparin caused a slight coalescence of the particles. In addition, the average diameter of heparin-modified PLGA NPs was between 70 and 220 nm. An increase in the weight percentage of lecithin or in the concentration of heparin enlarged the average diameter. Based on constant amount of surfactants, the loading efficiency of heparin on the particle surfaces reached a maximum when the weight percentage of lecithin was 50%. Moreover, the surface conductance of heparin-modified PLGA NPs was improved by an increased weight percentage of lecithin. A high concentration of heparin in microemulsion also promoted the loading efficiency and surface conductance of heparin-modified PLGA NPs. 相似文献

The objective of this study was to evaluate degradation behavior and the feasibility of biodegradable polymeric stents in common bile duct (CBD) repair and reconstruction. Various molar ratios of lactide (LA) and glycolide (GA) in poly(l-lactide-co-glycolide) (PLGA) were synthesized and processed into a circular tubing of ∼10.0 mm outer diameter and a wall thickness of about 2.0 mm. This tubing was cut into 40.0 mm length to form CBD stents. The stents were placed into human bile to determine the degradation behavior in vitro. The morphology, configuration, mass loss, water uptake, molecular weight and composition changes were examined. The PLGA with LA/GA = 71/29 exhibited an acceptable degradation life and was chosen as an in vivo stent material. These PLGA stents were used in common bile duct exploration (CBDE) and primary suturing for rats. Degradation status of the stents was examined and comparison was made between those before and after surgical procedure. The results showed that the polymer stents exhibited the same biomedical functions as T tubes and spontaneously disappeared from CBD in 4-5 weeks. Therefore, the PLGA stents fits the requirements in repair and reconstruction of CBD, to support the duct, guide bile drainage and reduce T-tube-related complications. 相似文献

The aim of the present work was to analyse the degradation rate of PGLA copolymer depending on a modifier (hydroxyapatite, carbon fibres) under in vivo conditions (rabbit femoral bone). Also, the influence of the implantation site on the degradation rate of a copolymer (rabbit mandible and femoral bone) was analyzed as a continuation of our previous research.The structural and phase changes of poly(lactide-co-glycolide) and its composite with hydroxyapatite and carbon fibres were determined on the basis of IR and NMR spectroscopy. Additionally, microscopic observations with elemental analyses were performed (SEM, EDS).The addition of a modifying phase accelerates implant degradation. However, modifying additives promote regeneration of the treated bone tissue simultaneously with faster polymer degradation. The compositions of a copolymer and composites are variable during degradation.The process of copolymer degradation in the femoral bone is much faster in comparison with previous studies on mandibles. This may be caused by higher activity of osteoblasts, differences in blood supply and oxygenation of tissue as well as different anatomical structure of these two kinds of bones.FTIR and NMR methods allow for following the changes occurring in the resorbable copolymer structure with time and they are very useful tools in analyzing the degradation mechanism and rate of aliphatic polyesters and their composites. 相似文献

Although poly(lactic acid) is known as a biodegradable polymer, its hydrolytic degradation is extremely slow, taking years in water and in the human body. In this study the effects of blending oligomeric poly(aspartic acid-co-lactide) (PALs) on the hydrolytic degradation of poly(l-lactic acid) (PLLA) were studied in detail. It was found that the addition of PAL did not accelerate the hydrolysis of the PLLA in air (25 °C, 60% relative humidity), but significantly accelerated it in a phosphate buffer solution. The degradation rate becomes higher for the blends containing PAL with higher molar ratios of lactide to aspartic acid units, [LA]/[Asp], when PLLA/PAL blends prepared with different PALs are compared at the same PAL concentration. TEM results, in which the distribution of PALs with higher [LA]/[Asp] occurs at a smaller scale in blends, imply that higher miscibility of the PAL with PLLA results in higher contact area between the components, thereby accelerating the degradation efficiently. 相似文献

Films of poly(l-lactic acid) (PLLA) with different number-average molecular weights (Mn) and d-lactide unit contents (Xd) were made amorphous and the effects of molecular weight and small amounts of d-lactide units on the hydrolytic degradation behavior in phosphate-buffered solution at 37 °C of PLLA were investigated. The degraded films were investigated using gravimetry, gel permeation chromatography, polarimetry, differential scanning calorimetry, X-ray diffractometry, and tensile testing. To exclude the effects of crystallinity on the hydrolytic degradation, the films were made amorphous by melt-quenching. The incorporation of small amounts of d-lactide units drastically enhanced the hydrolytic degradation of PLLA. In the period of 0-32 weeks, the hydrolytic degradation rate constant (k) of PLLA films increased with increasing Xd, while the k values did not depend on Mn. This means that the effects of Xd on the hydrolytic degradation rate of the films are higher than those of Mn. In contrast, in the period of 32-60 weeks neither Xd nor Mn was a crucial parameter to determine k values, probably because in addition to these parameters the differences in the amount of catalytic oligomers accumulated in films and crystallinity affect the hydrolytic degradation behavior of the films. The initially amorphous PLLA films remained amorphous even after the hydrolytic degradation for 60 weeks. 相似文献

This paper reports the preparation of bionanocomposites based on poly(d,l-lactide) and cellulose nanowhiskers (PDLLA/CNWs) and studies the influence of the CNWs on the hydrolytic degradation behavior of the polylactide. The hydrolytic degradation process was studied in a phosphate buffer medium through the sample weight loss and also by FTIR, DSC and TGA measurements. The presence of CNWs induced a strong delay in the hydrolytic degradation of the PDLLA, even when the concentration of the nanofillers was only 1%. This effect was related to the physical barrier created by the highly crystalline CNWs that inhibited water absorption and hence retarded the hydrolytic degradation of the bionanocomposites. In addition, the incorporation of cellulose nanocrystals in the PDLLA also made the biopolymer more thermally stable, increasing the initial temperature of mass loss even after the degradation in phosphate medium. The results presented here show the possibility of controlling the biodegradability and prolonging the service life of a polylactide through the incorporation of a small quantity of nanofillers obtained from renewable materials. 相似文献

Low molecular weight poly(lactic acid) was synthesized by direct polycondensation of lactic acid. The oligomers were characterized by viscometry, light scattering, and gel permeation chromatography (GPC). The swelling behaviour of tablets made of the above polymer immersed in buffer solutions at 37 °C was studied. In the same experiments, the hydrolytic stability of d,l-PLA was assessed by measuring the weight loss after drying the tablets. In order to inhibit any degradation due to bacteria, formaldehyde was added in the solution as biostatic factor. The effect of an incorporated drug on the swelling behaviour of d,l-PLA tablets was also considered. It was found that the incorporation of drug in d,l-PLA tablets increases their swelling index, probably due to the creation of additional porosity in the specimens or other interaction between drug and polymeric matrix. 相似文献

Surface properties and enzymatic degradation of poly(l-lactide) (PLLA) end-capped with hydrophobic dodecyl and dodecanoyl groups were investigated by means of advancing contact angle (θa) measurement, quartz crystal microbalance (QCM) and atomic force microscopy (AFM). The θa values of end-capped PLLA films were larger than those of non-end-capped PLLA films, suggesting that the hydrophobic dodecyl and dodecanoyl groups were segregated on the film surface. The weight changes of end-capped PLLA thin films during enzymatic degradation in the presence of proteinase K were monitored by using a QCM technique. The relatively fast weight loss of PLLA film occurred during first few hours of degradation, followed by a decrease in the erosion rate. The erosion rate of PLLA films at the initial stage of degradation was dependent on the chain-end structure of PLLA molecules, and the value decreased with an increase in the amount of hydrophobic functional groups. The surface morphologies of PLLA thin films before and after degradation were characterized by AFM. After the enzymatic degradation, the surface of non-end-capped PLLA films was blemished homogeneously. In contrast, the end-capped PLLA thin films were degraded heterogeneously by the enzyme, and many hollows were formed on the film surface. From these results, it has been concluded that the introduction of hydrophobic functional groups at the chain-ends of PLLA molecules depressed the erosion rate at the initial stage of enzymatic degradation. 相似文献

Poly(d,l-lactide) (PDLLA) degraded at processing temperature under air and nitrogen. A random chain scission model was established and used to determine the activation energy Ea, and FT-IR, 1H and 13C NMR were used to elucidate the degradation behavior under different atmospheres. Results showed that there were two to three stages. The 1st stage was dominated by the oligomers containing carboxylic acid groups and hydroxyl groups, during which oxygen and nitrogen had little effect on the degradation, thus they share similar Ea. When the oligomers were consumed over or evaporated, the 2nd stage began, and oxygen had a promoting effect on the thermo-oxidation process, resulting in the great decrease in Ea. The third stage of PDLLA was observed when it degraded under nitrogen over 200 °C, which was caused by the appearance of carboxylic acid substance. 相似文献

Poly(l-lactide) (PLLA) was melt-blended with poly(p-vinyl phenol) (PVPh) using a two-roll mill, and the miscibility between PLLA and PVPh and degradation of the blend films were investigated. It was found that PLLA/PVPh blend has miscibility in the amorphous state because only single Tg was observed in the DSC and DMA measurements. The Tg of the PLLA/PVPh blend could be controlled in the temperature range from 55 °C to 117 °C by changing the PVPh weight fraction. In alkaline solution, degradation rate of PLLA/PVPh blends was faster than that of neat PLLA because PVPh could dissolve in alkaline solution. The surface morphology of degraded PLLA and PLLA/PVPh blend were observed by SEM. The surface morphology of degraded PLLA/PVPh blend was finer than that of PLLA. Young's modulus of PLLA/PVPh blend increased with increasing PVPh content. Yield stress of PLLA/PVPh blends whose PVPh content was less than 30 wt% kept the level of about 55 MPa and that of PLLA/PVPh blend whose PVPh content was 40 wt% is much lower than that of neat PLLA. 相似文献

The surfaces of poly(l-lactide) (PLLA) microspheres were modified by chitosan via a method of hydrolysis and grafting-coating to improve their compatibility to chondrocytes. The PLLA microspheres with a diameter of 74-150mum were fabricated by an oil/water emulsion solvent evaporation method, followed by hydrolysis in alkaline solution to produce a larger number of carboxyl groups. Using water-soluble carbodiimide as a coupling reagent, chitosan was covalently grafted onto the microspheres. Due to the physical entanglement and insolubility at neutral pH, unbonded chitosan molecules were stably remained to yield a large amount of coated chitosan. Biological performance of the control PLLA and the chitosan-coated PLLA microspheres were assessed by in vitro culture of rabbit auricular chondrocytes. After 24h and 7d culture, the chitosan-coated PLLA microspheres, especially the ones with larger chitosan amount, exhibited stronger ability to promote cell attachment and proliferation, and maintain the secretion function of the chondrocytes. Therefore, the chitosan-coated PLLA microspheres can be potentially used as the injectable cell microcarriers for chondrogenesis in cartilage tissue engineering. 相似文献

Hyperbranched poly(amido amine)s containing vinyl and hydroxyl groups were successfully synthesized via Michael addition polymerization of triacrylamide (TT) and 3-amino-1,2-propanediol (APD) with equal molar ratio in feed. 1H, 13C and HSQC NMR techniques were used to clarify the structure of hyperbranched polymers and polymerization mechanism. The Michael addition reaction of hyperbranched poly(1TT-1APD)s with primary amine-terminated poly(ε-benzyloxycarbonyl-l-lysine)s [PLys(Z)] yielded a star-like hyperbranched polymers with poly(1TT-1APD) core and Plys(Z) shell. The Z groups in PLys(Z) were removed under acidolysis, and thus star-like hyperbranched polymers with hydroxyl groups inside and primary amine groups outside were obtained successfully. 相似文献

This article contains a detailed analysis of the crystallization behavior of poly(l-lactic acid) (PLLA). Crystallization rates of PLLA have been measured in a wide temperature range, using both isothermal and non-isothermal methods. The combined usage of multiple thermal treatments allowed to obtain information on crystallization kinetics of PLLA at temperatures almost ranging from glass transition to melting point. Crystallization rate of PLLA is very high at temperatures between 100 and 118 °C, showing a clear deviation from the usual bell-shaped curve. This discontinuity has been ascribed to a sudden acceleration in spherulite growth, and is not associated to morphological changes in the appearance of PLLA spherulites. Experimental data of spherulite growth rates of PLLA have been analyzed with Hoffman-Lauritzen method. Applicability and limitations of this theoretical treatment have been discussed. 相似文献

To obtain details of poly(l-lactic acid) (PLLA) photodegradation behavior, PLLA films were irradiated by UV-C light (λ = 253.7 nm) to directly excite carbonyl groups, resulting in a rapid decrease in the molecular weight accompanying a gradual decrease in the optical purity of monomeric units in the chains. The racemization during the photodegradation was first detected as a result of the chain scission by irradiation. From quantitative analyses of the molecular weight and the monomeric unit composition, it was found that the chain scission ratio and the d-lactate unit ratio increased in parallel during the irradiation, suggesting that approximately one d-lactate unit formed for every chain scission. From a mechanistic consideration, the racemization equilibrium was proposed to occur at both carboxyl and hydroxyl chain ends. 相似文献

Triptolide (TP), which has immunosuppressive effect, anti-neoplastic activity, anti-fertility function and severe toxicities on digestive, urogenital, blood circulatory system, was used as a model drug in this study. TP-loaded poly (d,l-lactic acid) (PLA) nanoparticles were prepared by the modified spontaneous emulsification solvent diffusion method (modified-SESD method). Dynamic light scattering system (DLS), transmission electron microscope (TEM), atomic force microscopy (AFM), differential scanning calorimetry (DSC), X-ray powder diffractometry and Fourier transform infra-red spectroscopy (FT-IR) were employed to characterize the nanoparticles fabricated for size and size distribution, surface morphology, the physical state of drug in nanoparticles, and the interaction between the drug and polymer. Encapsulation efficiency (EE) and the in vitro release of TP in nanoparticles were measured by the reverse phase high-performance liquid chromatography (RP-HPLC). The produced nanoparticles exhibited a narrow size distribution with a mean size of approximately 150 nm and polydispersity index of 0.088. The morphology of the nanoparticles exhibited a fine spherical shape with smooth surfaces without aggregation or adhesion. TP-entrapped in nanoparticles was found in the form of amorphous or semicrystalline. It was found that a weak interaction existed between the drug and polymer. In all experiments, more than 65% of EE were obtained. The in vitro release profile of TP from nanoparticles exhibited a typical biphasic release phenomenon, namely initial burst release and consequently sustained release. In this case, the particle size played an important role for the drug release. The modified-SESD method was a potential and advantage method to produce an ideal polymer nanoparticles for drug delivery system (DDS). 相似文献

(Lactic acid, ethylene glycol, malonic or succinic acid) copolymers [(LA-EG-MA) and (LA-EG-SA) copolymers] were synthesized with different monomer feed ratios by direct polycondensation. The copolymers were characterized in terms of various properties such as acid value and number average molecular weight. The aerobic biodegradation under controlled composting conditions of commercially available and laboratory synthesized poly(l-lactic acid) (PLA) and synthesized copolymers was carried out according to ISO 14855-1:2005. The biodegradability of tested materials was found to be strongly dependent on the lactic acid content, ranging from 94% (method A) and 104% (method B) to 43% (method A) and 46% (method B) over the 110-days of the 50 °C composting. 相似文献

Biodegradable poly(l-lactic acid) (PLLA) nanofiber was prepared by a carbon dioxide (CO2) laser supersonic drawing which was carried out by irradiating the laser on an as-spun fiber in a supersonic jet. The supersonic jet was generated by blowing off air into a vacuum chamber from a fiber supplying orifice. The flow velocity from the orifice can be estimated by applying Graham’s theorem from the pressure difference between the atmospheric pressure and the pressure of the vacuum chamber. The fastest flow velocity estimated was 396 m s−1 when the chamber pressure was 6 kPa. The PLLA nanofiber having an average diameter of 0.132 μm was obtained when the supersonic drawing was carried out by irradiating the laser at 177 W cm−2 on the as-spun fiber supplied at 0.1 m min−1 in the vacuum chamber at 6 kPa. The obtained nanofiber had a draw ratio of about 323,000 and a degree of crystallinity of 45%, and its diameter uniformity was high. The CO2 laser supersonic drawing was a new route for preparation of various nanofibers without using any solvent. 相似文献

The hydrolytic reactions between various Pt(II) complexes of the type [Pt(L)Cl2] and [Pt(L)(CBDCA-O,O′] (L is ethylenediamine, en; (±)-trans-1,2-diaminocyclohexane, dach; (±)-1,2-propylenediamine, 1,2-pn and CBDCA is the 1,1-cyclobutanedicarboxylic anion) and the N-acetylated l-methionylglycine dipeptide (MeCOMet-Gly) were studied by 1H NMR spectroscopy. All reactions were realized at 37 °C with equimolar amounts of the Pt(II) complex and the dipeptide at pH 7.40 in 50 mM phosphate buffer in D2O. Under these experimental conditions, a very slow cleavage of the Met-Gly amide bond was observed and this hydrolytic reaction proceeds through the intermediate [Pt(L)(H2O)(MeCOMet-Gly-S)]+ complex. In general, it can be concluded that faster hydrolytic cleavage of the MeCOMet-Gly dipeptide was observed in the reaction with the chloride complex than with corresponding CBDCA Pt(II) complexes. The steric effects of the Pt(II) complex on the hydrolytic cleavage of the amide bond in the MeCOMet-Gly dipeptide were also investigated by 1H NMR spectroscopy. It was found that the rate of hydrolysis decreases as the steric bulk of the CBDCA and chlorido Pt(II) complexes increase (en > 1,2-pn > dach). These results contribute to a better understanding of the toxic side effects of Pt(II) antitumor drugs and should be taken into consideration when designing new potential Pt(II) antitumor drugs with preferably low toxic side effects. 相似文献

Novel butanediamine-grafted poly(dl-lactic acid) polymers (BDPLAs) were synthesized via a series of chemical bulk modifications in this study. Briefly, maleic anhydride (MAH) was first grafted onto the side chain of poly(dl-lactic acid) (PDLLA) molecules via melt free radical copolymerization using benzoyl peroxide (BPO) as initiator to get maleic anhydride-grafted PDLLA polymers (MPLAs); thereafter butanediamine (BDA) was immobilized onto grafted anhydride groups in MPLAs via N-acylation reaction to obtain the desired BDPLAs. Gel permeation chromatography with multi-angle laser light scattering (GPC-MALLS), FT-IR, 13C NMR and XPS were employed to qualitatively characterize these synthesized polymers. Rhodamine-carboxyl interaction method and ninhydrin reaction were further used to quantitatively determine the graft ratio of MAH (MAH%) in MPLAs and the graft ratio of BDA (BDA%) in BDPLAs, respectively. The degradations of BDPLAs, PDLLA and MPLAs were investigated by observation of the changes of the pH value of incubation medium, molecular weight and weight loss ratio for a time interval of 12 weeks in vitro, respectively. The results revealed that grafting butanediamine onto PDLLA has weakened or neutralized the acidity of PDLLA degradation products. A uniform degradation of BDPLAs was observed in comparison with an acidity-induced auto-accelerating degradation featured by PDLLA and MPLAs. The biodegradation behaviors of BDPLAs are tunable by controlling the content of BDA. BDPLAs might be a new derivative of PDLLA-based biodegradable materials for medical applications without acidity-caused irritations and acidity-induced auto-accelerating degradation behavior as that of PDLLA. 相似文献

We developed a method for simultaneously determining l-citrulline and l-arginine levels in serum and tissue samples using RP-HPLC with ultraviolet (UV) detection. The serum was deproteinized by trichloroacetic acid and heat; the tissue was homogenized by trichloroacetic acid and deproteinized as the same as serum. Phenyl-isothiocyanate (PITC) solution was used as derivatization reagent and a gradient elution was carried out. The linearity for l-arginine and l-citrulline ranged from 0 to at least 1000 μmol/L. R2 values were above 0.9999 for both. LODs for l-arginine and l-citrulline were 0.0462 μmol/L and 0.0195 μmol/L, respectively, while LOQs were 0.530 μmol/L and 0.417 μmol/L, respectively. Intra- and inter-day CVs were less than 3.5% and 7.5% in serum, respectively. The average recovery was from 85.5% to 116.5% in serum. Intra-assay CVs were 3.8% and 10.3%, and inter-assay CVs were 13.7% and 10.7% for l-arginine and l-citrulline respectively in tissue. The average recovery was from 92.8% to 113.5% in tissue. This is a reliable and convenient analytical method which is suitable for most clinical laboratories. 相似文献
