Lipid characterization of bone marrow in vivo with proton magnetic resonance spectroscopy was performed using Spin-Echo Planar Spectroscopic Imaging sequences. The methods are shown capable of rapidly generating two-dimensional chemical shift imaging data sets suitable for measuring lipid indices that reflect unsaturation levels among triglycerides, as demonstrated in oil phantoms and bone marrow from a healthy volunteer. The volume coverage, spatial resolution, acquisition speed, and spectral characteristics of Spin-Echo Planar Spectroscopic Imaging should make it attractive for clinical studies of diseases affecting normal lipid chemical composition.  相似文献

A fast proton spectroscopic imaging pulse sequence based on the condition of steady-state free precession is presented. High 3D spatial and temporal resolution is achieved using simultaneous detection of both one spatial and one spectral dimension, with a time-dependent gradient cycle known from echo planar imaging. Additionally, in order to increase the spectral width of the measurement, an interleaved acquisition scheme is shown either for systems with limited gradient switching capabilities or applications with a wide chemical shift range. The pulse sequence is implemented on a standard 4.7-T nuclear magnetic resonance animal imaging system. Measurements with a total measurement time of less than 2.5 min and a nominal voxel size of 6.75 microl using a total of 64 x 32 x 16 voxels are performed on phantoms and healthy rat brain in vivo allowing the rapid detection of signals from both uncoupled and J-coupled spin systems with high signal-to-noise ratio.  相似文献

在超声背散射骨质评价中,不同测量感兴趣区域(ROI)的超声背散射信号会有波动,致使诊断准确度降低。该文目的是研究超声背散射信号随测量区域的变化规律。采用空间扫描方法离体测量了35块骨样本,将超声背散射参数从空间域变换到空间频率域。结果表明,超声背散射参数的主要空间频率成分集中于低频部分;空间频率域超声背散射参量最大值(MASF)与骨矿密度等参数具有中高度显著相关性(R2=0.45~0.83,p0.001);空间频率域超声背散射参量衰减系数(AC)也与松质骨密度及结构特征有显著相关性(R2=0.41~0.72,p0.001)。研究表明空间频率域变换方法有助于明确超声背散射信号随测量ROI的变化规律,空间频率域的超声背散射相关新参量评价松质骨状况具有可行性。  相似文献

Measurement of key parameters of the microstructure of trabecular bone is critical to the study of osteoporosis and bone strength. Density based methods cannot provide this information, and give only the total amount of bone present, and not its arrangement. Magnetic resonance imaging has shown the potential to provide information related to the microarchitecture of the trabecular bone matrix. Twelve samples (8 x 8 x 8 mm3 bone cubes) were cut from sheep vertebrae such that the trabeculae ran either parallel or perpendicular to each face. Detailed measurements of the structure of these bone cubes were made by histomorphometry, and compared to R'2 and R*2 measured with a spin and gradient-echo sequence, Partially Refocused Interleaved Multiple Echo, at 1.5 Tesla. The precision of the R'2 measurement (% coefficient of variation) was 8.7+/-5.1, and 7.7+/-4.3 for R*2. Uncorrected values of R'2 and R*2 were significantly correlated to density measured by quantitative computed tomography (r = 0.87, p = 0.0005, and r = 0.90, p = 0.0002, respectively), and trabecular bone area measured by histomorphometry (r = 0.80, p = 0.002, and r = 0.83, p = 0.0008, respectively). Density correction was effected by imaging the same slice of bone in two orientations (90 degrees and 0 degrees ) to the main magnetic field. For both R'2 and R*2 there was a significant difference between measurements in the 90 degrees and 0 degrees orientations (p < 0.01). The difference between the two values was used, and termed R'2net or R*2net. The net parameters were independent of bone mass. R'2net and R*2net were significantly correlated to trabecular separation (p < 0.05) with r = -0.58 and r = -0.62, respectively. These results demonstrate the ability of magnetic resonance imaging to characterize a key measure of the trabecular microstucture. An increase in trabecular separation has important biomechanical consequences in osteoporosis. This result also strengthens the hypothesis that the sensitivity of R'2 to osteoporosis-related bone changes is due to magnetic susceptibility effects in which rapid transitions between bone and marrow create local magnetic field inhomogeneities that result in an increase in R'2 values.  相似文献

A hybrid fluorescence–Raman confocal microscopy platform is presented, which integrates low‐wavenumber‐resolution Raman imaging, Rayleigh scatter imaging and two‐photon fluorescence (TPE) spectral imaging, fast ‘amplitude‐only’ TPE‐fluorescence imaging and high‐spectral‐resolution Raman imaging. This multi‐dimensional fluorescence–Raman microscopy platform enables rapid imaging along the fluorescence emission and/or Rayleigh scatter dimensions. It is shown that optical contrast in these images can be used to select an area of interest prior to subsequent investigation with high spatially and spectrally resolved Raman imaging. This new microscopy platform combines the strengths of Raman ‘chemical’ imaging with light scattering microscopy and fluorescence microscopy and provides new modes of correlative light microscopy. Simultaneous acquisition of TPE hyperspectral fluorescence imaging and Raman imaging illustrates spatial relationships of fluorophores, water, lipid and protein in cells. Magnetic resonance phase images can be used to assess trabecular bone by measuring the standard deviation of the phases in a region of interest. The standard deviation of regional phase measurements reflects the degree of magnetic field inhomogeneity caused by susceptibility differences between bone and marrow. A 3D computer model of trabecular bone was developed and then used to explore the influence of bone volume fraction and imaging parameters such as pixel size and slice width on the standard deviation of regional phase measurements. The results from these tests show that with appropriate selection of these parameters, phase spread strongly reflects variations in trabecular bone density (a correlation of R(2) = 0.98 with bone volume fraction between 0 and 10%). The technique was then applied in vivo on the radius of 25 patients who already had a bone density scan with peripheral quantitative tomography and a correlation between phase standard deviation and trabecular bone density was found (R(2) = 0.46).  相似文献

Photoinduced magnetization in a magnet based on organic species is reported for the first time. Upon optical excitation in the blue region of spectrum, Mn(tetracyanoethylene)(x)*y(CH2Cl2) (x approximately 2, y is approximately 0.8) exhibits increased magnetic susceptibility at temperatures as high as 75 K, accompanied with photoinduced absorption in the visible and infrared spectral regions. These effects are partially reversible by lower energy visible light and fully reversible by thermal treatment. The results suggest trapping of the photoexcited charge in a metastable state with enhanced exchange interaction.  相似文献

The hypercapnia induced by carbogen (95% O(2)/5% CO(2)) breathing, which is being re-evaluated as a clinical radiosensitiser, causes patient discomfort and hence poor compliance. Recent preclinical and clinical studies have indicated that the CO(2) content might be lowered without compromising increased tumour oxygenation and radiosensitisation. This preclinical study was designed to see if lower levels of hypercapnia could evoke similar decreases in the transverse relaxation rate R(2)* of rodent tumours to those seen with carbogen breathing. The response of rat GH3 prolactinomas to 1%, 212% and 5% CO(2) in oxygen, and 100% O(2) breathing, was monitored by non-invasive multi-gradient echo MRI to quantify R(2)*. As the oxygenation of haemoglobin is proportional to the blood p(a)O(2) and therefore in equilibrium with tissue pO(2), R(2)* is a sensitive indicator of tissue oxygenation. Hyperoxia alone decreased R(2)* by 13%, whilst all three hypercapnic hyperoxic gases decreased R(2)* by 29%. Breathing 1% CO(2) in oxygen evoked the same decrease in R(2)* as carbogen. The DeltaR(2)* response is primarily consistent with an increase in blood oxygenation, though localised increases in tumour blood flow were also identified in response to hypercapnia. The data support the concept that levels of hypercapnia can be reduced without loss of enhanced oxygenation and hence potential radiotherapeutic benefit.  相似文献

运用光谱技术研究了椎骨组织不同位置的特征识别因子.光谱采集系统由双光纤手钻一体式探头(光纤芯径200μm,中心距离0.5mm)、卤素光源(波长360~2 000nm)、光纤光谱仪(检测波长为200~1 100nm)和计算机组成,可以同时获得生物组织的漫反射光谱和约化散射系数.以猪椎骨为实验对象,测量椎弓根螺钉植入针道上不同骨组织的漫反射光谱和约化散射系数,并对光谱进行特定波长的峰值、面积、斜率分析,获得特性识别因子.研究发现,椎弓根钉植入针道上不同骨组织的光谱表现出不同的变化特性.其中峰值的变化比约化散射系数的变化高1.88倍,面积的变化比约化散射系数的变化高2.05倍.在495~505nm处,骨密质和骨疏质的光谱斜率都为正值;在520~535nm处,骨密质光谱的斜率为正值,而骨疏质光谱的斜率为负值.结果表明,通过光谱特性分析获得的峰值、面积和斜率因子能够有效地区分针道上骨密质与骨疏质的差异.  相似文献

PURPOSE: A newly developed contrast material, CH3-DTPA-Gd (NMS60), a trimer containing 3 Gd(3+) atoms per molecule, has been shown to offer greater enhancement and longer vascular retention than gadopentetate dimeglumine (Gd-DTPA) in animals. We report on our early phase II study on NMS60 in brain tumor patients together with supplementary investigations. METHODS AND MATERIALS: The longitudinal relaxation rate (R(1)=1/T(1)) and the transverse relaxation rate (R(2)*=1/T(2)*) of NMS60 and Gd-DTPA were determined at 20 degrees C in water at 1.5 T. An NMS60 dose of 0.1 or 0.2 mmol (Gd)/kg was randomly assigned and administered to 10 patients (five women, five men; mean age: 49 years) with brain tumors. Safety and contrast-enhancing ability of NMS60 were evaluated. Dual dynamic contrast-enhanced T(1) and R(2)* studies (DUCE imaging) were also carried out in two patients. RESULTS: Regarding the relaxivity per Gd, R(1) and R(2)* of NMS60 were 9.5 and 11.0 (mmol/L x s)(-1), respectively, compared to 4.8 and 7.2 (mmol/L x s)(-1) for Gd-DTPA. Although a transient slight increase of alanine aminotransferase was observed in one case, no other adverse reactions were observed after administration of NMS60. Contrast enhancement by NMS60 was excellent at both concentrations, and when tumor detectability was assessed with a five-point scale, the diagnostic usefulness was 4 or higher in all cases. In DUCE imaging, NMS60 appeared to show high signal intensity, when compared with the data obtained separately for Gd-DTPA. CONCLUSION: NMS60 had a high contrasting effect and little toxicity, and is expected to be clinically useful.  相似文献

The purpose of this study was to evaluate a magnetic resonance imaging (MRI) technique for quantifying the proton density water fraction (PDWF) as a biomarker of bone marrow cellularity. Thirty-six human bone marrow specimens from 18 donors were excised and subjected to different measurements of tissue composition: PDWF quantification using a multiple gradient echo MRI technique, three biochemical assays (triglyceride, total lipid and water content) and a histological assessment of cellularity. Results showed a strong correlation between PDWF and bone marrow cellularity from histology (r = 0.72). A strong correlation was also found between PDWF and the biochemical assay of water content (r = 0.76). These results suggest the PDWF is a predictor of bone marrow cellularity in tissues and can provide a non-invasive assessment of bone marrow changes in clinical patients undergoing radiotherapy.  相似文献

The purpose of this work is to develop a rapid MRI method amenable to profiling with minimal or no T(1) relaxation weighting. The behavior of a signal during a centric SPRITE acquisition is analyzed. It is shown that the technique can be made immune to a broad range of T(1) changes. In a properly executed measurement, only T(2)* and proton density parameters define the image intensity. A T(2)* mapping technique can be easily applied, separating T(2)* and proton density contributions to the image. A drying soil sample with low initial water content is experimentally studied as a demonstration of the technique. A characteristic baseline artifact is easily removed from the profiles by a simple operation.  相似文献

The modified Biot–Attenborough (MBA) model for acoustic wave propagation in porous media has been found useful to predict wave properties in cancellous bone. The present study is aimed at applying the MBA model to predict the dependence of phase velocity on porosity in cancellous bone. The MBA model predicts a phase velocity that decreases nonlinearly with porosity. The optimum values for input parameters of the MBA model, such as compressional speed cm of solid bone and phase velocity parameter s2, were determined by comparing the predictions with previously published measurements in human calcaneus and bovine cancellous bone. The value of the phase velocity parameter s2 = 1.23 was obtained by curve fitting to the experimental data for 53 human calcaneus samples only, assuming a compressional speed cm = 2500 m/s of solid bone. The root-mean-square error (RMSE) of the curve fit was 15.3 m/s. The optimized value of s2 for all 75 cancellous bone samples including 22 bovine samples was 1.42 with a value of 55 m/s for the RMSE of the curve fit. The latter fit was obtained by using of a value of cm = 3200 m/s. Although the MBA model relies on the empirical parameters determined from experimental data, it is expected that the model can be usefully employed as a practical tool in the field of clinical ultrasonic bone assessment.  相似文献

The limited bandwidths of volume selective RF pulses in localized in vivo MRS experiments introduce spatial artifacts that complicate spectral quantification of J-coupled metabolites. These effects are commonly referred to as a spatial interference or "four compartment" artifacts and are more pronounced at higher field strengths. The main focus of this study is to develop a generalized approach to numerical simulations that combines full density matrix calculations with 3D localization to investigate the spatial artifacts and to provide accurate prior knowledge for spectral fitting. Full density matrix calculations with 3D localization using experimental pulses were carried out for PRESS (TE=20, 70 ms), STEAM (TE=20, 70 ms) and LASER (TE=70 ms) pulse sequences and compared to non-localized simulations and to phantom solution data at 4 T. Additional simulations at 1.5 and 7 T were carried out for STEAM and PRESS (TE=20 ms). Four brain metabolites that represented a range from weak to strong J-coupling networks were included in the simulations (lactate, N-acetylaspartate, glutamate and myo-inositol). For longer TE, full 3D localization was necessary to achieve agreement between the simulations and phantom solution spectra for the majority of cases in all pulse sequence simulations. For short echo time (TE=20 ms), ideal pulses without localizing gradients gave results that were in agreement with phantom results at 4 T for STEAM, but not for PRESS (TE=20). Numerical simulations that incorporate volume localization using experimental RF pulses are shown to be a powerful tool for generation of accurate metabolic basis sets for spectral fitting and for optimization of experimental parameters.  相似文献

直接吸收光谱(DAS)可直接测量分子吸收率函数,并通过拟合吸收率函数确定待测气体参数.波长调制-直接吸收光谱(WM-DAS)在DAS基础上,结合了波长调制光谱(WMS)中谐波分析思想,利用傅里叶变换复现吸收率函数,可有效提高吸收率函数的测量精度.本文利用WM-DAS方法结合长光程气体吸收池,在室温低压条件下,对CO分子1567 nm处R5-R11近红外弱吸收谱线吸收率函数进行了精确复现,其拟合残差标准差低至3×10^-5,随后根据测得的吸收率函数对谱线的碰撞展宽、Dicke收敛以及速度依赖的碰撞展宽系数等光谱参数进行了高精度标定,并将其与高灵敏度的连续波腔衰荡光谱(CW-CRDS)测量结果进行了比较,实验结果表明该方法与CW-CRDS测量结果具有高度一致性,更具有系统简单、测量速度快、对环境要求低等优点.  相似文献

从分析对颗粒物PM2.5敏感的光谱特性入手,提出了一种基于差值指数的颗粒物PM2.5浓度反演新方法。使用Avafield-1光谱仪(测量范围300~1 100 nm)测量了典型地物植被、土壤在不同的颗粒物PM2.5污染条件下的光谱曲线,发现颗粒物使得植被和裸土的光谱曲线在红光波段反射率增加,在近红外波段反射率下降,且其变化量较为稳定,因此使用对颗粒物敏感的红光波段和近红外波段,构建差值指数(difference index,DI)以表征颗粒物PM2.5的浓度变化。以北京市为研究区,选择TM影像,求取差值指数,结合北京市及周边地区地面空气质量监测站提供的PM2.5逐时数据,反演了北京市的颗粒物PM2.5的浓度。结果表明,2016年3月1日(监测站PM2.5浓度均值为105.8)预测模型相关系数r为0.796,精度表现良好,12月14日(监测站PM2.5均值为15.8)预测模型相关系数r为0.628,即颗粒物污染程度较低情况下,差值指数模型预测精度低于颗粒物污染程度较高的情况,分析原因是颗粒物浓度较低时,由颗粒物引起的地物光谱特征变化比较不明显,造成差值指数模型精度较低。由于空气质量为重度及严重污染时,获取的遥感影像质量较差,影响颗粒物浓度反演,因此该方法适合轻度、中度污染情况下的颗粒物PM2.5浓度反演。该方法获取污染数据简单,反演结果空间分辨率较高(30m),且可根据需要选取包含敏感波段数据的遥感影像用以获取不同时间、空间分辨率的颗粒物PM2.5浓度分布,具有广泛的应用前景。  相似文献

The objective of this article is to determine telomere length, a measure of biological age, in true vocal fold (TVF), false vocal fold (FVF), and five other tissue types, to ascertain whether there is tissue-specific telomere shortening. The study design is that of a prospective, basic science study. Tissue samples were obtained from the TVF, FVF, skin from the back of hand, skin from thigh, aorta, blood, and bone marrow from 12 patients ages 54 to 76 years. Genomic DNA was isolated from each sample, and telomere lengths were calculated with real-time polymerase chain reaction. In our small age group, age was not significantly associated with telomere length across tissue types, nor were there any linear correlations within tissue types and age. Controlling for age, significant differences were found between the following tissues: aorta and blood (P < 0.000), aorta and bone marrow (P = 0.033), aorta and FVF (P = 0.015), aorta and hand skin (P = 0.004), blood and thigh skin (P = 0.012), and blood and TVF (P = 0.048). A significant linear correlation between telomere length and tissue type without considering donor age was established between bone marrow and hand skin (P < 0.05, R2 = 0.766), thigh skin and hand skin (P < 0.01, R2 = 0.926), TVF and blood (P < 0.01, R2 = 0.836), and thigh skin and TVF (P < 0.05, R2 = 0.624). Our findings indicate that surrogate tissue for measurement of telomere length of TVF includes FVF, bone marrow, skin, and aorta. These findings have implications for understanding vocal fold aging at the cellular level.  相似文献

在用选择离子流动管质谱(SIFT-MS)分析常用指甲油清洗垫发现大量4-丁酸内脂(γ-butyrolactone, GBL)和2-丁氧基乙醇(2-butoxy-1-ethanol)等挥发性气体后, 运用选择离子流动管(SIFT)对H3O , NO 和O ·2离子同九种烷氧基醇化合物(R1-O-R2OH)之间的反应进行了研究. 获得了这些反应在潮湿空气条件下进行的情况, 并运用产物离子水合物的种类和分布来确认产物离子的结构和反应机理. 还研究了在不同载气压力下进行的反应. 结果表明, 这些化合物同H3O 和NO 的反应都是先生成初生态离子-分子络合物, (H3O .M)*和(NO .M)*, 然后经不同反应渠道生成各种离子产物. 这些初生态络合物同反应体系中存在的气体分子(如氦气, 氮气和氧气分子)的碰撞对最终离子产物的形成和分布也有影响. 这些化合物同O2 ·反应会生成各种离解碎片离子, 但不能确定这些离子是否经由初生态络合物(O2 ·.M)*生成. 该项研究提供了用SIFT-MS在空气和潮湿气体中分析这些化合物所需的反应速率常数和离子产物等动力学数据, 并将进一步应用到药物成瘾及滥用和呼吸道疾病的诊断和分析等领域.  相似文献

We investigated the effects of transcranial magnetic stimulation (TMS) coils and electroencephalographic (EEG) electrodes on T(2)*-weighted echo-planar images (EPI) at 2.0 T (gradient-echo EPI, mean TE = 53 ms, 2x2x4 mm(3)). In comparison with anatomic gradient-echo images (3D FLASH, TE = 4 ms, 1x1x1 mm(3)), T(2)*-weighted EPI acquisitions of a water-filled spherical phantom revealed severe signal losses and geometric distortions in the vicinity of TMS coils. Even remote effects were observed for image orientations perpendicular to the coil plane. EEG electrodes and the fixation gel caused milder localized distortions. In humans, complications were avoided by the large distance between the TMS coil and the cortical surface and when using an EPI orientation parallel to the plane of the coil. It is concluded that T(2)*-weighted EPI studies of human brain function may be performed without distortions caused by TMS coils and EEG electrodes.  相似文献

An experimental model which can simulate physical changes that occur during aging was developed in order to evaluate the effects of change of mineral content and microstructure on ultrasonic properties of bovine cancellous bone. Timed immersion in hydrochloric acid was used to selectively alter the mineral content. Scanning electron microscopy and histological staining of the acid-treated trabeculae demonstrated a heterogeneous structure consisting of a mineralized core and a demineralized layer. The presence of organic matrix contributed very little to normalized broadband ultrasound attenuation (nBUA) and speed of sound. All three ultrasonic parameters, speed of sound, nBUA and backscatter coefficient, were sensitive to changes in apparent density of bovine cancellous bone. A two-component model utilizing a combination of two autocorrelation functions (a densely populated model and a spherical distribution) was used to approximate the backscatter coefficient. The predicted attenuation due to scattering constituted a significant part of the measured total attenuation (due to both scattering and absorption mechanisms) for bovine cancellous bone. Linear regression, performed between trabecular thickness values and estimated from the model correlation lengths, showed significant linear correlation, with R(2)=0.81 before and R(2)=0.80 after demineralization. The accuracy of estimation was found to increase with trabecular thickness.  相似文献
